Elevated levels of ovine reaginic antibody were induced by immunization with Ascaris suum antigens. The PCA activity of this antibody persisted in the skin of sheep for 20 days and was abolished by heating to 56 degrees C, suggesting that it was immunoglobulin E. An improved method for the detection of IgE antibody of defined specificity by ELISA using rat monoclonal anti-IgE antibodies is described. The innovation consists of coating the plates first by a monoclonal rat anti-murine IgE antibody, adding the sera to this antibody-coated plates and then adding the biotin-conjugated antigen after the sera. Four experimental procedures leading to lower antibody production, namely administration of a minute amount of antigen or excess of antigen, competition with an unrelated antigen and treatment with methotrexate, were studied for their effect on each of the forms of immunological responses in guinea-pigs to a DNP-BSA conjugate. It was found that delayed hypersensitivity and 2-ME sensitive antibody production were more reduced by administration of excess of antigen or by competition than by giving a minute amount of antigen or methotrexate. In mammals, ceramide kinase (CerK)-mediated phosphorylation of ceramide is the only known pathway to ceramide-1-phosphate (C1P), a recently identified signaling sphingolipid metabolite. To help delineate the roles of CerK and C1P, we knocked out the gene of CerK in BALB/c mice by homologous recombination. All in vitro as well as cell-based assays indicated that CerK activity is completely abolished in Cerk-/- mice.
