Evaluation of the designed multi-epitope protein of Brucella melitensis in guinea pigs.

OBJECTIVES: One of the causes of human and animal zoonotic infections is Brucella melitensis, which is transmitted to humans through dairy products. It seems for prevention of human infection we might protect the livestock by an efficient protein as a vaccine candidate. For this purpose, the use of immunogenic proteins of bacteria is able to create immunity the same as the traditional vaccines. MATERIALS AND METHODS: In this study, by finding the immunogenic antigens of this bacterium by 2-dimensional gel electrophoresis and MALDI-TOF methods and also the proteins reported in other studies, w

Enhancing humoral and mucosal immune response of PED vaccine candidate by fusing S1 protein to nanoparticle multimerization.

Porcine epidemic diarrhea virus (PEDV) is a highly infectious pathogen with a high mortality rate, which poses a serious threat to newborn piglets. A rapid, safe and effective vaccine is necessary for protecting pigs from PED infection. Nanoparticles have become molecular scaffolds for displaying soluble antigens due to their unique physical and chemical properties. Here, a vaccine candidate was based on the display of PEDV S1 protein on a mi3 nanoparticle platform using SpyTag/SpyCatcher technology. The size, zeta potential and microstructure of the S1-mi3 NPs were investigated, and their eff

An mRNA-based FimH nanoparticle vaccine against uropathogenic Escherichia coli confers protection in a mouse model of recurrent urinary tract infection.

BACKGROUND: Increasing antimicrobial resistance and the high health and economic burden of frequent, recurring urinary tract infections (rUTIs) drive the need for an efficacious UTI vaccine. Most UTIs are caused by uropathogenic E. coli (UPEC) expressing the virulence factor FimH, which mediates bladder colonisation. We have developed a lipid nanoparticle (LNP) mRNA vaccine, FimHDG-Ferritin, as a UPEC vaccine candidate. METHODS: FimHDG-Ferritin mRNA constructs were introduced into HeLa cells through mRNA-transfection to assess the generation of immune-accessible protein nanoparticles (PNPs) i

Vaccination with formulations targeting Eimeria maxima and Clostridium perfringens conferred comprehensive protection using a dual-infection challenge model of necrotic enteritis.

With increasing regulations restricting antibiotic use in animal feed, the need for alternative strategies to prevent and manage necrotic enteritis (NE) has become imperative. As a result, developing effective vaccines has emerged as a top priority for broiler chicken health management. Coccidial infections are a well-established predisposing factor for NE, underscoring the importance of controlling coccidiosis to help mitigate NE outbreaks. This research aimed to investigate the protective efficacy of vaccine preparations containing Eimeria maxima elongation factor-1α and a multicomponent ant

Immunization with a novel recombinant protein (YidR) reduced the risk of clinical mastitis caused by Klebsiella spp. and decreased milk losses and culling risk after Escherichia coli infections.

The primary objective of this study was to evaluate the protective efficacy of a novel recombinant subunit vaccine containing the protein YidR (rYidR) against clinical mastitis (CM) caused by Klebsiella spp. and Escherichia coli. Given that E. coli infection is known to cause metritis, we also evaluated the effect of rYidR vaccination on the incidence of metritis and conception at the first artificial insemination. Retained placenta and abortion incidence, milk production and composition, and serological responses to specific antigens were also evaluated. In total, 3,107 cows were blocked by p

Execution and Design of an Anti HPIV-1 Vaccine with Multiple Epitopes Triggering Innate and Adaptive Immune Responses: An Immunoinformatic Approach.

Human Parainfluenza Virus (HPIV) Type-1, which is an anti-sense ribonucleic acid (RNA) virus belonging to the paramyxoviridae family, induces upper and lower respiratory tract infections. The infections caused by the HPIV Type-1 virus are usually confined to northwestern regions of America. HPIV-1 causes infections through the virulence of the hemagglutinin-neuraminidase (HN) protein, which plays a key role in the attachment of the viral particle with the host's receptor cells. To the best of our knowledge, there is no effective antiviral drugs or vaccines being developed to combat the infecti

Monoclonal Antibodies Targeting an Opisthorchis viverrini Extracellular Vesicle Tetraspanin Protect Hamsters against Challenge Infection.

Opisthorchis viverrini causes severe pathology in the bile ducts of infected human hosts, and chronic infection can culminate in bile duct cancer. The prevention of infection by vaccination would decrease opisthorchiasis-induced morbidity and mortality. The tetraspanin protein, Ov-TSP-2, is located on the membrane of secreted extracellular vesicles (EVs), and is a candidate antigen for inclusion in a subunit vaccine. To address the role of anti-Ov-TSP-2 antibodies in protection, we assessed the protective capacity of anti-Ov-TSP-2 monoclonal antibodies (mAbs) against opisthorchiasis. Two anti-

Immunogenicity and efficacy in mice of an adenovirus-based bicistronic rotavirus vaccine expressing NSP4 and VP7.

NSP4 and VP7 are important functional proteins of rotavirus. Proper combination of viral gene expression is favorable to improving the protection effect of subunit vaccine. In the present study, We evaluated the immunogenicity and efficacy of the bicistronic recombinant adenovirus (rAd-NSP4-VP7) and two single-gene expressing adenoviruses (rAd-NSP4, rAd-VP7). The three adenovirus vaccines were used to immunize mice by intramuscular or intranasal administration. The data showed significant increases in serum antibodies, T lymphocyte subpopulations proliferation, and cytokine secretions of splen

The critical role of artificial intelligence and bioinformatics in accelerating peptide-based vaccine discovery for tackling global infectious diseases.

Peptide-based vaccines, enabled by bioinformatics and machine learning (ML), have emerged as one of the most promising approaches for rapid, safe, and cost-effective vaccine design against infectious diseases. Unlike conventional approaches that depend heavily on whole-pathogen cultures or recombinant protein expression, peptide vaccines can be designed in silico and synthesized quickly. Rational and targeted in silico approaches for the discovery of peptide-based vaccine candidates include B-cell and T-cell epitope prediction, immunogenicity, antigenicity, allergenicity, autoimmunity, populat

An Orthogonal Nanoadjuvant-Based RSV Subunit Vaccine Unlocks Potent Immunity through a High-Entropy-like Immunostimulatory Effect.

Respiratory syncytial virus (RSV) is one of the primary contributors to lower respiratory tract infections, especially in infants, children, and the elderly. The quest for a clinically acceptable and potent RSV vaccine constitutes a long-standing hurdle in clinical practice. Herein, we propose an orthogonal nanoadjuvant-based nanovaccine platform that addresses the long-standing challenge of insufficient cellular immunity in RSV F subunit vaccination. The vaccine is constructed based on Mn/Al-layered double hydroxide (Mn/Al-LDH) nanosheet functionalized with RSV F antigen and poly-CpG motifs.

Evaluation of candidate vaccine approaches for MERS-CoV.

The emergence of Middle East respiratory syndrome coronavirus (MERS-CoV) as a cause of severe respiratory disease highlights the need for effective approaches to CoV vaccine development. Efforts focused solely on the receptor-binding domain (RBD) of the viral Spike (S) glycoprotein may not optimize neutralizing antibody (NAb) responses. Here we show that immunogens based on full-length S DNA and S1 subunit protein elicit robust serum-neutralizing activity against several MERS-CoV strains in mice and non-human primates. Serological analysis and isolation of murine monoclonal antibodies revealed

Perspectives on novel vaccine development.

Vaccination is a common routine for prevention and control of human and animal diseases by inducing antibody responses and cell-mediated immunity in the body. Through vaccinations, smallpox and some other diseases have been eradicated in the past few years. The use of a patho- gen itself or a subunit domain of a protein antigen as immunogens lays the basis for traditional vaccine development. But there are more and more newly emerged pathogens which have expe- rienced antigenic drift or shift under antibody selective pressures, rendering vaccine-induced im- munity ineffective. In addition, vac

A ubiquitinated E2-based mRNA vaccine confers complete protection against lethal challenge with classical swine fever virus.

Classical swine fever (CSF), caused by classical swine fever virus (CSFV), continues to threaten the global swine industry. Current E2-based subunit vaccines are limited by their slow adaptation to emerging viral strains and insufficient cell-mediated immunity. To address these challenges, we developed mRNA-lipid nanoparticle (LNP) vaccines encoding modified forms of the CSFV E2 glycoprotein. In this study, we constructed three novel mRNA-LNP vaccines encoding engineered E2 proteins: E2-mRNA-LNP, XCL1-E2-mRNA-LNP containing the X-C motif chemokine ligand 1 (XCL1) to target dendritic cells, and

Comparative evaluation of protective immunity against Francisella tularensis induced by subunit or adenovirus-vectored vaccines.

Tularemia is a highly contagious disease caused by infection with Francisella tularensis (Ft), a pathogenic intracellular gram-negative bacterium that infects a wide range of animals and causes severe disease and death in people, making it a public health concern. Vaccines are the most effective way to prevent tularemia. However, there are no Food and Drug Administration (FDA)-approved Ft vaccines thus far due to safety concerns. Herein, three membrane proteins of Ft, Tul4, OmpA, and FopA, and a molecular chaperone, DnaK, were identified as potential protective antigens using a multifactor pro

Empty lipid nanoparticles (eLNPs) with adjuvant activity induce potent immune responses.

BACKGROUND AND PURPOSE: Achieving herd immunity via mass vaccination is recognized as the most effective strategy for curbing the transmission of contagious diseases. Among the various types of vaccines, subunit protein vaccines stand out for their favourable safety profile, proven efficacy and straightforward production processes. The development of next-generation vaccines is currently hindered by the scarcity of adjuvants capable of simultaneously amplifying both humoral and cellular immune responses. EXPERIMENTAL APPROACH: This study identified the physical characteristics of empty lipid

Nucleoside Hydrolase NH 36: A Vital Enzyme for the Leishmania Genus in the Development of T-Cell Epitope Cross-Protective Vaccines.

NH36 is a vital enzyme of the DNA metabolism and a specific target for anti-Leishmania chemotherapy. We developed second-generation vaccines composed of the FML complex or its main native antigen, the NH36 nucleoside hydrolase of Leishmania (L.) donovani and saponin, and a DNA vaccine containing the NH36 gene. All these vaccines were effective in prophylaxis and treatment of mice and dog visceral leishmaniasis (VL). The FML-saponin vaccine became the first licensed veterinary vaccine against leishmaniasis (Leishmune®) which reduced the incidence of human and canine VL in endemic areas. The NH3

Clinical development of plant-produced recombinant pharmaceuticals: vaccines, antibodies and beyond.

In the last few years, plants have become an increasingly attractive platform for recombinant protein production. This builds on two decades of research, starting with transgenic approaches to develop oral vaccines in which antigens or therapeutics can be delivered in processed plant biomass, and progressing to transient expression approaches whereby high yields of purified targets are administered parenterally. The advantages of plant-based expression systems include high scalability, low upstream costs, biocontainment, lack of human or animal pathogens, and ability to produce target proteins

Ferritin nanoparticle complex with porcine epidemic diarrhea virus spike protein induces neutralizing antibody response against PEDV in mouse models.

As the spike protein is the major antigen that contains various neutralizing epitopes against porcine epidemic diarrhea virus (PEDV), numerous vaccine trials employing the spike protein have been established. In this study, we developed a ferritin-based nanoparticle vaccine for PEDV by combining gene delivery functions of recombinant adenoviruses. To generate nanoparticles, the S1 subunit of the spike protein was genetically linked to the N-terminus of the human ferritin heavy chain (hFTHC), and recombinant adenoviruses were generated to deliver the genetic material. The efficacy of S1 conjuga

Single mucosal immunization of recombinant adenovirus-based vaccine expressing F1 protein fragment induces protective mucosal immunity against respiratory syncytial virus infection.

Respiratory syncytial virus (RSV) is a major cause of severe lower respiratory tract disease in infancy and early childhood. Despite its importance as a pathogen, there is no licensed vaccine against RSV. The fusion (F) protein of RSV is a potentially important target for protective antiviral immune responses. Here, we studied the immune responses elicited by recombinant replication-deficient adenovirus (rAd)-based vaccines expressing the soluble F1 fragment of F protein (amino acids 155-524) in murine model. The expression of secreted F1 fragment by rAd was significantly increased by codon op
