Specific Anti-biofilm Activity of Carbon Quantum Dots by Destroying P. gingivalis Biofilm Related Genes.

INTRODUCTION: Biofilms protect bacteria from antibiotics and this can produce drug-resistant strains, especially the main pathogen of periodontitis, Porphyromonas gingivalis. Carbon quantum dots with various biomedical properties are considered to have great application potential in antibacterial and anti-biofilm treatment. METHODS: Tinidazole carbon quantum dots (TCDs) and metronidazole carbon quantum dots (MCDs) were prepared by a hydrothermal method with the clinical antibacterial drugs tinidazole and metronidazole, respectively. Then, TCDs and MCDs were characterized by transmission elect…

Gut microbiota and lipid metabolism alterations in mice induced by oral cadmium telluride quantum dots.

The potential toxicity of cadmium-containing quantum dots (QDs) has received much attention because of increasing biomedical applications. However, little has been known about how cadmium telluride (CdTe) QDs influence the gut microbiota and lipid metabolism. In this study, mice were exposed orally to CdTe QDs (200 μL of 0.2, 2, 20 or 200 μm; twice per week) for 4 weeks. The oral experiments showed CdTe QD exposure led to a decrease of the Firmicutes/Bacteroidetes (F/B) ratio of gut microbiota, which highly negatively correlated with the low-density lipoprotein (LDL), triglyceride (TG) and tot…

Peptide-enabled receptor-binding-quantum dots for enhanced detection and migration inhibition of cancer cells.

We report the efforts to construct active targeting quantum dots using receptor-binding peptide for enhanced detection and migration inhibition of cancer cells. Peptide E5 has specific binding with chemokine receptor 4 (CXCR4), which is a transmembrane G-coupled receptor involved in the metastasis of various types of cancers. E5 was introduced to the surface of CdSe/ZnS quantum dots via biotin-streptavidin interactions. The constructed CXCR4-targeting quantum dots (E5@QDs) was observed to display improved detection sensitivity and significantly enhanced binding affinity for CXCR4 over-expresse…

Nephrotoxicity Evaluation of Indium Phosphide Quantum Dots with Different Surface Modifications in BALB/c Mice.

InP QDs have shown a great potential as cadmium-free QDs alternatives in biomedical applications. It is essential to understand the biological fate and toxicity of InP QDs. In this study, we investigated the in vivo renal toxicity of InP/ZnS QDs terminated with different functional groups-hydroxyl (hQDs), amino (aQDs) and carboxyl (cQDs). After a single intravenous injection into BALB/c mice, blood biochemistry, QDs distribution, histopathology, inflammatory response, oxidative stress and apoptosis genes were evaluated at different predetermined times. The results showed fluorescent signals fr

PEGylated CuInS2/ZnS quantum dots inhibit neurite outgrowth by downregulating the NGF/p75NTR/MAPK pathway.

The widespread application of cadmium-free CuInS2/ZnS QDs has raised great concern regarding their potential toxicity to humans. To date, toxicological data related to CuInS2/ZnS QDs are scarce. Neurons play extraordinary roles in regulating the activities of organs and systems, and serious consequences occur when neurons are damaged. Currently, the potential toxicity of CuInS2/ZnS QDs on neurons has not been fully elucidated. Here, we investigate the neurotoxicity of PEGylated CuInS2/ZnS (CuInS2/ZnS-PEG) QDs on neuron-like PC12 cells. We found that CuInS2/ZnS-PEG QDs were taken up by PC12 cel

Mixed Redox-Couple-Involved Chalcopyrite Phase CuFeS2 Quantum Dots for Highly Efficient Cr(VI) Removal.

Iron-based nanosized ecomaterials for efficient Cr(VI) removal are of great interest to environmental chemists. Herein, inspired by the "mixed redox-couple" cations involved in the crystal structure and the quantum confinement effects resulting from the particle size, a novel type of iron-based ecomaterial, semiconducting chalcopyrite quantum dots (QDs), was developed and used for Cr(VI) removal. A high removal capacity up to 720 mg/g was achieved under optimal pH conditions, which is superior to those of the state-of-the-art nanomaterials for Cr(VI) removal. The mechanism of Cr(VI) removal wa…

Nitrogen-Doped Carbon Quantum Dots for Preventing Biofilm Formation and Eradicating Drug-Resistant Bacteria Infection.

The development of novel antimicrobial agents is a top priority in the fight against drug-resistant bacteria. Here, we synthesized a green nanoantibiotic, nitrogen-doped carbon quantum dots (N-CQDs) from bis-quaternary ammonium salt (BQAS) as carbon and nitrogen sources. The as-obtained N-CQDs possess high antibacterial activity (>99%) against both methicillin-resistant Staphylococcus aureus (MRSA) and Ampicillin-resistant Escherichia coli bacteria in vitro than some known clinical antibiotics (vancomycin and gentamicin). The N-CQDs can kill MRSA pathogens without inducing resistance, prevent …

Ligand-assisted synthesis and cytotoxicity of ZnSe quantum dots stabilized by N-(2-carboxyethyl)chitosans.

Here we report a green synthesis of ZnSe quantum dots (QDs) in aqueous solution of polyampholyte chitosan derivative - N-(2-carboxyethyl)chitosan (CEC) with substitution degrees (DS) from 0.7 to 1.3 and molecular weight (MW) of 40 kDa and 150 kDa. We have shown that the maximum intensity of photoluminescence (PL) is exhibited by ZnSe QDs synthesized in solutions of CEC with DS 1 at Se:Zn molar ratio 1:2.5. The defect-related band was predominant in the PL spectra of ZnSe QDs obtained at room temperature; however, hydrothermal treatment at 80-150 °C during 1-2 h significantly increased contribu…

Ratiometric sensor based on imprinted quantum dots-cationic dye nanohybrids for selective sensing of dsDNA.

A ratiometrically responsive sensor for dsDNA is reported, based on molecularly imprinted polymer coated quantum dots (MIP-QDs). A new platform is described for probing dsDNA by tracing the "turn on" fluorescence signal of malachite green (MG) as a cationic dye and "turn off-on" room temperature phosphorescence (RTP) signal of MIP-QDs/MG nanohybrids. The interaction between MIP-QDs surface and MG discloses an intense quenching in RTP (turning off) by a phosphorescence resonance energy transfer (PRET) process. After the addition of dsDNA, MG molecules escape from the MIP-QDs surface and interca…

A novel quartz-crystal microbalance humidity sensor based on solution-processible indium oxide quantum dots.

Having a large surface area, like the quantum confinement effect also caused by the nano-level size of quantum dots (QDs), creates fantastic potential for humidity sensing. A high concentration of surface adsorption sites initiates an increased response. Porosity between QDs allows fast water vapor penetration and outflow. Here, a quartz-crystal microbalance (QCM) humidity sensor was prepared using indium oxide (In2O3) QDs, synthesized via a solvothermal method. After the In2O3 QDs were directly spin-coated onto the QCM, an annealing process removed organic long chains and exposed more moistur…

Rapid detection of aflatoxin B1 by dummy template molecularly imprinted polymer capped CdTe quantum dots.

A novel and sensitive fluorescent sensor was synthesized for the rapid and specific recognition of aflatoxin B1 (AFB1) by our combining molecular imprinting techniques with quantum dot technology. Molecularly imprinted polymers coated CdTe quantum dots (MIP@CdTe QDs) were prepared through the Stöber method with 5,7-dimethoxycoumarin as a dummy template. 3-Aminopropyltriethoxysilane was selected as the functional monomer, and tetraethyl orthosilicate was used as the cross-linking agent. The best molar ratio of 5,7-dimethoxycoumarin to functional monomer to cross-linker was 4:20:15. The MIP@CdTe…

Changes in metallothionein transcription levels in the mussel Mytilus galloprovincialis exposed to CdTe quantum dots.

Quantum dots (QDs) are a class of engineered nanoparticles (ENPs) with several biomedical, industrial and commercial applications. However, their metabolism and detoxification process in aquatic invertebrates and environmental health hazards remain unclear. This study investigate the transcriptional changes of metallothioneins (MTs) isoforms (mt10IIIa and mt20IV) induced by CdTe QDs, in comparison with its dissolved counterpart, in the marine mussel Mytilus galloprovincialis. Mussels were exposed to CdTe QDs and to the same Cd concentration (10 µg Cd L-1) of dissolved Cd for 14 days and mt tra…

Interaction of carboxylated CdSe/ZnS quantum dots with fish embryos: Towards understanding of nanoparticles toxicity.

Due to colloidal instability even with protective coatings, nanoparticles tend to aggregate in complex environments and possibly interact with biota. In this study, visualization of quantum dots (QDs) interaction with rainbow trout (Oncorhynchus mykiss) embryos was performed. Studies on zebrafish (Danio rerio) and pearl gourami (Trichogaster leerii) embryos have shown that QDs interact with embryos in a general manner and their affects are independent on the type of the embryo. It was demonstrated that carboxylated CdSe/ZnS QDs (4 nM) were aggregating in accumulation media and formed agglomera…

A novel CdTe quantum dots probe amplified resonance light scattering signals to detect microRNA-122.

We report a rapid and facile resonance light scattering (RLS) technique that utilizes CdTe quantum dots (CdTe QDs) probe to detect microRNA-122. The RLS sensor is ingeniously designed with P1 and P2, two cDNA sequence probes with partially complementary sequences to miRNA-122. The amine-modified P1 and P2 are coupled to the surface of QDs to form functional QDs-P1 and QDs-P2 conjugates, which are collectively referred to as QDs-P. The cDNAs hybridize with the target miRNA to rapidly induce the self-assembly of QDs probe and change RLS intensity. The proposed technique can detect miRNA-122 with…

Covalent Protein Labeling and Improved Single-Molecule Optical Properties of Aqueous CdSe/CdS Quantum Dots.

Semiconductor quantum dots (QDs) have proven to be superior probes for single-molecule imaging compared to organic or genetically encoded fluorophores, but they are limited by difficulties in protein targeting, their larger size, and on-off blinking. Here, we report compact aqueous CdSe/CdS QDs with significantly improved bioconjugation efficiency and superior single-molecule optical properties. We have synthesized covalent protein labeling ligands (i.e., SNAP tags) that are optimized for nanoparticle use, and QDs functionalized with these ligands label SNAP-tagged proteins ∼10-fold more effic…

The stability and removal of water-dispersed CdSe/CdS core-shell quantum dots from water.

The increasingly wide use of semiconductor nanocrystals inevitably leads to their release into aquatic environment. The aggregation behaviors of 3-mercaptopropionic acid-capped CdSe/CdS core-shell quantum dots (MPA-QDs) under various water chemistry conditions were examined and their removal using Fe3+ and Al3+ coagulants was evaluated. Cationic species rather than concentrations affected the stability of MPA-QDs. Adding 2 mM Ca2+ led to a much larger ζ-potential decrease and particle size increase than adding 150 mM K+ at each tested solution pH. This indicated that complexation and depletion…

Sensitive and Selective Detection of Oxo-Form Organophosphorus Pesticides Based on CdSe/ZnS Quantum Dots.

A rapid, sensitive and enzyme-based optical biosensor was applied for the determination of seven organophosphorus pesticides (OPPs), including the oxo forms (malaoxon, paraoxon, dibrom, and dichlorvos), the thio forms (malathion and parathion) and the mixed form (demeton) in Panax ginseng. The principal of the proposed method is that the fluorescence quenching effect of quantum dots (QDs) can be observed by enzyme-generated H₂O₂. The active centers of acetylcholinesterase (AChE) could be inhibited in the presence of pesticides, which caused decrease of the generated H₂O₂. Then, the inhibition …

CuFeS2 Quantum Dots and Highly Luminescent CuFeS2 Based Core/Shell Structures: Synthesis, Tunability, and Photophysics.

We report the synthesis of copper iron sulfide (CuFeS2) quantum dots (QDs). These materials exhibit a tunable band gap that spans the range 0.5-2 eV (600-2500 nm). Although the as-prepared material is nonemissive, CuFeS2/CdS core/shell structures are shown to exhibit quantum yields that exceed 80%. Like other members of the I-III-VI2 family QDs, CuFeS2 based nanoparticles exhibit a long-lived emission that is significantly red-shifted compared to the band gap. CuFeS2 QDs are unique in terms of their composition. In particular, these QDs are the only band-gap-tunable infrared chromophore compos…
