Tenascin-C orchestrates radiotherapy-induced head and neck tumor regression.

Given that head and neck squamous cell carcinoma (HNSCC) patients have poor survival outcomes, a better understanding of the therapeutic benefits of ionizing irradiation (IR), the major treatment modality besides surgery, is needed. A confounding factor is the immunosuppressive tumor microenvironment determined by tenascin-C (TNC), a highly abundant extracellular matrix molecule upregulated by IR. We investigated the roles of TNC on radio-induced tumor regression in a murine oral HNSCC model expressing or lacking TNC. While tumors in a TNC-expressing host were radiosensitive, they were radiore

KLK10 Upregulation Drives Aggressiveness and Radioresistance and Has a Negative Prognostic Impact on Rectal Cancer.

PURPOSE: Rectal cancer is a heterogeneous disease with widely variable treatment responses. Beyond the TNM staging system, no reliable biomarkers currently exist to predict the efficacy of concurrent chemoradiotherapy (CCRT) in rectal cancer, limiting the personalization of curative-intent treatment. Proteolytic enzymes promote extracellular matrix degradation and tumor progression, with serine proteases playing a key role. Consequently, this study intended to investigate their potential to predict preoperative CCRT response and survival in rectal cancer. MATERIALS AND METHODS: Associations b

Inhibition of SIRT7 Overcomes Radioresistance in Pancreatic Neuroendocrine Tumors by Reactivating MEN1 Expression.

Pancreatic neuroendocrine tumors (PanNETs) frequently exhibit loss or reduced expression of the tumor suppressor MEN1, a key regulator of tumor progression and DNA damage response (DDR). However, the upstream mechanisms driving MEN1 silencing remain unclear. Here, a pooled epigenetic CRISPR-Cas9 screen identified the NAD+-dependent deacetylase SIRT7 as a critical suppressor of MEN1 expression. Mechanistically, SIRT7 interacts with DNMT1 and promotes catalytic activity-dependent recruitment of DNMT1 to the MEN1 promoter, leading to promoter hypermethylation and transcriptional repression. Radiation Adaptive Response Enhances Hematopoietic Stem and Progenitor Cells Radioresistance in Mice: IL-17 Signaling Pathway Activation as the Underlying Mechanism.

Radiation adaptive response (RAR) significantly improves resistance to subsequent high-dose radiation exposure. While hematopoietic stem and progenitor cells (HSPCs) are known to be crucial for radiation protection, their specific roles and response mechanisms in RAR remain unclear. Systematic investigation of these processes could reveal valuable therapeutic targets for developing HSPCs-focused radiation protection strategies. We establish a radiation-adaptive mouse model by administering a priming dose of 0.5 Gy γ rays after a 14-day interval and then investigate the protective effects and u

Isoform-specific expression of peroxiredoxins in human cancer cell lines with various radioresistance.

To investigate the role of peroxiredoxin isoforms (Prdx) in the radioresistance of cancer cells, the expression of Prdx1-6, DNA repair genes, and apoptosis regulators was studied in human cancer cell lines with varying radiosensitivities (A549, Caco-2, and MCF-7) after exposure to ionizing radiation. A correlation was found between high constitutive Prdx1-6 expression levels and increased radioresistance. Predominantly cytosolic isoforms Prdx2 and Prdx6 demonstrated pronounced induction after irradiation, indicating their critical role in protecting against radiation-induced oxidative stress.

Nanomechanical Single-Cell Profiling Reveals Mechanical Dormancy Underlying Radiation Resistance in Polyploid Giant Cancer Cells.

Radiation therapy induces DNA damage primarily through reactive oxygen species, leading to cancer cell apoptosis. However, intratumoral heterogeneity and spatial dose variations often result in the survival of polyploid giant cancer cells (PGCCs), a therapy-resistant subpopulation characterized by multinucleation, genetic instability, and stem-like features. Particularly in malignant breast cancer, PGCCs contribute to recurrence by adopting a dormant yet invasive phenotype. Despite their clinical relevance, reliable tools to identify or characterize these cells remain lacking. Here, we present

Mapping radiosensitivity in glioblastoma using MR elastography and biomechanical modeling.

Glioblastoma multiforme (GBM) remains a devastating disease with a poor prognosis. GBM progression generates mechanical forces that compromise blood vessels' functionality, impair perfusion, and create hypoxic regions, which in turn reduce radiosensitivity. While the link between hypoxia and radiosensitivity is known, a quantitative methodology to predict this effect is lacking. Here, we developed a patient-specific mechanistic mathematical model of radiotherapy that integrates Magnetic Resonance Elastography (MRE) imaging. The model can translate MRE-derived stiffness into spatial maps of mec

Unraveling the effects of FLASH and conventional irradiation on retinal pigment epithelial cells: in vitro and in vivo studies.

The retinal pigment epithelium (RPE) is a fundamental monolayer of pigmented cells that supports visual function, situated between the neural retina and choroidal blood vessels. Radiotherapy is a common treatment for ocular tumors such as uveal melanoma; however, the RPE is inevitably exposed to radiation during treatment. FLASH radiotherapy, characterized by ultra-high dose-rates, has emerged as a promising approach to minimize normal tissue toxicity while maintaining tumor control. Despite its potential, few preclinical studies have explored the effects of FLASH irradiation on the RPE, and n

HEYL-mediated activation of LAMA3 influences radiotherapy response in esophageal cancer.

BACKGROUND: Esophageal cancer (EC) is a major cause of cancer-related mortality. This study aims to investigate how hairy/enhancer-of-split is related to YRPW motif-like protein (HEYL), which regulates laminin subunit alpha 3 (LAMA3) and its impact on EC radiotherapy via the epithelial-to-mesenchymal transition (EMT) pathway. METHODS: Differentially expressed genes (DEGs) related to radiotherapy sensitivity in EC cells were screened. Radioresistant cells were constructed using X-ray treatment at different doses. LAMA3 expression in radioresistant and parental cells was validated using RT-qPCR

Multi-proteomics investigation of the early response to X-rays and carbon ion irradiations of HeLa cells.

Despite the considerable decline of cervical cancer incidence in developed countries, the disease remains a public health problem in low-income and middle-income countries due to the low popularity of human papillomavirus vaccination and cervical cancer screening. Mainly treated with radiotherapy, the number of recurrences linked to radioresistance increases in women suffering from this disease and constitutes major obstacle. Here, we perform a combined proteomic and phosphoproteomic profiling of HeLa cervical cancer cells after in vitro treatment with X-rays and carbon ions. We observed diffe

In vitro study of radiosensitivity in colorectal cancer cell lines associated with Lynch syndrome.

INTRODUCTION: Lynch syndrome patients have an inherited predisposition to cancer due to a deficiency in DNA mismatch repair (MMR) genes which could lead to a higher risk of developing cancer if exposed to ionizing radiation. This pilot study aims to reveal the association between MMR deficiency and radiosensitivity at both a CT relevant low dose (20 mGy) and a therapeutic higher dose (2 Gy). METHODS: Human colorectal cancer cell lines with (dMMR) or without MMR deficiency (pMMR) were analyzed before and after exposure to radiation using cellular and cytogenetic analyses i.e., clonogenic assay

Borrelia burgdorferi radiosensitivity and Mn antioxidant content: antigenic preservation and pathobiology.

The bacterium responsible for Lyme disease, Borrelia burgdorferi, accumulates high levels of manganese without iron and possesses a polyploid genome, characteristics suggesting potential extreme resistance to radiation. Contrary to expectations, we report that wild-type B. burgdorferi B31 cells are radiosensitive, with a gamma-radiation survival limit for 106 wild-type cells of <1 kGy. Thus, we explored B. burgdorferi radiosensitivity through electron paramagnetic resonance (EPR) spectroscopy by quantitating the fraction of Mn2+ present as antioxidant Mn2+ metabolite complexes (H-Mn). Tumor hypoxia: a new PET imaging biomarker in clinical oncology.

Tumor hypoxia is associated with tumor progression and resistance to various treatments. Noninvasive imaging using positron emission tomography (PET) and F-18-labeled fluoromisonidazole (FMISO) was recently introduced in order to define and quantify tumor hypoxia. The FMISO uptake was closely correlated with pimonidazole immunohistochemistry and hypoxia-inducible factor 1 expression in basic studies. Tumor hypoxia in head and neck cancers and other tumors in a clinical setting may also indicate resistance to radiation and/or chemotherapy. Hypoxic imaging may thus play a new and important role

Radiosensitization of yeast cells by inhibition of histone h4 acetylation.

Deletion of genes for proteins involved in histone H4 acetylation produces sensitivity to DNA-damaging agents in both Saccharomyces cerevisiae and mammalian cells. In the present studies, we show that treating wild-type yeast cells with histone acetyl transferase (HAT) inhibitors, which are chemicals that cause a global decrease in histone H4 acetylation, sensitizes the cells to ionizing radiation. Using HAT inhibitors, we have placed histone H4 acetylation into the RAD51-mediated homologous recombination repair pathway. We further show that yeast cells with functionally defective HAT proteins

Tolerance of normal human brain to boron neutron capture therapy.

Data from the Harvard-MIT and the BNL Phase I and Phase I/II clinical trials, conducted between 1994 and 1999, have been analyzed and combined, providing the most complete data set yet available on the tolerance of the normal human brain to BPA-mediated boron neutron capture therapy. Both peak (1cm(3)) dose and average whole-brain dose show a steep dose-response relationship using somnolence syndrome as the clinical endpoint. Probit analysis indicates that the doses associated with a 50% incidence for somnolence (ED(50)+/-SE) were 6.2+/-1.0 Gy(w) for average whole-brain dose and 14.1+/-1.8 Gy(

Mucocutaneous side effects of antineoplastic chemotherapy.

All structures of the skin may be affected by side effects of antineoplastic chemotherapy. The most commonly described effects concern skin adnexes, especially hair with alopecia. Nails are also frequently involved. Eccrine sweat or sebaceous gland involvement is more rarely reported. Mucous membranes, particularly in the mouth, are frequently altered by several mechanisms; direct cytotoxicity, infection, and a decrease in polymorphonuclear or platelet counts. Among cutaneous side effects, hyperpigmentation is very common and may have different clinical patterns; generalised, figurated, or loc

Resistance to radiation-induced apoptosis in Burkitt's lymphoma cells is associated with defective ceramide signaling.

Increased sensitivity to ionizing radiation has been shown to be due to defects in double-strand break repair and mutations in the proteins that detect DNA damage. However, it is now recognized that the cellular radiation response is complex and that radioresistance/radiosensitivity may also be regulated at different levels in the radiation signal transduction pathway. Here, we describe a direct relationship between resistance to radiation-induced apoptosis and defective ceramide signaling. Radiation sensitivity in human tumor cells correlated with the immediate accumulation of the second mess
