We report a case of fixed sporotrichosis in a 2-year-old girl who showed a reddish nodule on her left cheek for 1 year without trauma. Fungi flora-Y staining showed fungal elements from the exudate onthe erosion of this nodule. Sporothrix schenckii was isolated from a culture of the fungus taken from the biopsy specimen. Treatment with itraconazole 4.5 mg/kg/day for 16 weeks resulted in an atrophic scar after 2.1 mg/kg/day for 8 weeks without side effects. Six months later, her eruption recurred and wastreated with potassium iodide for 21 weeks with complete resolution. During the 1 year follo…

Congenitally athymic nude (nu/nu) mice showed higher sensitivity to intratestical infection of Sporothrix schenckii than phenotypically normal littermates (nu/+). Active immunization with viable cells enhanced the resistance to intravenous (i.v.) infection of this fungus in BALB/c mice. Nu/nu mice transferred with immune spleen cells acquired the enhancement of resistance to the infection with S. schenckii, but not ones with normal spleen cells. Pre-treatment of OK-432 (Picibanil), one of the macrophage activating agents, enhanced resistance to i.v. infection of this fungus in BALB/c mice. On …

BACKGROUND: Sporotrichosis is a subcutaneous mycosis caused by the traumatic inoculation of dimorphic fungi from the genus Sporothrix. Traditional diagnostic methods, including direct microscopy and fungal culture, are time-consuming, require specialized expertise, and may yield false-negative results.
AIMS: This systematic review and meta-analysis aimed to evaluate the diagnostic accuracy of the enzyme-linked immunosorbent assay (ELISA) technique in detecting sporotrichosis in humans and animals.
METHODS: A comprehensive literature search was conducted across PubMed, LILACS, EMBASE, Scopus,…

Twenty-five years have passed since the initial observation of endemic zoonotic sporotrichosis in Rio de Janeiro, Brazil. Since then, this disease has spread throughout South America. Accompanying the emergence of this mycosis, some progress has been made, including the expansion of a research network in this field and higher visibility of sporotrichosis within government authorities and funding agencies. However, there are still some challenges to curbing the expansion of this disease in the coming years. These include the development of rapid and accurate diagnostic tests, new antifungal dru

Vertebrate animal models are essential in research; however, efforts need to be made to decrease animal suffering as much as possible. It could be useful to determine humane endpoints that could serve as surrogates for a fatal outcome. We address this issue with respect to infectious diseases. We propose a humane endpoint for studies of Sporothrix brasiliensis infection. BALB/c mice were inoculated subcutaneously in the footpad. To define a humane endpoint, we considered two groups: animals who died during the experiment, and those euthanized at the end of the experiment. The groups were compa…

BACKGROUND: Systemic sporotrichosis occurs when organs, other than subcutaneous tissues and lymph nodes, are infected. Interestingly, systemic sporotrichosis in apparently immunocompetent individuals is increasing in Brazil, but data on clinical manifestations and risk factors are scarce. Most of the existing data on such condition relate to people living with HIV. We aimed to study the risk factors associated with systemic sporotrichosis among HIV-negative and HIV-positive patients.
METHODS: We performed a retrospective cross-sectional study with 80 patients from Brazil, diagnosed between 20

The aim of this study was to develop a microemulsion (ME) formulation containing an association of itraconazole (ITC) and clotrimazole (CLT) … T, indicating that CLT acts as a promoter enhancer. In the in vitro antifungal activity assay using Sporothrix brasiliensis yeast, the inhibition halo produced by ITC/CLT-ME exhibited a mean diameter of 43.67 ± 2.31 mm. The ITC/CLT-ME formulation did not cause skin irritation in mice. The results suggest that ITC/CLT-ME is a promising tool for the transderma …

We excised surgically a feline granulomatous lesion and performed histopathological, mycological and molecular examinations. As a result, it was diagnosed as sporotrichosis, which was the second recorded case of a cat so afflicted in Japan. After the operation, we recognized another nodule on the lymph node. Histopathological examination was therefore performed, but no fungi were detected. To prevent recurrence, the cat was administered a antimycotic drug, itraconazole. As a result, no recurrence was found. Excision of the lesion is the treatment of choice for feline sporotrichosis.

We report on a case of sporotrichosis in which lesions occurred bilaterally on upper limbs at different periods. The patient was an 84-year-old farmer living in Tsukuba City, Japan. The first lesion appeared on the left upper arm and remained untreated. The second lesion appeared on the back of the right hand 5 years later. Findings suggesting internal metastasis could not be confirmed in various examinations. We judged that the lesion of the left upper arm was primary and the lesion at the back of the right hand was caused by autoinoculation.

Phagocytosis plays an important role as a protective mechanism against infections, since polymorphonuclear leukocytes (PMN) and macrophages are the first cellular lines opposed to aggressive microorganisms. In patients with sporotrichosis a diminished capability of killing engulfed yeast by their PMN has been described, but the origin of this deficiency remains unknown. In this work, partial aspects of the oxidative metabolism of PMN leukocytes and peritoneal macrophages of mongolian gerbils experimentally infected with sporotrichosis were studied. For this purpose the nitroblue tetrazolium (N

Nodular lymphangitis, characterized by inflammatory nodules along the lymphatics draining a primary skin infection, most commonly follows superficial inoculation with Sporothrix schenckii, Nocardia brasiliensis, Mycobacterium marinum, Leishmania (Viannia) panamensis/guyanensis, and Francisella tularensis. Epidemiologic context, clinical presentation, and presumed incubation period help to predict the specific etiologic microorganism. Sporotrichosis, often occurring in gardeners, remains the most recognized cause of nodular lymphangitis. Injuries sustained in marine environments suggest Mycobac…

Sporotrichosis is a zoonotic implantation mycosis caused by dimorphic fungi of the genus Sporothrix. Transmission occurs through traumatic inoculation of the fungus and may involve different routes, including sapronotic transmission from contaminated soil, plants, and organic matter, zoonotic transmission mainly associated with infected felines, and, less frequently, contaminated fomites. The disease remained neglected for many years; however, several regions have recently reported outbreaks that are difficult to control. Thus, this study aimed to describe the emergence of sporotrichosis in th…
