Background ion shift induces microbial compensatory and structural remodeling in anammox granules.

Stable municipal anammox treatment requires the retention of slow-growing anammox bacteria within structurally robust granules. However, the role of background cation availability in maintaining mineral-associated granule stability remains unclear. Here, parallel continuous-flow anammox reactors were operated under contrasting cation conditions to determine how influent hydrochemistry affects granule persistence and reactor performance. The control reactor (R1) maintained stable and efficient removal of nitrogen (over 80%), whereas the cation-limited reactor (R2) showed progressive deteriorati

Impact of palatopharyngeal sizes changing on pharyngeal airflow fluctuation and airway vibration in a pediatric airway.

Snoring is common in children and is associated with many adverse consequences. One must study the relationships between pharyngeal morphology and snoring physics to understand snoring progression. Although some model studies have provided fluid-structure interaction dynamic descriptions for the correlation between airway size and snoring physics, the descriptions still need to be further investigated in patient-specific airway models. Fluid-structure interaction studies using patient-specific airway structures complement the above model studies. Based on reported cephalometric measurement met

1,4-Disubstituted 1H-1,2,3-Triazole Containing Peptidotriazolamers: A New Class of Peptidomimetics With Interesting Foldamer Properties.

Peptidotriazolamers are hybrid foldamers with features of peptides and triazolamers, containing alternation of amide bonds and 1,4-disubstituted 1H-1,2,3-triazoles with conservation of the amino acid side chains. We report on the synthesis of a new class of peptidomimetics, containing 1,4-disubstituted 1H-1,2,3-triazoles in alternation with amide bonds and the elucidation of their conformational properties in solution. Based on enantiomerically pure propargylamines bearing the stereogenic center in the propargylic position and α-azido esters, building blocks were obtained by copper-catalyzed a

Structurally- and dynamically-driven allostery of the chymotrypsin-like proteases of SARS, Dengue and Zika viruses.

Coronavirus 3C-like and Flavivirus NS2B-NS3 proteases utilize the chymotrypsin fold to harbor their catalytic machineries but also contain additional domains/co-factors. Over the past decade, we aimed to decipher how the extra domains/co-factors mediate the catalytic machineries of SARS 3C-like, Dengue and Zika NS2B-NS3 proteases by characterizing their folding, structures, dynamics and inhibition with NMR, X-ray crystallography and MD simulations, and the results revealed: 1) the chymotrypsin fold of the SARS 3C-like protease can independently fold, while, by contrast, those of Dengue and Zik

A structure-specific nucleic acid-binding domain conserved among DNA repair proteins.

SMARCAL1, a DNA remodeling protein fundamental to genome integrity during replication, is the only gene associated with the developmental disorder Schimke immuno-osseous dysplasia (SIOD). SMARCAL1-deficient cells show collapsed replication forks, S-phase cell cycle arrest, increased chromosomal breaks, hypersensitivity to genotoxic agents, and chromosomal instability. The SMARCAL1 catalytic domain (SMARCAL1(CD)) is composed of an SNF2-type double-stranded DNA motor ATPase fused to a HARP domain of unknown function. The mechanisms by which SMARCAL1 and other DNA translocases repair replication

The Modified Alar Batten Graft as a Tip-Alar Superstructure: A Technique to Restore Soft Triangle Stability and External Valve Support.

Lateral nasal wall insufficiency and external nasal valve collapse are frequent causes of both functional impairment and aesthetic deformity following primary or previous nasal surgery. Traditional alar batten grafts provide structural reinforcement but have a limited impact on alar margin contour and tip-alar harmony. We present a modification of the classic batten graft, placed over the lateral crura as a superstructure following tip framework construction during open rhinoplasty, through an illustrative case. The graft is carved in a trapezoidal configuration and secured at the domal level,

Spatial-Bioenergetic-Immune Axis in Type 2 Diabetes Mellitus-Associated Sarcopenia: A Review from Pathogenesis to Targeted Reconstruction.

Type 2 diabetes mellitus (T2DM)-associated sarcopenia extends beyond isolated metabolic dysregulation; consequently, standard glucose-centric therapeutic strategies are insufficient to arrest myofibrillar atrophy. In this review, we characterize this pathogenesis through a novel spatial-bioenergetic-immune axis, stratifying disease progression across three hierarchical levels. First, within the tissue niche, the proliferation of CD90⁺ fibro-adipogenic progenitors (FAPs) establishes a pro-inflammatory microenvironment, driving extracellular matrix fibrosis that physically constrains the myogeni

The Erlangen Program in lateral occipital cortex: Hierarchical encoding of emergent features.

Emergent features are fundamental concepts in Gestalt psychology, yet the neural encoding of these features, particularly a quantitative understanding of their relative superiority, remains elusive. This study bridges this gap by conceptualizing emergent features through geometric transformations within the Erlangen Program, which provides a principled framework to quantify their hierarchical relationships. We propose that the lateral occipital cortex (LOC) encodes these emergent features in accordance with the geometric hierarchies defined by this program. Using fMRI and multivariate pattern

Fine-tuning FAM161A gene augmentation therapy to restore retinal function.

For 15 years, gene therapy has been viewed as a beacon of hope for inherited retinal diseases. Many preclinical investigations have centered around vectors with maximal gene expression capabilities, yet despite efficient gene transfer, minimal physiological improvements have been observed in various ciliopathies. Retinitis pigmentosa-type 28 (RP28) is the consequence of bi-allelic null mutations in the FAM161A, an essential protein for the structure of the photoreceptor connecting cilium (CC). In its absence, cilia become disorganized, leading to outer segment collapses and vision impairment.

Ultrasound Shear Wave Elastography of the Arch-Supporting Structures in Symptomatic Flatfoot: A Pilot Study.

BACKGROUND: The posterior tibial tendon (PTT), deltoid ligament, and spring ligament are often torn or attenuated in patients with progressive collapsing foot deformity. The goal of this pilot study was to measure the ultrasound shear wave velocity (SWV) of these arch-supporting structures in feet with varying degrees of deformity to improve our understanding of their role in the progression of deformity. METHODS: Two observers measured the SWV of the supramalleolar and inframalleolar PTT in long and short axes, in the tibiospring portion of the deltoid ligament, and in the superomedial band

Electrosprayed hierarchical mesoporous Mn0.5Ti2(PO4)3@C microspheres as promising High-Performance anode for Potassium-Ion batteries.

NASICON-structured Ti-based polyanion compounds benefit from a stable structural framework, large ion channels, and fast ion mobility. However, the large radius of potassium and its poor electronic conductivity restrict its use in potassium-ion batteries. Herein, hierarchical mesoporous Mn0.5Ti2(PO4)3@C microspheres have been successfully synthesized using a simple electrospraying method. These microspheres consist of Mn0.5Ti2(PO4)3 nanoparticles evenly embedded in three-dimensional mesoporous carbon microspheres. The hierarchical mesoporous micro/nanostructure facilitates the rapid insertion

Anatomical Brain Changes and Cognitive Abilities in Patients with Obstructive Sleep Apnea Syndrome and Nonalcoholic Fatty Liver Disease.

Obstructive sleep apnea (OSA) is characterized by repetitive complete or partial collapse of the upper airway and reduction of airflow during sleep. It is associated with significantly increased daytime muscle sympathetic nerve activity thought to result from the repetitive intermittent periods of hypoxemia during sleep and brain alterations that are likely to result. Different brain regions are affected by subsequent hypoxia/anoxia. Neurodegenerative processes result in measurable atrophy of cortical gray matter in the temporal lobes and posterior cingulate cortex, as well as in subcortical s

New findings on the functional morphology of the male genitalia of the blood-feeding bug Rhodnius prolixus in the light of a different observation on living tissue.

Among insect species, the male genitalia evolved many different anatomical features to ensure reproductive success. Males of primitive insects do not copulate but transfer sperm packaged in a spermatophore to the female, whereas males of the most advanced insects copulate and deliver free sperm to the storage organ, the spermatheca, in the female reproductive tract. Based on previous studies on several species of triatomine bugs (Rhodnius prolixus included), Triatominae are thought to use an intermediate form of sperm transfer in which the male genitalia have evolved to carry out both copulati

Intermittent brugada syndrome presenting with syncope in an adult female.

Background. Brugada syndrome accounts for 4-12% of all sudden deaths worldwide and at least 20% of sudden deaths in patients with structurally normal hearts. Case Report. A 48-year-old female presented to the emergency department after two witnessed syncopal episodes. While awaiting discharge had a third collapse followed by cardiac arrest with shockable rhythm. Initial electrocardiogram showed wide QRS complex with left axis deviation, ST-segment elevation of <1 mm in V1 and V2, and flattening of T waves in V1. The angiogram did not demonstrate obstructive coronary disease. The electrocardiog

Crystal structures of Plasmodium falciparum cytosolic tryptophanyl-tRNA synthetase and its potential as a target for structure-guided drug design.

Malaria, most commonly caused by the parasite Plasmodium falciparum, is a devastating disease that remains a large global health burden. Lack of vaccines and drug resistance necessitate the continual development of new drugs and exploration of new drug targets. Due to their essential role in protein synthesis, aminoacyl-tRNA synthetases are potential anti-malaria drug targets. Here we report the crystal structures of P. falciparum cytosolic tryptophanyl-tRNA synthetase (Pf-cTrpRS) in its ligand-free state and tryptophanyl-adenylate (WAMP)-bound state at 2.34 Å and 2.40 Å resolutions, respectiv

Functional and structural alterations in the endoplasmic reticulum and mitochondria during apoptosis triggered by C2-ceramide and CD95/APO-1/FAS receptor stimulation.

C(2)-ceramide (C(2)-cer) and binding of the CD95/APO-1/FAS (aCD95) receptor are acknowledged inducers of apoptosis. In spite of that, their effects on the endoplasmic reticulum (ER) and mitochondria during early phases of apoptotic onset are poorly characterized. Here, by employing various approaches, we followed structural and functional modifications of these organelles at the beginning of cellular demise. In detail, we observed that C(2)-cer, but not CD95 activation, markedly modifies the morphology of the ER and promotes Ca(2+) release. Accordingly, mitochondria of C(2)-cer-treated, but no

Overexpression of PPK-1, the Caenorhabditis elegans Type I PIP kinase, inhibits growth cone collapse in the developing nervous system and causes axonal degeneration in adults.

Growth cones are dynamic membrane structures that migrate to target tissue by rearranging their cytoskeleton in response to environmental cues. The lipid phosphatidylinositol (4,5) bisphosphate (PIP(2)) resides on the plasma membrane of all eukaryotic cells and is thought to be required for actin cytoskeleton rearrangements. Thus PIP(2) is likely to play a role during neuron development, but this has never been tested in vivo. In this study, we have characterized the PIP(2) synthesizing enzyme Type I PIP kinase (ppk-1) in Caenorhabditis elegans. PPK-1 is strongly expressed in the nervous syste

Role of Cone-Beam Computed Tomography (CBCT) in Obstructive Sleep Apnea (OSA): A Comprehensive Review.

Obstructive sleep apnea (OSA) is characterized by recurrent partial or complete upper airway collapse during sleep. Accurate assessment of airway anatomy is crucial for risk stratification, diagnosis, and treatment planning. While polysomnography (PSG) is considered the gold standard for OSA diagnosis, it provides limited anatomical insights. Cone-beam computed tomography (CBCT) has emerged as a valuable tool with lower radiation dose for three-dimensional (3D) assessment of the upper airway space and craniofacial structures. CBCT enables precise measurement of critical airway parameters inclu

A deep eutectic solvent-assisted green synthesis of advanced three-dimensional covalent organic framework based stationary phase for dual-mode liquid chromatography.

Two-dimensional covalent organic frameworks (2D COFs) are widely used as stationary phases in liquid chromatography, yet their simple pore structures often mandate surface modifications to attain the necessary separation selectivity. In contrast, three-dimensional COFs (3D COFs) feature customizable hierarchical channel systems with complex pore structures, high specific surface areas, and spatially diversified active sites, enabling superior separation selectivity and enhanced column efficiency. Furthermore, their rigid frameworks prevent structural collapse of stationary phase, offering uniq
