The effect of iron substrate on mitochondrial haem synthesis in copper deficiency.

Studies of iron utilization and haem synthesis were carried out with hepatic mitochondria obtained from copper-deficient and pair-fed control rats. Ferric chloride can be used as Fe substrate for mitochondrial haem synthesis in the presence of succinate. Utilization is further enhanced by the addition of FMN. Ferritin does not support haem synthesis in the presence of succinate alone, but does support haem synthesis when FMN is added. Mitochondrial haem synthesis is impaired in Cu deficiency when either FeCl3 or homologous ferritin is used as Fe substrate. The results of the present study sugg

Factors Influencing Internal Medicine Resident Beta-Blocker Discontinuation in Acute Decompensated Heart Failure.

BACKGROUND: Heart failure is one of the leading causes of hospital admissions in North America. Although guidelines support the continuation of beta blockers on admission, hemodynamic considerations and mechanistic reasoning may prompt beta blocker discontinuation even in the absence of contraindications. Resident physicians often face this dilemma and are an important group in which to evaluate this decision-making. METHODS: Internal medicine residents at two institutions were presented with two scenarios: 1) whether to continue outpatient metoprolol succinate for a patient without evidence

Fractionation of dicarboxylic acids produced by Rhizopus oryzae using reactive extraction.

Fumaric, malic, and succinic acids have been selectively separated from their mixture obtained by Rhizopus oryzae fermentation using reactive extraction with Amberlite LA-2 dissolved in three solvents with different dielectric constants (n-heptane, n-butyl acetate, and dichloromethane). This technique allows recovering preferentially fumaric acid from the mixture, the raffinate containing only malic and succinic acids. The extractant concentration and organic phase polarity control the efficiency and selectivity of acids extraction. The increase of aqueous phase viscosity reduces the extractio

Intermediate metabolism in Trypanosoma cruzi.

Epimastigotes of Trypanosoma cruzi, the causative agent of Chagas disease, catabolize proteins and amino acids with production of MH3, and glucose with production of reduced catabolites, chiefly succinate and L-alanine, even under aerobic conditions. This "aerobic fermentation of glucose" is probably due to both the presence of low levels of some cytochromes, causing a relative inefficiency of the respiratory chain for NADH, reoxidation during active glucose catabolism, and the lack of NADH dehydrogenase and phosphorylation site I, resulting in the entry of reduction equivalents into the chain

Transcriptome analysis of a dog model of congestive heart failure shows that collagen-related 2-oxoglutarate-dependent dioxygenases contribute to heart failure.

Fibrosis is an important pathological mechanism in heart failure (HF) and is associated with poor prognosis. We analyzed fibrosis in HF patients using transcriptomic data. Genes differentially expressed between normal control and congestive HF (CHF) dogs included P3H1, P3H2, P3H4, P4HA2, PLOD1 and PLOD3, which belong to the 2-oxoglutarate-dependent dioxygenases (2OGD) superfamily that stabilizes collagen during fibrosis. Quantitative polymerase chain reaction analysis demonstrated 2OGD gene expression was increased in CHF samples compared with normal left ventricle (LV) samples. 2OGD gene expr

Iron-sulphur centres in mitochondria from Arum maculatum spadix with very high rates of cyanide-resistant respiration.

X-band electron-paramagnetic-resonance spectroscopy at 4.2--77K combined with measurements of oxidation-reduction potential was used to identify iron--sulphur centres in Arum maculatum (cuckoo-pint) mitochondria. In the oxidized state a signal with a derivative maximum at g = 2.02 was assigned to succinate dehydrogenase centre S-3. Unreduced particles showed additional signals at g = 2.04 and 1.98 (at 9.2 GHz), which may be due to a spin-spin interaction. In the reduced state a prominent signal at g = 1.93 and 2.02 was resolved into at least three components that could be assigned to centres S

The action of trialkyltin compounds on mitochondrial respiration. The effect of pH.

1. Inhibition of 2,4-dinitrophenol-stimulated respiration by trialkyltins is dependent on the presence of Cl(-) in the assay medium and is only apparent at acid pH values. It appears to be a result of the Cl(-)-OH(-) exchange mediated by trialkyltins. 2. In a KCl medium at alkaline pH values, the maximum rate of respiration produced by uncouplers is further increased by the presence of trialkyltins. 3. The inhibition of uncoupled succinate oxidation at acid pH values is not reversed by increasing the external substrate concentration, suggesting that depletion of intramitochondrial succinate is

Induction by oxygen of respiration and phosphorylation of anaerobically grown Escherichia coli.

The changes occurring in the respiratory enzymes of anaerobically grown Escherichia coli strain B and E. coli 15 T(-)A(-)U(-)bar during exposure to oxygen were studied. Reduced nicotinamide adenine dinucleotide (NADH) oxidase activity reached its peak soon after O(2) exposure; cytochrome content and succinate oxidase activity increased more slowly, and these increases paralleled each other. The activities of isocitrate and malate dehydrogenases also increased, but the increase was less than that of the succinate and NADH oxidases; exposure to O(2) had no effect on the succinate and NADH dehydr

PHEOCHROMOCYTOMA: A GENETIC AND DIAGNOSTIC UPDATE.

OBJECTIVE: Pheochromocytomas and paragangliomas (PPGLs) are neuroendocrine tumors derived from adrenal or extra-adrenal locations, respectively. Upon suspicion of PPGL, specific metabolomic, molecular, biochemical, imaging, and histopathologic studies are performed to prove, localize, treat, and monitor disease progression. Improved diagnostic tools allow physicians to accurately diagnose PPGL, even in patients presenting with small (<1 cm) or biochemically silent tumors, which previously delayed proper detection and treatment. METHODS: This review outlines the most updated approach to PPGL p

A multi-heme flavoenzyme as a solar conversion catalyst.

The enzyme flavocytochrome c3 (fcc3), which catalyzes hydrogenation across a C═C double bond (fumarate to succinate), is used to carry out the fuel-forming reaction in an artificial photosynthesis system. When immobilized on dye-sensitized TiO2 nanoparticles, fcc3 catalyzes visible-light-driven succinate production in aqueous suspension. Solar-to-chemical conversion using neutral water as the oxidant is achieved with a photoelectrochemical cell comprising an fcc3-modified indium tin oxide cathode linked to a cobalt phosphate-modified BiVO4 photoanode. The results reinforce new directions in th

Reye's syndrome: preservation of mitochondrial enzymes in brain and muscle compared with liver.

The activities of five mitochondrial enzymes tested in liver from patients with Reye's syndrome were measured. Citrate synthase, glutamic dehydrogenase, succinic dehydrogenase, pyruvate carboxylase, and pyruvate dehydrogenase were all outside of the range shown by control samples and well below them in activity. The activity of two extramitochondrial enzymes, glucose-6-phosphatase, which is a microsomal enzyme, and fructose-1,6-diphosphatase, which is a soluble enzyme, were in the normal range in samples from Reye's syndrome patients. In both muscle and brain the activities of the mitochondria

Anaerobic metabolic responses of sludge and mine wastewater-derived consortia to winery wastewater-related compounds.

Winery wastewater contains readily biodegradable organic matter, but some of its components can inhibit anaerobic microbial activity. This study compares two enriched microbial consortia, derived from anaerobic digester sludge (ADS) and mining-process wastewater (MPW), exposed to alcohols, carboxylic acids, and phenolics. Assays were conducted in acetate-fumarate-based media containing low or high concentrations of each compound class and a combined condition containing the compounds at their highest levels. Growth, fumarate consumption, succinate production, acetate turnover, and compound con

Oncometabolites in Cancer Metabolism: Mechanistic Insights and Biomarker Potential- A Narrative Review.

BACKGROUND AND AIMS: Cancer cells undergo adaptation in hostile tumor microenvironments and go through extensive metabolic reprogramming for flourishing. Oncogenic transformation caused by mutations in some metabolic enzymes results in the generation of some metabolic intermediates called onco-metabolites. A growing body of evidence highlights the role of oncometabolites such as 2-hydroxyglutarate (2-HG), fumarate, and succinate in driving oncogenesis through epigenetic remodeling, altered redox balance, and interrupted cellular signaling. These metabolites are usually accumulated due to mutat

Endothelial responses to angiogenic modulators highlight metabolic mechanisms underlying vascular dysfunction in preeclampsia.

INTRODUCTION: An elevated sFlt-1/PlGF ratio in preeclampsia (PE) directly impairs endothelial cell (EC) metabolism, leading to increased lactate dehydrogenase (LDH) release, lipid peroxidation, and mitochondrial dysfunction. This may reflect a distinct metabolite profile which correlates with systemic maternal metabolic dysregulation, as evidenced by the association between circulating sFlt-1 and succinate/glycine levels. METHODS: A cross-sectional case-control study was performed on plasma of PE patients (n = 54) in addition to an in vitro study using ECs. We correlated plasma metabolite pro

Most infants with dilating vesicoureteral reflux can be treated nonoperatively.

PURPOSE: Dilating vesicoureteral reflux provokes concern for physicians and parents that often leads to corrective surgery in young children. Since there are limited data describing the natural history of dilating vesicoureteral reflux in infants, we identified factors predictive of resolution/improvement in infants initially treated nonoperatively. MATERIALS AND METHODS: We reviewed the medical records of 90 infants 6 months old or younger from 2004 to 2010 who were referred for prenatal hydronephrosis or initial febrile urinary tract infection and found to have dilating vesicoureteral reflu

In situ determination of different dehydrogenase activity profiles in the linings of odontogenic keratocysts and radicular cysts.

The levels of succinate, lactate, glutamate, glycerophosphate and glucose-6-phosphate dehydrogenases within the linings of keratinizing and non-keratinizing odontogenic cysts were investigated using static end-point and continuously monitored Nitroblue Tetrazolium-based histochemical methods. The use of TV image analysis for quantification of formazan final reaction products was validated by demonstrating significant relationships between the integrated absorbance at 585 nm and the amount of formazan in, and thickness of, gelatin films containing reduced tetrazolium salt (r = 1.0, p < 0.001).

Enhancement of tumor cell susceptibility to lymphokine-activated killer cells by treatment with the streptococcal preparation OK432.

We investigated whether tumor cell lysis by LAK cells was augmented by treatment with OK432 in vitro. NK and LAK activity against K562 cells was not enhanced by their treatment with OK432. In contrast, the susceptibility of OK432-treated Daudi and KATO-III cells to lysis by LAK cells was enhanced. Succinate dehydrogenase activity and RNA synthesis were impaired in Daudi and KATO-III cells by treatment with OK432, and moreover the expression of HLA Class I antigen and beta 2-microglobulin was inhibited in OK432-treated KATO-III cells. Thus, it is suggested that the enhancement of the susceptibi

Catabolism of 4-aminobutyrate (GABA) and proteinogenic amino acids sustains the tricarboxylic acid cycle in Arabidopsis thaliana (L.) Heynh. plants under salinity stress.

Glutamate decarboxylases1,2,4 (GAD1,2,4) are calcium/calmodulin-activated enzymes that produce 4-aminobutyrate (GABA) and eventually lead to succinate (GABA shunt), which can theoretically be incorporated into the tricarboxylic acid cycle (TCAC) during abiotic stress. We tested the hypothesis that the GABA shunt is involved in the adaptive response of the TCAC to salinity stress. Quantitative analyses of amino and organic acids, as well as GAD1,2,4 expression, in shoots and roots of soil-grown, late-vegetative-stage Arabidopsis wild-type plants and GAD1,2,4 single, double and triple loss-of fu

Differential metabolic responses to hydrogen peroxide-induced oxidative stress in parotid and submandibular gland acinar cell lines.

This study explored how oxidative stress induced by hydrogen peroxide alters metabolite levels in submandibular and parotid salivary gland acinar cells. We investigated the impact of oxidative stress on the metabolite profiles of two salivary gland cell lines and examined differences in response. Rat-derived immortalized acinar epithelial cell lines from the parotid and submandibular salivary glands were exposed to 50 μM and 150 μM hydrogen peroxide for 24 h. Metabolite levels were then analyzed using liquid chromatography mass spectrometry, and succinate levels were independently assessed usi
