Integrative systems biology identifies PSMB2 as a core oxidative stress-associated target in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) represents one of the most aggressive and therapeutically challenging subtypes of breast cancer, characterized by the absence of estrogen receptor, progesterone receptor, and HER2 expression. Due to the lack of specific molecular targets, treatment options for TNBC remain limited, resulting in higher recurrence rates, increased metastatic potential, and poorer overall survival compared with other breast cancer subtypes. Therefore, identifying novel therapeutic targets and potential bioactive compounds is essential for improving TNBC management. In this stud

Integrative Analyses Identify a cGAS-STING Pathway-Driven Signature With Context-Dependent Roles in Systemic Lupus Erythematosus.

The cGAS-STING pathway is emerging as an essential driver in systemic lupus erythematosus (SLE). Here, we characterize the key signature of cGAS-STING pathway and its roles in SLE by leveraging large-scale transcriptomics, cell-based assays, and two lupus-like mouse models. We identify a STING-dependent gene signature termed M7core, enabling quantitative assessment of cGAS-STING pathway activity in SLE. M7core reveals widespread cGAS-STING pathway activation in 70.4% of 3,180 SLE samples and predicts therapeutic response to STING antagonists in 74.1% of patients, with higher activity indicatin

Applying a Complexity Lens to Policy Implementation: How Feedback Loops Help to Understand Systems Change in Integrated Healthcare.

BACKGROUND: Although a complexity theory lens can help to understand national policy implementation at a local government level, application of this lens often remains at a metaphorical level. We illustrate how complexity concepts, such as adaptation, feedback, emergence and co-evolution can be used practically in research on policy implementation using a UK case study (Future in Mind; FiM), aimed at improving school children's access to mental health services in a metropolitan borough in the North England. METHODS: We re-analysed interview data with staff from local government, National Heal

An integrated microfluidic system for automatic and self-validated analysis of cervical extracellular vesicle markers PD-L1 and ERBB3.

The early and precise diagnosis of gynecological malignancies, such as cervical cancer, is critical for improving patient treatments. Extracellular vesicles (EVs), such as exosomes, which carry molecular signals from their parental cells, offer a promising method for non-invasive liquid biopsy, however, conventional detection methods are often complex, high in reagent consumption, and susceptible to environmental fluctuations. To address this, we present an integrated, self-validated microfluidic system for the rapid, on-chip isolation and multiplexed identification of the gynecological EV mar

Refining Human Phenotype Ontology (HPO) to enable better phenotype-genotype integration in systemic autoimmune rheumatic diseases.

BACKGROUND: The Human Phenotype Ontology (HPO) provides a standardised framework for disease-phenotype associations. Given the complexity of systemic autoimmune rheumatic diseases (SARDs) and the absence of prior evaluations, we aimed to assess the completeness of HPO terms capturing SARD-related phenotypic features and SARD-phenotype associations. METHODS: Whole-exome sequencing was performed in 111 individuals from the LEAP (Lupus Extended Autoimmune Phenotype) cohort, a cohort of adult and paediatric SARD patients, who had at least one feature suggestive of a monogenic disease. We evaluate

Delphi consensus validation of an integrated climate sensitive diabetes management model for climate vulnerable health systems in Iran.

Type 2 diabetes mellitus (T2DM) is rising rapidly in Iran, where climate-related hazards such as heatwaves, sand and dust storms, and disruptions to food and medicine supply chains challenge continuity and quality of care. We sought to validate a climate-sensitive, health system-oriented management model for T2DM using a Delphi consensus process. Eighteen experts in diabetes, noncommunicable diseases and health systems rated 51 statements derived from the preliminary Integrated Iranian Climate-Sensitive Diabetes Management Model (ICDMM), mapped onto six domains aligned with WHO health system b

Advanced radiotherapy and systemic therapy in head and neck adenoid cystic carcinoma: Current progress and future integrated strategies.

Head and neck adenoid cystic carcinoma (HNACC) remains a therapeutic challenge because of its infiltrative behavior, perineural tropism, and high propensity for distant metastasis. Although surgery combined with radiotherapy remains the standard local approach, treatment outcomes remain suboptimal in patients with unresectable tumors, advanced local disease, or anatomically complex lesions. Advances in radiotherapy, including MRI-guided radiotherapy, FLASH radiotherapy, and proton or carbon-ion therapy, are reshaping local treatment by improving dose precision, normal tissue sparing, and oppor

Integrative post-GWAS analysis prioritizes immune regulatory pathways and candidate effector signals in systemic lupus erythematosus.

BACKGROUND: Systemic lupus erythematosus (SLE) has a complex polygenic architecture, but translating genome-wide association signals into biologically interpretable candidates remains challenging. We applied an integrative post-GWAS framework to refine SLE-associated loci and prioritize candidate regulatory mechanisms. METHODS: European-ancestry SLE GWAS summary statistics from FinnGen and Bentham et al. were meta-analysed, comprising 8417 cases and 354,277 controls. After quality filtering, 6,782,131 SNPs were retained. Downstream analyses included LAVA regional prioritization, Bayesian colo

[Neuroendocrine regulation of the reproductive system: integration of research of neuropeptide and experimental pathophysiology of amenorrhea of different genesis (literature review)].

Neuroendocrine regulation of reproductive function represents a complex system based on the integration of signals between the central nervous system and peripheral organs. In recent years, particular attention has been given to the role of neuropeptides - such as kisspeptin, brain-derived neurotrophic factor (BDNF), and orexins - in the pathogenesis of disorders associated with menstrual irregularities. This review provides a detailed analysis of the molecular mechanisms underlying neuropeptide regulation in functional hypothalamic amenorrhea (FHA), primary ovarian insufficiency (POI), and po
