Molecular characterization of the VP2 gene of infectious pancreatic necrosis virus (IPNV) isolates from Mexico.

Infectious pancreatic necrosis virus (IPNV) is one of the most important viruses in the Pacific salmon Oncorhynchus spp., Atlantic Salmon Salmo salar, and Rainbow Trout O. mykiss industry. This virus has been shown to produce high mortality among salmonid fry and juveniles, and survivors might become carriers. Since 2000, IPNV has affected Mexican Rainbow Trout culture, resulting in considerable economic losses. In the current study, molecular characterization of the VP2 gene of a number of Mexican IPNV isolates was done and the virus's phylogenetic relationships to IPNV reference strains were…

Bioaccumulation patterns of methyl mercury and essential fatty acids in lacustrine planktonic food webs and fish.

Organisms of the planktonic food web convey essential nutrients as well as contaminants to animals at higher trophic levels. We measured concentrations of methyl mercury (MeHg) and essential fatty acids (EFAs, key nutrients for aquatic food webs) in four size categories of planktonic organisms - seston (10-64 microm), micro-(100-200 microm), meso-(200-500 microm), and macrozooplankton (>500 microm) - as well as total mercury (THg) and EFAs in rainbow trout (Oncorhynchus mykiss) in coastal lakes. We demonstrate that, in all lakes during this summer sampling, MeHg concentrations of planktonic or…

New Copper(I) Complex with a Coumarin as Ligand with Antibacterial Activity against Flavobacterium psychrophilum.

A new copper (I) complex, [Cu(NN1)2](ClO4), was synthesized, where NN1 was a imine ligand 6-((quinolin-2-ylmethylene)amino)-2H-chromen-2-one obtained by a derivatization of natural compound coumarin. The structural characterization in solution was done by NMR techniques, UV-Vis and cyclic voltammetry. The potential antibacterial effect of [Cu(NN1)2](ClO4), was assessed for F. psychrophilum isolated 10094. F. psychrophilum is a Gram-negative bacterium which causes diseases such as bacterial cold-water disease and rainbow trout fry syndrome, causing large economic losses in the freshwater salmon…

Selenium bioaccumulation in stocked fish as an indicator of fishery potential in pit lakes on reclaimed coal mines in Alberta, Canada.

Pit lakes are a common reclamation strategy for open pit mines; however, there is a concern about their water quality and suitability as fish habitat because they are often contaminated by metals or metalloids. This study assessed the exposure of fish and invertebrates to selenium (Se) and other metals and metalloids in pit lakes formed by open pit coal mining in Tertiary (thermal coal) and in Cretaceous (metallurgical coal) bedrock. Juvenile hatchery rainbow trout, Oncorhynchus mykiss, and brook trout, Salvelinus fontinalis, were stocked into two thermal coal pit lakes (water Se < 2 μg/L, low…

Metabolic zonation in teleost gastrointestinal tract. Effects of fasting and cortisol in tilapia.

Activities of several metabolic enzymes show distinct patterns of zonation along the intestinal tract of tilapia (Oreochromis niloticus), rainbow trout (Oncorhynchus mykiss) and copper rockfish (Sebastes caurinus). Zonation is species and enzyme specific, with different metabolic activities concentrated in specific areas, and few generalizations can be made. The rockfish show the smallest degree of zonation, with highest activities in the third quarter of the intestine, and shallow gradients to either side, and a general upswing in activity towards the distal end. In the trout, mitochondrial e…

An in vitro binucleated blocked hepatic cell technique for genotoxicity testing in fish.

The main objective of this work was to develop and use a binucleated method which depends on the cytokinesis blocking of hepatic cells in fish to enable the scoring of micronuclei in cells that have completed only one division after genotoxic insult. The use of this novel technique in a study of the influence of some pollutants in vitro (selenium, mercury, methylmercury, and their mixtures) allows evaluation of the risk from exposure to these water pollutants under field and laboratory conditions. The frequencies of micronuclei in trout hepatic cells in vitro were elevated in a dose-dependent …

Heme proteins: quantum yield determined by the pulse method.

We report results of the application of the "pulse method" to the study of photodissociation of various ligands from several heme proteins. By use of this technique, which allows the determination of the quantum yield of photodissociation accurately and rapidly, several ligands (CO, O(2), isocyanides) have been investigated for sperm-whale myoglobin and trout hemoglobin I. In agreement with previous results, the new data lead to the conclusion that no simple relationship exists between the quantum yield and the affinity constant in the ground state. For trout hemoglobin I, the experiments were…

First report of Yersinia ruckeri biotype 2 in the USA.

A polyphasic characterization of atypical isolates of Yersinia ruckeri (causative agent of enteric redmouth disease in trout) obtained from hatchery-reared brown trout Salmo trutta in South Carolina was performed. The Y. ruckeri isolates were biochemically and genetically distinct from reference cultures, including the type strain, but were unequivocally ascribed to the species Y. ruckeri, based on API 20E, VITEK, fatty acid methyl ester profiles, and 16S rRNA gene sequencing analysis. These isolates were nonmotile and unable to hydrolyze Tween 20/80 and were therefore classified as Y. ruckeri…

Wild salmonids and sea louse infestations on the west coast of Scotland: sources of infection and implications for the management of marine salmon farms.

The sea louse (Lepeophtheirus salmonis Krøyer) is a major health problem for both farmed and wild salmonids. This paper investigates louse epidemiology and management in the salmon-farming zone of western Scotland. Based on a review of the marine ecology of wild salmon (Salmo salar L) and sea trout (Salmo trutta L), and catch and farm production statistics, best estimates were made for numbers of wild and farm hosts present in coastal waters in March-June 2000. Applying data for ovigerous female louse infections and fecundity, the sources and risks of larval transmission to wild salmon and sea…

Physiological vulnerability drives species-specific toxicity of 6PPD-Q in salmonids revealed by a damage-coupled biokinetic model.

The tire-wear transformation product 6PPD-quinone (6PPD-Q) is a primary driver of "Urban Runoff Mortality Syndrome" by inducing acute and species-specific mortality in salmonids, yet the underlying toxicokinetic mechanisms remain poorly understood. In this study, we investigated the multi-tissue biokinetics of 6PPD-Q in sensitive rainbow trout (Oncorhynchus mykiss) and relatively tolerant masu salmon (Oncorhynchus masou) using novel damage-coupled biokinetic models. Surprisingly, results revealed a non-linear, accelerating accumulation trajectory across all tissues, with the brain and liver se…

Important variations in parvalbumin content in common fish species: a factor possibly contributing to variable allergenicity.

BACKGROUND: Although 95% of fish-allergic patients are sensitized to the major fish allergen parvalbumin, clinical reactions to different fish species vary considerably in symptoms, intensity and frequency in allergic subjects. This study aimed at the quantification of parvalbumin levels in salmon, trout, cod, carp, mackerel, herring, redfish and tuna. METHODS: Fish muscle extracts were separated by SDS-PAGE and parvalbumin content was estimated by densitometric band quantification. Individual antisera were raised in BALB/c mice against parvalbumins purified from seven fish species. Parvalbum…

Arsenic and Mercury Distribution in an Aquatic Food Chain: Importance of Femtoplankton and Picoplankton Filtration Fractions.

Arsenic (As) and mercury (Hg) were examined in the Yellowstone Lake food chain, focusing on two lake locations separated by approximately 20 km and differing in lake floor hydrothermal vent activity. Sampling spanned from femtoplankton to the main fish species, Yellowstone cutthroat trout and the apex predator lake trout. Mercury bioaccumulated in muscle and liver of both trout species, biomagnifying with age, whereas As decreased in older fish, which indicates differential exposure routes for these metal(loid)s. Mercury and As concentrations were higher in all food chain filter fractions (0.1…

Comparison of two aquatic alphaviruses, salmon pancreas disease virus and sleeping disease virus, by using genome sequence analysis, monoclonal reactivity, and cross-infection.

Cell culture isolates of salmon pancreas disease virus (SPDV) of farmed Atlantic salmon and sleeping disease virus (SDV) of rainbow trout were compared. Excluding the poly(A) tracts, the genomic nucleotide sequences of SPDV and SDV RNAs include 11,919 and 11,900 nucleotides, respectively. Phylogenetic analysis places SPDV and SDV between the New World viruses of Venezuelan equine encephalitis virus and Eastern equine encephalitis virus and the Old World viruses of Aura virus and Sindbis virus. When compared to each other, SPDV and SDV show 91.1% nucleotide sequence identity over their complete…

Stress and food deprivation: linking physiological state to migration success in a teleost fish.

Food deprivation is a naturally occurring stressor that is thought to influence the ultimate life-history strategy of individuals. Little is known about how food deprivation interacts with other stressors to influence migration success. European populations of brown trout (Salmo trutta) exhibit partial migration, whereby a portion of the population smoltifies and migrates to the ocean, and the rest remain in their natal stream. This distinct, natural dichotomy of life-history strategies provides an excellent opportunity to explore the roles of energetic state (as affected by food deprivation) …

Evaluation of an exposure assay to measure uptake of sediment PAH by fish.

The ecological risks of polynuclear aromatic hydrocarbons (PAH) in aquatic sediments will vary with both toxicity and bioavailability to aquatic biota. While there are standardized protocols to test the acute toxicity of sediment-borne compounds to aquatic invertebrates, there are none for assessing bioavailability to fish. We found that sediment-borne PAH caused an exposure-dependent induction of cytochrome P450 (CYP1A) enzymes in rainbow trout (Oncorhynchus mykiss) fingerlings exposed in semi-static 96 h bioassays, as shown by increased activity of ethoxyresorufin-o-deethylase (EROD).
