TDP-43 in familial and sporadic frontotemporal lobar degeneration with ubiquitin inclusions.

TAR DNA-binding protein 43 (TDP-43) is a major pathological protein of sporadic and familial frontotemporal lobar degeneration with ubiquitin-positive, tau-negative inclusions (FTLD-U) with or without motor neuron disease (MND). Thus, TDP-43 defines a novel class of neurodegenerative diseases called TDP-43 proteinopathies. We performed ubiquitin and TDP-43 immunohistochemistry on 193 cases of familial and sporadic FTLD with or without MND. On selected cases, immunoelectron microscopy and biochemistry were performed. Clinically defined frontotemporal dementias (FTDs) included four groups: 1) fa

Ubiquitin-proteasome system alterations in a striatal cell model of Huntington's disease.

Huntington's disease (HD) is a progressive, autosomal dominant neurodegenerative disease caused by an abnormally expanded CAG repeat in the HD gene. Ubiquitylated aggregates containing mutant huntingtin protein in neurons are hallmarks of HD. Misfolded mutant huntingtin monomers, oligomers, or aggregates may be a result of, and cause, ubiquitin- proteasome dysfunction. To investigate the ubiquitin-proteasome system we designed a series of firefly luciferase reporters targeted selectively to different points along this pathway. These reporters were used to monitor ubiquitin-proteasome system fu

Activation of the ubiquitin-proteasome pathway in the diaphragm in chronic obstructive pulmonary disease.

RATIONALE: Studies show that the myosin content of the diaphragm in patients with mild to moderate chronic obstructive pulmonary disease (COPD) is reduced, compromising diaphragm contractile performance. The mechanisms for reduced contractile protein content are unknown. In the present study we hypothesized that the loss of contractile protein content is associated with activation of the ubiquitin-proteasome pathway in the diaphragm of patients with mild to moderate COPD. METHODS: Proteolytic activity of isolated 20S proteasomes was determined in diaphragm biopsies from patients with and with

Role of the ubiquitin-proteasome system in carotid plaque instability in diabetic patients.

In order to define the role of the ubiquitin-proteasome system in atherosclerotic plaque rupture in patients with type 2 diabetes mellitus (T2DM), we evaluated the amount of this system, of the main inflammatory cells, of the collagen content and some indexes indicative of oxidative stress in the carotid plaques of both diabetic and non-diabetic asymptomatic patients. Plaques were obtained from 31 type 2 diabetic and 27 non-diabetic patients undergoing endoterectomy. Both were examined for macrophages, T-lymphocytes, ubiquitin/proteasome 20S activity, NFkB, IkB-b, nitrotyrosine, matrix metallo

Ubiquitin-binding domains in Y-family polymerases regulate translesion synthesis.

Translesion synthesis (TLS) is the major pathway by which mammalian cells replicate across DNA lesions. Upon DNA damage, ubiquitination of proliferating cell nuclear antigen (PCNA) induces bypass of the lesion by directing the replication machinery into the TLS pathway. Yet, how this modification is recognized and interpreted in the cell remains unclear. Here we describe the identification of two ubiquitin (Ub)-binding domains (UBM and UBZ), which are evolutionarily conserved in all Y-family TLS polymerases (pols). These domains are required for binding of poleta and poliota to ubiquitin, thei…

The ubiquitin/proteasome system in Epstein-Barr virus latency and associated malignancies.

Epstein-Barr virus (EBV) is a human herpesvirus associated with lymphoid and epithelial malignancies. Three viral proteins, the EBV nuclear antigen 1 and the latent membrane proteins-1 and -2A, regulate viral latency by manipulating ubiquitin-dependent proteolysis. The activation of cellular oncogenes is instrumental for the progression of EBV infected cells to full malignancy. Constitutively activation of c-myc correlates with decreased proteasomal activity and upregulation of compensatory proteolytic pathways in Burkitt's lymphomas. Knowledge of these multiple strategies of interference with

Disease-specific accumulation of mutant ubiquitin as a marker for proteasomal dysfunction in the brain.

Molecular misreading of the ubiquitin-B (UBB) gene results in a dinucleotide deletion in UBB mRNA. The resulting mutant protein, UBB+1, accumulates in the neuropathological hallmarks of Alzheimer disease. In vitro, UBB+1 inhibits proteasomal proteolysis, although it is also an ubiquitin fusion degradation substrate for the proteasome. Using the ligase chain reaction to detect dinucleotide deletions, we report here that UBB+1 transcripts are present in each neurodegenerative disease studied (tauo- and synucleinopathies) and even in control brain samples. In contrast to UBB+1 transcripts, UBB+1

Ubiquitin-protein ligase WWP2 binds to and downregulates the epithelial Na(+) channel.

The epithelial Na(+) channel (ENaC) is a critical component of the pathway maintaining salt and water balance. The channel is regulated by members of the Nedd4 family of ubiquitin-protein ligases, which bind to channel subunits and catalyze channel internalization and degradation. ENaC mutations that abolish this interaction cause Liddle's syndrome, a genetic form of hypertension. Here, we test the hypothesis that WW domain-containing protein 2 (WWP2), a member of the Nedd4 family of ubiquitin-protein ligases, is a candidate to regulate ENaC. Consistent with this hypothesis, we found that WWP2

Ubiquitin-immunohistochemical investigation of atypical Pick's disease without Pick bodies.

Six cases of atypical Pick's disease (PD) without Pick bodies (PB) were examined immunohistochemically. These cases showed severe neuronal loss with gliosis predominantly in the temporal cortices. Ubiquitin immunohistochemistry revealed ubiquitin-positive intraneuronal inclusions in the dentate gyrus and ubiquitin-positive neurites in the cerebral cortex. In the dentate gyrus, the dendrites in the stratum moleculare as well as the intraneuronal inclusions in the granular cells were positively stained. Both structures were composed of ubiquitin-positive ribosome-like granular components and a f

Heat-shock protein 90 and ubiquitin: developmental regulation during myogenesis.

Heat-shock/stress proteins are constitutive and stress-inducible proteins, regulated by a number of factors including developmental processes. The 90-kD heat-shock protein (hsp90) and ubiquitin are up-regulated in regenerating fibers and diseased fibers of Duchenne muscular dystrophy. The aim of the present study was to investigate whether the heat-shock response in regenerating fibers is developmentally regulated or disease-associated. Immunohistochemistry and immunoblot analysis were employed to compare the expression of hsp90 and ubiquitin in normal immature muscle from infants and regenera

A case of primary progressive aphasia with abnormally ubiquitinated neurites in the cerebral cortex.

We report the histopathological and immunohistochemical findings in a patient with primary progressive aphasia and abnormally ubiquitinated neurites in the cerebral cortex. Neuropathological examination showed severe neuronal loss and astrocytosis with a spongy change in the frontal cortex and neostriatum. Immunohistochemistry for ubiquitin antibody showed many immunoreactive dystrophic neurites in the superficial layer of the affected cortices and putamen. Those neurites were neither argentophilic nor stained with other antibodies against neurofilament, tau, or microtubule-associated protein-

Motor neuron disease/amyotrophic lateral sclerosis--lessons from ubiquitin.

Ubiquitin is a stress protein implicated in the degradation of short-lived and abnormal proteins. In a neuropathological study of 43 cases with motor neuron disease/amyotrophic lateral sclerosis (MND/ALS) and 44 control cases the distribution and specificity of Bunina bodies and ubiquitin-reactive inclusions (UBRI) were investigated. The primary motor area showed nerve cell loss in 67%, Bunina bodies in Betz cells (10%) and UBRI in small pyramidal cells (17%). Degeneration of anterior horn cells in all cases coincided with Bunina bodies (84%) and UBRI (98%) in the same location; the motor nucl

E3 ubiquitin ligase Pellino1 suppresses acinar cell necroptosis and alleviates severe acute pancreatitis by promoting ubiquitin-dependent receptor-interacting protein kinase 3 (RIP3) degradation.

BACKGROUND AND PURPOSE: Severe acute pancreatitis (SAP) is a critical abdominal condition with high mortality rates. The activation of necroptosis in acinar cells is an critical mechanism for SAP. Pellino1 (PELI1), as an E3 ubiquitin ligase, is involved in pathogenic mechanisms in various diseases. Its ubiquitination function also can regulate necroptosis. However, in SAP, there is a lack of research on upstream regulatory mechanisms and small molecule-targeted drug therapy of necroptosis. Therefore, it is of great significance to study the specific mechanism of PELI1 regulating necroptosis in…

CHIP overexpression in cardiomyocytes obviously inhibited LPS-initiated release of TNF-α and IL-6 by promoting KPNA2 degradation, reducing NF-κB translocation into the nucleus. Consistent with the in vitro results, data obtained f …

MDA-7/IL-24, a novel tumor suppressor/cytokine is ubiquitinated and regulated by the ubiquitin-proteasome system, and inhibition of MDA-7/IL-24 degradation enhances the antitumor activity.

Steady-state protein levels are determined by the balance between protein synthesis and degradation. Protein half-lives are determined primarily by degradation, and the major degradation pathways involve either lysosomal destruction or an ATP-dependent process involving ubiquitination to target proteins to the proteosome. Studies have shown that multiple tumor-suppressor proteins are ubiquitinated and degraded by the 26S proteasome. In the present study, we investigated whether the tumor suppressor/cytokine melanoma differentiation-associated gene-7/interleukin-24 gene (MDA-7/IL-24) protein is…

RNF207 promotes colorectal cancer growth by regulating the Hippo-YAP pathway via enhanced MST1 ubiquitination and degradation.

BACKGROUND: Colorectal cancer ranks second in global mortality rates, yet its molecular pathogenesis remains incompletely understood. Accumulating evidence indicates that E3 ubiquitin ligases modulate the activity and stability of critical oncoproteins via ubiquitination, positioning these enzymes as promising therapeutic targets for cancer intervention. The E3 ligase Ring finger protein 207 (RNF207), characterized as a regulator involved in heart disease, has not been investigated in colorectal cancer. METHODS: We constructed RNF207 overexpression and knockdown colorectal cancer cell lines, …

MaMPK3-2 Phosphorylating and Stabilizing Transcriptional Repressor MaAS1 by Counteracting MaRNF4-Mediated Ubiquitination During Banana Fruit Ripening.

The ripening of fleshy fruits is strongly regulated by a complex interplay between transcriptional regulation and post-translational modific … ppressing fruit ripening. In contrast, the ethylene-induced E3 ubiquitin ligase MaRNF4 mediates the ubiquitination and degradation of MaAS1, thereby promoting ripening. Notably, phosphorylation by MaMPK3-2 antagonises MaRNF4-mediated ubiquitination, maintaining MaAS1 levels and regulating ripening progression. Collectively, these findings reveal a regulatory mod …

Computational design and experimental validation of peptide inhibitors targeting the p53-COP1 interaction to suppress p53 ubiquitination.

Breast cancer, as one of the most prevalent malignant tumors among women, is a leading cause of female mortality. Inhibiting the ubiquitination of p53 protein would be helpful to control the development and progression of breast cancer, as p53 is crucial for regulating cell proliferation, DNA damage repair, and apoptosis. Herein, peptide inhibitors were designed to inhibit the interaction between p53 protein and ubiquitin ligase COP1. Molecular dynamics (MD) simulations with combined Molecular Mechanics/Poisson-Boltzmann Surface Area (MM-PBSA) calculations were used to explore the molecular in…
