Characterization of a tumor-associated gene, a member of a novel family of genes encoding membrane glycoproteins.

To isolate genes involved in tumor formation and in embryogenesis, a subtracted cDNA library was constructed from a c-myc-induced mouse brain tumor. A gene isolated in this screen, named TMP (tumor-associated membrane protein), codes for a putative glycoprotein with four transmembrane domains. The TMP gene was found to be highly expressed in brain tumor cells but not in normal brain. It is also expressed at high levels in undifferentiated embryonic stem cells, but markedly down-regulated in these cells after their differentiation into embryoid bodies. The TMP amino acid sequence bears high hom

Matrix permissiveness regulates 3D confinement of fibroblasts in biosynthetic hydrogel microenvironments.

The extracellular matrix (ECM) is a major regulator of cellular behavior, fate, and various mechanisms underlying homeostasis, development, and disease. Biophysical and biochemical properties of the ECM are known to affect three-dimensional (3D) cellular behavior and phenotype that regulate a wide range of pathological conditions. Tunable biomimetic hydrogels are extensively employed to investigate cell-matrix interactions in defined 3D microenvironments. However, the dynamics of these interactions in complex coupled multiparametric microenvironments has been relatively less studied. This stud

Orthogonal alignment of multilayered MC3T3-E1 cells induced by cyclic stretch.

When cyclic stretch is applied to a monolayer of cells cultured on an elastic substrate, many types of cells align in the direction perpendicular to the stretch or along the direction of minimal substrate strain. However, the behavior of multilayer cells under cyclic stretch remains unclear. In this study, we cultured MC3T3-E1 osteoblast-like cells at high density to form multilayer cells and subjected them to cyclic stretch with an amplitude of 10% at 1 Hz. We found that the lower layer cells aligned in the direction of the stretch after 12 h, whereas the upper layer cells aligned perpendicul

Melt Electrowriting of Elastic Scaffolds Using PEOT-PBT Multi-block Copolymer.

Melt electrowriting (MEW) is a powerful additive manufacturing technique to produce tissue engineering scaffolds. Despite its strength, it is limited by a small number of processable polymers. Therefore, to broaden the library of materials for MEW, we investigated the printability of poly(ethylene oxide terephthalate)-poly(butylene terephthalate) (PEOT-PBT), a thermoplastic elastomer. The effect of different printing parameters and material thermal degradation are studied. It is observed that the material is stable for >60 min at a printing temperature of 195 °C in a nitrogen environment. Next

Design, synthesis and biological evaluation of cajanonic acid A analogues as potent PPAR γ antagonists.

Peroxisome proliferator-activated receptor γ (PPAR γ) antagonists are a key instrument of insulin sensitizers since they have the ability to sensitize insulin and can avoid adverse reactions caused by receptor agonist. In this paper, two series of 28 novel Cajanonic acid A (CAA) derivatives were designed and synthesized. The biological activity showed that a novel CAA derivative 9f was identified as a potential PPAR γ antagonist by medicinal chemistry efforts. The results in vitro displayed that compound 9f could improve the PPAR γ antagonist activity (96.2 % / 50.2 % decrease in PPAR γ transa

Differential effects of polyunsaturated fatty acids on chemosensitivity of NIH3T3 cells and its transformants.

Polyunsaturated fatty acids (PUFAs) have been suggested, on the basis of animal-model studies, to be related not only to cancer development … ts revealed that the sensitivity of SIC transformants to mitomycin C (MC) was lower than that of NIH3T3 cells cultured in 10% calf-serum DMEM without PUFA supplementation. When cells were cultured in DMEM supplemented with eicosapentaenoic acid (EPA) at a concentration (2 micrograms/ml) that does not influence cell proliferation, the sensitivity of SIC tran …

Autophagosomes accumulate in differentiated and hypertrophic adipocytes in a p53-independent manner.

Autophagy is induced by several kinds of stress, including oxidative, genotoxic, endoplasmic reticulum and nutrient stresses. The tumor suppressor p53, which is a stress sensor, plays a critical role in the regulation of autophagy. Although p53 is required for starvation (nutrient deficient stress)-induced autophagy, it is still not clear whether p53 is also required for the autophagy observed in differentiated and hypertrophic adipocytes, which accumulate excessive amounts of nutrients in the form of triglycerides. In this study, we demonstrated that starvation induces autophagy in p53-profic

Recent advances in cultivated epithelial transplantation.

Currently, cultivated epithelial transplantation usually uses ex vivo-expanded epithelial cells, with or without biological carriers. Our group has carried out approximately 100 such procedures and has conducted research in this area. The results of a retrospective study indicate that cultivated limbal epithelial transplantation with amniotic membrane (AM) and airlifting may be beneficial at avoiding sight-threatening complications in patients with severe chronic cicatricial keratoconjunctivitis. Our experience in cultivated oral mucosal epithelial transplantation indicates that this technique

Anti-tumor therapy of glycyrrhetinic acid targeted liposome co-delivery of doxorubicin and berberine for hepatocellular carcinoma.

During the development of hepatocellular carcinoma (HCC), hepatic stellate cells undergo activation and transform into cancer-associated fibroblasts (CAFs) due to the influence of tumor cells. The interaction between CAFs and tumor cells can compromise the effectiveness of chemotherapy drugs and promote tumor proliferation, invasion, and metastasis. This study explores the potential of glycyrrhetinic acid (GA)-modified liposomes (lip-GA) as a strategy for co-delivery of berberine (Ber) and doxorubicin (Dox) to treat HCC. The characterizations of liposomes, including particle size, zeta potenti

Substrates with engineered step changes in rigidity induce traction force polarity and durotaxis.

Rigidity sensing plays a fundamental role in multiple cell functions ranging from migration, to proliferation and differentiation1-5. During migration, single cells have been reported to preferentially move toward more rigid regions of a substrate in a process termed durotaxis. Durotaxis could contribute to cell migration in wound healing and gastrulation, where local gradients in tissue rigidity have been described. Despite the potential importance of this phenomenon to physiology and disease, it remains unclear how rigidity guides these behaviors and the underlying cellular and molecular mec

The arginine metabolite agmatine protects mitochondrial function and confers resistance to cellular apoptosis.

Agmatine, an endogenous metabolite of arginine, selectively suppresses growth in cells with high proliferative kinetics, such as transformed cells, through depletion of intracellular polyamine levels. In the present study, we depleted intracellular polyamine content with agmatine to determine if attrition by cell death contributes to the growth-suppressive effects. We did not observe an increase in necrosis, DNA fragmentation, or chromatin condensation in Ha-Ras-transformed NIH-3T3 cells administered agmatine. In response to Ca(2+)-induced oxidative stress in kidney mitochondrial preparations,…

Preparation of Extracellular Matrices and Cell/Matrix Functional Units Produced by Cultured and Primary Fibroblasts.

Fibroblast-derived matrices (FDMs) provide a biologically relevant in vitro alternative to conventional two-dimensional (2D) substrates by enabling fibroblasts to assemble and organize their own extracellular matrix (ECM) environment, closely mimicking the physiological stromal microenvironment. This unit describes methods for generating interstitial matrices in which fibroblasts assemble their own multilayered extracellular scaffold, termed a fibroblast/ECM functional unit, that yields a cell-free FDM anchored to the culture surface, following decellularization. The resulting FDM is a polymer

Effects of topographical guidance cues on osteoblast cell migration.

Cell migration is a fundamental process that is crucial for many biological functions in the body such as immune responses and tissue regeneration. Dysregulation of this process is associated with cancer metastasis. In this study, polydimethylsiloxane platforms with various topographical features were engineered to explore the influence of guiding patterns on MC3T3-E1 osteoblast cell migration. Focusing on the guiding effects of grating patterns, variations such as etch depth, pattern discontinuity, and bending angles were investigated. In all experiments, MC3T3-E1 cells on patterned surfaces

New cationic nanovesicular systems containing lysine-based surfactants for topical administration: Toxicity assessment using representative skin cell lines.

Cationic nanovesicles have attracted considerable interest as effective carriers to improve the delivery of biologically active molecules into and through the skin. In this study, lipid-based nanovesicles containing three different cationic lysine-based surfactants were designed for topical administration. We used representative skin cell lines and in vitro assays to assess whether the cationic compounds modulate the toxic responses of these nanocarriers. The nanovesicles were characterized in both water and cell culture medium. In general, significant agglomeration occurred after 24h incubati

Speciation, Protein Binding, Biotransformation, and Cytotoxicity of a VV-Lactate Complex.

We studied the speciation, protein binding, biotransformation, and cytotoxicity of the dioxidovanadium(V) lactate complex Cs2[VV2O4(lact)2]·2H2O and compared the results with those obtained for the analogous malate compound. 51V NMR and ESI-MS results show that Cs2[VV2O4(lact)2]·2H2O forms [VVO2]+, [H2VVO4]-, [H2VV2O7]2-, [VV2O4(lact)2]2-, [VV3O7(lact)2]3-, [VV4O12]4-, [VV5O15]5-, [VVO2(lact)(H2O)]-, [VVO2(lact)(OH)]2-, and [VV10O28]6- species in aqueous solution. In the presence of lysozyme, the amounts of [VV10O28]6- and [VVO2(lact)(H2O)]- significantly decrease and protein adducts with [VV2

Identification of a novel MIPEP splice variant with altered substrate-binding properties.

Mitochondrial intermediate peptidase (MIPEP) is a mitochondrial signal peptidase that removes N-terminal amino acids from mitochondrial matrix proteins. We have identified a novel Mipep splice variant that lacks exons 15 and 16, which we termed "ΔMIPEP". We characterized the molecular features of ΔMIPEP by investigating its expression level in numerous mouse tissues and by performing a computer simulation that allows the prediction of protein structures and substrate-binding properties. ΔMipep mRNA was detected in all mouse tissues examined but at much lower levels than full-length Mipep. Stru
