American Association of Veterinary Parasitologists Hookworm Task Force review on best practices for diagnosis and treatment of multidrug-resistant hookworms.

Ancylostoma caninum, the canine hookworm, is the most common and clinically important gastrointestinal nematode parasite infecting dogs in the US. Common clinical signs associated with acute infections include anemia, melena, hematochezia, dehydration, and poor weight gain or weight loss. In the past few years, multiple anthelmintic drug resistance to fenbendazole, moxidectin, and pyrantel pamoate has been documented in A caninum in the US, and cases of drug-resistant canine hookworms are increasingly being reported in the US and Canada. Clinical and genetic evidence strongly suggests that mul

Retrospective analysis of canine fecal flotation and coproantigen immunoassay hookworm positive results in Greyhounds and other dog breeds.

Recent studies demonstrated that Greyhounds are commonly infected with Ancylostoma caninum and these infections have been shown to be resistant to anthelmintics. This study evaluated samples submitted to a commercial reference laboratory (IDEXX Laboratories) for canine fecal flotation zinc sulfate centrifugation and coproantigen immunoassay between January 1, 2019, and July 30, 2023 for evidence that Greyhounds were more often positive for Ancylostoma spp. (hookworms) compared to other breeds. The purpose of the study was to determine if Greyhounds were more likely to be hookworm-positive comp

Of dogs and hookworms: man's best friend and his parasites as a model for translational biomedical research.

We present evidence that the dog hookworm (Ancylostoma caninum) is underutilised in the study of host-parasite interactions, particularly as a proxy for the human-hookworm relationship. The inability to passage hookworms through all life stages in vitro means that adult stage hookworms have to be harvested from the gut of their definitive hosts for ex vivo research. This makes study of the human-hookworm interface difficult for technical and ethical reasons. The historical association of humans, dogs and hookworms presents a unique triad of positive evolutionary pressure to drive the A. caninu

Prevalence of zoonotic helminths in Italian house dogs.

INTRODUCTION: Dogs may act as potential sources of zoonotic parasites, e.g. intestinal helminths like Toxocara spp., Ancylostoma spp., Echinococcus spp. In particular circumstances, the environment contaminated by parasitic elements represents a source of infection for people and animals. The present study has evaluated the presence of zoonotic helminths in house dogs from central and north-eastern Italy. METHODOLOGY: Stool samples from 493 dogs were examined by a qualitative copromicroscopic technique and differences in prevalence of zoonotic parasites were statistically examined in relation

Prevalence of Ancylostoma braziliense in cats in three northern counties of Florida, United States.

A convenience sampling of fecal specimens from 40 cats in northern Florida was examined for the presence of Ancylostoma braziliense eggs by using centrifugal sugar flotation and polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP). Of the 40 samples, 26 (65%) contained hookworm eggs. DNA from 24 samples was successfully amplified using PCR; using RFLP, 10 samples were identified as containing DNA of A. braziliense (41.7% of the 24 samples that successfully amplified). Of these, 6 samples contained DNA of both Ancylostoma tubaeforme and A. braziliense, and 4 samples c

The biological control of Ancylostoma spp. dog infective larvae by Duddingtonia flagrans in a soil microcosm.

Experiments to evaluate the potential ability of the nematode-trapping fungus Duddingtonia flagrans (Isolate CG768) to prey on the Ancylostoma spp. dog infective larvae (L(3)) in pasteurized soil were performed through several laboratory assays. A microcosm approach was used with increasing fungal concentrations in an inoculum of a chlamydospore water suspension. The highest fungal concentrations provide a more consistent larval reduction than the lowest concentrations, but no difference was observed from 10,000 to 25,000 chlamydospores per grain of soil. When using D. flagrans in a water susp

Identification of specific male and female genes in adult Ancylostoma caninum.

The hookworm Ancylostoma canium represents a serious health problem, not only for animals but also for humans. These blood-feeding parasites produce various proteolytic enzymes in order to digest the host hemoglobin. The female worm ingests more blood than does the male. It is not known whether this difference is accompanied by expression of sex-specific proteinases. The identification of new genes related either to the developmental process of maturation of each sex or to the proteinases secreted by these worms could provide researchers with new tools to be used in control programs for this i

Immunochemical characterization of Ancylostoma caninum antigens.

Adult worms of Ancylostoma caninum were dissected and manually separated into cephalic glands, cervical glands, intestine, esophagus and cuticula. These fractions as well as third stage larvae were fractionated with Triton X-114 into water soluble (hydrophilic), Triton soluble (hydrophobic) and unsoluble proteins. These fractions were characterized by immunoblotting with serum from rabbits immunized either with a pool of cervical, cephalic glands and intestine, or the esophagus fraction as well as with sera from percutaneously infected dogs and rabbits. Immunodominant antigens were found that

Resistance of the hookworms Ancylostoma ceylanicum and Necator americanus to intestinal inflammatory responses induced by heterologous infection.

Experiments were carried out to ascertain whether the acute inflammatory phase of the intestinal response of hamsters to infection with Trichinella spiralis would adversely affect hookworms in concurrently infected animals. The survival and growth of hookworms were unaffected. However, the presence of hookworms reduced the establishment of T. spiralis, the initial growth of female worms and their fecundity. The expulsion of T. spiralis was also significantly slower in concurrently infected animals and there was significant depression of the serum IgG antibody response to muscle stage and adult

Hepatic microsomal cytochrome P450 system during experimental hookworm infection.

Experimental infection of golden hamsters with the hookworm, Ancylostoma ceylanicum, caused a profound decline in the hepatic microsomal cytochrome P450 content. Concomitant decrease was also noticed in aminopyrine N-demethylase and benzo[a]pyrene hydroxylase activities. However, aniline hydroxylase activity was only marginally elevated during the infection. Microsomal markers, viz., cytochrome b5, NADH-cytochrome-c reductase, and glucose-6-phosphatase, were not significantly altered. Hepatic tissue exhibited an accumulation of lipids, especially phospholipids, triglycerides, and cholesterol,

Anthelmintic efficacy of febantel combined with praziquantel against Ancylostoma tubaeforme, Toxocara cati, and Taenia taeniaeformis in cats.

Forty cats, each harboring 2 or 3 parasitic infections (Ancylostoma tubaeforme, Toxocara cati, and/or Taenia taeniaeformis), were used to titrate the anthelmintic efficacy of a paste containing 3.4% febantel and 0.34% praziquantel. The cats were allotted into 4 groups (10 cats/group). For 3 consecutive days, the cats were given febantel/praziquantel at 5/0.5 mg/kg/day, 10/1 mg/kg/day, 15/1.5 mg/kg/day, or a blank paste vehicle (control) at 0.29 g/kg of body weight. The recommended dosage of 10 mg of febantel and 1 mg of praziquantel/kg cleared greater than or equal to 98% of the 3 helminth spe

[Variation of helminthiasis incidence in Reunion as a function of different parameters].

Four years after Picot's experiments on parasitic diseases in Réunion Island, and after a large sanitation campaign, the authors valued the effect of this campaign on parasitic incidence. They showed that it had significantly decreased. At the same time, they pointed out the leading part of water quality in parasitic incidence; particularly, they showed that there are twice less Ascaridiasis when water is bacteriologically and chemically safe.
