Relationships between biological age, distance from aquatic habitats and pyrethroid resistance status of Anopheles funestus mosquitoes in south-eastern Tanzania.

BACKGROUND: Malaria transmission can be highly heterogeneous between and within localities, and is influenced by factors such as survival and biting frequencies of Anopheles mosquitoes. This study investigated the relationships between the biological age, distance from aquatic habitats and pyrethroid resistance status of Anopheles funestus mosquitoes, which currently dominate malaria transmission in south-east Tanzania. The study also examined how such relationships may influence malaria transmission and control. METHODS: Female An. funestus were collected in houses located 50-100 m, 150-200 …

Plasmodium falciparum LipB mutants display altered redox and carbon metabolism in asexual stages and cannot complete sporogony in Anopheles mosquitoes.

Malaria is still one of the most important global infectious diseases. Emergence of drug resistance and a shortage of new efficient antimalarials continue to hamper a malaria eradication agenda. Malaria parasites are highly sensitive to changes in the redox environment. Understanding the mechanisms regulating parasite redox could contribute to the design of new drugs. Malaria parasites have a complex network of redox regulatory systems housed in their cytosol, in their mitochondrion and in their plastid (apicoplast). While the roles of enzymes of the thioredoxin and glutathione pathways in par

Bendiocarb resistance in Anopheles gambiae s.l. populations from Atacora department in Benin, West Africa: a threat for malaria vector control.

BACKGROUND: Owing to pyrethroid resistance in An. gambiae, the carbamate and organophosphate insecticides are currently regarded as alternatives or supplements to pyrethroids for use on mosquito net treatments. Resistance monitoring is therefore essential to investigate the susceptibility of An. gambiae s.l to these alternative products. METHODS: Two to three day old adult female Anopheles mosquitoes were reared from larvae collected in the five districts (Kouandé, Natitingou, Matéri, Péhunco, Tanguiéta) of the Atacora department. Mosquitoes were then exposed to WHO impregnated papers. The fo…

First report on co-occurrence knockdown resistance mutations and susceptibility to beta-cypermethrin in Anopheles sinensis from Jiangsu Province, China.

The increasing prevalence of insecticide resistance in Anopheles sinensis, a major vector of malaria in Jiangsu province in eastern China, threatens to compromise the successful use of insecticides in malaria control strategies. It is therefore vital to understand the insecticide resistance status of An. sinensis in the region. This study examined the nucleotide diversity of the para-sodium channel and knockdown resistance (kdr) in five field populations of adult An. sinensis mosquitoes collected in Jiangsu province, identifying the L1014F and L1014C substitutions for the first time. Studies of Anopheles gambiae s.l (Diptera: Culicidae) exhibiting different vectorial capacities in lymphatic filariasis transmission in the Gomoa district, Ghana.

BACKGROUND: Two lymphatic filariasis endemic communities Mampong and Hwida in Ghana have been regularly monitored for impact on transmission after annual mass drug administration (MDA) with albendazole and ivermectin. After six MDAs even though the ABR for Mampong was 55883/person/year and that of Hwida was 2494/person/year, they both had ATPs of 15.21 infective larvae/person/year. Interestingly the human microfilaraemia levels had reduced significantly from 14% to 0% at Mampong and 12% to 3% at Hwida. In an attempt to understand this anomaly, we collected mosquitoes over a 5-month period usin

Resistance of early midgut stages of natural Plasmodium falciparum parasites to high temperatures in experimentally infected Anopheles gambiae (Diptera: Culicidae).

We studied the effects of high temperature, 30 and 32 versus 27 C on early Plasmodium falciparum development in Anopheles gambiae experimentally infected with gametocytes from 30 volunteers with mean density of 264.1 gametocytes/microl blood (range: 16-1,536/microl). From several batches of mosquitoes, fed by membrane feeding, midguts of individual mosquitoes were dissected at 24 hr for ookinete enumeration and at 7 days to quantify oocysts. There were temperature-related differences in mean ookinete intensity per mosquito midgut, with 9.71 +/- 1.6 at 27 C, 9.85 +/- 2.32 at 30 C, and 3.89 +/- …

Isolates of Bacillus thuringiensis Berliner with toxicity to Anopheles darlingi Root, 1926 (Diptera: Culicidae) larvae, the main malaria vector in Brazil.

Bacillus thuringiensis Berliner 1911 has high effectiveness against important mosquito vectors of human pathogens; however, there are few studies on the selection of Brazilian isolates of B. thuringiensis with activity against Anopheles darlingi Root, 1926, the main transmitter of human malaria parasites in the Amazon region. The present study sought to select native isolates of B. thuringiensis acquired from soil samples from three Brazilian biomes, with pathogenicity to An. darlingi larvae under laboratory conditions. Fifteen isolates of B. thuringiensis from the Maranhão Entomopathogenic Ba…

Efficacy of injectable versus topical formulation of ivermectin against Anopheles stephensi mosquitoes feeding on different body locations of treated Holstein calves.

BACKGROUND: Malaria is a major public health concern and is transmitted to humans by the bite of infected Anopheles mosquitoes. One strategy to reduce populations of zoophagic Anopheles (i.e., likely to feed on other animals as well as humans) is the use of systemic veterinary parasiticides. The most widely systemic parasiticide used for this purpose is ivermectin. Ivermectin is available for livestock in two formulations; injectable and topical "pour-on." The purpose of this study was to evaluate the survival and fecundity of a zoophagic species, Anopheles stephensi, when fed on calves treate…

Cymbopogon citratus and Lippia multiflora essential oils as alternative repellent to DEET against Anopheles gambiae s.l. in Bobo-Dioulasso, Burkina Faso.

Personal protective tools, such as repellents, play a vital role in preventing mosquito-borne disease transmission. Although N,N-diethyl-meta-toluamide (DEET), a synthetic repellent, is effective against insects, it may cause allergic reactions. Therefore, there is a need to explore effective alternative repellents exhibiting fewer side effects. Here, we assessed the repellent activity of essential oils (EOs) from Lippia multiflora and Cymbopogon citratus alone and in combination, against Anopheles populations, using the Human Landing Catch (HLC) method in Dogona, Burkina Faso. For this purpos…

First detection and entomological characterisation of invasive malaria vector Anopheles stephensi in sympatry with other vectors in Gayi Southern Niger Republic.

Anopheles stephensi, an invasive vector is aggressively expanding its geographic range across Africa, posing significant threat to malaria control. Surveillance of its spread is crucial for mitigating its impact on malaria transmission. Here, we report for the first time incursion of An. stephensi into Gayi, a rural area in southern Niger Republic. A combination of morphological identification, end point PCR and sequencing of fragments of COXI and ITS2 genes confirmed An. stephensi. This species was found resting indoor together with other mosquitoes, including An. coluzzii, An. gambiae s.s., …

Insecticide resistance and its intensity in urban Anopheles arabiensis in Kisumu City, Western Kenya: Implications for malaria control in urban areas.

BACKGROUND: The rise of insecticide resistance poses a growing challenge to the effectiveness of vector control tools, particularly in rural areas. However, the urban setting has received comparatively less focus despite its significance in attracting rural to urban migration. Unplanned urbanization, often overlooked, exacerbates insecticide resistance as Anopheles mosquitoes adapt to the polluted environments of rapidly expanding cities. This study aimed to assess the insecticide susceptibility status of malaria vectors and identify potential underlying mechanisms across three distinct ecolog…

Anopheles mosquito exposure is associated with age, gender and bed net use in areas in Uganda experiencing varying malaria transmission intensity.

OBJECTIVES: The number of Anopheles mosquito bites a person receives determines the risk of acquiring malaria and the likelihood of transmitting infections to mosquitoes. We assessed heterogeneity in Anopheles biting and associated factors in two settings in Uganda with different endemicity. METHODS: Plasmodium falciparum parasites in blood-fed indoor caught Anopheles mosquitoes were quantified using qPCR targeting the Pf18S rRNA gene. Human DNA in dried blood spots from household occupants and mosquito blood meals was profiled using 15 short-tandem repeats (STRs) and analysed using a log-lik…

The effect of novel mosquito bite prevention tools on Anopheles minimus landing and key secondary endpoints: semi-field evaluations in Thailand.

BACKGROUND: The Greater Mekong Subregion (GMS) aims to eliminate all human malaria by 2030 and is making substantial progress toward this go … smission overall. These population groups are exposed to malaria vector bites during the day due to Anopheles daytime biting, and during the night, due to low bed net use and open sleeping structures. Such characteristics limit the effectiveness of the WHO core vector control strategies [indoor residual spraying (IRS), insecticide-treated nets (ITNs)], whic …

Insecticidal roof barriers mounted on untreated bed nets can be as effective against Anopheles gambiae as regular insecticide-treated bed nets.

Barrier bednets (BBnets), regular bednets with a vertical insecticidal panel to target mosquitoes above the bednet roof, where they are most active, have the potential to improve existing Insecticidal Treated Bednets (ITNs), by reducing the quantity of insecticide required per net, reducing the toxic risks to those using the net, potentially increasing insecticide choice. We evaluated the performance of PermaNet 3.0 (P3) and untreated (Ut) bed nets with and without pyrethroid and piperonyl butoxide roof barriers in killing pyrethroid-resistant and susceptible Anopheles gambiae, simultaneously …

Larvicidal property and active compound profiling of Annona squamosa leaf extracts against two species of diptera, Aedes aegypti and Anopheles stephensi.

BACKGROUND & OBJECTIVES: Vector control measures are important in lowering the spread of infections spread by mosquito. Synthetic pesticides used to suppress vector populations during the larval stage have had adverse impacts on people and the environment. The early III instar larvae of Aedes aegypti and Anopheles stephensi were the targets of the current experiment, which assessed the larvicidal ability of petroleum ether, chloroform, methanol, and aqueous extracts of Annona squamosa leaves. METHODS: Using the standard World Health Organization (WHO) larval bioassay test, leaf extracts were …

Evaluation of the interaction between insecticide resistance-associated genes and malaria transmission in Anopheles gambiae sensu lato in central Côte d'Ivoire.

BACKGROUND: There is evidence that the knockdown resistance gene (Kdr) L1014F and acetylcholinesterase-1 gene (Ace-1R) G119S mutations involved in pyrethroid and carbamate resistance in Anopheles gambiae influence malaria transmission in sub-Saharan Africa. This is likely due to changes in the behaviour, life history and vector competence and capacity of An. gambiae. In the present study, performed as part of a two-arm cluster randomized controlled trial evaluating the impact of household screening plus a novel insecticide delivery system (In2Care Eave Tubes), we investigated the distribution …
