Neuroendocrine and stress-related aspects of endometriosis.

Endometriosis is a model of a benign gynecologic disease associated with two major symptoms: pain and infertility. When becomes chronic, severe psychological and neuroendocrine changes may occur. The high levels of perceived stress caused by symptoms cause a neuroendocrine disequilibrium thus contributing to the progression of the disease. Elevated stress levels alter hormonal secretions, mood and behavior, sexual disorders and appetite custom. Inflammatory comorbidities may be associated with elevated stress in endometriotic patients (inflammatory bowel disease, fibromyalgia, chronic fatigue)

Evidence-based approaches to other symptoms in advanced cancer.

Dyspnea, nausea and vomiting, anorexia, fatigue, and sleep disturbances are common and distressing in advanced cancer. We updated previous systematic reviews of how these symptoms can be alleviated with targeted literature searches. The approach to these symptoms requires comprehensive symptom assessment; treating underlying causes when benefits exceed risks; prioritizing treatment, as patients usually have many symptoms; and addressing psychosocial and spiritual distress. For dyspnea, evidence supports systemic opioids and nonpharmacological treatments such as a fan. The strongest evidence su

Hemithyroidectomy in a horse with confirmed hyperthyroidism.

A 23-year-old Quarter Horse gelding was admitted to the hospital because of cachexia and hyperactive behavior of 1 year's duration. At admission the horse was severely emaciated, tachycardic with a grade V/VI diastolic murmur, pyrexic, polydipsic, enophthalmic, and alopecic. The right lobe of the thyroid gland was noticeably larger than typical. The horse was also hyperexcitable and had a ravenous appetite. A presumptive diagnosis of hyperthyroidism was made on the basis of clinical signs and high plasma thyroid hormone concentrations. Confirmation of the diagnosis was made on the basis of res

Overview of adrenergic anorectic agents.

Adrenergic anorexic agents of the amphetamine class suppress appetite and reduce body weight via activation of beta-adrenergic and/or dopaminergic receptors within the perifornical hypothalamus (PFH). Although phenylpropanolamine (PPA) is often considered to be a member of the amphetamine class of anorexiants, this drug is an atypical adrenergic anorexiant. Unlike amphetamine, microinjection of PPA into the PFH does not suppress feeding. Moreover, PPA anorexia is not reversed by the dopamine antagonist haloperidol. The anorexic action of PPA may result, in part, from its interaction with alpha

Trajectories of appetitive traits and dietary patterns from childhood to adolescence: findings from the Generation XXI birth cohort.

PURPOSE: This study examined the association between appetitive trait profiles and dietary patterns from childhood to early adolescence. METHODS: We included 5040 participants from the Generation XXI cohort (Porto, Portugal). Appetitive traits were assessed at ages 7, 10, and 13 with the validated Children’s Eating Behaviour Questionnaire. Six trajectory-based profiles were previously identified: “Smallest appetite”, “Small appetite but increasing”, “Small to moderate appetite”, “Moderate appetite”, “Increasing appetite”, and “Avid appetite”. A validated Food Frequency Questionnaire collected

Ovulatory cycle shifts in human motivational prioritisation of sex and food.

Previous research on the endogenous effects of ovarian hormones on motivational states in women has focused on sexual motivation. The Motivational Priority Shifts Hypothesis has a broader scope. It predicts a shift from somatic to reproductive motivation when fertile. In a highly powered preregistered online diary study across 40 days, we tested whether 390 women report such an ovulatory shift in sexual and eating motivation and behaviour. We compared 209 naturally cycling women to 181 women taking hormonal contraceptives (HC) to rule out non-ovulatory changes across the cycle as confounders.

Berberine for Appetite Suppressant and Prevention of Obesity.

Berberine (BBR), a natural plant product, has been shown to have antidiabetic, cholesterol-reducing effects. To investigate the action of BBR as appetite suppressants, two experimental protocols were performed. In the first experiment, the mice were fed either a normal-chow diet or a high-fat diet (HF). The mice received daily intraperitoneal injections of BBR (10 mg/kg or saline at 1 ml/kg) for 3 weeks. To determine the antiobesity effects of BBR, the food consumption, body weight, fat contents, serum leptin, and glucose level were investigated. In the second experiment, we set out to validat

Effects of ghrelin in energy balance and body weight homeostasis.

Ghrelin is a gut peptide composed of 28 amino acids mostly secreted in the gastric fundus mucosa. It was isolated and described in 1999 by Kojima et al. and only three years later its specific receptor, GHSR1a, was also identified. Ghrelin, the endogenous ligand for the GH secretagogue receptor, is the only peripheral orexigenic hormone that activates the receptors to be found especially in the appetite center (hypothalamus and pituitary gland). Ghrelin is present in human plasma in two forms: an inactive form known as deacylated ghrelin, and an active form called acylated ghrelin synthesized

[Conservative obesity treatment - when and how?].

With a prevalence of 24 %, obesity is a frequent disease in the general population in Germany. Obesity requires an effective prevention and treatment, mainly because it significantly increases the risk of developing type 2 diabetes, cardiovascular, orthopaedic, psychologic and other disorders. The indication for the treatment of obesity is based on BMI and body fat distribution, but also includes considerations based on comorbidities. Main treatment goals include body weight reduction and weight maintenance after weight loss. To achieve these goals, a conservative treatment strategy is primari

Targeting cocaine versus heroin memories: divergent roles within ventromedial prefrontal cortex.

In the search for novel treatments for addiction, most research has been propelled by the hope for a 'magic bullet' that would cure all forms of addiction. More recently, the field has started to appreciate the differences between psychostimulants versus opiates. Recent data suggest that the ventromedial prefrontal cortex (vmPFC) may fundamentally serve different roles in cocaine versus heroin addiction: acting as a neural OFF switch for cocaine seeking, but an ON switch for heroin seeking. We discuss the relevance of this distinction in relationship to three main functions of the vmPFC: (i) e

The role of gut hormones and the hypothalamus in appetite regulation.

The World Health Organisation has estimated that by 2015 approximately 2.3 billion adults will be overweight and more than 700 million obese. Obesity is associated with an increased risk of diabetes, cardiovascular events, stroke and cancer. The hypothalamus is a crucial region for integrating signals from central and peripheral pathways and plays a major role in appetite regulation. In addition, there are reciprocal connections with the brainstem and higher cortical centres. In the arcuate nucleus of the hypothalamus, there are two major neuronal populations which stimulate or inhibit food in

Pharmacotherapy for obesity.

The current obesity pandemic imposes a major global disease burden. However, sustained weight loss of between 5 and 10% in the obese confers marked health benefits. Currently available pharmacotherapies, orlistat and sibutramine, can induce weight loss of between 5 and 10% over 2 years or more. However, in these trials, drug induced weight loss tends to be only 2-4 kg greater than that produced by placebo control. Despite this, in the XENDOS trial, the modest placebo-subtract weight loss produced by orlistat (2.8 kg) reduced the incidence of diabetes by over a third. Recent data on the potenti

Olfaction in the domestic fowl: a critical review.

It has been known for some time that many species of birds, including domestic fowl Gallus domesticus, have an olfactory sense. However, the functional significance of avian olfaction is less clear. We review neurobiological, embryological and behavioral evidence relevant to the question of how domestic fowl use the sense of smell. Evidence suggests a potential role for olfaction in the formation of attachments to familiar objects or environments; in the elicitation of fear responses by alarm and predator-related odors; in the control of feeding and drinking; and in avoidance of noxious substa

Healthy eating in infancy.

Dietary prescriptions meant to reduce the risk of adult diseases could retard growth and development in infants and young children. Parents who pay attention to their infant's hunger or satiety signals will avoid under-feeding or over-feeding their infant. A useful guiding principle is that, as long as overall growth compares favourably with the standards for age, it is likely that all is well. Nutrition priorities during infancy should reflect the promotion of growth and development rather than the prevention of diseases later in life. For this reason, fat restriction is not appropriate under

Eating attitudes in seasonal affective disorder and bulimia nervosa.

Markedly increased appetite and eating are commonly found in seasonal affective disorder (SAD) and bulimia nervosa (BN). To investigate dysfunctional eating attitudes, we administered the Eating Disorders Inventory (EDI), a self-rating questionnaire that assesses abnormal eating attitudes and behaviours, to 30 female SAD patients, 30 matched BN patients, and 30 non-clinical subjects. The SAD patients had higher scores on most EDI subscales compared to the non-clinical subjects; the BN patients had higher scores than the SAD patients on only two of three EDI subscales that are specific to eatin

The effects of reinforcement frequency and response requirements on the maintenance of behavior.

Six rats responded under fixed-interval and tandem fixed-interval fixed-ratio schedules of food reinforcement. Basic fixed-interval schedules alternated over experimental conditions with tandem fixed-interval fixed-ratio schedules with the same fixed-interval value. Fixed-interval length was varied within subjects over pairs of experimental conditions; the ratio requirement of the tandem schedules was varied across subjects. For both subjects with a ratio requirement of 10, overall response rates and running response rates typically were higher under the tandem schedules than under the corresp

Peripheral and central mechanisms of action of serotoninergic anorectic drugs.

Several pieces of evidence indicate that brain serotoninergic systems play an inhibitory role in feeding, being specifically involved in regulating satiety and food selection. The anorectic drug fenfluramine has been shown to exert its effects by activating serotoninergic mechanisms. Since fenfluramine influences both central and peripheral serotonin stores, it is difficult to establish the relative contributions of the central and peripheral serotoninergic mechanisms in the regulation of feeding behaviour. In the present paper evidence is presented that changes in feeding come about In the pr
