Enhanced chromatin compaction is associated with de novo expression of a nuclear microprotein, global loss of H3 acetylation and local transcriptional changes in retinal rod photoreceptors.

We have limited understanding of how aging alters gene expression and remodels cellular architecture in post-mitotic neurons. The inverted nuclear organization of mouse rod photoreceptors provides a unique model to gain mechanistic insights into age-associated decline in neuronal function. We have generated and integrated multi-omic datasets including 3D-genome topology, histone modifications, chromatin accessibility, DNA methylation and transcriptome of rod photoreceptors from young- and aged-mice. We show that aging drives global chromatin compaction, with regional alterations enriched at ac

Folding the message: mRNA structure as a regulatory layer of human mitochondrial gene expression.

Mammalian mitochondrial gene expression operates within an unusually compact genomic architecture in which most regulatory information must be encoded within or immediately adjacent to protein-coding sequences. In this context, mitochondrial mRNAs function not merely as templates for translation but as structured molecules whose folding landscape contributes to multiple stages of gene expression. Recent advances in chemical probing, mutational profiling, and mitoribosome profiling have begun to disclose the human mitochondrial mRNA structurome in its native organellar context, revealing a tran

Ensemble deep learning architectures for detecting pulmonary tuberculosis in chest X-rays.

Tuberculosis (TB) remains a major global health challenge, causing approximately 1.4 million deaths annually. In many high-burden regions, limited access to expert radiological interpretation leads to delayed or missed diagnoses. To address this, we propose a cost-effective, automated TB screening method suitable for under-resourced settings. Our method integrates a Convolutional Autoencoder Neural Network and a Multi-Scale Convolutional Neural Network with deep layer aggregation into an ensemble learning architecture for robust TB detection from chest radiographs. The framework was evaluated

A robust deep learning classifier for screening multiple retinal diseases on optical coherence tomography.

Retinal diseases are among the leading causes of visual impairment worldwide, where timely diagnosis and management are critical to prevent irreversible vision loss and blindness, especially in regions with limited access to ophthalmologists. While artificial intelligence (AI) has shown remarkable potential for screening ocular diseases, many existing models are heavily dependent on the datasets used for their development and often experience significant performance drops when tested on external datasets. These limitations in robustness and generalizability reduce their practical applicability

The evolution of ovarian somatic cells characterized by transcriptome and chromatin accessibility across rodents, monkeys, and humans.

The ovary plays a crucial role in the reproductive system of female mammals by producing mature oocytes through folliculogenesis. Non-human model organisms are extensively utilized in research on human ovarian biology, thus necessitating the investigation of conservation and divergence in molecular mechanisms across species. In this study, we employed integrative single-cell analysis of transcriptome and chromatin accessibility to identify the evolutionary conservation and divergence patterns of ovaries among humans, monkeys, mice, rats, and rabbits. Our analyses revealed that theca cells exhi

Cost-effective and natural-inspired lotus root/GelMA scaffolds enhanced wound healing via ROS scavenging, angiogenesis and reepithelialization.

Skin wounds, prevalent and fraught with complications, significantly impact individuals and society. Wound healing encounters numerous obstacles, such as excessive reactive oxygen species (ROS) production and impaired angiogenesis, thus promoting the development of chronic wound. Traditional clinical interventions like hemostasis, debridement, and surgery face considerable challenges, including the risk of secondary infections. While therapies designed to scavenge excess ROS and enhance proangiogenic properties have shown effectiveness in wound healing, their clinical adoption is hindered by h

Autonomous Trajectory Generation Comparison for De-Orbiting with Multiple Collision Avoidance.

Over the past four decades, space debris has been identified as a growing hazard for near-Earth space systems. With limited access to space debris tracking databases and only recent policy advancements made to secure a sustainable space environment and mission architecture, this manuscript aims to establish an autonomous trajectory maneuver to de-orbit spacecrafts back to Earth using collision avoidance techniques for the purpose of decommissioning or re-purposing spacecrafts. To mitigate the risk of colliding with another object, the spacecraft attitude slew maneuver requires high levels of p

Wi-Fi Access Point Design Concept Targeting Indoor Positioning for Smartphones and IoT.

Indoor positioning systems (IPS) have been regarded as essential for many applications, particularly for smartphones, during the past decade. With the internet of things (IoT), and especially device-to-device (D2D) cases, the client is supposed to have a very simple structure and low cost. It is also desirable that the client contains minimal software modules specifically for IPS purposes. This study proposes a new IPS technique that satisfies these conditions. The evaluation of the technique was previously executed based on a manual procedure. This technique utilizes Wi-Fi technology in addit

Identification of piRNA disease associations using deep learning.

Piwi-interacting RNAs (piRNAs) play a pivotal role in maintaining genome integrity by repression of transposable elements, gene stability, and association with various disease progressions. Cost-efficient computational methods for the identification of piRNA disease associations promote the efficacy of disease-specific drug development. In this regard, we developed a simple, robust, and efficient deep learning method for identifying the piRNA disease associations known as piRDA. The proposed architecture extracts the most significant and abstract information from raw sequences represented in a

Nanofiber Topographies Enhance Platelet-Fibrinogen Scaffold Interactions.

The initial contact with blood and its components, including plasma proteins and platelets, directs the body's response to foreign materials. Natural scaffolds of extracellular matrix or fibrin contain fibrils with nanoscale dimensions, but how platelets specifically respond to the topography and architecture of fibrous materials is still incompletely understood. Here, planar and nanofiber scaffolds are fabricated from native fibrinogen to characterize the morphology of adherent platelets and activation markers for phosphatidylserine exposure and α-granule secretion by confocal fluorescence mi

Effect of architecture disorder on the elastic response of two-dimensional lattice materials.

We examine how disordering joint position influences the linear elastic behavior of lattice materials via numerical simulations in two-dimensional beam networks. Three distinct initial crystalline geometries are selected as representative of mechanically isotropic materials with low connectivity, mechanically isotropic materials with high connectivity, and mechanically anisotropic materials with intermediate connectivity. Introducing disorder generates spatial fluctuations in the elasticity tensor at the local (joint) scale. Proper coarse-graining reveals a well-defined continuum-level scale e

A Hybrid Wired/Wireless Deterministic Network for Smart Grid.

With the rapid growth of time-critical applications in smart grid, robotics, autonomous vehicles, and industrial automation, demand for high reliability, low latency and strictly bounded jitter is sharply increasing. High-precision time synchronization communications, such as Time Triggered Ethernet (TTE), have been successfully developed for wired networks. However, the high cost of deploying additional equipment and extra wiring limits the scalability of these networks. Therefore, in this article, a hybrid wired/wireless high-precision time synchronization network based on a combination of h

CHARGE syndrome protein CHD7 regulates epigenomic activation of enhancers in granule cell precursors and gyrification of the cerebellum.

Regulation of chromatin plays fundamental roles in the development of the brain. Haploinsufficiency of the chromatin remodeling enzyme CHD7 causes CHARGE syndrome, a genetic disorder that affects the development of the cerebellum. However, how CHD7 controls chromatin states in the cerebellum remains incompletely understood. Using conditional knockout of CHD7 in granule cell precursors in the mouse cerebellum, we find that CHD7 robustly promotes chromatin accessibility, active histone modifications, and RNA polymerase recruitment at enhancers. In vivo profiling of genome architecture reveals th

Development of surface imprinted core-shell nanoparticles and their application in a solid-phase dispersion extraction matrix for methyl parathion.

Applying molecular imprinting techniques to the surface of functionalized SiO2 allows the preparation of molecularly imprinted polymers (MIPs) with accessible, high affinity and surface exposed binding sites. This paper demonstrates a new strategy for producing such hybrid organic-inorganic surface imprinted silica nanoparticles for specific recognition of methyl parathion. The technique provides surface grafting imprinting in chloroform using amino modified silica nanoparticles as supports, acrylamide as the functional monomer, γ-methacryloxypropyl trimethoxy silane as the grafting agent, and

The science and practice of micronutrient supplementations in nutritional anemia: an evidence-based review.

Nutritional anemia is the most common type of anemia, affecting millions of people in all age groups worldwide. While inadequate access to food and nutrients can lead to anemia, patients with certain health status or medical conditions are also at increased risk of developing nutritional anemia. Iron, cobalamin, and folate are the most recognized micronutrients that are vital for the generation of erythrocytes. Iron deficiency is associated with insufficient production of hemoglobin. Deficiency of cobalamin or folate leads to impaired synthesis of deoxyribonucleic acid, proteins, and cell divi
