Surveillance for Ixodes pacificus and the tick-borne pathogens Anaplasma phagocytophilum and Borrelia burgdorferi in birds from California's Inner Coast Range.

We investigated the involvement of birds in the ecology of the western black-legged tick, Ixodes pacificus, and its associated zoonotic bacteria, Borrelia burgdorferi and Anaplasma phagocytophilum, at two interior coast-range study sites in northern California. Anaplasma phagocytophilum, the agent of granulocytic anaplasmosis (GA), and B. burgdorferi s.s., the agent of Lyme disease (LD), are tick-borne pathogens that are well established in California. We screened blood and ticks from 349 individual birds in 48 species collected in 2011 and 2012 using pathogen-specific PCR. A total of 617 imma

Needle-stick injuries and blood contacts during invasive radiologic procedures: frequency and risk factors.

OBJECTIVE: Possible nosocomial transmission of blood-borne pathogens is a serious concern for health care workers and patients alike. Needle-stick injuries and blood contacts pose a risk of pathogen transmission during procedures. We performed a study to determine the frequency of injuries and blood contacts during invasive radiologic procedures and to examine procedure-related factors that may increase risk of injury or exposure. SUBJECTS AND METHODS: A prospective study of needle-stick injuries and blood contacts during invasive radiologic procedures was performed. During a 10-week period, …

Transfusion-related Epstein-Barr virus infection among stem cell transplant recipients: a retrospective cohort study in children.

BACKGROUND: Blood safety warrants strict screening measures to minimize the risk of transmitting blood-borne pathogens. However, transfusion-transmitted infections for which testing is not currently performed continue to be a concern. Among these untested agents is Epstein-Barr virus (EBV) which, in the transplant setting, is associated with lymphoproliferative disease, a potentially fatal cancer. The aim of this study was to analyze the incidence of posttransplant EBV infection and its association with administration of blood products in children receiving a hematopoietic stem cell (HSC) graf…

A newly developed multiplex RT-PCR enables detection of blood-associated viral nucleic acids in cats.

Feline immunodeficiency virus (FIV), feline leukemia virus (FeLV), domestic cat hepadnavirus (DCHBV), felis catus gammaherpesvirus 1 (FcaGHV1), and severe fever with thrombocytopenia syndrome virus (SFTSV) are significant blood-associated pathogens impacting feline health. Because these viruses often co-circulate, rapid and simultaneous detection is essential for effective diagnosis and surveillance. A multiplex reverse transcription polymerase chain reaction (RT-PCR) assay was developed for simultaneous detection of viral and proviral nucleic acids associated with feline blood-borne viruses.

Senescence Alters Antimicrobial Peptide Expression and Induces Amyloid-β Production in Retinal Pigment Epithelial Cells.

Age-related retinal degeneration, such as diabetic retinopathy and age-related macular degeneration, are major causes of blindness in modern society. Recent studies suggest that dysbiosis and intraocular translocation of bacteria from the blood circulation are critically involved in retinal degeneration. We hypothesise that the blood-retinal barrier (BRB) cells can protect the neuroretina from blood-borne pathogens by producing antimicrobial peptides (AMPs). The antimicrobial activity may decline during ageing, putting the retina at risk of low-degree chronic inflammation and degeneration. Her…

Development of Pentaplex Reverse Transcription Droplet Digital PCR Assay for Simultaneous Detection and Absolute Quantification of HIV-1, HIV-2, HCV, and HBV With Internal Control.

This study presents the development of a pentaplex reverse transcription droplet digital polymerase chain reaction (RT-ddPCR) assay for the simultaneous detection and absolute quantification of human immunodeficiency virus (HIV)-1, HIV-2, hepatitis C virus (HCV), hepatitis B virus (HBV), and an internal control. These viruses are important blood-borne pathogens for diagnostics, blood screening, and the validation of reference materials. Commercial kits used for blood screening allow multiplex detection but rely on relative, cycle threshold-based quantification approaches; consequently, the res…

A nationwide seroprevalence study of Francisella tularensis in Denmark.

BACKGROUND: Francisella tularensis is the causative agent of tularaemia. The incidence of tularaemia has increased in Europe in recent years. Due to the non-specific symptoms and diverse clinical manifestations, potential underreporting may exist. Additionally, the recent expansion in tick-borne pathogens could have increased the risk of F. tularensis transmission. We therefore aimed to estimate the seroprevalence of F. tularensis in Danish blood donors and tick-exposed individuals. METHODS: Plasma samples from healthy blood donors collected in 2022 (n=999) from all five administrative region

Establishing a laboratory colony of the human flea, Pulex irritans: methods for collecting, rearing, and feeding.

BACKGROUND: Colonizing fleas under laboratory conditions is a crucial step to studying their biology, conducting bioassays, and evaluating their ability to transmit pathogens. Starting a colony implies collecting and maintaining wild-caught specimens to obtain the next generations. Here we describe methods to collect, safely transport, and maintain adult and immature stages, and for the first time, to produce viable next generations of Pulex irritans, the human flea in the insectary. METHODS: Adult fleas were collected using human landing catches, while immature stages were obtained using the

Occupational exposure to hepatitis C virus infection and associated factors among healthcare workers in Fattouma Bourguiba University Hospital, Tunisia.

BACKGROUND: Healthcare workers (HCWs) are at high risk of hepatitis C virus (HCV) infection. Indeed, they are exposed to blood and body fluid which put them at an important risk of transmission of various blood borne pathogens including HCV. The goal of this study was to determine the magnitude of occupational exposure to hepatitis C virus infection as well as the factors associated to this exposure among HCWs at a Tunisian University Hospital in 2017. METHODS: A hospital-based cross-sectional study was carried out at Fattouma Bourguiba University Hospital in Monastir Governorate (Tunisia) fr

Hepatitis G virus in Saudi blood donors and chronic hepatitis B and C patients.

INTRODUCTION: Screening blood donors for blood-borne pathogens is very critical for the recipient's safety. Similar to hepatitis B and C infections, hepatitis G infection is transmitted through contaminated blood and causes acute and chronic hepatitis. Previous reports have shown that the prevalence of hepatitis G virus (HGV) RNA among healthy Saudi donors was 1%-2%. However, the exposure rate of this virus has never been studied. We hypothesized that the prevalence of HGV infection may have changed overtime due to socio-economic and environmental factors. Since hepatitis B and C infections ar…

Peritrophic matrix formation and Brugia malayi microfilaria invasion of the midgut of a susceptible vector, Ochlerotatus togoi (Diptera: Culicidae).

The mosquito midgut is the first site that vector-borne pathogens contact during their multiplication, differentiation, or migration from blood meal to other tissues before transmission. After blood feeding, the mosquitoes synthesize a chitinous structure called peritrophic matrix (PM) that envelops the blood meal and separates the food bolus from the midgut epithelium. In this study, a systematic investigation of the PM formation and the interaction of Brugia malayi within the midgut of a susceptible vector, Ochlerotatus togoi, were performed using scanning electron microscopy (SEM). SEM anal

Infection of the hip joint by Serratia marcescens.

A 72-year-old male patient presented to the hospital because of sudden inability to bear weight and without a history of trauma. A fracture of the head of the femur was identified on CT scan of the pelvis. In his history, the patient had a hospital admission 3 months earlier, during which he had a urinary catheter, and a urine specimen was analysed. The same pathogen was found in the patient urine and in the head of the femur specimen. This is a report of blood-borne spread of Serratia marcescens infection from the urothelium to the hip joint, responsible for spontaneous fracture of the femora

[Ocular manifestations during hepatitis C infection].

Hepatitis C wirus is one of the major blood-born pathogens of huge epidemic value. Due to its easy transmission, lack of routinely pursued tests and long non-symptomatic period makes it crucial to be alert for all co-morbidites suggesting HCV infection. Consequently to reduce its farther transmission large knowledge of HCV should be spread as there is still no effective means of prophylaxis and its therapy is much costly. This article describes ophthalmic symptoms of hepatitis C virus infection such a mere keratoconiunctivitis sicca to ischemic retinopathy, macular edema and ischemic neuropath

There Is a Method to the Madness: Strategies to Study Host Complement Evasion by Lyme Disease and Relapsing Fever Spirochetes.

Lyme disease and relapsing fever are caused by various Borrelia species. Lyme disease borreliae, the most common vector-borne pathogens in both the U.S. and Europe, are transmitted by Ixodes ticks and disseminate from the site of tick bites to tissues leading to erythema migrans skin rash, arthritis, carditis, and neuroborreliosis. Relapsing fever borreliae, carried by ticks and lice, trigger reoccurring fever episodes. Following transmission, spirochetes survive in the blood to induce bacteremia at the early stages of infection, which is thought to promote evasion of the host complement syste

The tale of two flaviviruses: subversion of host pathways by RNA shapes in dengue and hepatitis C viral RNA genomes.

Pathogenic RNA viruses continue to emerge owing to their rapid evolutionary rates. The family of the Flaviviridae contains enveloped, single-stranded, positive-sense RNA viruses that include mosquito borne viruses such as dengue virus and the blood-borne hepatitis C virus. Upon infection, the genomic viral RNA needs to first compete with a sea of host mRNAs for host ribosomes that synthesize the viral proteins. Then, the positive-sense template needs to be amplified and packaged into newly assembled virions. To accomplish these tasks, the virus subverts several biochemical machineries from the

Novel Ehrlichia and Hepatozoon genotypes in white-eared opossums (Didelphis albiventris) and associated ticks from Brazil.

White-eared opossums (Didelphis albiventris) are well adapted to anthropized areas. The increased contact with domestic animals and humans mediates the transmission of arthropod-borne pathogens. Despite the worldwide occurrence of tick-borne Anaplasmataceae and Hepatozoidae species in a variety of vertebrates, few studies reported serological evidence or molecular detection of theses agentes in marsupials. Up to now, while Ehrlichia/Anaplasma spp. have only been detected in marsupials from Brazil, Hepatozoon spp. have been reported in marsupials from Chile, Australia and Brazil. The present wo

Osteomyelitis of the Distal Phalanx of the Thumb due to Parvimonas micra and Fusobacterium nucleatum: A Case Report.

We herein report a case of osteomyelitis of the distal phalanx of the thumb of a 55-year-old man caused by Parvimonas micra and Fusobacterium nucleatum. Osteomyelitis often occurs in long bones and rarely occurs in the bones of the fingers. In addition, osteomyelitis of the finger frequently occurs after trauma or surgery, and blood-borne infection is very rare. P. micra and F. nucleatum, normal flora of the oral cavity, are very rare pathogenic bacteria of osteomyelitis except in periodontal disease, and there are no previous reports regarding the occurrence of osteomyelitis due to P. micra a

Human T cell lymphotropic virus type 1 infection among U.S. thalassemia patients.

Thalassemia is an inherited genetic disorder requiring multiple transfusions to treat anemia caused by low hemoglobin levels. Thus, thalassemia patients are at risk for infection with blood-borne pathogens, including human T cell lymphotropic viruses (HTLV) that are transmitted by transfusion of cellular blood products. Here, we examined the prevalence of HTLV among 234 U.S. thalassemia patients using sera collected in 2008. Sera were tested for antibodies to HTLV-1/2 using enzyme immunoassay (EIA) and a confirmatory western blot (WB) that differentiates between HTLV-1 and HTLV-2. Demographic …
