Exploring the heart-brain and brain-heart axes: Insights from a bidirectional Mendelian randomization study on brain cortical structure and cardiovascular disease.

INTRODUCTION: The bidirectional relationship between the brain cortex and cardiovascular diseases (CVDs) remains inadequately explored. METHODS: This study used bidirectional Mendelian randomization (MR) analysis to explore the interactions between nine phenotypes associated with hypertension, heart failure, atrial fibrillation (AF), and coronary heart disease (CHD), and brain cortex measurements. These measurements included total surface area (SA), average thickness (TH), and the SA and TH of 34 regions defined by the Desikan-Killiany atlas. The nine traits were obtained from sources such as…

Elimination of senescent cells by treatment with Navitoclax/ABT263 reverses whole brain irradiation-induced blood-brain barrier disruption in the mouse brain.

Whole brain irradiation (WBI), a commonly employed therapy for multiple brain metastases and as a prophylactic measure after cerebral metastasis resection, is associated with a progressive decline in neurocognitive function, significantly impacting the quality of life for approximately half of the surviving patients. Recent preclinical investigations have shed light on the multifaceted cerebrovascular injury mechanisms underlying this side effect of WBI. In this study, we aimed to test the hypothesis that WBI induces endothelial senescence, contributing to chronic disruption of the blood-brain…

African trypanosomiasis: haematogenic brain parasitism early in experimental infection through bypassing the blood-brain barrier, with considerations on brain trypanosomiasis in man.

A hematogenic invasion of the brain in suckling NMRI mice infected with Trypanosoma brucei rhodesiense was initiated by means of a mechanical damage of the blood-brain barrier. The brain was punctured after development of a blood infection. Brain infection was found in 31 out of 32 animals examined. Trypanosomes are initially capable of rapid multiplication. The number of parasites was highest during the 1st week. From the middle of the 2nd week the number of parasites decreased continuously, alongside increasing atrophy. In the 3rd and 4th week only rare degenerating or ghost trypanosomes wer…

Comparison of whole brain radiation therapy for synchronous brain metastases with irradiation protecting the hippocampus versus whole brain radiotherapy for sequential brain metastases to boost irradiation in the treatment of brain metastases from SCLC: study protocol for a randomized controlled trial.

BACKGROUND: This study is in regard to the comparison of whole brain radiation therapy for synchronous brain metastases with irradiation protecting the hippocampus versus whole brain radiotherapy for sequential brain metastases to boost irradiation in the treatment of brain metastases from small cell lung cancer (SCLC). Therapeutically, they have notably varying dose distributions. Based on theoretical and model studies, it has long been speculated that these modes may result in different prognostic outcomes. We aim to assess the efficacy of tomotherapy in the treatment of SCLC brain metastase…

Bridging the blood-brain barrier: strategies to improve delivery of biologics to tumors in the brain.

The unique anatomical and physiological features of the blood-brain barrier (BBB), while essential for maintenance of normal brain health, present significant challenges in development of effective therapies for neurological disorders from a drug delivery perspective. This is particularly the case for biotherapeutics such as monoclonal antibodies, which have a restricted brain distribution following systemic administration. While the BBB is often presumed to be leaky in tumors within the brain, it is important to recognize that the degree of compromise is highly variable with certain regions s…

The effects of blast-induced traumatic brain injury on brain cellular mechanics and differentiation.

Blast-induced traumatic brain injury (bTBI) causes significant disruptions in cellular and subcellular structures within the central nervous system (CNS) when an extremely large force is applied. The corresponding changes in biomechanical properties and cellular functionalities of neuronal and glial cells due to bTBI remain largely unexplored. In this work, high blast overpressures (BOPs) of 14.5 psi (single shockwave) and 29.0 psi (double shockwave) were applied to adult hippocampal progenitor cells (AHPCs) in two different directions (overpressure applied from 'top-to-bottom' and 'bottom-to-…

Brain Abscesses as a Complication of Brain Hematomas in Infective Endocarditis: A Rare Case Report.

INTRODUCTION: Brain abscesses developing within a nonoperated intracranial hematoma are even rarer. We report brain abscesses developing within 2 hematomas and complicating a methicillin-sensitive Staphylococcus aureus (MSSA) infectious endocarditis (IE). CASE: A previously healthy 26-year-old man without risk factors was diagnosed at hospital day (HD) 2 with a parietal hematoma (24 mm). Angiography did not reveal any aneurysm. He underwent a biological aortic valve replacement at HD4. Valve culture was positive for MSSA. Subsequently, a frontal hematoma appeared, an enlargement of both hemat…

The gut-brain ConNECtion: exploring the developmental impact of necrotizing enterocolitis on the neonatal brain.

Necrotizing enterocolitis (NEC) is the leading cause of death from gastrointestinal disease within premature infants, posing a significant concern to infants admitted to the neonatal intensive care unit. In those patients who overcome the disease, there are significant long-term effects that include gastrointestinal effects as well as neurodevelopmental deficits. The pathogenesis of necrotizing enterocolitis and its related brain injury that results in these neurodevelopmental deficits is complex and multifaceted, with much research supporting the idea that NEC-induced inflammation is a major …

Brain State of the Newborn as a Biomarker for Brain Injury in Infants with Hypoxic-Ischemic Encephalopathy.

OBJECTIVE: To evaluate the use of a fully automated trend measure of cortical activity, Brain State of the Newborn (BSN), in early stratification of infants for add-on neuroprotective therapies during therapeutic hypothermia (TH). STUDY DESIGN: Our retrospective cohort study included 167 infants with moderate-to-severe hypoxic-ischemic encephalopathy who underwent TH and continuous electroencephalography monitoring. The BSN trends were computed using fully automated pipelines, and we used a priori-defined thresholds at 6, 12, 24, and 36 hours after birth to assess prediction of an adverse pos…

Assessing brain involvement in Fabry disease with deep learning and the brain-age paradigm.

While neurological manifestations are core features of Fabry disease (FD), quantitative neuroimaging biomarkers allowing to measure brain involvement are lacking. We used deep learning and the brain-age paradigm to assess whether FD patients' brains appear older than normal and to validate brain-predicted age difference (brain-PAD) as a possible disease severity biomarker. MRI scans of FD patients and healthy controls (HCs) from a single Institution were, retrospectively, studied. The Fabry stabilization index (FASTEX) was recorded as a measure of disease severity. Using minimally preprocessed…

Arsenic induced neurotoxicity in the brain of ducks: The potential involvement of the gut-brain axis.

BACKGROUND: Arsenic is a widely distributed ecotoxic pollutant that has been found to cause neurotoxicity in a variety of species. Gut-brain axis is a two-way information network between the gut microbiome and the brain, which is closely related to organismal health. However, the role of the gut-brain axis in arsenic-induced neurotoxicity remains largely unknown. METHODS: In order to explore whether there is a relationship between brain and gut microbiota of meat ducks, we performed molecular biological detection including RT-qPCR and Western blot, as well as morphological detection including…

Brain functional connectivity in children with a mild traumatic brain injury: A scoping review.

INTRODUCTION: The occurrence of mild traumatic brain injury(mTBI) is estimated at 0,2-0,3% cases annually. Following a mTBI, some children experience persistent symptoms, and functional connectivity(FC) changes may be implicated. However, characteristics of FC have not been widely described in this population. This scoping review aimed to identify and understand the impacts of mTBI on EEG-measured FC in children, provide an overview of the available literature, detail analysis techniques, and describe gaps in the research. METHODS: PubMed, Web of Science, Medline, Embase, ProQuest and CINAHL …

Is Short Sleep Bad for the Brain? Brain Structure and Cognitive Function in Short Sleepers.

Many sleep less than recommended without experiencing daytime sleepiness. According to prevailing views, short sleep increases risk of lower brain health and cognitive function. Chronic mild sleep deprivation could cause undetected sleep debt, negatively affecting cognitive function and brain health. However, it is possible that some have less sleep need and are more resistant to negative effects of sleep loss. We investigated this using a cross-sectional and longitudinal sample of 47,029 participants of both sexes (20-89 years) from the Lifebrain consortium, Human Connectome project (HCP) and…

Brain stimulation competes with ongoing oscillations for control of spike timing in the primate brain.

Transcranial alternating current stimulation (tACS) is a popular method for modulating brain activity noninvasively. In particular, tACS is often used as a targeted intervention that enhances a neural oscillation at a specific frequency to affect a particular behavior. However, these interventions often yield highly variable results. Here, we provide a potential explanation for this variability: tACS competes with the brain's ongoing oscillations. Using neural recordings from alert nonhuman primates, we find that when neural firing is independent of ongoing brain oscillations, tACS readily ent…

The role of brain inflammation and abnormal brain oxygen homeostasis in the development of hepatic encephalopathy.

Hepatic encephalopathy (HE) is a frequent complication of chronic liver disease (CLD) and has a complex pathogenesis. Several preclinical and clinical studies have reported the presence of both peripheral and brain inflammation in CLD and their potential impact in the development of HE. Altered brain vascular density and tone, as well as compromised cerebral and systemic blood flow contributing to the development of brain hypoxia, have also been reported in animal models of HE, while a decrease in cerebral metabolic rate of oxygen and cerebral blood flow has consistently been observed in patie…

The role of the default mode network in longitudinal functional brain reorganization of brain gliomas.

The study of patients after glioma resection offers a unique opportunity to investigate brain reorganization. It is currently unknown how the whole-brain connectomic profile evolves longitudinally after surgical resection of a glioma and how this may be associated with tumor characteristics and cognitive outcome. In this longitudinal study, we investigate the impact of tumor lateralization and grade on functional connectivity (FC) in highly connected networks, or hubs, and cognitive performance. Twenty-eight patients (17 high-grade, 11 low-grade gliomas) underwent longitudinal pre/post-surgery…

Impact of obesity and diet on brain structure and function: a gut-brain-body crosstalk.

Most societies witness an ever increasing prevalence of both obesity and dementia, a scenario related to often underestimated individual and public health burden. Overnutrition and weight gain have been linked with abnormal functionality of homoeostasis brain networks and changes in higher cognitive functions such as reward evaluation, executive functions and learning and memory. In parallel, evidence has accumulated that modifiable factors such as obesity and diet impact the gut-brain axis and modulate brain health and cognition through various pathways. Using neuroimaging data from epidemiol…

The brain at war: effects of stress on brain structure in soldiers deployed to a war zone.

In search of the neural basis of severe trauma exposure and post-traumatic stress disorder (PTSD), a multitude of cross-sectional studies have been conducted, most of them pointing at structural deficits in the hippocampus and medial prefrontal cortex including the anterior cingulate cortex (ACC) and ventromedial prefrontal cortex (vmPFC). Since cross-sectional studies are silent to causality, the core question remains: which brain structural alterations constitute a risk factor for disease and therewith precede the stressor, and which brain regions may undergo alterations as a consequence of …
