Acute infection with venezuelan equine encephalitis virus replicon particles catalyzes a systemic antiviral state and protects from lethal virus challenge.

The host innate immune response provides a critical first line of defense against invading pathogens, inducing an antiviral state to impede the spread of infection. While numerous studies have documented antiviral responses within actively infected tissues, few have described the earliest innate response induced systemically by infection. Here, utilizing Venezuelan equine encephalitis virus (VEE) replicon particles (VRP) to limit infection to the initially infected cells in vivo, a rapid activation of the antiviral response was demonstrated not only within the murine draining lymph node, where

Divergent effects of ischemia/reperfusion and nitric oxide donor on TNFalpha mRNA accumulation in rat organs.

We previously showed that serum TNFalpha bioactivity in rats is proportional to the extent of graded tissue injury caused by laparotomy, intestinal ischemia, and reperfusion and that the spleen is an important source of TNFalpha secretion in this condition. TNFalpha production varies, depending on the type and duration of tissue injury. It is also affected by other mediators, such as nitric oxide (NO). TNFalpha is known to increase NO production, but the effect of NO on the production of TNFalpha has not yet been fully elucidated. In this study we determined the levels of TNFalpha mRNA in rat

High-resolution MR venography of cerebral arteriovenous malformations.

The purpose of this study was to evaluate the diagnostic potential of a high-resolution magnetic resonance (MR) venography technique in patients with cerebral arteriovenous malformations (AVMs). A high-resolution 3D gradient echo sequence was used with a long echo time TE to obtain venous information down to sub-pixel sized vessel diameters of several hundred microns. The method is based on the paramagnetic property of deoxyhemoglobin, and the resulting developing phase difference between veins and brain parenchyma at long echo times which leads to signal cancellation. The reconstructed venogr

Plasma exchange in patients with septic shock including acute renal failure.

In patients with septic shock and multiorgan failure including acute renal failure, the prognosis is poor. This study evaluated the effects of plasma exchange as adjunctive therapy to the conventional treatment in such severely ill patients and compared the results with the expected outcome according to data from other studies. A total of 25 patients (17 men and 8 women, mean age 47, range 15-74 years) were treated by a median of 3 (range 1-10) plasma exchanges, mainly by centrifugation technique. The main replacement fluid was liquid-stored plasma and albumin (in a few cases fresh frozen plas

[Lateral ventricle tumors. (Part 2) -Neuroradiological analyses of 30 cases- (author's transl)].

In the first report, the clinical manifestations of 30 cases of the lateral ventricle tumor were reviewed. This report summarizes neuroradiological findings of the same 30 cases, in which 26 cases were examined by cerebral angiograms and 3 cases by CT scan. For the radiological analyses, the tumors of the lateral ventricle are classified into two groups, as follows: 1. Intraventricular tumors arise in the projection of the choroid plexus, the tela and the ependyma and grow in the lateral ventricle. 2. Paraventricular tumors arise from the wall of the neuroglia not only bulge into ventricle, bu

Acquired Chiari malformation Type I and holocord syringomyelia associated with a high-flow supratentorial fistulous arteriovenous malformations: A case report and literature review.

BACKGROUND: Chiari malformation Type I (CMI) is generally considered a congenital lesion and typically associated with syringomyelia. Acquired CMI or adult Chiari malformation caused by intracranial mass is extremely rare. Brain arteriovenous malformations (AVMs) are characteristically symptomatic due to seizure, intracranial hemorrhage, or neurological deficit. We report an extremely rare case of an acquired CMI and extensive syringomyelia associated with a large supratentorial AVM. CASE DESCRIPTION: A 35-year-old woman was referred to our institute after a diagnosis of CMI and extensive syr

Time-of-Arrival Parametric Maps and Virtual Bolus Images Derived From Contrast-Enhanced Time-Resolved Radial Magnetic Resonance Angiography Improve the Display of Brain Arteriovenous Malformation Vascular Anatomy.

OBJECTIVES: Time-of-arrival (TOA) maps can be derived from high-resolution 4-dimensional (4D) contrast-enhanced magnetic resonance angiography (MRA) data sets to provide a quantitative description of contrast material arrival time in each voxel. This information can further be processed to create a compressed time evolution curve that virtually shortens the contrast bolus (virtual bolus [VB]). The purpose of this project was to determine whether TOA-enhanced 4D MRA and/or VB imaging improve the display of contrast kinetics in patients with vascular disease. METHODS: High-resolution whole-brai

Sunset Sign Due to Intraventricular Tension Pneumocephalus: A Key Clue to Evaluating Delayed Emergence After General Anesthesia.

Prompt emergence from general anesthesia is crucial after neurosurgical procedures, such as craniotomies, to facilitate timely neurological evaluation for identification of intraoperative complications. Delayed emergence can be caused by residual anesthetics, metabolic imbalances, and intracranial pathology, for which an eye examination can provide early diagnostic clues. The sunset sign (or setting sun sign), characterized by a downward deviation of the eyes, can be an early indicator of raised intracranial pressure (ICP) or midbrain compression, as is commonly observed in states of hydroceph

Decoding brain arteriovenous malformations: from genetic insights to modeling the vascular maze.

Brain arteriovenous malformations (bAVMs) are complex cerebrovascular anomalies characterized by a tangle of dysplastic vessels, including feeding arteries, a nidus, and draining veins, with the absence of an intervening capillary bed. bAVMs pose a significant risk of severe complications, such as intracranial hemorrhage, seizures, and progressive neurological deficits, leading to substantial morbidity and mortality. While advancements in treatments, including microsurgical resection, endovascular embolization, and stereotactic radiosurgery, have improved patient outcomes, the underlying patho

The thymus gland is a target and maintainer of amyotrophic lateral sclerosis pathophysiology.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by the selective loss of motor neurons in the brain and spinal cord with evidence of local neuroinflammation. Regulatory T cells (Treg) exert neuroprotective effects and correlate with disease progression, but central mechanisms governing Treg development during ALS remain unclear. We investigated the thymic architecture and T cell differentiation in SOD1-G93A (mSOD1) ALS mice at multiple disease stages. Thymocyte and thymic epithelial cells (TEC) subpopulations, as well as stages of Treg differentiation and

Maximizing safety in the treatment of a large choroidal fissure arteriovenous malformation: The essential role of anatomy, staged embolization, and microsurgical technique.

BACKGROUND: Arteriovenous malformations (AVMs) are complex congenital pathologies that require a deep understanding of their angioarchitecture and the involved brain anatomy. The ones within the temporal horn of the lateral ventricle present significant challenges for microsurgical treatment due to their deep location and proximity to critical neurovascular structures. These rare AVMs typically manifest with seizures or hemorrhage. They have afferents from the choroidal arteries, with arterial supply often arising from both the anterior and posterior circulations through the choroidal arteries

ROCKETS - a novel one-for-all toolbox for light sheet microscopy in drug discovery.

Advancing novel immunotherapy strategies requires refined tools in preclinical research to thoroughly assess drug targets, biodistribution, safety, and efficacy. Light sheet fluorescence microscopy (LSFM) offers unprecedented fast volumetric ex vivo imaging of large tissue samples in high resolution. Yet, to date laborious and unstandardized tissue processing procedures have limited throughput and broader applications in immunological research. Therefore, we developed a simple and harmonized protocol for processing, clearing and imaging of all mouse organs and even entire mouse bodies. Applyin

Robotic external ventricular drain placement for acute neurosurgical care in low-resource settings: feasibility considerations and a prototype design.

OBJECTIVE: Emergency neurosurgical care in lower-middle-income countries faces pronounced shortages in neurosurgical personnel and infrastructure. In instances of traumatic brain injury (TBI), hydrocephalus, and subarachnoid hemorrhage, the timely placement of external ventricular drains (EVDs) strongly dictates prognosis and can provide necessary stabilization before transfer to a higher-level center of care that has access to neurosurgery. Accordingly, the authors have developed an inexpensive and portable robotic navigation tool to allow surgeons who do not have explicit neurosurgical train

Early resection of ectopic bone in patients with heterotopic ossification about the hip after trauma.

OBJECTIVES: Heterotopic ossification (HO) is a common complication in patients who have sustained high-energy trauma to the hip region. Traditionally, resection is performed after ectopic bone maturation. We hypothesized that early HO resection in patients with hip ankylosis after trauma can be performed with little chance of recurrence. DESIGN: Retrospective clinical cohort. SETTING: Level I Trauma Center PATIENTS/PARTICIPANTS: 14 patients with resection of HO about the hip performed by the senior author during a six-year period. INTERVENTION: Early resection of Brooker Class III or IV HO.

Comparative pharmacokinetic and biodistribution study of two distinct squalene-containing oil-in-water emulsion adjuvants in H5N1 influenza vaccines.

BACKGROUND: In recent years, there has been great interest from academia, industry and government scientists for an increased understanding of the mode of action of vaccine adjuvants to characterize the safety and efficacy of vaccines. In this context, pharmacokinetic (PK) and biodistribution studies are useful for quantifying the concentration of vaccine adjuvants in mechanistically or toxicologically relevant target tissues. METHODS: In this study, we conducted a comparative analysis of the PK and biodistribution profile of radiolabeled squalene for up to 336 h (14 days) after intramuscular
