The role of transient receptor potential cation channels in Ca2+ signaling.

The 28 mammalian members of the super-family of transient receptor potential (TRP) channels are cation channels, mostly permeable to both monovalent and divalent cations, and can be subdivided into six main subfamilies: the TRPC (canonical), TRPV (vanilloid), TRPM (melastatin), TRPP (polycystin), TRPML (mucolipin), and the TRPA (ankyrin) groups. TRP channels are widely expressed in a large number of different tissues and cell types, and their biological roles appear to be equally diverse. In general, considered as polymodal cell sensors, they play a much more diverse role than anticipated. Fun

TPCs: FROM PLANT TO HUMAN.

In 2005, the Arabidopsis thaliana two-pore channel TPC1 channel was identified as a vacuolar Ca2+-release channel. In 2009, three independent groups published studies on mammalian TPCs as nicotinic acid adenine dinucleotide phosphate (NAADP)-activated endolysosomal Ca2+ release channels, results that were eventually challenged by two other groups, claiming mammalian TPCs to be phosphatidylinositol-3,5-bisphosphate [PI(3,5)P2]-activated Na+ channels. By now this dispute seems to have been largely reconciled. Lipophilic small molecule agonists of TPC2, mimicking either the NAADP or the PI(3,5)P2

The regulatory roles of calcium channels in tumors.

Calcium (Ca2+) and its relevant transmembrane and intracellular calcium channels were previously thought to be chiefly associated with the regulation of cardiovascular and neuronal systems. Nowadays, an increasing evidence shows those calcium channels are also responsible for tumorigenesis and progression. However, the general underlying mechanisms and the involving signaling transduction pathways still remain unclear. Therefore, in this mini-review, we are mainly focusing on the linkage between calcium channels and major characteristics of tumors such as multi-drug resistance (MDR), metastasi

Promoting Autophagy Mitigates Stress-Induced Remodeling in Patient iPSC-CMs with the Phospholamban R9C Mutation.

Phospholamban (PLN) is a key regulator of adrenergic signaling and calcium homeostasis in cardiomyocytes. The PLN R9C mutation causes early-onset dilated cardiomyopathy (DCM) and premature death, yet the mechanisms underlying its pathogenic remodeling remain unclear. In this study, patient-specific induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs) harboring the PLN R9C and their isogenic corrected controls are generated to investigate the molecular and functional consequences of the mutation. At baseline, PLN R9C iPSC-CMs exhibit near-normal calcium handling but blunt β-adrenergi

Arsenic induces chicken liver injury by inhibiting endoplasmic reticulum-lysosomal calcium ion transport and lysosomal autophagy activity via the IP3R/TRPML1 pathway.

Arsenic is a toxic substance widely present in the environment, and arsenic-containing pesticides and fertilizers are widely used in agricul … s well as organelle function and cell signaling. However, studies on the mechanism of intracellular calcium signaling in arsenic-induced liver injury are lacking. We found that arsenic impeded the endoplasmic reticulum-lysosome calcium transport process and induced impaired lysosomal activity and lysosome-associated autophagy blockage through this pathway, which ul …

The calcium channel blocker verapamil inhibits oxidative stress response in Candida albicans.

Candida albicans is a common opportunistic fungal pathogen, causing both superficial candidiasis and life-threatening systemic infections in immune-compromised individuals. Calcium signaling is responsible for this pathogen in responding to several stresses, such as antifungal drugs, alkaline pH and membrane-perturbing agents. Our recent study revealed that it is also involved in oxidative stress response. In this study, we investigated the effect of verapamil, an L-type voltage-gated calcium channel blocker, on oxidative stress response in this fungus. The addition of verapamil resulted in in…

Finerenone Improves Albuminuria via MR-TRPC Signaling in Diabetic Kidney Disease.

BACKGROUND: Recent studies have confirmed the protective effects of nonsteroidal MR (mineralocorticoid receptor) antagonists in diabetic kidney disease. However, the physiological mechanisms underlying their albuminuria-reducing effects remain incompletely defined. We hypothesized that inhibition of the MR could protect podocytes by limiting excessive calcium influx via TRPC (transient receptor potential canonical) 5, thereby reducing albuminuria. METHODS: We evaluated the effects of the nonsteroidal MR antagonist finerenone on albuminuria, podocyte morphology, and glomerular function in diab

"Calcium receptors" on eukaryotic cells with special reference to the osteoclast.

There is a growing list of cells that are capable of detecting and responding to changes in the concentration of extracellular calcium. The two classic examples of this behaviour are the calcitonin-secreting parafollicular cells of the thyroid and parathyroid hormone-secreting chief cells of the parathyroid gland. A more recent addition to this list is the renin-secreting juxtaglomerular cell of the kidney. Particularly intriguing has been independently the discovery by two laboratories, that the resorptive cell of bone, the osteoclast, is capable of detecting changes in ambient calcium. A com

Mitigation of Stress-induced Structural Remodeling and Functional Deficiency in iPSC-CMs with PLN R9C Mutation by Promoting Autophagy.

BACKGROUND: Phospholamban (PLN) is a key regulator of cardiac function connecting adrenergic signaling and calcium homeostasis. The R9C mutation of PLN is known to cause early onset dilated cardiomyopathy (DCM) and premature death, yet the detailed mechanisms underlie the pathologic remodeling process are not well defined in human cardiomyocytes. The aim of this study is to unravel the role of PLN R9C in DCM and identify potential therapeutic targets. METHODS: PLN R9C knock-in (KI) and patient-specific induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs) were generated and comprehe

Glycin-proline-hydroxyproline modified N-terminal 20 peptide from CEMP1 blocks calcium ion influx to alleviate inflammatory response in gingival fibroblasts.

Gingival fibroblasts play a pivotal role in the progression of periodontitis, and targeted inhibition of calcium ion influx in gingival fibr … macrophages. This specific binding significantly blocked intracellular calcium influx and aberrant calcium signaling activation, and effectively abrogated LPS-induced pro-inflammatory responses in these cells. Mechanistic analyses further revealed that the dual functions of GPH-N20, its collagen-targeting capacity and calcium-chelating activity, synergistically su …

The CAGE Framework (Calcium-Gated Excitability): From Single Neurotransmitters to Calcium States in the Tumor Microenvironment.

The tumor microenvironment (TME) is an innervated and neurochemically active setting. To date, its cholinergic and GABAergic signaling have largely been analyzed as separate systems, contributing to inconsistent findings across the literature. We propose a Calcium-Gated Excitability (CAGE) model, in which α7 nicotinic acetylcholine receptor (nAChR), α4β2 nAChR, GABA type A (GABAA) and type B (GABAB) receptors, and α2δ-1 converge on a single downstream variable: Ca2+ flux. α7 conducts Ca2+ directly; α4β2 may redirect cholinergic input toward GABA release; GABAA and GABAB set the direction of th

Mechanotransduction Failure and Molecular Rescue in Gastric Cancer: Kinetotherapy Across the IL-6/STAT3-Myostatin/ACVR2B-Akt/mTOR Axis.

Muscle wasting associated with gastric cancer represents a complex, multifactorial systems disorder involving inflammatory, anabolic, mechanosensory, calcium-regulatory, mitochondrial, and proteostatic disruption. This review synthesizes current evidence regarding the cellular and physiological mechanisms involved in skeletal muscle dysfunction in gastric cancer and provides a unifying framework centered on loss of signaling coherence. Specifically, it examines IL-6/STAT3 and NF-κB inflammatory signaling, the myostatin-activin-ACVR2B-SMAD pathway, PI3K/Akt/mTOR signaling, mechanotransduction,

Functional coupling between peripheral nociceptor terminals in vivo is enhanced during acute inflammation.

Primary nociceptors are essentially characterized as afferent neurons carrying noxious sensory information from the periphery to the central … ry cytokines, the efferent signaling along nociceptive terminals increases, culminating in enhanced calcium signaling in the remote unstimulated terminals. This increase in intraterminal calcium could trigger an enhanced release of inflammatory mediators, affecting wider areas and terminals from adjacent unstimulated receptive fields, leading to the expansion of hy …

Piezo1 Agonist Yoda1 Induces Rapid Relaxation in Cultured Airway Smooth Muscle Cells and Bronchodilation in Mouse Models.

Bronchodilators that relax airway smooth muscle cells (ASMCs) are essential for treating constrictive airway diseases such as asthma. However, the existing bronchodilators are often unable to control symptoms of severe asthmatic patients, leaving a pressing need to search for alternatives. Recent studies indicate that the transmembrane mechanosensitive channel, Piezo1, may provide a novel target for bronchodilation, as it mediates ASMC relaxation by means of calcium signaling and activation of large-conductance, calcium-activated potassium channels, and the Piezo1-specific agonist Yoda1 has be…

Serum Calcium is Related to the Degree of Artery Stenosis in Acute Ischemic Stroke.

BACKGROUND/AIMS: Acute ischemic stroke is caused by stenosis of artery supplying to brain. We aimed to detect some metabolites in the serum … ed genes were enriched in a phosphatidylinositol signaling system, which is closely involved in the calcium signaling pathway. CONCLUSION: Our results suggest that the concentration of serum calcium is higher in patients with more severe artery stenosis lesions and that the phosphatidylinositol signaling system is a key biological pathway involved in this process.

Danger signals activate a putative innate immune system during regeneration in a filamentous fungus.

The ability to respond to injury is a biological process shared by organisms of different kingdoms that can even result in complete regeneration of a part or structure that was lost. Due to their immobility, multicellular fungi are prey to various predators and are therefore constantly exposed to mechanical damage. Nevertheless, our current knowledge of how fungi respond to injury is scarce. Here we show that activation of injury responses and hyphal regeneration in the filamentous fungus Trichoderma atroviride relies on the detection of two danger or alarm signals. As an early response to inj

Herpes simplex virus pUL56 abolishes neuronal activity by removing voltage-gated ion channels from the plasma membrane.

Herpes simplex virus 1 (HSV-1) infections of the central nervous system cause encephalitis and are associated with increased risk of neurode … voltage-gated sodium, potassium, and calcium channels, resulting in a profound loss of synchronous calcium signaling. We identify the viral ubiquitin-ligase adaptor pUL56 as the principal effector of this process: pUL56-dependent degradation of ion channels abolishes coordinated calcium signaling, whereas mutation of its E3-ligase-binding motifs preserves synchron …

Ultralow calcium requirements of fungi facilitate use of calcium regulating agents to suppress host calcium-dependent defenses, synergizing infection by a mycoherbicide.

Infection by many fungi activates a variety of calcium dependent defenses in the hosts, slowing or suppressing the attacker and limiting the efficacy of mycoherbicides. The calcium requirement for fungal growth is so low that it could only be implied based on fungi containing calcium-dependent signaling enzymes. Analytical grade media contain <2 microM calcium, and the addition of specific chelators does not affect fungal growth. Hydrophobic derivatives of the calcium-specific chelator BAPTA designed to traverse plant cuticles were synthesized in order to chelate calcium internally during fung

Calcium-Handling Defects and Neurodegenerative Disease.

Calcium signaling is critical to neuronal function and regulates highly diverse processes such as gene transcription, energy production, protein handling, and synaptic structure and function. Because there are many common underlying calcium-mediated pathological features observed across several neurological conditions, it has been proposed that neurodegenerative diseases have an upstream underlying calcium basis in their pathogenesis. With certain diseases such as Alzheimer's, Parkinson's, and Huntington's, specific sources of calcium dysregulation originating from distinct neuronal compartmen…
