Mammalian aquaglyceroporin function in metabolism.

Aquaglyceroporins are integral membrane proteins that are permeable to glycerol as well as water. The movement of glycerol from a tissue/organ to the plasma and vice versa requires the presence of different aquaglyceroporins that can regulate the entrance or the exit of glycerol across the plasma membrane. Actually, different aquaglyceroporins have been discovered in the adipose tissue, small intestine, liver, kidney, heart, skeletal muscle, endocrine pancreas and capillary endothelium, and their differential expression could be related to obesity and the type 2 diabetes. Here we describe the

Glucosuria and albuminuria in diabetic nephropathy: a consideration at nanolevel.

In diabetes, the appearance of albumin in the urine has long been recognized as a cardinal feature of kidney disease. Physiologically, the glomerular capillary wall provides a barrier to protein in blood not to cross into urine. However, in the pathological condition, the change in the glomerular permeability, protein can be detected in urine. Here, the author reviewed the reported size of the glomerular pore size as well as the molecular size of glucose and albumin in the literature. According to this study, it can conclude that the appearance of glucosuria in diabetic patient is due to the i

Intensive care treatment of ovarian hyperstimulation syndrome (OHSS).

Ovarian hyperstimulation syndrome (OHSS) is a morbid, iatrogenic, and potentially lethal condition caused by the pharmacological induction of ovulation. In OHSS, the ovaries are the site of significant stromal edema with bilateral cystic ovarian enlargement. It has been suggested that ascitic fluid accumulation may be due to increased capillary permeability, which results in fluid shift from the intravascular to the extravascular compartment. While the exact etiology is not clear, several biochemical markers are considered to be possible mediators. There is an incidence of OHSS in about 3% to …

[Acute respiratory distress syndrome: definitions, mechanisms and treatment].

Acute respiratory distress syndrome is a secondary acute respiratory insufficiency caused by an inflammatory syndrome which is characterized by an increased of permeability pulmonary edema, associated with many other clinic anomalies, radiological and pathophysiological not directly caused by, but with which it could coexist, a left atrial hypertension. The illness, characterized by refractory hypoxemia, recognizes several causes, which have direct or indirect harm on the cells of the membrane alveolus-capillary. In spite of the improvements in the therapeutic approach, during these last 40 ye

Pulmonary edema associated with hyperbaric oxygen therapy.

We report three cases of pulmonary edema associated with hyperbaric oxygen therapy, including one fatality. All three patients had cardiac d … e right and left hearts, inducing bradycardia with concomitant LV dysfunction, increasing pulmonary capillary permeability, or by causing pulmonary oxygen toxicity. We advise caution in the use of hyperbaric oxygen therapy in patients with heart failure or in patients with reduced cardiac EFs.

Transcriptome clarifies mechanisms of lesion genesis versus progression in models of Ccm3 cerebral cavernous malformations.

Cerebral cavernous malformations (CCMs) are dilated capillaries causing epilepsy and stroke. Inheritance of a heterozygous mutation in CCM3/PDCD10 is responsible for the most aggressive familial form of the disease. Here we studied the differences and commonalities between the transcriptomes of microdissected lesional neurovascular units (NVUs) from acute and chronic in vivo Ccm3/Pdcd10ECKO mice, and cultured brain microvascular endothelial cells (BMECs) Ccm3/Pdcd10ECKO.We identified 2409 differentially expressed genes (DEGs) in acute and 2962 in chronic in vivo NVUs compared to microdissected

[Reexpansion pulmonary edema].

Reexpansion pulmonary edema is an uncommon complication which sometimes occurs after evacuation of a large amount of air or fluid from the pleural space. We report two cases that illustrate the diversity of the clinical expression, severe in one case and latent in the other. The pathophysiology of reexpansion pulmonary edema remains obscure. Increased pulmonary capillary permeability, favored by previous atelectatic parenchyma and rapid reexpansion appears to be the main cause. Treatment is basically preventive. Curative treatment is based on adequate oxygenation and circulation. Lower aspirat…

Localization of the atrial natriuretic peptide binding sites in the inner ear tissue--possibly an additional regulating system.

Specific high affinity atrial natriuretic peptide (ANP) binding sites were identified and localized by autoradiography (125J-ANP 1-28) in the inner ear of guinea pigs. ANP is a cardiac hormone known to mediate increased capillary permeability, vasodilation, and natriuresis. Since specific receptors for ANP exist in both eye and brain, possibly playing a role in regulation of fluid and pressure (14, 15), we hypothetized that ANP might be involved also in inner ear fluid dynamics via interaction with other volume/electrolyte balance regulating hormones including vasopressin, renin, and aldostero…

Arterial and venous effects of atrial natriuretic peptide in the human forearm.

Atrial natriuretic peptide (ANP) has direct vasodilating properties in addition to its diuretic and natriuretic effects. Furthermore, vascular permeability may be influenced. Because relatively little is known about the venous and capillary actions of ANP in humans, we investigated the spectrum of vascular actions of ANP in the human forearm. In seven healthy subjects, ANP was infused intraarterially in consecutive doses of 0, 10, 50, and 250 ng/100 mL tissue/min together with vehicle or norepinephrine 1 ng/kg/min. In seven other subjects, 0.5 ng/kg/min angiotensin II and 10 ng/kg/min serotoni

Estimating the Starling equation parameters in living humans by using volume kinetic analysis.

The transcapillary filtration of fluid is governed by the Starling forces. The purpose of the present study was to simultaneously estimate all Starling parameters in living humans by using likelihood mathematics. First, the capillary filtration was estimated by volume kinetic analysis at 3,015 time points during 193 intravenous infusion experiments using crystalloid and colloid fluid in adult volunteers and surgical patients placed in the supine position. Second, the obtained capillary filtration and the measured plasma colloid osmotic pressure were entered into a new kinetic analysis to estim

Transient weakening of the blood-brain barrier in neonatal mice.

BACKGROUND: Tight junctions (TJs) are a major structural component of the blood-brain barrier (BBB), contributing to brain homeostasis by restricting paracellular diffusion. Although BBB maturation begins during embryogenesis, the timing and dynamics of functional barrier maturation perinatally remain unclear. This question is particularly relevant for the striatum, a metabolically demanding brain region that undergoes rapid postnatal maturation and is vulnerable to neonatal injury. METHODS: We investigated developmental dynamics of BBB tracer permeability in mouse striatum using in situ micr

Assessment of Angiogenic Properties of Malignant Pleural Effusion (MPE) Fluid.

Patients with various malignancies can develop malignant pleural effusion (MPE). MPE is the abnormal accumulation of fluid in the pleural cavity. This fluid usually contains immune cells, tumor cells, and high levels of plasma proteins. This condition is associated with impaired pleural fluid drainage and is considered an immune and angiogenic manifestation of tumors metastasized in the pleura from other thoracic and non-thoracic areas. Tumor-induced pleural vascular hyperpermeability and neoangiogenesis are crucial to pleural fluid accumulation and could be therapeutically targeted to minimiz

Secretome of brain microvascular endothelial cells promotes endothelial barrier tightness and protects against hypoxia-induced vascular leakage.

Cell-based therapeutic strategies have been proposed as an alternative for brain and blood vessels repair after stroke, but their clinical application is hampered by potential adverse effects. We therefore tested the hypothesis that secretome of these cells might be used instead to still focus on cell-based therapeutic strategies. We therefore characterized the composition and the effect of the secretome of brain microvascular endothelial cells (BMECs) on primary in vitro human models of angiogenesis and vascular barrier. Two different secretome batches produced in high scale (scHSP) were anal

High-Throughput Bioprinting Method for Modeling Vascular Permeability in Standard Six-well Plates with Size and Pattern Flexibility.

Vascular permeability is a key factor in developing therapies for disorders associated with compromised endothelium, such as endothelial dysfunction in coronary arteries and impaired function of the blood-brain barrier. Existing fabrication techniques do not adequately replicate the geometrical variation in vascular networks in the human body, which substantially influences disease progression; moreover, these techniques often involve multi-step fabrication procedures that hinder the high-throughput production necessary for pharmacological testing. This paper presents a bioprinting protocol fo

Neuropilin 1 Regulation of Vascular Permeability Signaling.

The vascular endothelium acts as a selective barrier to regulate macromolecule exchange between the blood and tissues. However, the integrity of the endothelium barrier is compromised in an array of pathological settings, including ischemic disease and cancer, which are the leading causes of death worldwide. The resulting vascular hyperpermeability to plasma molecules as well as leukocytes then leads to tissue damaging edema formation and inflammation. The vascular endothelial growth factor A (VEGFA) is a potent permeability factor, and therefore a desirable target for impeding vascular hyperp

Pharmacological interventions to reduce edema following cardiopulmonary bypass: A systematic review and meta-analysis.

PURPOSE: To compare the effectiveness of different types of pharmacological agents to reduce organ specific edema following cardiopulmonary bypass (CPB). METHODS: Pubmed, Embase.com and Cochrane were searched for studies administrating a pharmacological agent before CPB. Primary outcome was postoperative edema. RESULTS: Forty-four studies (clinical n = 6, preclinical n = 38) fulfilled eligibility criteria. Steroids were used in most clinical studies (n = 5, 83%) and reduced postoperative edema in 4 studies, however heterogeneity precluded meta-analysis. In preclinical studies, a total of 31

Adipose-derived stem cells attenuate pulmonary microvascular hyperpermeability after smoke inhalation.

BACKGROUND: Pulmonary edema is a hallmark of acute respiratory distress syndrome (ARDS). Smoke inhalation causes ARDS, thus significantly increasing the mortality of burn patients. Adipose-derived stem cells (ASCs) exert potent anti-inflammatory properties. The goal of the present study was to test the safety and ecfficacy of ASCs, in a well-characterized clinically relevant ovine model of ARDS. METHODS: Female sheep were surgically prepared. ARDS was induced by cooled cotton smoke inhalation. Following injury, sheep were ventilated, resuscitated with lactated Ringer's solution, and cardiopul

Intravascular staining for discrimination of vascular and tissue leukocytes.

Characterization of the cellular participants in tissue immune responses is crucial to understanding infection, cancer, autoimmunity, allergy, graft rejection and other immunological processes. Previous reports indicate that leukocytes in lung vasculature fail to be completely removed by perfusion. Several studies suggest that intravascular staining may discriminate between tissue-localized and blood-borne cells in the mouse lung. Here we outline a protocol for the validation and use of intravascular staining to define innate and adaptive immune cells in mice. We demonstrate application of thi
