The psychological impact of genetic testing in childhood cancer: A systematic review.

OBJECTIVE: Cancer predisposition syndromes are being more frequently recognized in the etiology of pediatric oncology and genetic-related technologies are evolving rapidly, leading to an increasing availability of genetic testing for families. This systematic review assessed the psychological impact of genetic testing on children and parents in the context of childhood cancer. METHODS: Searches were performed using three databases (Web of Science, Pubmed and Embase) to identify relevant empirical studies. Following Cochrane guidelines, we screened 3838 articles and identified 18 eligible stud

Impact of presymptomatic genetic testing on young adults: a systematic review.

Presymptomatic and predictive genetic testing should involve a considered choice, which is particularly true when testing is undertaken in early adulthood. Young adults are at a key life stage as they may be developing a career, forming partnerships and potentially becoming parents: presymptomatic testing may affect many facets of their future lives. The aim of this integrative systematic review was to assess factors that influence young adults' or adolescents' choices to have a presymptomatic genetic test and the emotional impact of those choices. Peer-reviewed papers published between Januar

A combined data mining approach for infrequent events: analyzing HIV mutation changes based on treatment history.

Many biological databases contain a large number of variables, among which events of interest may be very infrequent. Using a single data mining method to analyze such databases may not find adequate predictors. The HIV Drug Resistance Database at Stanford University stores sequential HIV-1 genotype-test results on patients taking antiretroviral drugs. We have analyzed the infrequent event of gene mutation changes by combining three data mining methods. We first use association rule analysis to scan through the database and identify potentially interesting mutation patterns with relatively hig

[Studies on the association of genes with language disorders in the post-genomic era].

Since genetic research entered the post-genomic era, the high heritability of language disorders has been confirmed. A variety of genetic-related diseases may cause various types of language disorders in children and/or adults. This article has summarized language disorders and their underlying mechanisms by searching the Web of Science database over the last decade, and combed the genetic researches for dyslexia, frontotemporal degeneration, specific language disorder, childhood speech apraxia and other single diseases that are strongly associated with the language disorders. It also provided

Pierre Robin sequence: Subdivision, data, theories, and treatment - Part 2: Syndromic and nonsyndromic Pierre Robin sequence.

CONTEXT: The disorder currently accepted as Pierre Robin syndrome/anomaly/sequence (PRS) has been plagued by controversy ever since initially being described. Controversy exists not only about the appropriate terminology and etiopathogenesis of the disorder but also about its management. Clinical findings and treatment outcomes of a large database of 266 PRS cases were compared with the current state of knowledge in the scientific literature, relating to history, clinical description, diagnostic criteria, epidemiology, theories of oligohydramnios, mandibular catch-up growth, midfacial hyperpla

Genetic risk factors and causal associations with sudden cardiac arrest: A Mendelian randomization study.

Sudden cardiac arrest (SCA) is a major cause of mortality, with complex etiology involving multiple genetic and environmental factors. In this study, we aimed to identify genetic risk factors for SCA and evaluate their causal associations using Mendelian randomization (MR) analysis. We retrieved summarized genome-wide association study data for various exposures and the SCA outcome from public databases. Four MR methods, including inverse variance weighted (IVW), Egger, weighted median, and weighted mode, were applied to estimate causal effects. All MR methods revealed significant positive cau

A Homozygous Variant in DMRTB1 Is Associated With Non-Obstructive Azoospermia in Humans.

Non-obstructive azoospermia (NOA) represents the most severe form of male infertility, and the pathogenesis in partial patients can be attributed to endocrine dysfunction or heritable genetic variants. Although the application of whole-exome sequencing (WES) has facilitated the identification of numerous pathogenic genes associated with NOA, the genetic etiology of a significant proportion of cases remains elusive. In this study, we identified an NOA patient carrying a novel variant in DMRTB1 (c.792-797del, p.265-266del). The variant was rare in public databases and predicted to be likely path

Clinical and molecular characterization of Krabbe disease in Iranian patients: case report and literature review.

Krabbe disease (KD, OMIM #245200) is a rare autosomal recessive lysosomal storage disorder characterized by severe demyelination affecting both the central and peripheral nervous systems. Here we report the clinical and molecular findings of two unrelated Iranian patients with KD, originating from consanguineous families. Genetic analysis was initially performed using whole-exome sequencing (WES), then validated by Sanger sequencing. WES identified a novel homozygous variant in the GALC, c.836T > G (p.L279X), in patient-1. In patient-2, WES detected a previously reported homozygous variant, c.

NF-κB1 haploinsufficiency due to a novel c.104delT variant in two patients with common variable immunodeficiency.

Common variable immunodeficiency (CVID) is the most frequent clinically relevant inborn error of immunity. Genetic defects can be identified in one-third of cases. The identification of novel mutations remains crucial for understanding disease mechanisms and genotype-phenotype correlations. We report a 19-year-old female with recurrent infections, hepatomegaly, and lymphadenopathy that began one year before the assessment. Laboratory evaluation revealed neutropenia, lymphopenia, anemia, thrombocytopenia, hypogammaglobulinemia, and a reduced percentage of switched memory B cells. Next-generatio

[Case report of a cystic lung disease: From a rarity to the discovery of an unknown genetic variant].

INTRODUCTION: Cystic lung diseases are rare, with numerous differential diagnoses. Iconographic discovery consequently necessitates medical examinations in view of proposing an etiological orientation. CASE REPORT: A 57-year-old woman consulted in pulmonology following fortuitous detection of a cystic lung disease on an abdominal CT scan. Complementary medical examinations did not allow orientation towards a particular diagnosis. During a follow-up consultation, the patient informed her pulmonologist of the recent detection of a monoallelic variant of a FAT4 gene in one of her daughters, who

Potential role of fibroblast senescence in malignant transformation of oral submucous fibrosis.

Oral Submucous Fibrosis (OSMF) is a disorder that occurs primarily due to the usage of betel nut and paan masala. The rate of malignant transformation from OSMF is 26 %. There are several markers to predict this malignant transformation such as inflammatory mediators, hypoxia, Cell cycle alteration, alteration in oncosuppressor genes, angiogenesis-related molecules, genetic susceptibility, and senescence. In carcinogenesis, multiple mutations occur leading to genetic damage. This affects the normal cell cycle and DNA repair. One of the reasons for the genetic damage and mutation to result is b

Meta-analysis of human leukocyte antigen genetic polymorphisms and susceptibility to chronic myelogenous leukemia in Chinese population.

Human leukocyte antigen (HLA) genetic polymorphisms are assumed to be correlated to the risk of chronic myelogenous leukemia (CML) in various ethnicities. Up to now, no clear consensus has been reached. Our goal is to address this issue in Chinese population. By searching the data in PubMed, Embase and four Chinese databases (prior to July 2010), the association of HLA genetic polymorphisms with CML has been fixed as the research objective. We studied a totality of 12 studies, comprising 2281 CML cases and 41000 health controls. The data demonstrated that HLA-A*11, A*74, HLA-B*40, B*47, B*55 a

When brachytherapy met genetic oncology. Can radiation oncologists improve the detection of hereditary non-polyposis colorectal cancer?

Between January 1994 and December 2004, 696 patients with localized endometrial carcinoma have been treated at the Institute Jean-Godinot. Patients were selected on the following criteria: histologically proven adenocarcinoma of the endometrium; age at onset under 60 years; patient not deceased at the time of the study. One hundred twelve patients met these criteria and received a mailed specific questionnaire to establish their pedigree. Thirty-one patients (35.5%) were eventually found eligible for a genetic counselling but only 13 patients agreed to be informed later on. According to the ob

The Roles of Genetic and Epigenetic Abnormalities in Essential Thrombocythemia.

Essential thrombocythemia (ET) is a Ph(-) myeloproliferative neoplasm characterized by elevated platelet counts. Patients most often have one of the three following driver mutations: JAK2, MPL, or CALR. However, these mutations are not detected in approximately 10-15% of patients, who are thus referred to as "triple-negative" (TN). Background/Objectives: In recent years, there have been breakthroughs in the genetic diagnosis of hematological diseases and progress in understanding their epigenetic basis. Aim: This study aimed to review the available literature and systematize the current knowle

The Influence of Pharmacogenetic Factors on the Pharmacokinetics of Morphine and Its Metabolites in Pediatric Patients: A Systematic Review.

Morphine is a potent analgesic used for treating surgical and cancer pain. Despite being the drug of choice for the management of severe pain in children, the high interindividual variability in morphine pharmacokinetics limits its clinical utility to effectively relieve pain without adverse effects. This review was conducted to identify and describe all studies that have assessed the effect of genetic factors on the pharmacokinetics of morphine and its main metabolites in children. Embase and Medline databases were used to conduct the literature search, and the systematic review was conducted

Socioeconomic position, genetic susceptibility, and epigenetic profiles in obesity across the life course: a systematic review.

BACKGROUND: While genetic studies have identified hundreds of loci associated with adiposity, the phenotypic expression of this individual genetic risk is not fixed but dependent on environmental circumstances. Socioeconomic Position (SEP), which includes education, income, and area-level deprivation, serves as an important environmental modulator of genetic liability. However, the literature on Gene-Environment (G×E) interactions remains fragmented, particularly regarding how these evolve across the life course. AIM: Provide a comprehensive overview of available literature on how SEP factors
