[miRNA in epigenetics of rheumatic diseases].

Epigenetic modifications are heritable changes in genome function that occur without a change in DNA sequence. They turn genes on or off by small chemical modifications. Thereby, they change the structure of the DNA making them accessible or not for transcription factors or other DNA binding molecules. MicroRNAs are a new class of regulatory non-coding RNAs that modulate gene expression and influences epigenetic mechanisms. miRNAs offer novel mechanisms of therapeutic strategies for cancer and autoimmune diseases. Here, we summarize which miRNAs play a role in rheumatic diseases and their biol

Epigenetic dysregulation in follicular lymphoma.

The adoption of next-generation sequencing technologies has led to a remarkable shift in our understanding of the genetic landscape of follicular lymphoma. While the disease has been synonymous with the t(14;18), the prevalence of alterations in genes that regulate the epigenome has been established as a pivotal hallmark of these lymphomas. Giant strides are being made in unraveling the biological consequences of these alterations in tumorigenesis opening up new opportunities for directed therapies.

Maternal choline and prebiotic supplementation ameliorate PCOS traits and reduce intergenerational transmission in rats.

Polycystic ovary syndrome (PCOS) is a common endocrine disorder with strong heritability, driven by complex metabolic, epigenetic, and gut-microbial dysregulation. While both epigenetic and microbial axes are implicated in PCOS, their integration and contribution to intergenerational transmission remain poorly understood. We investigated whether maternal choline (methyl-donor) and galactooligosaccharide (GOS, prebiotic) supplementation could restore gut-epigenome homeostasis and prevent vertical transmission of PCOS traits in rats. PCOS was induced using dehydroepiandrosterone plus high-fat di

A Vegan Diet Epigenetically Modulates Inflammatory Pathways and Biological Aging: Genome-Wide DNA Methylation Analysis of a One-Month Isocaloric Vegan Versus Meat-Rich Dietary Intervention.

We explored the epigenomic effects of vegan diet (VD) versus meat-rich diet (MR) and identified mechanisms that help to explain how VD affects epigenetic gene regulation. Genome-wide DNA methylation profiles in 48 healthy individuals were investigated after a 1-month randomized isocaloric dietary intervention comparing effects of a VD versus a MR. Genome-wide DNA methylation analysis revealed changes in differentially methylated positions following dietary intervention, with the VD group showing a higher degree of gene-promoter silencing in cancer-related pathways and cell growth-associated pa

Advances and challenges of precision epigenetic therapy in treating genomic imprinting diseases.

Genomic imprinting disorders (IDs) arise from the disruption of parent-of-origin-specific gene expression, a process governed by heritable epigenetic marks. Conventional therapies are largely symptomatic and fail to correct the underlying defect in dynamic gene regulation. Epigenome editing, using programmable DNA-targeting tools to rewrite epigenetic information, has emerged as a powerful therapeutic strategy that directly addresses this molecular pathology. This review synthesizes the transformative progress of the past decade, arguing that epigenome editing is poised to revolutionize the tr

Genome-wide DNA methylation association study of recent and accumulated ozone exposure in the National Longitudinal Study of Adolescent to Adult Health (Add Health) Study.

Ground-level ozone exposure poses a significant public health threat, linked to respiratory problems, cardiovascular disease, and cognitive decline. Emerging evidence suggests that DNA methylation (DNAm) alterations, a form of epigenetic modification, may contribute to these health effects. Here, we conducted an epigenome-wide association study (EWAS) involving 4500 adults aged 32-42 years from diverse populations across the United States to explore how recent (past year) and accumulated (past 14 years) ozone exposure relate to DNAm patterns. We identified six CpGs that reached epigenome-wide

Dietary impacts on the human methylome - focus on vitamin intake in context of health and disease risk.

Diet is a key modifiable component of health, and a major risk factor for chronic disease in humans. At the molecular level diet can affect DNA methylation (DNAm), an epigenetic process that is involved in gene expression regulation. Multiple studies have linked DNAm profiles to changes in diet across different layers. Specifically, intake of nutrients such as folate and vitamin B12 has been associated with changes in human DNAm patterns genome-wide, linking dietary intakes to the potential modulation of gene activity. In this review we present a focused overview of recent findings from epigen

Epigenetic reprogramming in autoimmune and immune-mediated skin disease.

BACKGROUND: Autoimmune skin diseases exhibit chronicity and site-specific relapse that are not fully explained by genetic susceptibility or cytokine signaling alone. Emerging evidence suggests that epigenetic reprogramming of skin-resident immune cells contributes to disease persistence and recurrence. METHODS: We conducted a structured narrative review of the human literature by searching PubMed for studies published from database inception through March 1, 2026. The literature search was performed between February 1 and March 1, 2026 and focused on epigenetic mechanisms across major autoimm

Human brain organoids record the passage of time over multiple years.

The human brain develops and matures over an exceptionally prolonged period of time that spans nearly two decades of life. Processes that govern species-specific aspects of human postnatal brain development are difficult to study in animal models1. While human brain organoids offer a promising in vitro model, they have thus far been shown to largely mimic early stages of brain development. Here we develop human brain organoids for 5 years in culture, optimizing growth conditions to extend excitatory neuron viability beyond previous limits. Using maturation-associated modules derived from endog

Chromatin Organization Governs Transcriptional Response and Plasticity of Cancer Stem Cells.

Chromatin organization regulates transcription to influence cellular plasticity and cell fate. We explored whether chromatin nanoscale packing domains are involved in stemness and response to chemotherapy. Using an optical spectroscopic nanosensing technology we show that ovarian cancer-derived cancer stem cells (CSCs) display upregulation of nanoscale chromatin packing domains compared to non-CSCs. Cleavage under targets and tagmentation (CUT&Tag) sequencing with antibodies for repressive H3K27me3 and active H3K4me3 and H3K27ac marks mapped chromatin regions associated with differentially exp

Methylation variability and LINE-1 activation in multiple myeloma.

Multiple myeloma (MM) is a type of hematological cancer that arises from uncontrolled proliferation of plasma cells. In addition to frequent genetic mutations, malignant plasma cells are characterized by alterations to the epigenome. Myeloma cells display a genome-wide loss of DNA methylation and a corresponding increase in 'active' chromatin modifications. The dramatic epigenetic remodeling that occurs in cancer genomes is associated with loss of silencing at transposable elements, which can impact genome regulation. Through paired epigenome and transcriptome profiling of patient derived MM s

The clinical use of epigenetics in psychiatry: a narrative review of epigenetic mechanisms, key candidate genes, and precision psychiatry.

The etiology of psychiatric disorders is complex, involving both genetic and environmental factors with emerging evidence suggesting that epigenetic modifications, including DNA methylation, histone modifications, and non-coding RNA regulation, significantly contribute to mental health. The epigenome influences the development of psychiatric disorders and human behavior and may be considered in clinical observations. Epigenetic changes have been well-established in BDNF, COMT, FKBP5, NR3C1, SLC6A4, and DRD2, genes associated with psychiatric disorders, including schizophrenia, major depressive

Placental PFAS concentrations are associated with perturbations of placental DNA methylation.

The placenta is crucial for fetal development, is affected by PFAS toxicity, and evidence is accumulating that gestational PFAS perturb the epigenetic activity of the placenta. Gestational PFAS exposure can adversely affect offspring, yet individual and cumulative impacts of PFAS on the placental epigenome remain underexplored. Here, we conducted an epigenome-wide association study (EWAS) to examine the relationships between placental PFAS levels and DNA methylation in a cohort of mother-infant dyads in Arkansas (N = 151). Associations between placental hydroxymethylation and birthweight.

5-hydroxymethylcytosine (5hmC), formed through the ten-eleven translocation (TET) methylcytosine dioxygenase mediated oxidation of 5-methylcytosine (5mC) at cytosine-phosphate-guanine (CpG) dinucleotides, is believed to mainly serve as an intermediate in the DNA demethylation pathway, though recent evidence suggests that 5hmC may also play a functionally relevant role. We have conducted an epigenome-wide association study (EWAS) to assess the association between placenta 5hmC, obtained through parallel bisulfite and oxidative bisulfite modification of DNA and array-based assessment, and newbor

Comparison of CcrM-dependent methylation in Caulobacter crescentus and Brucella abortus by nanopore sequencing.

Bacteria rely on DNA methylation for restriction-modification systems and epigenetic control of gene expression. Here, we use direct detection of methylated bases by nanopore sequencing to monitor global DNA methylation in Alphaproteobacteria, where use of this technique has not yet been reported. One representative of this order, Caulobacter crescentus, relies on DNA methylation to control cell cycle progression, but it is unclear whether other members of this order, such as Brucella abortus, depend on the same systems. We addressed these questions by first measuring CcrM-dependent DNA methyl
