Interleukin-35 Inhibits TNF-α-Induced Osteoclastogenesis and Promotes Apoptosis via Shifting the Activation From TNF Receptor-Associated Death Domain (TRADD)-TRAF2 to TRADD-Fas-Associated Death Domain by JAK1/STAT1.

Over-activated osteoclasts derived from myeloid or peripheral blood monocytes by inflammatory cytokines results in osteoporosis, osteoarthritis, and other bone erosion-related diseases. Interleukin 35 (IL-35) is a novel anti-inflammatory and immunosuppressive factor. This study investigated the effect of IL-35 on TNF-α-induced osteoclastogenesis. In the presence of IL-35, this process was detected by Tartrate-Resistant Acid Phosphatase (TRAP) staining, F-actin staining, and bone resorption assays. The effects of IL-35 on TNF-α-induced apoptosis were demonstrated by TUNEL staining, cell viabili

Free flow electrophoresis separation and AMS quantitation of C-naphthalene-protein adducts.

Naphthalene is a volatile aromatic hydrocarbon to which humans are exposed from a variety of sources including mobile air sources and cigarette smoke. Naphthalene produces dose- (concentration) dependent injury to airway epithelial cells of murine lung which is observed at concentrations well below the current occupational exposure standard. Toxicity is dependent upon the cytochrome P450 mediated metabolic activation of the parent substrate to unstable metabolites which become bound covalently to tissue proteins. Nearly 70 proteins have been identified as forming adducts with reactive naphthal

Modeling and experimental analysis of CO2 methanation reaction using Ni/CeO2 monolithic catalyst.

In this study, the effect of the cell density of monolithic catalysts was investigated and further mathematically modeled on cordierite supports used in CO2 methanation. Commercial cordierite monoliths with 200, 400, and 500 cpsi cell densities were coated by immersion into an ethanolic suspension of Ni/CeO2 active phase. SEM-EDS analysis confirmed that, owing to the low porosity of cordierite (surface area < 1 m2 g-1), the Ni/CeO2 diffusion into the walls was limited, especially in the case of low and intermediate cell density monoliths; thus, active phase was predominantly loaded onto the ch

Injectable thermosensitive hydrogel incorporating exosome-loaded chitosan microspheres for immunomodulation and cartilage regeneration.

BACKGROUND: Chronic inflammation and impaired immune microenvironment are key barriers to effective cartilage regeneration. Herein, we developed an integrated, injectable thermosensitive hydrogel system termed CS@Gel-Mg. This system modularly combines exosome-loaded chitosan microspheres (CM@Exo) embedded within a Mg2+-coordinated poly(N-isopropylacrylamide)-based ionic liquid hydrogel (Gel-Mg), aiming to modulate local immune response and promote cartilage repair. METHODS: Hydroxypropyl-modified chitosan microspheres were synthesized via emulsion crosslinking and effectively loaded with bio

Reduction of tumor hypoxia by anti-PD-1 therapy assessed using pimonidazole and [18F]FMISO.

INTRODUCTION: Hypoxia is common in solid tumors and creates an immunosuppressive environment that leads to resistance to immunotherapy, such as an anti-programmed death receptor-1 (PD-1) therapy. It has been suggested that anti-PD-1 therapy may reduce tumor hypoxia by remodeling the tumor vasculature; however, it is unclear how anti-PD-1 therapy reduces hypoxia over time. Therefore, we investigated the relationship between hypoxia and immune activation by anti-PD-1 therapy in murine cancer models. METHODS: Anti-PD-1 antibody was injected to CT26- and MC38-tumor-bearing mice on days 0 and 5. T

Determination of Essential and Toxic Elements by ICP-MS in Herbal Medicines.

This review synthesizes applications of inductively coupled plasma-mass spectrometry (ICP-MS) and hyphenated techniques for elemental determination in herbal medicines. The herbal medicine analysis has become increasingly important due to their widespread use in traditional and modern therapies. Due to the elements naturally present or contaminants (toxic elements) in herbal medicines and daily mineral requirements at the microgram level, analytical methods capable of precisely determining elemental concentrations in herbal medicines are essential. Analyses of several species have been conduct

Enhanced selectivity and sensitivity of sequential injection chromatography: determination of bioflavonoids in nutraceuticals.

The gradual development of sequential injection chromatography (SIC) has aimed to increase separation selectivity and sensitivity. In this work, an HPLC column, a sub-2 μL flow cell detector, and advanced processing of UV-vis spectral data were used to determine the concentration of closely related analytes, such as diosmin and hesperidin, in nutraceuticals. The improvements resulted in narrower peaks, higher resolution, and more raw data acquired for effective noise reduction with the Savitzky-Golay filter. This was followed by an automated calculation of peak characteristics, which facilitat

The permeability of shale exposed to supercritical carbon dioxide.

Permeability is a critical parameter of tight reservoir rocks and one of the important parameters for characterizing fluid flow and production from reservoirs. It determines the feasibility of its commercial development. SC-CO2 has been used in shale gas exploitation for efficient fracturing and the added benefit of CO2 geo-storage. And SC-CO2 plays an important role in permeability evolution of shale gas reservoirs. In this paper, Firstly, the permeability characteristics of shale under CO2 injection are discussed. The experimental results show that the relationship between permeability and g

Arginase 1+ microglia reduce Aβ plaque deposition during IL-1β-dependent neuroinflammation.

BACKGROUND: Neuroinflammation has long been considered a driver of Alzheimer's disease progression. However, experiments developed to explore the interaction between neuroinflammation and Alzheimer's disease (AD) pathology showed a surprising reduction in amyloid beta (Aβ) plaque deposition. We sought to understand this unexpected outcome by examining microglia phenotypes during chronic neuroinflammation. METHODS: Using an adeno-associated virus vector carrying hIL-1β cDNA, inflammation was induced in one hippocampus of 8-month-old amyloid precursor protein (APP)/PS1 mice for 4 weeks, while t

Macrophage depletion in inflamed rat knees prevents the activation of synovial mesenchymal stem cells by weakening Nampt and Spp1 signaling.

BACKGROUND: Macrophages and mesenchymal stem cells (MSCs) engage in crucial interplay during inflammation and have significant roles in tissue regeneration. Synovial MSCs, as key players in joint regeneration, are known to proliferate together with macrophages in synovitis. However, the crosstalk between synovial MSCs and macrophages remains unclear. In this study, we investigated changes in the activation of synovial MSCs in inflamed rat knees following selective depletion of macrophages with clodronate liposomes. METHODS: Acute inflammation was induced in rat knee joints by injection of car

Report of the Radionuclides in Nephrourology Committee for evaluation of transplanted kidney (review of techniques).

Comprehensive evaluation of renal transplants has been important in differential diagnosis of medical and surgical complications in the early post-transplantation period and in the long-term follow-up. If performed well, it yields excellent functional and good anatomic information about the graft that can be effectively used in the patient. That includes selection of patients for biopsy and for various drug regimens. This is true especially in patients with anuric acute tubular necrosis (ATN) and in patients with developing chronic rejection. Improving indices of renal function (effective rena

Postoperative control of functional muscle flaps for facial palsy reconstruction: Ultrasound guided tissue monitoring using contrast enhanced ultrasound (CEUS) and ultrasound elastography.

BACKGROUND AND OBJECTIVES: Facial paralysis causes excruciating impairments including facial asymmetry, limited eye closure, oral incontinence and social dysfunction. Modern plastic surgical reconstructions render favorable results with well-perfused dynamic muscle flaps. Post-operative tissue monitoring is a critical determinant for success. Contrast-enhanced Ultrasound (CEUS) and elastography have proven superior properties to evaluate tissue perfusion in various organs. We evaluated their role for functional muscle flaps positioned at the sub-skin level in facial palsy patients. METHODS: F

Expression of intercellular adhesion molecule 1 (ICAM-1) is reduced in permanent focal cerebral ischemic mouse brain using an adenoviral vector to induce overexpression of interleukin-1 receptor antagonist.

Our previous studies have demonstrated that overexpression of recombinant human interleukin-1 receptor antagonist protein (IL-1ra) via gene transfer can reduce ischemic brain injury. However, the mechanism of action of IL-1ra in ischemia is unclear. Since interleukin-1 can up-regulate intercellular adhesion molecules in endothelium, the present study was designed to determine whether overexpression of the IL-1ra can reduce the expression of intercellular adhesion molecule-1 (ICAM-1) after ischemic injury. Normal saline or adenovirus vector (1x109 particles) encoding the human IL-1ra gene (Ad.R

Phenanthrene Exposure Induces Immune Dysfunction and Metabolic Disruption in Mice.

Polycyclic aromatic hydrocarbons (PAHs) are widespread environmental pollutants that may impair immune function. However, the immunotoxic mechanisms of PAH exposure remain largely unexplored. The present study was designed as a mechanistic, effect-oriented investigation, in which male C57BL/6J mice were exposed to phenanthrene via intraperitoneal injection for six consecutive weeks. We assessed toxicological effects of phenanthrene exposure in mice through urinary metabolite profiling, hematological and immunological tests, and serum metabolomics. Urinary concentrations of phenanthrene metabol
