Assessment of Sorbitol as a Pharmaceutical Excipient in Oral Liquid Pediatric Formulations Submitted to the U.S. FDA.

Sorbitol is a commonly used excipient in pediatric oral liquid formulations. Previous research revealed that sorbitol as an excipient can diminish the bioavailability of concomitant drugs. The reported cases and amount of sorbitol in pediatric formulations has not been explored, thus the aims of this study were to (1) examine the number of reports of sorbitol as an excipient in orally administered drugs with liquid formulations used in pediatrics, and (2) estimate the maximum amount of sorbitol that may be consumed per day for each drug. A compilation of drug products containing sorbitol as an

Development and Characterization of Cannabidiol Self-Emulsifying Drug Delivery System: In Vitro and In Vivo Evaluation.

Cannabidiol (CBD) is a non-psychoactive phyto-cannabinoid with numerous pharmacological potentials. CBD is a lipophilic drug with poor and varied bioavailability due to its low water solubility and extensive first-pass metabolism, and it is highly affected by the presence of food. A self-emulsifying drug delivery system (SEDDS) was developed to improve the aqueous solubility and oral bioavailability of CBD. The formulation strategy involved incorporating excipients that maintain drug solubility under both fasted and fed conditions, while potentially mitigating first-pass metabolism to enhance

Rheological and Technological Design of Texture-Modified Food Systems for Dysphagia Management in Amyotrophic Lateral Sclerosis.

Amyotrophic lateral sclerosis (ALS) is frequently accompanied by progressive dysphagia, weight loss, and hypermetabolism, which complicate the maintenance of adequate nutritional status. For these patients, food systems must not only provide sufficient energy and protein within a limited volume but also exhibit textural and rheological properties that support safe swallowing. This review analyzes technological and rheological approaches to the design of texture-modified food systems for nutritional support in ALS. Particular attention is given to apparent viscosity, shear-thinning behavior, yi

Cascade-triggered oral gelatin-alginate micro-nano carriers integrate dual targeting and microbiota restoration for anti-Salmonella therapy.

Foodborne pathogens such as Salmonella enterica serovar Typhimurium remain a major threat to public health and food safety. Conventional antibiotic therapy is often ineffective due to intracellular persistence and biofilm protection. We developed a cascade-responsive oral micro-nano carrier (EQM@MS) that combines the food-derived flavonoid quercetin and enrofloxacin within dual-targeted micelles encapsulated by gelatin-alginate microspheres. The system provides gastric protection, enzyme-triggered intestinal release, and ligand-mediated targeting of epithelial cells and macrophages. EQM@MS ach

Development and characterization of a carvacrol nanoemulsion and evaluation of its antimicrobial activity against selected food-related pathogens.

Carvacrol has been recognized as an efficient growth inhibitor of food pathogens. However, carvacrol oil is poorly water-soluble and can be oxidized, decomposed or evaporated when exposed to the air, light, or heat. To overcome these limitations, a carvacrol nanoemulsion was developed and its antimicrobial activity against food pathogens evaluated in this study. The nanoemulsion containing 3% carvacrol oil, 9% surfactants (HLB 11) and 88% water, presented good stability over a period of 90 days. In general, the carvacrol nanoemulsion (MIC: 256 µg ml-1 for E. coli and Salmonella spp., 128 µg ml

Adjuvant-driven modulation of epitope recognition and protective immunity in Bm86 vaccinated Holstein-Friesian cattle.

Livestock production is vital to the economies and food security of African countries. Rising global demand for livestock-derived products intensifies the challenge of managing ticks and tick-borne diseases. This study aimed to optimize a Bm86-based vaccine for controlling Rhipicephalus microplus. Commercial Bm86-based vaccines show variable efficacy (0-100 %), reflecting incomplete understanding of the antigen and the immune response elicited. To address this, homologous challenge was conducted in Holstein-Friesian calves with Bm86 formulated with Montanide™ ISA 71 VG (referred to as Montanid

Differential activation of chemically identified neurons in the caudal nucleus of the solitary tract in non-entrained rats after intake of satiating vs. non-satiating meals.

Satiety signals arising from the gastrointestinal (GI) tract and related digestive organs during food ingestion and digestion are conveyed by vagal sensory afferents to the hindbrain nucleus of the solitary tract (NST). Two intermingled but chemically distinct NST neuronal populations have been implicated in meal size control: noradrenergic (NA) neurons that comprise the A2 cell group, and glucagon-like peptide-1 (GLP-1)-positive neurons. Previous results indicate that A2 neurons are activated in a meal size-dependent manner in rats that have been acclimated/entrained to a feeding schedule in

Effect of bowel preparation and a fiber-free liquid diet on expression of transforming growth factor and procollagen in colonic tissue preoperatively and postoperatively.

PURPOSE: Dehiscence of colonic anastomoses is prevalent and potentially fatal. In an attempt to reduce the likelihood of anastomotic dehiscence, the colon is cleansed before surgery and fiber-free diets are prescribed postoperatively. However, fiber-free diets induce colonic atrophy and impair healing. This study was designed to investigate the effect of bowel preparation and postoperative fiber-free diet on the local gene expression of transforming growth factor-beta 1 and procollagen type I. METHODS: Four Sprague-Dawley rats underwent bowel preparation with a fiber-free liquid diet and poly

Bioavailability and Health Impact of Ingested Amyloid-like Protein Fibrils and their Link with Inflammatory Status: A Need for More Research?

The use of amyloid-like protein fibrils (ALFs) in food formulations looks very promising in terms of improving techno-functional properties, but raises some concerns in terms of food safety, because of their structural resemblance to disease-related endogenous amyloids. This review focuses on the biological fate and potential health implications of ingested ALF structures in both healthy and predisposed individuals. A comprehensive overview of ALF gastrointestinal digestion, intestinal absorption, and systemic dissemination is provided, in addition to a thorough assessment of potential ALF cro

Polylactide/Montmorillonite Hybrid Latex as a Barrier Coating for Paper Applications.

We developed a paper coating for the potential application in food packaging based on polylactide and montmorillonite. It is applied to the paper in the form of a stable, water-based latex with a solid content of 25⁻28 wt %. The latex is prepared from a commercially available polylactide, surfactants, montmorillonite, a plasticizer, chloroform (to be removed later) and water by an emulsion/solvent evaporation procedure. This coating formulation is applied to the paper substrate by bar-coating, followed by hot-pressing at 150 °C. The coated papers achieved up to an 85% improvement in water vapo

Development and Validation of an Evaporative, Low-Flammability, and Low-Moisture Antimicrobial Liquid for Dry Cleaning and Sanitization of Food Processing Equipment.

Environmentally persistent organisms such as Salmonella spp. and Cronobacter spp. have caused several high-profile outbreaks associated with the consumption of low-moisture foods. Sanitary operations in dry food-processing environments are challenging as excess moisture elevates the risk of microbial harborage and transfer. In this work, the evaporation kinetics of selected polar and nonpolar solvents was characterized with dynamic vapor sorption. A low-moisture, volatile antimicrobial liquid was formulated using 3% v/v water-in-cyclomethicone emulsion as the carrier for 200 mM acetic acid, wh

DaxibotulinumtoxinA-lanm (Daxxify™): A Comprehensive Overview.

Botulinum toxin A (BoNTA) is produced by Clostridium botulinum and widely used for aesthetic indications requiring neuromuscular blockade. For dynamic facial lines, BoNTA is effective and safe, but also temporary, requiring repeat injections approximately every 3-4 months for maintenance of effects. There is a desire by both patients and providers for a longer-lasting neurotoxin to prevent periods of suboptimal correction. Approved by the US Food and Drug Administration (FDA) in September 2022, daxibotulinumtoxinA for injection (DAXI or Daxxify™) is the first long-lasting BoNTA formulated with

Impact of intervention strategies on listeriosis risk in cooked ham: Biopreservation and high pressure processing.

Biopreservation and post-lethality treatments (PLT) are the basis of operating procedures applied to reduce the levels of Listeria monocytogenes in ready-to-eat food. The combination of these strategies must be wisely selected, and their interaction should be considered in quantitative microbial risk assessments (QMRA) assessing the listeriosis risk. The aim of the present study was to develop a QMRA model to assess the impact of intervention strategies, including formulation with lactate, bioprotective cultures and the use of high-pressure processing (HPP), on reducing the listeriosis risk as
