Four-stranded G4 DNA Formation During Synthesis Phase of the Cell Cycle Protects Human Cells from Radiation Induced DNA Breaks.

The breaks induced during exposure to ionizing radiation are generally random. However, some studies show differences in radioprotection among the species within the same genus due to factors like DNA repair efficiency, genome complexity, chromatin organization, etc. A recent study linked the formation of G4 structures to differential radiosensitivity of the human genome. The present study investigates whether radiation-induced DNA damage varies across different phases of the cell cycle and explores the molecular basis for this. Neutral comet and immunofluorescence assays demonstrated that cel

The design and synthesis of selective and potent selenium-containing KRASG12D inhibitors.

KRASG12D is the most common KRAS mutation and a promising therapeutic target for various type cancers, particularly pancreatic cancer. In th … R)-5a suppressed KRAS downstream signaling, including ERK/MAPK pathway and induced apoptosis and G0/G1 phase arrest in a dose-dependent manner. In addition, compound (R)-5a has good pharmacokinetic properties compared to MRTX1133. These findings demonstrate (R)-5a as a promising lead compound for the development of KRASG12D selective inhibitor.

Theobromine as a molecular switch: targeting Nrf2 pathway in invasive and non-invasive bladder cancer.

Naturally occurring compounds like Theobromine, found in tea, coffee, and cocoa, may have anticancer effects. This study explores how Theobromine induces cell death and apoptosis in bladder cancer cells and its impact on NRF2-related gene activity. Two human bladder cancer cell lines-RT112 (non-invasive) and HTB9 (invasive)-were employed. Cell viability was measured using the WST-1 assay, whereas apoptosis and caspase 3/7 activation were determined through flow cytometry. qPCR was performed to measure the expression of NRF2, KEAP1, and P62 genes. Theobromine exhibited a dose-dependent reductio

Melatonin sensitizes human breast cancer cells to ionizing radiation by downregulating proteins involved in double-strand DNA break repair.

Radiation and adjuvant endocrine therapy are nowadays considered a standard treatment option after surgery in breast cancer. Melatonin exert … ed G2 -M phase arrest compared with irradiation alone, with a higher percentage of cells in the G0 -G1 phase and a lower percentage of cells in S phase. Radiation alone diminished RAD51 and DNA-protein kinase (PKcs) mRNA expression, two main proteins involved in double-strand DNA break repair. Treatment with melatonin for 7 days before radiation led to a s …

The farnesyl transferase inhibitor RPR-130401 does not alter radiation susceptibility in human tumor cells with a K-Ras mutation in spite of large changes in ploidy and lamin B distribution.

BACKGROUND: Growth inhibition by RPR-130401, a non-peptidomimetic farnesyltransferase inhibitor, was investigated without or with combined exposure to ionizing radiation in three human tumor cell lines (HCT-116, MiAPaCa-2 and A-549) bearing a point mutation in the K-Ras gene. RESULTS: RPR-130401 inhibited cell growth with an IC50 of 50 nM (HCT-116), 120 nM (MiAPaCa-2) and 710 nM (A-549), with a poor incidence of apoptosis. The drug brought about G1 and S phase depletion together with arrest of cells in G2 phase and induced a significant accumulation of hyperploid cells showing active S phase

Abrogation by novobiocin of cytotoxicity due to the topoisomerase II inhibitor amsacrine in Chinese hamster cells.

Using cultured V79 Chinese hamster cells, we found that novobiocin (or 2,4-dinitrophenol) can abrogate, almost completely, the cytotoxicity due to the topoisomerase II inhibitor amsacrine (mAMSA). V79 cells were sensitive to mAMSA killing at all stages in the cell cycle but mainly in S phase followed by late G1 phase; however, novo rescued cells of all ages. The properties of two kinds of radiation-sensitive Chinese hamster cells were also examined, i.e., the line of V79 cells that can be rescued by caffeine, designated S-10 (H. Utsumi and M.M. Elkind, Radiat. Res., 96: 348-358, 1983); and Chi…

Opposing effects of the UV lesion repair protein XPA and UV bypass polymerase eta on ATR checkpoint signaling.

An essential component of the ATR (ataxia telangiectasia-mutated and Rad3-related)-activating structure is single-stranded DNA. It has been suggested that nucleotide excision repair (NER) can lead to activation of ATR by generating such a signal, and in yeast, DNA damage processing through the NER pathway is necessary for checkpoint activation during G1. We show here that ultraviolet (UV) radiation-induced ATR signaling is compromised in XPA-deficient human cells during S phase, as shown by defects in ATRIP (ATR-interacting protein) translocation to sites of UV damage, UV-induced phosphorylati

CCNE1: A Cell Cycle Regulator That Influences Tumor Progression.

Cyclin E1 (CCNE1), a pivotal member of the cyclin family, governs the G1/S phase transition of the cell cycle by binding to and activating cyclin-dependent kinase 2 (CDK2), thereby initiating DNA replication and driving S-phase entry. In a broad spectrum of human malignancies-including breast, ovarian, gastric, and non-small cell lung cancers-amplification or overexpression of CCNE1 disrupts the orderly regulation of cell cycle progression, leading to uncontrolled proliferation, impaired DNA damage repair, and genomic instability. These oncogenic consequences are intimately associated with inc

Risankizumab in Adults with Psoriatic Arthritis.

Psoriatic arthritis (PsA) is a chronic inflammatory musculoskeletal disease associated with psoriasis. Its major clinical domains include peripheral and axial arthritis, enthesitis, dactylitis and skin and nail involvement. Approximately 30% of patients with psoriasis develop psoriatic arthritis. The pathophysiology of PsA is complex and involves a dysregulated immune response. In particular, interleukin (IL)-23 is a major regulatory cytokine that has been implicated in PsA, including bone remodeling, enthesitis, synovitis and psoriatic lesions. Risankizumab is a humanized immunoglobulin G1 mo

CCND1 polymorphism and age of onset of hepatoblastoma.

Cyclin D1, encoded by the gene CCND1, is a major regulator of the cell cycle transition from G1 phase to S phase. A CCND1 polymorphism (G to A) at codon 242, the boundary of exon 4 and intron 4, affects splicing such that exon 5 is not expressed in the A allele. Since exon 5 is involved in rapid turnover, the variant cyclin D1 corresponding to the A allele may have a longer half-life. A previous study demonstrated that in families with hereditary nonpolyposis colorectal cancer, the age of onset of colorectal cancer varied according to variation at this polymorphic site. We examined this CCND1p…

Mcm2 deficiency results in short deletions allowing high resolution identification of genes contributing to lymphoblastic lymphoma.

Mini-chromosome maintenance (Mcm) proteins are part of the replication-licensing complex that is loaded onto chromatin during the G1-phase of the cell cycle and required for initiation of DNA replication in the subsequent S-phase. Mcm proteins are typically loaded in excess of the number of locations that are used during S-phase. Nonetheless, partial depletion of Mcm proteins leads to cancers and stem cell deficiencies. Mcm2 deficient mice, on a 129Sv genetic background, display a high rate of thymic lymphoblastic lymphoma. Here array comparative genomic hybridization is used to characterize t

Catestatin alleviates PASMC phenotypic switching-mediated pulmonary arterial remodeling in a rat model of MCT-induced pulmonary arterial hypertension by promoting endothelium-derived no synthesis.

BACKGROUND: Enhancing the nitric oxide (NO) signaling pathway is an effective strategy for treating pulmonary arterial hypertension (PAH). P … xogenous NO effectively inhibited rPASMC phenotypic switching by arresting the cell cycle at the G0/G1 phase. CONCLUSION: CST enhances endothelial NO synthesis through the PI3K/Akt/eNOS pathway, which subsequently acts on rPASMCs to inhibit their phenotypic switching via the NO/cGMP/PKG signaling pathway, thereby alleviating pulmonary arterial remodeling …

Periplocin inhibits growth of lung cancer in vitro and in vivo by blocking AKT/ERK signaling pathways.

Periplocin is one of cardenolides isolated from cortex periplocae which is used for treatment of rheumatoid arthritis and reinforcement of bones and tendons in traditional medicine. Here, we investigated the anti-tumor activity of periplocin against lung cancer cells bothin vitro and in vivo, and explored its anti-cancer mechanism. Periplocin inhibited the growth of lung cancer cells and induced their apoptosis in time- and dose-dependent manners by cell cycle arrest in G0/G1 phase. Periplocin exhibited anti-tumor activity both in human (A549) and mouse (LL/2) lung cancer xenograft models. Imm…

G1/S transcriptional networks modulated by the HOX11/TLX1 oncogene of T-cell acute lymphoblastic leukemia.

The HOX11/TLX1 homeobox gene is aberrantly expressed in a subset of T-cell acute lymphoblastic leukemia (T-ALL). Here, we employed oligonucleotide microarrays to compare the expression profiles of the K3P and Sil leukemic cell lines originating from patients with HOX11+ T-ALL to that of Jurkat cells, which originated from a distinct subtype of T-ALL (TAL1+). To distinguish potential HOX11 target genes from those characteristic of the stage of HOX11 leukemic arrest, we also performed gene expression analysis on Jurkat cells, genetically engineered to express exogenous HOX11. The resulting HOX11
