Plants actively suppress growth and development upon activation of immunity. In turn, when pathogen attack has subsided, immune responses are suppressed again. Phytohormones play an important role in regulating this balance and the growth-immunity trade-off in general. The trade-off is evident in the Arabidopsis dmr6 dlo1 mutant, which accumulates the immune-activating phytohormone salicylic acid (SA) to high levels, resulting in high disease resistance but repression of growth. Little is known about the SA-induced growth trade-off mechanism. In this study, we performed a genetic suppressor sc

MdBZR1 directly binds to the promoter of MdABI5 and suppresses its expression to mediate ABA response. The plant hormones brassinosteroids (BRs) and abscisic acid (ABA) antagonistically regulate various aspects of plant growth and development. However, the association between BR and ABA signaling is less clear. Here, we identified MdBZR1 in apple (Malus domestica) and demonstrated that it was activated by BRs and could respond to ABA treatment. Overexpression of MdBZR1 in apple calli and Arabidopsis reduced ABA-hypersensitive phenotypes, suggesting that MdBZR1 negatively regulates ABA signalin

BACKGROUND: Plants have inducible defenses to combat attacking organisms. Hence, some herbivores have adapted to suppress these defenses. Suppression of plant defenses has been shown to benefit herbivores by boosting their growth and reproductive performance.
RESULTS: We observed in field-grown tomatoes that spider mites (Tetranychus urticae) establish larger colonies on plants already infested with the tomato russet mite (Aculops lycopersici). Using laboratory assays, we observed that spider mites have a much higher reproductive performance on russet mite-infested plants, similar to their pe

MicroRNA159/160 (miR159/160) has been implicated in plant development and stress resistance in annual crops and Arabidopsis, yet their functional roles in perennial cool-season forage and turf grass remain elusive. Tall fescue (Festuca arundinacea Schreb.) is one main perennial cool-season forage and turfgrass species worldwide. Here, we found that Fa-miR159/160 has 7/4 copies in tall fescue, respectively. Knockdown of both Fa-miR159 and Fa-miR160 (STTM159/160) using the short tandem target mimics (STTM) system resulted in a dwarf phenotype with higher tiller density, lower growth rate levels,

3-Hydroxy-3-methylglutaryl coenzyme A reductase (HMGR) serves as a key rate-limiting enzyme in the mevalonate pathway and plays a central regulatory role in the biosynthesis of tanshinones. To date, four HMGR family members (SmHMGR1-4) have been identified in Salvia miltiorrhiza. Here, we cloned and identified a novel member, SmHMGR5, by integrating multiple genomic datasets. Genomically, SmHMGR5 formed an inverted repeat with SmHMGR3 (98.04% homology) and phylogenetically clustered with SmHMGR2. Based on the expression patterns of the five HMGR genes, we further generated SmHMGR2 and SmHMGR5

Extensive laboratory experimentation has revealed conserved molecular pathways controlling growth and stress responses in plants, yet how these programs operate in natural settings remains poorly understood. We have investigated the organization of the transcriptome in wild populations of Arabidopsis thaliana by sampling plants across 60 natural sites in Europe and North America over two seasons. Transcriptomes varied extensively among individuals and were only weakly structured by geography or season. No groups of plants stood out due to clear biotic or abiotic stress signatures known from th

Paspalum distichum L. was tested to evaluate its ability to phytoremediate mercury (Hg) contaminated soil over a 60-d period by analysis of the total Hg concentrations in roots and leaves. Hg concentration in Hg-contamination soil decreased by 70.0 μg g-1 after 60 day of grass cultivation and Hg was readily taken up by the roots (4.51 ± 1.90 μg g-1) rather than the leaves (0.35 ± 0.02 μg g-1). In addition, a comparative proteomic study was performed to unravel the protein expression involved in the Hg stress response in P. distichum L. A total of 49 proteins were classified as differentially p

Elicitor-induced sanguinarine accumulation in opium poppy (Papaver somniferum) cell cultures provides a responsive model system to profile modulations in gene transcripts and metabolites related to alkaloid biosynthesis. An annotated expressed sequence tag (EST) database was assembled from 10,224 random clones isolated from an elicitor-treated opium poppy cell culture cDNA library. The most abundant ESTs encoded defense proteins, and enzymes involved in alkaloid metabolism and S-adenosylmethionine-dependent methyl transfer. ESTs corresponding to 40 enzymes involved in the conversion of sucrose

Artemisia annua L. is a medicinal plant valued for its ability to produce artemisinin, a molecule used to treat malaria. Plant nutrients, especially phosphorus (P), can potentially influence plant biomass and secondary metabolite production. Our work aimed to explore the genetic and metabolic response of A. annua to hardly soluble aluminum phosphate (AlPO4, AlP), using soluble monopotassium phosphate (KH2PO4, KP) as a control. Liquid chromatography-mass spectrometry (LC-MS) was used to analyze artemisinin. RNA sequencing, gene ontology (GO), and the Kyoto Encyclopedia of Genes and Genomes (KEG

Expression of OXIDATIVE SIGNAL-INDUCIBLE1 (OXI1) is induced by a number of stress conditions and regulates the interaction of plants with pathogenic and beneficial microbes. In this work, we generated Arabidopsis OXI1 knockout and genomic OXI1 overexpression lines and show by transcriptome, proteome, and metabolome analysis that OXI1 triggers ALD1, SARD4, and FMO1 expressions to promote the biosynthesis of pipecolic acid (Pip) and N-hydroxypipecolic acid (NHP). OXI1 contributes to enhanced immunity by induced SA biosynthesis via CBP60g-induced expression of SID2 and camalexin accumulation via

Sclerotinia sclerotiorum (Lib.) de Bary is a necrotrophic fungal pathogen that causes a disease known as white mold, which is a major problem for dry bean (Phaseolus vulgaris L.) and other crops in many growing areas in Brazil. To investigate the role of methyl jasmonate (MeJA) in defending dry bean plants against S. sclerotiorum, we used suppression subtractive hybridization (SSH) of cDNA and identified genes that are differentially expressed during plant-pathogen interactions after treatment. Exogenous MeJA application enhanced resistance to the pathogen, and SSH analyses led to the identifi

Pepper is an important vegetable and economic crop, and the MYB family is one of the most numerous transcription factor families in plants, extensively participating in various biological processes such as plant growth, development, and stress resistance. In this study, CcMYB330 is identified as a differentially expressed gene in the pepper fruit, and CcMYB330 is expressed with higher expression levels in the placenta and pericarp at different development stages of pepper fruit. Analysis of the promoter cis-elements revealed that this gene contains not only core elements but also environmental

BACKGROUND: The regulatory network governing triacylglycerol (TAG) and fatty acid (FA) accumulation in hexaploid Camellia oleifera kernels remains unclear due to the lack of an appropriate reference genome. In this study, we combined oil trait measurements with multi-omics analyses (global transcriptomics, miRNA sequencing, proteomics, and metabolomics) across 3 harvest stages, using the hexaploid C. oleifera 'Changlin 40' genome as reference.
RESULTS: Integrated analysis of oil traits and multi-omics data demonstrated that premature harvesting (1 week before the recommended date) decreased o

OBJECTIVE: Peach brachytic dwarfism determined by Dwarf gene (Dw) is an undesired trait segregating in some peach breeding programs. Recently, a single nucleotide polymorphism (SNP) mutation in the gibberellin insensitive dwarf 1 (GID1) peach gene causing brachytic dwarfism was described. In this research we wanted to validate this marker in an F2 population of the 'Nectavantop' peach cultivar (Nv) to include it as a marker assisted selection tool for peach breeding programs.
RESULTS: The observed segregation of the trait was in agreement with that of a recessive gene, the individuals homozyg

Cucumber, a very important vegetable crop worldwide, is easily damaged by pests. Aphids (Aphis gossypii Glover) are among the most serious pests in cucumber production and often cause severe loss of yield and make fruit quality get worse. Identifying genes that render cucumbers resistant to aphid-induced damage and breeding aphid-resistant cucumber varieties have become the most promising control strategies. In this study, a Illumina Genome Analyzer platform was applied to monitor changes in gene expression in the whole genome of the cucumber cultivar 'EP6392' which is resistant to aphids. Nin

NPR1 is required for systemic acquired resistance, and there are five NPR1 paralogs in Arabidopsis. Here we report knockout analysis of two of these, NPR3 and NPR4. npr3 single mutants have elevated basal PR-1 expression and the npr3 npr4 double mutant shows even higher expression. The double mutant plants also display enhanced resistance against virulent bacterial and oomycete pathogens. This enhanced disease resistance is partially dependent on NPR1, can be in part complemented by either wild-type NPR3 or NPR4, and is not associated with an elevated level of salicylic acid. NPR3 and NPR4 int

Increasing attention has been focused on the health of vegetables and grains grown in the contaminated agricultural soil, it is thus meaningful to find ways to reduce the heavy metals (HMs) accumulation in plants. As sulfur is considered to be an essential macronutrient for plant stress defenses, the important role of sulfur assimilation in plants responding to HMs stress has been followed. However, the potential mechanism of the only sulfur-containing gasotransmitter hydrogen sulfide (H2S) and its main endogenously generated substrate, cysteine (Cys), in plant defense is poorly understood. Branching architecture is a key agronomic and ornamental trait in woody plants; however, the molecular mechanisms coordinating hormone-mediated axillary bud formation and outgrowth remain largely unclear. Clerodendrum japonicum (Thunb.) Sweet is an important ornamental woody species with strong branching potential, but the regulatory network underlying its shoot branching has not been characterized. In this study, we investigated the regulatory basis of hormone-induced branching in Clerodendrum japonicum and identified a LATERAL SUPPRESSOR homolog, CjLAS, as a key candidate regulator. Combined

Tobacco (Nicotiana tabacum L.) is a globally important industrial crop, yet its productivity is severely threatened by bacterial wilt caused by R. solanacearum. In this study, integrative metabolomics and transcriptomics profiling of tobacco plants infected with R. solanacearum identified 571 differentially accumulated metabolites (DAMs), peaking at 96 h postinoculation (hpi). Among 88 alkaloid DAMs, six plumerane alkaloids (indole, indole-3-acetic acid (IAA), 3-indoleacetonitrile (IAN), methoxyindoleacetic acid (MIAA), N-acetyl-5-hydroxytryptamine, and N-acetylisatin) showed the most pronounc

Limited selenium (Se) accumulation in agricultural products contributes to widespread dietary Se deficiency, posing a global nutritional threat. Efficient agronomic strategies to enhance Se biofortification in crops are still lacking. Here, we reveal the synergistic effects of shading and Se nanoparticles application on Se biofortification in tea leaves. Field experiments demonstrate that shading enhances Se accumulation in tea leaves under sufficient Se supply, through activating Se metabolism pathway. We identify CsAPR1 functions at the early stage of Se assimilation, catalyzing the reductio
