Genomic insights into multidrug - resistant Salmonella enterica isolates from pet dogs and cats.

Companion animals are recognized as potential reservoirs and transmitters of antimicrobial resistance (AMR) within the One Health framework. However, in-depth knowledge on AMR in pet animals remains limited. This study aimed to characterize Salmonella from companion dogs and cats using Whole Genome Sequencing (WGS). A total of 25 Salmonella obtained from clinically healthy household dogs and cats were serotyped and had their antimicrobial susceptibility tested. A discrepancy between the serovars identified by traditional slide agglutination tests and those determined by WGS analysis was observ

Colistin-resistance genes in Escherichia coli isolated from patients with urinary tract infections.

BACKGROUND: The incidence of antimicrobial resistance is alarmingly high because it occurs in humans, environment, and animal sectors from a "One Health" viewpoint. The emergence of plasmid-carried mobile colistin-resistance (MCR) genes limits the efficacy of colistin, which is the last-line treatment for multidrug resistance (MDR) against gram-negative infections. OBJECTIVES: The current study aimed to investigate emergence of colistin-resistance (MCR 1-5) genes in E. coli isolated from patients with urinary tract infections (UTIs) in Jordan. METHODS: E. coli (n = 132) were collected from u

Broiler Farms and Carcasses Are an Important Reservoir of Multi-Drug Resistant Escherichia coli in Ecuador.

Antimicrobial resistance (AMR) is a major health threat for public and animal health in the twenty-first century. In Ecuador, antibiotics have been used by the poultry industry for decades resulting in the presence of multi-drug resistant (MDR) bacteria in the poultry meat production chain, with the consequent risk for public health. This study evaluated the prevalence of ESBL/AmpC and mcr genes in third-generation cephalosporin-resistant Escherichia coli (3GC-R E. coli) isolated from broiler farms (animal component), broiler carcasses (food component), and human enteritis (human component) in

Antibacterial and antibiofilm properties of 1,3,4-oxadiazoles against multidrug-resistant Pseudomonas aeruginosa isolated from burn infections - an in vitro study.

BACKGROUND: Pseudomonas aeruginosa is a major causative agent of burn wound infections, frequently characterised by multidrug resistance (MDR) and robust biofilm formation, which limit the efficacy of conventional antibiotics. 1,3,4-Oxadiazole derivatives have emerged as potential antimicrobial and antibiofilm agents; however, comprehensive evaluations against clinical MDR isolates from burn patients remain limited. The objective of this study was to investigate the in vitro antibacterial and antibiofilm activities of a synthesised 1,3,4-oxadiazole compound against clinical MDR P. aeruginosa i

Virulome-resistome convergence in swine-associated multidrug-resistant Escherichia coli from Hungary: virulence marker profiles and zoonotic potential.

BACKGROUND: Swine production can sustain dense reservoirs of multidrug-resistant (MDR) Escherichia coli in which mobilizable resistance may co-occur with virulence modules relevant to animal health and One Health risk. We profiled the virulome of Hungarian swine-associated MDR E. coli and assessed whether key virulence signatures co-segregate with selected high-impact resistance flags. METHODS: In late 2023, E. coli isolates were obtained from four large-scale Hungarian pig farms through routine veterinary diagnostic/surveillance sampling. Phenotypic antimicrobial susceptibility testing was p

Clade-specific evolution and genome plasticity underlying antimicrobial resistance in Enterococcus faecium.

A commensal organism has transformed into the leading multidrug resistant (MDR) hospital acquired pathogen, with serious implication on global public health. The ability of Enterococcus faecium to rapidly adapt through its great genomic plasticity, horizontal gene transfer and under selective pressures created by heavy antibiotic application both clinically and non-clinically is responsible for the phenomenon. Comparative genome studies have reported the presence of different community acquired and hospital acquired clades; hospital acquired clones being particularly the clonal complex 17 have

A POC label-free biosensor to detect point mutations in the rpoB gene of Mycobacterium tuberculosis from RR-TB raw sputum samples.

Rifampicin-resistant and multidrug-resistant tuberculosis (RR/MDR-TB) remain pressing global health concerns. Conventional diagnostics such as culture and GeneXpert are limited by high cost, infrastructure requirements, and lengthy procedures, highlighting the need for rapid and reliable molecular detection platforms. We developed an electrochemical biosensor for the selective detection of wild-type and clinically relevant point mutations (codons 516 and 531) in the rpoB gene of Mycobacterium tuberculosis. Thiolated gene-specific ssDNA probes were immobilized on gold nanoparticle-modified carb

A hypothetical adhesin protein induces anti-biofilm antibodies against multi-drug resistant Acinetobacter baumannii.

The increase in multidrug-resistant (MDR) Acinetobacter baumannii strains in hospital environments has generated great concern around the world. Biofilm is one of the forms of bacterial adaptation that is increasingly leading to antimicrobial resistance and therapeutic failure. The search for alternative therapeutic strategies, especially non-antibiotic-based, is urgently needed. In this study, we produce polyclonal antibodies (pAbs) in murine models against recombinant CAM87009.1 antigen, an A. baumannii fimbriae protein. The pAbs produced were isotyped and anti-biofilm activity evaluated in

Resistance of bcr-abl-positive acute lymphoblastic leukemia to daunorubicin is not mediated by mdr1 gene expression.

In vitro resistance to anthracyclines is thought to be a poor prognosis in achieving long-term remission in patients with acute lymphoblastic leukemia (ALL). Expression of a multidrug resistance gene (mdr1) that codes for 170 Kd transmembrane glycoprotein is responsible for conferring resistance to malignant cells to anthracyclines. The t(9:22) translocation, resulting in bcr-abl fusion gene, is commonly found in B-lineage ALL and is known to be a poor prognostic factor for long-term remission. To investigate whether resistance to anthracyclines contributes to poor prognosis in bcr-abl-positiv

The prevalence and mechanism of triclosan resistance in Escherichia coli isolated from urine samples in Wenzhou, China.

BACKGROUND: The widespread application of triclosan contributes to its residual deposition in urine, which provides an environment of long-term exposure to triclosan for the intestinal Escherichia coli. We determined the triclosan and antibiotic resistance characteristics of E. coli strains isolated from urine samples and further investigated the resistance mechanism and molecular epidemic characteristics of triclosan-resistant E. coli isolates. METHODS: A total of 200 non-repetitive E. coli strains were isolated from urine samples and then identified. The minimum inhibitory concentrations (M

Antibacterial activities of hexadecanoic acid methyl ester and green-synthesized silver nanoparticles against multidrug-resistant bacteria.

Antibacterial drug resistance is considered one of the biggest threats to human health worldwide, and the overuse of antibiotics accelerates this problem. Multidrug-resistant (MDR) bacteria are becoming harder to treat as the antibiotics used to treat them become less effective. Therefore, it is necessary to evaluate novel methods to control MDR bacteria. In this study, 40 bacterial isolates were collected from diabetic patients. The sensitivity of 40 bacterial isolates to seven antibiotics was evaluated. Four bacterial isolates were resistant to all antibiotic groups. The MDR pathogenic bacte

Genomic and functional characterization of six novel phages targeting multidrug-resistant Escherichia coli: promising candidates for clinical phage therapy.

Antimicrobial resistance in multidrug-resistant Escherichia coli has severely compromised the efficacy of conventional antibiotics, particularly in hospital-acquired infections. Bacteriophage therapy offers a targeted approach, yet the rapid emergence of phage-resistant mutants during monotherapy limits its durability. However, phage cocktail therapy can overcome this issue. In this study, we isolated and characterized six novel lytic bacteriophages from hospital wastewater that specifically infect MDR E. coli clinical isolates. All six phages exhibited strictly lytic lifestyles, and genomic a

Conjugative transmission of virulence plasmid in Klebsiella pneumoniae mediated by a novel IncN-like plasmid.

Klebsiella pneumoniae (Kp) is increasingly recognized as a reservoir for a range of antibiotic resistance genes and a pathogen that frequently causes severe infections in both hospital and community settings. In this study, we have identified a novel mechanism of conjugative transfer of a non-conjugative virulence plasmid through the formation of a fusion plasmid between the virulence plasmid and a novel 59,162 bp IncN- plasmid. This plasmid was found to be a multidrug-resistance (MDR) plasmid and carried a T4SS cluster, which greatly facilitated the efficient horizontal transfer of the fusion

The Gut of Healthy Infants in the Community as a Reservoir of ESBL and Carbapenemase-Producing Bacteria.

The recent rapid rise of multi-drug resistant Enterobacteriaceae (MDR-E) is threatening the treatment of common infectious diseases. Infections with such strains lead to increased mortality and morbidity. Using a cross-sectional study, we aimed to estimate the prevalence of gut colonization with extended spectrum beta-lactamase (ESBL) producing Enterobacteriaceae among healthy infants born in Pakistan, a setting with high incidence of MDR-E infections. Stool samples were collected from 104 healthy infants between the ages of 5 and 7 months. Enterobacteriaceae isolates were screened for resista
