The c-Jun N-terminal kinase (JNK) signaling pathway plays a critical role in insect stress responses and immunity; however, its downstream transcriptional mechanisms remain poorly understood in coleopteran species. The kayak gene, a Fos homolog that functions as a JNK-responsive transcription factor, has been well studied in Drosophila melanogaster, but its role in beetles remains unexplored. In this study, we examined the functional role of TmKayak in the immune response of Tenebrio molitor using RNA interference. Upon TmKayak knockdown, we assessed the expression patterns of 15 antimicrobial

Primary sclerosing cholangitis (PSC) is a chronic autoimmune cholestatic liver disease characterized by progressive inflammation and fibrosis, but the role of neutrophils in PSC remains poorly understood. To define neutrophil states in the PSC liver microenvironment, we performed single-cell RNA sequencing of leukocytes isolated from liver perfusate obtained from healthy donor grafts and explanted PSC livers. We identified a marked expansion of transcriptionally diverse neutrophil subclusters in PSC liver perfusate. Importantly, PSC livers were enriched for four neutrophil subclusters: RHOB-hi

Leydig cells play a crucial role in male reproductive physiology, and their dysfunction is often associated with male infertility. Hypoxia negatively affects the structure and function of Leydig cells. This study aimed to investigate the impact of melatonin on the c-Jun N-terminal kinase (Jnk), P38, and extra-cellular signal-regulated kinases 1 and 2 (Erk1/2) mitogen-activated protein kinase (MAPK) signaling pathways in TM3 mouse Leydig cells under hypoxia induced by cobalt (II) chloride (CoCl2). The TM3 cell line was utilized as a subject of research, and 100 μM CoCl2 was employed to induce h

Valproic acid (VPA) is well-known as a histone deacetylase (HDAC) inhibitor. It has been reported that HDAC inhibitors enhance basal and aryl hydrocarbon receptor (AhR) ligand-induced aryl hydrocarbon receptor-responsive gene expression. Other studies suggested that HDAC inhibition might significantly activate the NF-E2-related factor-2 (Nrf2). Moreover, VPA activates mitogen-activated protein kinases (MAPKs). MAPK pathways regulate Nrf2 transactivation domain activity. Uridine 5'-diphospho-glucuronosyltransferase (UGT) 1A6 is one of the important isoforms to affect drug pharmacokinetics. UGT1

In rat in vivo, both paracetamol (APAP) and carbon tetrachloride (CCl4) induce liver necrosis, but long-term treatment with CCl4, in contrast to paracetamol, causes liver fibrosis. The aim of this study was to perform transcriptomic analysis to compare the early changes in mRNA expression profiles induced by APAP and CCl4 in the rat precision-cut liver slice model (PCLS) and to identify early markers that could predict fibrosis-inducing potential. Microarray data of rat PCLS exposed to APAP andCCl4was generated using a toxic dose based on decrease in ATP levels. Toxicity pathway analysis using

Chronic obstructive pulmonary disease (COPD) is associated with persistent inflammation and oxidative stress in susceptible individuals. Using microarray analysis of bronchial biopsy samples from patients with COPD and controls, we identified Wnt4 as being up-regulated in COPD. Analysis of bronchial biopsy samples showed a very strong correlation between Wnt4 and IL8 gene expression, suggesting that Wnt4 plays a role in chronic lung inflammation. In vitro, Wnt4 induced proliferation and inflammation in human epithelial cells (BEAS-2B) and normal primary human bronchial epithelial cells in a co

Human type II deiodinase is a master regulator of thyroid hormone activation in several tissues. In placenta, type II deiodinase mRNA levels and enzymatic activity are elevated only during the first trimester of pregnancy and then progressively decline. During this early stage, mitogens such as epidermal growth factor (EGF) have been shown to promote the proliferation of the trophoblast by acting through multiple mechanisms. Here we show that EGF modulates transcription of human type II deiodinase gene (Dio2) through distinct signaling pathways, leading to the assembly of a heterogeneous trans

Sequential and transient expression of c-fos, c-jun, c-myc, c-Ha-ras and c-Ki-ras proto-oncogene RNA transcripts with zonal heterogeneity was demonstrated in virtually all hepatocytes of adult rat liver by in situ hybridization with single-stranded, [35S]-labeled cRNA probes at various time points after intraperitoneal administration of a single dose of carbon tetrachloride (CCl4). After a brief interval, elevated RNA levels of these genes were also observed in nonparenchymal cells. A second phase of proto-oncogene expression was characterized by high RNA levels in only a fraction of parenchym

The association between multiple trace elements exposure and liver fibrosis, along with potential biological correlates, remains poorly understood. We conducted a longitudinal panel study among 205 adults with four repeated visits in Enshi, China. Liver stiffness measurement (LSM) and controlled attenuation parameter (CAP) were assessed using transient elastography, and serum trace elements and untargeted metabolomic profiles were measured. Linear mixed-effect models and mixture exposure models were applied to assess the individual and mixture associations, while a meet-in-the-middle (MITM) ap

Residual contamination of the type II pyrethroid lambda-cyhalothrin (LCT) is documented in coastal environments, posing potential risks to benthic fauna such as Tachypleus tridentatus. We integrated acute toxicity bioassays, histopathology, Na+/K+-ATPase and Ca2+-ATPase activity assays, and transcriptomic profiling to evaluate LCT-induced effects and mechanisms in first-instar larvae. The 48-h LC50 was 41.99 μg/L. Exposed larvae displayed characteristic neurotoxicity, including motor inhibition and sustained contraction of limbs and gills. Enzyme assays showed rapid and persistent suppression

INTRODUCTION: Therapy-induced senescence (TIS) is an important response to anticancer treatments in ovarian cancer, which can trigger the senescence-associated secretory phenotype (SASP) that may contribute to epithelial-mesenchymal transition (EMT), tumor migration, invasion, and adhesion. This review delineates the molecular mechanisms through linking TIS-mediated SASP to EMT and evaluates emerging therapeutic opportunities.
AREAS COVERED: We discuss key regulatory pathways including cGAS-STING, NF-κB, p53, and STAT3 that orchestrate SASP components such as IL-6, IL-8, and TGF-β to activate

Peripheral nerve injuries (PNIs) present a significant clinical challenge, often resulting in impaired motor and sensory function. While the peripheral nervous system possesses regenerative capacity, severe injuries necessitate intervention to promote effective recovery. This study investigated the efficacy of novel PCL/Tragacanth scaffolds functionalized with amino acid-modified carbon quantum dots (CQDs) for peripheral nerve regeneration. The CQD was designed to mimic the action of nerve growth factor (NGF) by binding to TrkA receptors and activating the downstream Ras-MAPK/ERK signaling pat

Immunomodulatory (IMiD) agents like lenalidomide and pomalidomide induce the recruitment of IKZF1 and other targets to the CRL4CRBN E3 ubiquitin ligase, resulting in their ubiquitination and degradation. These agents are highly active in B-cell lymphomas and a subset of myeloid diseases but have compromised effects in T-cell lymphomas (TCLs). Here, we show that 2 factors determine resistance to IMiDs among TCLs. First, limited CRBN expression reduces IMiD activity in TCLs but can be overcome by newer-generation degrader CC-92480. Using mass spectrometry, we show that CC-92480 selectively degra

Sestrin2 is a stress-inducible metabolic regulator and a conserved antioxidant protein which has been implicated in the pathogenesis of several diseases. Sestrin2 can protect against atherosclerosis, heart failure, hypertension, myocardial infarction, stroke, spinal cord injury neurodegeneration, nonalcoholic fatty liver disease (NAFLD), liver fibrosis, acute kidney injury (AKI), chronic kidney disease (CKD), and pulmonary inflammation. Oxidative stress and cellular damage signals can alter the expression of Sestrin2 to compensate for organ damage. Different stress signals such as those mediat

Revision operations have become a new issue after successful artificial joint replacements, and periprosthetic osteolysis leading to prosthetic loosening is the main cause of why the overactivation of osteoclasts (OCs) plays an important role. The effect of biochanin A (BCA) has been examined in osteoporosis, but no study on the role of BCA in prosthetic loosening osteolysis has been conducted yet. In this study, we utilised enzyme-linked immunosorbent assay, computed tomography imaging, and histological analysis. Results showed that BCA downregulated the secretion levels of tumor necrosis fac

The unbalanced crosstalk between osteoclasts and osteoblasts could lead to disruptive bone homeostasis. Herein, we investigated the therapeutic effects of rubidium chloride (RbCl) on ovariectomized (OVX) and titanium (Ti) particle-induced calvaria osteolysis mouse models, showing that non-toxic RbCl attenuated RANKL-stimulated osteoclast formation and functionality while significantly enhancing osteogenesis in vitro. The expressions of osteoclast-specific genes were downregulated considerably by RbCl. Despite the direct inhibition of RANKL-induced activation of MAPK signaling, RbCl was able to

Resistance to the breakpoint cluster region-abelson (BCR-ABL) tyrosine kinase inhibitor (TKI), imatinib, poses a major problem in the treatment of chronic myeloid leukemia (CML). Imatinib resistance often results from a secondary mutation in BCR-ABL1. However, the basis of this BCR-ABL1-independent resistance in the absence of such mutation remains to be elucidated. The aim of the present study is to identify the mechanism of imatinib resistance in CML. To gain insight into BCR-ABL1-independent imatinib resistance mechanisms, we performed an array-based comparative genomic hybridization. We id

Polycystic ovary syndrome (PCOS) is the most common type of endocrine disorder, affecting 5‑11% of women of reproductive age worldwide. Transcription factors (TFs) and microRNAs are considered to have crucial roles in the developmental process of several diseases and have synergistic regulatory actions. However, the effects of TFs and microRNAs, and the patterns of their cooperation in the synergistic regulatory network of PCOS, remain to be elucidated. The present study aimed to determine the possible mechanism of PCOS, based on a TF‑microRNA synergistic regulatory network. Initially, the dif

Hepatocarcinogenesis is a complex process involving chronic liver injury, inflammation, unregulated wound healing, subsequent fibrosis and carcinogenesis. To decipher the molecular mechanism underlying transition from chronic liver injury to dysplasia, we investigated the oncogenic role of gankyrin (PSMD10 or p28GANK) during malignant transformation in a transgenic mouse model. Here, we find that gankyrin increased in patients with cirrhosis. In addition to more severe liver fibrosis and tumorigenesis after DEN plus CCl4 treatment, hepatocyte-specific gankyrin-overexpressing mice (gankyrinhep)

Previously, we characterized the biological properties of Akbu-LAAO, a novel L-amino acid oxidase from Agkistrodon blomhoffii ussurensis snake venom (SV). Current work investigated its in vitro anti-tumor activity and underlying mechanism on HepG2 cells. Akbu-LAAO inhibited HepG2 growth time and dose-dependently with an IC50 of ~38.82 μg/mL. It could induce the apoptosis of HepG2 cells. Akbu-LAAO exhibited cytotoxicity by inhibiting growth and inducing apoptosis of HepG2 as it showed no effect on its cell cycle. The inhibition of Akbu-LAAO to HepG2 growth partially relied on enzymatic-released
