SWAN scale for ADHD trait-based genetic research: a validity and polygenic risk study.

BACKGROUND: Population-based samples with valid, quantitative and genetically informative trait measures of psychopathology could be a powerful complement to case/control genetic designs. We report the convergent and predictive validity of the parent- and self-report versions of the Strengths and Weaknesses of ADHD Symptoms and Normal Behavior Rating Scale (SWAN). We tested if SWAN scores were associated with ADHD diagnosis, ADHD polygenic risk, as well as traits and polygenic risk for disorders that co-occur with ADHD: anxiety and obsessive-compulsive disorder (OCD). METHODS: We collected pa

Haplotype function score improves biological interpretation and cross-ancestry polygenic prediction of human complex traits.

We propose a new framework for human genetic association studies: at each locus, a deep learning model (in this study, Sei) is used to calculate the functional genomic activity score for two haplotypes per individual. This score, defined as the Haplotype Function Score (HFS), replaces the original genotype in association studies. Applying the HFS framework to 14 complex traits in the UK Biobank, we identified 3619 independent HFS-trait associations with a significance of p < 5 × 10-8. Fine-mapping revealed 2699 causal associations, corresponding to a median increase of 63 causal findings per t

Age-Specific Associations of Polygenic Risk Scores With Advanced Fibrosis and Histological Activity in Biopsy-Proven MASLD.

BACKGROUND AND AIMS: The genetic susceptibility to MASLD histological severity remains incompletely defined. We assessed the associations between recently developed genome-wide association studies-derived polygenic risk scores (PRSs) and advanced fibrosis in MASLD. We also evaluated PRSs' associations with histological activity and selected PRSs' predictive ability for advanced fibrosis. METHODS: We analysed 2149 adults and 900 children with biopsy-proven MASLD from the NASH Clinical Research Network studies. Genotyping was performed by the Regeneron Genetics Center using whole-exome sequenci

Clinical, Dietary, Lifestyle and Genetic Factors Associated With Age at Onset of Esophageal Adenocarcinoma.

BACKGROUND: Esophageal adenocarcinoma (EAC) represents one of the most increasing malignancies in Western countries. The disease is multifactorial, involving modifiable risk factors and genetic susceptibility variants. These variants can be aggregated to a polygenic risk score (PRS) that reflects individual genetic risk. Investigation of the effects of lifestyle factors, PRS, and co-medication on EAC age at onset (AAO) is critical for shaping prevention strategies. METHODS: A detailed questionnaire was used to assess pre-diagnostic exposure to lifestyle factors and clinical information from a

Joint effects of childhood adversity and genetic risk for psychosis on psychopathology in the UK Biobank.

BACKGROUND: The individual effects of genetic factors and adverse childhood experiences (ACEs) on risk of psychosis, including schizophrenia (SCZ) and bipolar disorder (BIP), have been widely acknowledged, but their interaction effects on individual psychopathological symptoms remain unclear. METHODS: Based on data from 163,704 individuals in the UK Biobank, we investigated the joint effects of polygenic risk scores (PRSs) of SCZ and BIP and ACEs on psychopathology. ACEs status and 55 psychopathological symptoms from seven domains were measured retrospectively using an online mental health qu

Functional polygenic risk score of glucocorticoid-dependent regulatory element activity and its relation to clinical and stress-related phenotypes.

BACKGROUND: Chronic stress and adverse life events are major contributors to depression and anxiety disorders. Genetic variants that influence glucocorticoid (GC) hormone responses may moderate the impact of stress on psychiatric outcomes. Here, we use a functional polygenic risk score (fPRS) based on GC-responsive gene regulatory elements (GREs) following dexamethasone (DEX) exposure (DEX-fPRS) and examined its association with clinical and biological stress-related phenotypes. METHODS: We analyzed data from 2286 participants in the Dutch NESDA cohort, with baseline, 2-year, and 6-year follo

Polygenic risk scores and help-seeking behaviour in young people with recent onset of mood and psychotic disorders.

OBJECTIVES: We examined associations between polygenic risk scores (PRS) for depression (PRS-MDD), psychosis (PRS-SCZ), bipolar disorders (PRS-BD) and neuroticism (PRS-NEU) and (i) help-seeking, and (ii) new onset cases of full-threshold mood or psychotic disorders in youth. METHODS: Help-seeking for mental health problems was assessed by self-report and mood and psychotic disorders were identified using the Composite International Diagnostic Interview. A principal component analysis of the four selected PRS identified two dimensions (BD-SCZ; MDD-NEU) that accounted for 69.9 % of the explaine

Type 2 diabetes pathway-specific polygenic risk scores elucidate heterogeneity in clinical presentation, disease progression and diabetic complications in 18,217 Chinese individuals with type 2 diabetes.

AIMS/HYPOTHESIS: Type 2 diabetes is a complex and heterogeneous disease and the aetiological components underlying the heterogeneity remain unclear in the Chinese and East Asian population. Therefore, we aimed to investigate whether specific pathophysiological pathways drive the clinical heterogeneity in type 2 diabetes. METHODS: We employed newly developed type 2 diabetes hard-clustering and soft-clustering pathway-specific polygenic risk scores (psPRSs) to characterise individual genetic susceptibility to pathophysiological pathways implicated in type 2 diabetes in 18,217 Chinese patients f

Inflammation-Mediated Lipid Metabolism in Endocrine Autoimmune Diseases: A Genetic Distance-Based PRS Approach Integrating HLA Region.

Background: Endocrine autoimmune diseases (AIDs) exhibit special polygenic characteristics in human leucocyte antigen (HLA) region. Current understanding of their association with lipid metabolism remains constrained by imprecise polygenic risk score (PRS) modeling. Advanced analytical approaches are needed to elucidate the association between genetic susceptibility and lipid metabolic dysregulation. Methods: We proposed a genetic distance-based clumping gPRS to account for linkage disequilibrium in the HLA region. gPRS and pathway gPRS were constructed for individuals diagnosed with type I di

Performance Metrics for Selecting Single Nucleotide Polymorphisms in Late-onset Alzheimer's Disease.

Previous genome-wide association studies using P-values to select single nucleotide polymorphisms (SNPs) have suffered from high false-positive and false-negative results. This case-control study recruited 713 late-onset Alzheimer's disease (LOAD) cases and controls aged ≥65 from three teaching hospitals in northern Taiwan from 2007 to 2010. Performance metrics were used to select SNPs in stage 1, which were then genotyped to another dataset (stage 2). Four SNPs (CPXM2 rs2362967, APOC1 rs4420638, ZNF521 rs7230380, and rs12965520) were identified for LOAD by both traditional P-values (without c

Genome-wide investigations of the lipid accumulation product index pinpoints polygenic determinants underlying pancreatitis susceptibility.

The lipid accumulation product index (LAP) is a robust marker of lipid burden. However, its genetic architecture and causal relevance to pancreatitis remain poorly characterized. We performed a genome-wide association study (GWAS) of LAP in 388,213 European from UK Biobank, followed by colocalization, transcriptome-wide association studies (TWAS), LD score regression and Mendelian randomization for causal relationships. Polygenic risk scores (PRS) were constructed and tested against chronic pancreatitis (CP), hypertriglyceridemic acute pancreatitis (HTG-AP), and acute pancreatitis (AP) using r

Genetic determinants of childhood onset systemic lupus erythematosus.

INTRODUCTION: Childhood-onset systemic lupus erythematosus (cSLE) is associated with significant morbidity and mortality. While numerous variants have been associated with adult-onset SLE, limited data exist on genetic variation within cSLE. We aimed to investigate genetic factors of early-onset cSLE, defined as onset of cSLE prior to age 10. METHODS: Employing a case-only design, we performed whole genome sequencing analysis on 37 subjects with early onset cSLE. We hypothesised that rare, functional variants with large effects in genes associated with SLE contribute to the risk of early-onse

Brain volume trajectories in young children are associated with polygenic scores for late-onset Alzheimer's disease risk.

INTRODUCTION: Polygenic risk scores (PRSs) for Alzheimer's disease (AD) capture an individual's genetic susceptibility to AD. Although thoroughly studied in older populations, there exists a notable gap in comprehensively exploring the association of AD PRS with early brain development. METHODS: We examined longitudinal brain magnetic resonance imaging (MRI) data from 348 typically developing children in the RESONANCE cohort. Proportional cerebrospinal fluid (pCSF), white matter (pWM), and gray matter (pGM) volumes were analyzed using functional concurrent regression and Riemannian functional

Variability in epilepsy polygenic risk prediction across Taiwanese population and clinical cohorts.

OBJECTIVE: This study was undertaken to evaluate the predictive, stratification, and prognostic utility of polygenic risk scores (PRSs) for epilepsy in three Taiwanese cohorts comprising more than 7000 cases among more than 600 000 individuals, thereby addressing the limited evidence on PRS performance in non-European populations. METHODS: Leveraging results from the latest multiancestry genome-wide association study (GWAS), we calculated PRS for all epilepsy, genetic generalized epilepsy (GGE), and focal epilepsy (FE) in two population cohorts-Taiwan Biobank-National Health Insurance Researc

Polygenic risk score and age: an extra help in the cardiovascular prevention of the young?

All major guidelines recommend assessing the risk of atherosclerotic cardiovascular disease (ASCVD) using risk scores. In fact, it has been shown that their use at the population level increases the accuracy of event prediction and facilitates the choice of strategies to be adopted in primary prevention. In fact, their use in clinical practice is far from optimal and their predictive ability on an individual level is not excellent. Our genetic heritage is substantially stable from birth and determines a 'baseline risk' on which external influences act. Genetic information therefore has the pot

Genome-wide association study of alcohol use disorder identification test (AUDIT) scores in 20 328 research participants of European ancestry.

Genetic factors contribute to the risk for developing alcohol use disorder (AUD). In collaboration with the genetics company 23andMe, Inc., we performed a genome-wide association study of the alcohol use disorder identification test (AUDIT), an instrument designed to screen for alcohol misuse over the past year. Our final sample consisted of 20 328 research participants of European ancestry (55.3% females; mean age = 53.8, SD = 16.1) who reported ever using alcohol. Our results showed that the 'chip-heritability' of AUDIT score, when treated as a continuous phenotype, was 12%. No loci reached

Genome-wide association study of antisocial personality disorder diagnostic criteria provides evidence for shared risk factors across disorders.

INTRODUCTION: While progress has been made in determining the genetic basis of antisocial behaviour, little progress has been made for antisocial personality disorder (ASPD), a condition that often co-occurs with other psychiatric conditions including substance use disorders, attention deficit hyperactivity disorder (ADHD), and anxiety disorders. This study aims to improve the understanding of the genetic risk for ASPD and its relationship with other disorders and traits. METHODS: We conducted a genome-wide association study (GWAS) of the number of ASPD diagnostic criteria data from 3217 alco
