Comparison of physiological and pharmacological tests of growth hormone function in children with short stature.

We studied 30 short-statured children to investigate their 24-h integrated serum concentration of GH (ICGH) and its diurnal pattern and to compare this data with their responses to standard stimulation tests for GH release. Eight patients with normal stimulation tests had a mean (+/- SD) ICGH value of 8.0 +/- 3.8 ng/ml (range, 4.5--16.4), and all had several secretory peaks greater than 12 ng/ml. Nineteen patients with abnormal responses (peak, less than 10 ng/ml) had a mean ICGH value of 2.7 +/- 1.9 ng/ml (range, less than 0.8 to 7.0); this was significantly lower than that of control subject

Sleep and sleep disorders in children and adolescents.

Pediatric sleep disorders are common, affecting approximately 25% to 40% of children and adolescents. Although there are several different types of sleep disorders that affect youth, each disorder can have a significant impact on daytime functioning and development, including learning, growth, behavior, and emotion regulation. Researchers are only beginning to uncover the interaction between sleep and psychiatric disorders in children and adolescents, including depression, attention-deficit/hyperactivity disorder, and autism. This article reviews normal sleep and sleep disorders in children an

Pediatric otorhinolaryngologic manifestations of endocrinological pathologies.

OBJECTIVE: Aim of the present review is to offer a concise but complete discussion about pathologies with pediatric otorhinolaryngological interest recognizing: (i) growth disorders, (ii) chromosomal disorders, and (iii) tumors. METHODS: Pathologies characterized by a growth anomaly have been grouped, underlining the characteristics of otorhinolaryngological interest. Numerous syndromes typical of the pediatric age have been reported and classified according to the ailments of the orofacial district. Tumoral pathologies, for which the otorhinolaryngological operation is fundamental, have been…

Congenital optic atrophy and brachytelephalangy: the Berk-Tabatznik syndrome.

Here we report on a prepubertal boy with the Berk-Tabatznik syndrome. Manifestations included short stature, congenital optic atrophy, and hypoplasia of the cervical vertebral bodies and distal phalanges. Prognosis was adversely affected by visual and auditory difficulties. The cause of this condition remains unknown.

Monogenic causes of familial short stature.

Genetic factors play a crucial role in determining human height. Short stature commonly affects multiple family members and therefore, familial short stature (FSS) represents a significant proportion of growth disorders. Traditionally, FSS was considered a benign polygenic condition representing a subcategory of idiopathic short stature (ISS). However, advancements in genetic research have revealed that FSS can also be monogenic, inherited in an autosomal dominant manner and can result from different mechanisms including primary growth plate disorders, growth hormone deficiency/insensitivity o…

GM2 ganglioside and pyramidal neuron dendritogenesis.

GM2 ganglioside, although scarce in normal adult brain, is the predominant ganglioside accumulating in several types of lysosomal disorders, most notably Tay-Sachs disease. Pyramidal neurons of cerebral cortex in Tay-Sachs, as well as many other types of neuronal storage disorders, are known to exhibit a phenomenon believed unique to storage disorders: growth of ectopic dendrites. Recent studies have shown that a common metabolic abnormality shared by storage diseases with ectopic dendrite growth is the abnormal accumulation of GM2 ganglioside. The correlation between increased levels of GM2 a

Overlap of quantitative trait loci for early growth rate, and for body weight and age at onset of sexual maturity in chickens.

Critical age, weight and body composition have been suggested as necessary correlates of sexual maturity. A genome scan to identify quantitative trait loci (QTL) for age and body weight at first egg (AFE and WFE) was conducted on 912 birds from an F(2) broiler-layer cross using 106 microsatellite markers. Without a covariate, QTL for body WFE were detected on chromosomes 2, 4, 8, 27 and Z and a single QTL for AFE was detected on chromosome 2. With AFE as a covariate, additional QTL for body WFE were found on chromosomes 1 and 13, with abdominal fat pad as covariate a QTL for body WFE was found

Pituitary growth hormone from human cadavers: neurologic disease in ten recipients.

Creutzfeldt-Jakob disease (CJD) has been reported in three US patients previously treated with human growth hormone derived from large pools of human cadaver pituitary glands (pit-hGH). Neurologic disorders other than CJD occurred in 10 growth hormone-deficient patients treated with pit-hGH. These 10 cases could have been chance events or true syndromes; some cases may have been caused by transmissible agents contained in the pooled growth hormone product.

[Growth and development patterns of Noonan syndrome and advances in the treatment of short stature].

Patients with Noonan syndrome (NS) are born with normal or slightly lower body length and weight compared to the normal ranges. However, their height gradually falls behind that of the general population, leading to growth retardation and delayed puberty. In China, the incidence of short stature in patients with NS is approximately 65%. Short stature in these patients arises from multiple causes, including feeding difficulties in infancy, comorbidities such as congenital heart disease, genetic heterogeneity, and disorders of the growth hormone/insulin-like growth factor-1 axis. Growth hormone

A Rare Presentation of HIST1H1E Syndrome with Short Stature and Multiple Pituitary Hormone Deficiencies.

Histone Gene Cluster 1 Member E (HIST1H1E) syndrome is a rare autosomal dominant disorder resulting from a heterozygous variation in the H1-4 gene located on chromosome 6p22.2. Mental retardation, recognizable facial features, skeletal abnormalities and overgrowth are the main clinical manifestations of this syndrome. A 17-year-old male presented to the pediatric endocrinology clinic due to short stature. He had a characteristic facial appearance with neurodevelopmental delay. Genetic analysis revealed a heterozygous pathogenic variation in the H1-4 gene located on chromosome 6p22.2 which conf

Use of a 3D Model for the Correction of a Complex Madelung Deformity in a Teenager: A Case Report.

CASE: The aim of the article is to report on a case of a teenager affected by Madelung deformity treated with a double osteotomy, planned by means of a 3D model. Using a custom-made cutting guide, the radial osteotomy was performed, and after the reorientation, a shortening ulnar osteotomy completed the procedure. Postoperative clinical assessment showed a normal alignment of the ulna with increased range of motion wrist motion. CONCLUSIONS: Using a 3D model when planning a multidirectional correction of a Madelung deformity may be advantageous to achieve a more accurate and precise realignme

Failures of Endochondral Ossification in the Mucopolysaccharidoses.

PURPOSE OF REVIEW: The mucopolysaccharidoses (MPS) are a group of inherited lysosomal storage disorders characterized by abnormal accumulation of glycosaminoglycans (GAGs) in cells and tissues. MPS patients frequently exhibit failures of endochondral ossification during postnatal growth leading to skeletal deformity and short stature. In this review, we outline the current understanding of the cellular and molecular mechanisms underlying failures of endochondral ossification in MPS and discuss associated treatment challenges and opportunities. RECENT FINDINGS: Studies in MPS patients and anim

[Current problems in diagnosis and treatment of hereditary disturbances of growth and development in children].

The authors' long-term experience in diagnosing and treating childhood hereditary growth and developmental disturbances, such as genetic diseases of connective tissue, amino acid metabolic disturbances, rickets-like diseases, mitochondrial abnormalities, Rett syndrome, and fragile X syndrome is presented. The findings suggest that multimodality treatment is highly effective in treating children with hereditary growth and developmental disturbances in genetic care.
