Advancing throat cancer detection using an iterative multimodal machine learning approach for enhanced diagnostic accuracy.

Malignant growths in the tonsils, larynx, or pharynx are associated with throat cancer. Current diagnostic techniques are limited by subjectivity and clinician variability and rely on invasive procedures. This study examines sophisticated models for detecting throat cancer that use machine learning and multimodal analysis. Recurrent neural networks (RNNs), convolutional neural networks (CNNs), and hybrid models that combine these with other machine learning techniques are examples of state-of-the-art methods. These models enable data-driven improvements in diagnostic accuracy by utilizing a va

Automatic classification framework for ventricular septal defects: a pilot study on high-throughput mouse embryo cardiac phenotyping.

Intensive international efforts are underway toward phenotyping the entire mouse genome by modifying all its [Formula: see text] genes one-by-one for comparative studies. A workload of this scale has triggered numerous studies harnessing image informatics for the identification of morphological defects. However, existing work in this line primarily rests on abnormality detection via structural volumetrics between wild-type and gene-modified mice, which generally fails when the pathology involves no severe volume changes, such as ventricular septal defects (VSDs) in the heart. Furthermore, in e

Elucidating the pathogenic mechanism of a pedigree with complex rearrangements on chromosome 4 using optical genome mapping technology: a study on the genetics and functional pathways in a child with developmental delay.

INTRODUCTION: Chromosomal structural variations (SVs) are important causes of neurodevelopmental disorders in children, but traditional detection techniques often fail to accurately resolve the precise breakpoints and pathogenic genes of complex rearrangements. To apply optical genome mapping (OGM) detects SVs across the whole genome by high-throughput labeling of ultra-long (>150 kb) DNA molecules, high-resolution fluorescence imaging, and alignment algorithms using a reference genome. Its resolution is up to 500 bp and is especially effective in finding exact breakpoints and orientations of

Design of the Genetics of Early Onset Cardiovascular Disease (GENECARD) study.

BACKGROUND: Coronary artery disease (CAD) is the leading cause of death and a leading cause of disability in the developed world. Early onset (premature) coronary artery disease (EOCAD) is known to have a particularly strong genetic component. However, the actual genes leading to this increased risk of CAD remain obscure. METHODS: The primary goal of the Genetics of Early Onset Cardiovascular Disease (GENECARD) study is to perform a genetic linkage study in 920 families with at least 1 sibling pair having EOCAD. The study sites include a US network of 15 cardiology practices and 5 additional

Association of common genetic variants in GPCPD1 with scaling of visual cortical surface area in humans.

Visual cortical surface area varies two- to threefold between human individuals, is highly heritable, and has been correlated with visual acuity and visual perception. However, it is still largely unknown what specific genetic and environmental factors contribute to normal variation in the area of visual cortex. To identify SNPs associated with the proportional surface area of visual cortex, we performed a genome-wide association study followed by replication in two independent cohorts. We identified one SNP (rs6116869) that replicated in both cohorts and had genome-wide significant associatio

Case Report: Novel molecular characterization of rare primary mucinous adenocarcinoma of the renal pelvis.

Mucinous adenocarcinoma of the renal pelvis (MAC-RP) is an exceedingly rare condition, accounting for less than 1% of malignancies originating from this epithelial site. We report a case of MAC-RP in a woman presenting with right flank pain from obstructing stones, recurrent urinary tract infections, and ureteral stents. Imaging showed a right kidney mass consistent with xanthogranulomatous pyelonephritis (XGP) with an abscess surgically removed. Pathology after radical nephrectomy unexpectedly revealed in situ MAC-RP with intestinal metaplasia at the margin. Surveillance testing showed positi

Progressive Polycystic Kidney Disease in an Infant Girl With TSC2/PKD1 Contiguous Gene Syndrome.

TSC2/PKD1 contiguous gene syndrome is caused by deletions involving the TSC2 and PKD1 genes that lead to tuberous sclerosis complex and autosomal dominant polycystic kidney disease. It is characterized by early-onset severe cystic kidney disease with progressive enlargement of the kidneys and the cysts. As it can lead to early hypertension and an accelerated decline of kidney function, early genetic testing is needed for early diagnosis of this syndrome, and more frequent imaging-based examinations are necessary to assess disease progression and determine appropriate management. We report the

Genome-wide analyses identify 60 loci associated with liver triglyceride content.

We performed a large-scale genome-wide association study meta-analysis of magnetic resonance imaging-derived liver triglyceride content across 71,488 European-ancestry participants from the UK Biobank, approximately doubling the sample size of previous genome-wide association studies1. We identified 60 independent loci, including 31 not previously reported, and estimated the single nucleotide polymorphism-based heritability of liver triglyceride content at 25%. Integrative fine-mapping and gene-prioritization analyses nominated candidate effector genes for 48 loci. Of the 31 newly identified v

Detect de novo expressed ORFs in transcriptomes with DESwoMAN.

De novo gene emergence refers to the process by which new genes arise from mutations in previously non-coding genomic regions. Investigating early stages of de novo gene emergence is essential for understanding the mechanisms that enable gene formation from scratch. Several workflows have been developed to detect and study the early stage of de novo gene emergence in specific species. However, no software currently exists that can automatically identify and characterize novel, unannotated open reading frames from a transcriptome, and analyse their mutations and fixation patterns within or acro

Genetic Testing in Inherited Retinal Disease: Current Strategies and Future Directions.

Inherited retinal diseases (IRDs) are a major cause of visual impairment worldwide, marked by extensive genetic and phenotypic heterogeneity. Recent estimates from the U.S. suggest a prevalence of nearly 1 in 1000 individuals, reflecting both disease burden and improved diagnostic recognition. This review traces the shift from linkage analysis and Sanger sequencing to high-throughput next-generation sequencing, including panel-based, whole-exome, and whole-genome sequencing. Phenotype-driven testing strategies and standardized variant interpretation frameworks, such as the American College of

Antenatal Presentation of MRPS22-Related Mitochondrial Disease Confirmed With Rapid Proteomics.

MRPS22-related mitochondrial disease (MIM#611719) is a rare autosomal recessive disorder caused by defects in the mitochondrial ribosomal protein S22, a component of the small mitoribosomal subunit essential for mitochondrial translation. Of the few reported cases, most present antenatally with a severe phenotype, conveying a poor prognosis. We describe a fetus with severe antenatal-onset MRPS22-related mitochondrial disease and the use of multi-omics in the molecular diagnosis. A primigravida underwent termination of pregnancy following identification of multiple congenital anomalies (hydrops

Neisseria lactamica meningitis as the initial presentation of a rhino-cerebrospinal fluid fistula in an adult with remote cranial trauma.

BACKGROUND: Neisseria lactamica is a commensal of the upper respiratory tract in children and is rarely isolated in adults. Its genome shares approximately 60% homology with Neisseria meningitidis but lacks the virulence genomic islands essential for blood-brain barrier invasion. Meningitis due to Neisseria lactamica is exceptionally rare and few cases are reported in literature, mainly among pediatric patients. To date, only one case of Neisseria lactamica meningitis in an adult, associated with head trauma, has been reported in the literature (1991). CASE PRESENTATION: We report the case of

Long-read sequencing identifies FGF14 repeat expansions in Parkinson's disease.

Pathogenic GAA repeat expansions in FGF14 are an established cause of late-onset cerebellar ataxia, but have not been linked to Parkinson's disease. Given emerging evidence that repeat expansions in ataxia-associated genes like RFC1 can contribute to atypical or familial forms of Parkinson's disease, we investigated whether FGF14 expansions might play a similar role. Using long-read whole-genome sequencing, we analysed 411 individuals with Parkinson's disease and 197 neurologically healthy controls from the Parkinson's Progression Markers Initiative (PPMI) cohort, together with 1429 additional

Integrating Radiogenomics and CSF-Based Liquid Biopsy Sequencing for Precision Neuro-Oncology.

Glioblastoma and diffuse gliomas pose major therapeutic challenges due to marked intratumoral heterogeneity, limited tissue accessibility, and the blood-brain barrier. Tissue-based next-generation sequencing (NGS) remains essential for WHO CNS5 molecular classification, yet it is invasive and poorly suited to serial monitoring. Two complementary non- or minimally invasive approaches have advanced rapidly: radiogenomics, which correlates multiparametric MRI features with genomic alterations, and cerebrospinal fluid (CSF) liquid biopsy sequencing, which detects circulating tumor DNA with high ti
