Getting rid of the bad apple: inflammasome-induced extrusion of Salmonella-infected enterocytes.

Two reports in this issue of Cell Host & Microbe (Sellin et al., 2014; Knodler et al., 2014) establish the cell-intrinsic inflammasome-induced extrusion of infected enterocytes as a general defense mechanism against acute bacterial infections.

Inflammasome-mediated inhibition of Listeria monocytogenes-stimulated immunity is independent of myelomonocytic function.

Activation of the Nlrc4 inflammasome results in the secretion of IL-1β and IL-18 through caspase-1 and induction of pyroptosis. L. monocytogenes engineered to activate Nlrc4 by expression of Legionella pneumophilia flagellin (L. monocytogenes L.p.FlaA) are less immunogenic for CD8(+) T cell responses than wt L. monocytogenes. It is also known that IL-1β orchestrates recruitment of myelomonocytic cells (MMC), which have been shown to interfere with T cell-dendritic cells (DC) interactions in splenic white pulp (WP), limiting T cell priming and protective immunity. We have further analyzed the r

Downregulation of the Na/K-ATPase pump by leptospiral glycolipoprotein activates the NLRP3 inflammasome.

Leptospira interrogans is responsible for a zoonotic disease known to induce severe kidney dysfunction and inflammation. In this work, we demonstrate that L. interrogans induces NLRP3 inflammasome-dependent secretion of IL-1β through the alteration of potassium transport in bone marrow-derived macrophages. Lysosome destabilization also contributed to the IL-1β production upon stimulation with live, but not dead, bacteria. Using bone marrow-derived macrophages from various TLRs and nucleotide-binding oligomerization domain-deficient mice, we further determined that IL-1β production was dependen

The AIM2 inflammasome is essential for host defense against cytosolic bacteria and DNA viruses.

Inflammasomes regulate the activity of caspase-1 and the maturation of interleukin 1beta (IL-1beta) and IL-18. AIM2 has been shown to bind DNA and engage the caspase-1-activating adaptor protein ASC to form a caspase-1-activating inflammasome. Using Aim2-deficient mice, we identify a central role for AIM2 in regulating caspase-1-dependent maturation of IL-1beta and IL-18, as well as pyroptosis, in response to synthetic double-stranded DNA. AIM2 was essential for inflammasome activation in response to Francisella tularensis, vaccinia virus and mouse cytomegalovirus and had a partial role in the…

Auranofin protects against anthrax lethal toxin-induced activation of the Nlrp1b inflammasome.

Anthrax lethal toxin (LT) is the major virulence factor for Bacillus anthracis. The lethal factor (LF) component of this bipartite toxin is a protease which, when transported into the cellular cytoplasm, cleaves mitogen-activated protein kinase kinase (MEK) family proteins and induces rapid toxicity in mouse macrophages through activation of the Nlrp1b inflammasome. A high-throughput screen was performed to identify synergistic LT-inhibitory drug combinations from within a library of approved drugs and molecular probes. From this screen we discovered that auranofin, an organogold compound with

Caspase-7 activation by the Nlrc4/Ipaf inflammasome restricts Legionella pneumophila infection.

Legionella pneumophila (L. pneumophila), the causative agent of a severe form of pneumonia called Legionnaires' disease, replicates in human monocytes and macrophages. Most inbred mouse strains are restrictive to L. pneumophila infection except for the A/J, Nlrc4(-/-) (Ipaf(-/-)), and caspase-1(-/-) derived macrophages. Particularly, caspase-1 activation is detected during L. pneumophila infection of murine macrophages while absent in human cells. Recent in vitro experiments demonstrate that caspase-7 is cleaved by caspase-1. However, the biological role for caspase-7 activation downstream of

Critical function for Naip5 in inflammasome activation by a conserved carboxy-terminal domain of flagellin.

Inflammasomes are cytosolic multiprotein complexes that sense microbial infection and trigger cytokine production and cell death. However, the molecular components of inflammasomes and what they sense remain poorly defined. Here we demonstrate that 35 amino acids of the carboxyl terminus of flagellin triggered inflammasome activation in the absence of bacterial contaminants or secretion systems. To further elucidate the host flagellin-sensing pathway, we generated mice deficient in the intracellular sensor Naip5. These mice failed to activate the inflammasome in response to the 35 amino acids …

CHK1 phosphorylates TRIM28 at Ser473 to drive NLRP3 inflammasome assembly and inflammatory responses.

The NLRP3 inflammasome is a central player in innate immunity and is implicated in various inflammatory diseases, but its kinase-mediated regulatory networks remain incompletely defined. Here, we identify CHK1 as a positive regulator of NLRP3 inflammasome activation. Pharmacological inhibition or siRNA-mediated knockdown of CHK1 suppressed NLRP3-dependent caspase-1 activation, IL-1β secretion, and pyroptosis, without affecting AIM2 or NLRC4 inflammasomes. CHK1 promoted NLRP3 oligomerization and ASC speck formation in a kinase activity-dependent manner. Mechanistically, CHK1 phosphorylated the …

NLRP3 inflammasome priming and activation is involved in depression: A systematic review and meta-analysis.

Depression is characterised by a low mood, loss of interest or pleasure, pessimism, impaired concentration, decreased energy, and fatigue. A strong relationship between inflammatory processes and the pathophysiology of depression had been identified. Studies have suggested that the NLRP3 inflammasome is a key contributor to the pathogenesis of depression. This systematic review aimed to synthesise evidence from animal models of depression to evaluate both priming and activation of the inflammasome, with a specific focus on clarifying the role in depressive pathology. An N-terminal CDC42 T43I variant reveals the mechanism of pyrin inflammasome activation.

Heterozygous carboxyl-terminal variants in the RHO guanosine triphosphatase (GTPase) CDC42 are known to cause severe autoinflammatory syndromes. Here, we identified a heterozygous amino-terminal p.T43I (Thr43→Ile) CDC42 variant in patients with autoinflammation and uncovered a molecular link between CDC42 and the inflammasome sensor pyrin, mutated in the hereditary autoinflammatory syndrome familial Mediterranean fever. We demonstrate that the region surrounding residue T43 of CDC42 interacts with the carboxyl-terminal B30.2 domain of pyrin and regulates its localization and activation. The p.

Metrnl ameliorates ulcerative colitis by inhibiting NLRP3 inflammasome activation via ASC.

Ulcerative colitis (UC) is characterized by the dysregulation of inflammatory pathways, leading to extensive tissue damage and chronic inflammation. NLRP3 inflammasome promotes inflammatory cytokine maturation and pyroptosis. Meteorin-like protein (Metrnl) has anti-inflammatory effects; however, its specific mechanisms in regulating these pathways remain unclear. This study investigated the effect of Metrnl on UC via regulation of the NLRP3 inflammasome through regulation of its key component, apoptosis-associated speck-like protein containing CARD (ASC). NCM460 and FHC cells were treated with

The role of NLRP3 inflammasome in age-related macular degeneration: mechanisms and therapeutic prospects.

Age-related macular degeneration (AMD) is a fundus oculi disease that progressively impairs the central vision of patients. To date, its pathogenesis has not been fully elucidated, and therapeutic options for dry AMD remain limited. Recently, chronic low-grade inflammation has been recognized as an important pathogenic factor in various neurodegenerative diseases, including AMD. The NLRP3 inflammasome, a key component of the innate immune system, has emerged as a critical integrator of retinal stress signals. This review first delineates the molecular architecture and activation modalities of

Elevated NLRP1 and IL-1β Expression Supports Inflammasome Activation in SJS/TEN.

BACKGROUND: Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) are rare, life-threatening mucocutaneous reactions most commonly triggered by medications. While cytotoxic T-cell-mediated keratinocyte death is considered central to disease pathogenesis, the contribution of innate immune pathways, particularly NLRP1-mediated inflammasome activation, remains insufficiently characterized. METHODS: In this retrospective study, we analyzed skin biopsy specimens from 22 patients with histopathologically confirmed SJS/TEN and 18 controls with normal skin. Immunohistochemical staining

Targeting the NLRP3 Inflammasome: Novel Inhibitors for Cardiovascular Diseases Management - A Narrative Review.

The innate immune system is the first line of defense against pathogens and intracellular danger signals, providing a rapid nonspecific response to eliminate the infection and maintain tissue homeostasis. NLRP3 (NLR family pyrin domain containing 3) inflammasome, as a critical component of the innate immune system, plays a pivotal role in the inflammatory response. Many research studies have highlighted the implication of NLRP3 in the pathogenesis of various cardiovascular diseases including atherosclerosis, myocardial infarction, hypertension and heart failure. Activation of NLRP3 elicits a r

Characterizing the Activity of Inflammasome-Related Genes and Their Association With Oncological Outcomes in Prostate Cancer.

BACKGROUND: Inflammation plays a critical role in cancer cell proliferation; however, the specific role of inflammasomes, multiprotein complexes that regulate inflammation-associated signaling pathways, in prostate cancer (PCa) remains insufficiently explored. This study aims to characterize the expression of inflammasome-related genes in PCa and evaluate their association with clinical outcomes. METHODS: De-identified transcriptome data from the Decipher GRID RP, a cohort of 52,266 radical prostatectomy (RP) samples tested (2016-2024) with the Decipher prostate genomic classifier (Veracyte, …

Neddylation Targets and Stabilizes NLRP3 to Augment Inflammasome-Mediated Colitis and Mood Disorder.

The activation of NLRP3 inflammasome contributes to the development of numerous chronic inflammatory diseases, including ulcerative colitis and stress-induced anxiety. Recent studies have revealed that NLRP3 K48-linked ubiquitination by several E3 ligases, including Trim31, leads to degradation. Neddylation is a process highly similar to ubiquitination by covalently conjugating NEDD8 to lysines in specific substrate proteins. Neddylation of substrate proteins alters their subcellular localization, stability, and activity. The role of neddylation in the NLRP3 inflammasome remains elusive. Here,

Discovery of a brain penetrant NLRP3 inflammasome inhibitor that reduces neuropathic pain in mice.

In our continuing campaign to discover novel inhibitors of the nucleotide-binding oligomerization domain (NOD)-like receptor (NLR) protein 3 (NLRP3), an emerging drug target that tightly regulates the innate immune responses, a 1H-1,2,3-triazole-4-carboxamide scaffold was designed and optimized. Iterative optimization led to the identification of a lead inhibitor, 22, with an IC50 of 70 nM to suppress the release of interleukin (IL)-1β upon NLRP3 activation. Further in vivo studies supported its selective suppression of the NLRP3 inflammasome. Pharmacokinetic studies of 22 and PET imaging stud

Streptococcus anginosus activates the NLRP3 inflammasome to promote inflammatory responses from macrophages.

Chronic inflammation and oral dysbiosis are common features of oral squamous cell carcinoma (OSCC). The commensal bacterium Streptococcus anginosus is increased in oral diseases, including OSCC. Our previous work revealed that S. anginosus promotes inflammatory responses from macrophage cell lines; however, the molecular mechanism by which S. anginosus interacts with macrophages to instigate this response remains to be investigated. Here, we found S. anginosus activated primary bone marrow-derived macrophages (BMMs), which presented increased NF-κB activation and downstream inflammatory cytoki

KCNJ2 is Required for NLRP3 Inflammasome Activation That Drives Allergic Airway Inflammation and Remodeling.

Airway inflammation and remodeling are cardinal features of asthma pathogenesis. Genome-wide association studies have shown that several SNPs of KCNJ2, a member of the inwardly rectifying potassium channel family, are associated with asthma in patients. However, the role of KCNJ2 in airway inflammation and remodeling in asthma remains unknown. Here, we demonstrate that the Kcnj2 serves as a critical regulator of airway epithelial inflammation and remodeling. KCNJ2 expression is significantly reduced in the airway epithelium of asthmatic patients, which is associated with goblet cell metaplasia

NLRP3 inflammasome characterization in first-trimester, preterm, and term human villous placenta.

Precise regulation of inflammatory signaling at the maternal-fetal interface is essential for a healthy pregnancy, yet how the NLRP3 inflammasome is regulated in the placenta during pregnancy and in preterm birth remains poorly understood. We characterized NLRP3 inflammasome-associated signaling in human villous placental tissue from first-trimester (8-11 weeks), preterm (28-<37 weeks), and term (38-40 weeks) pregnancies. Gene expression, protein abundance, caspase-1 processing, and tissue cytokine concentrations were assessed using qPCR, western blot, and ELISA, alongside analysis of upstream
