Isolation and characterization of an arylalkylamine N-acetyltransferase from Drosophila melanogaster.

The enzyme arylalkylamine N-acetyltransferase (aaNAT) catalyzes the rate-limiting step in melatonin formation in the vertebrate pineal gland. Numerous attempts to purify this highly unstable enzyme from vertebrates have been unsuccessful. Here, we report the purification of an aaNAT enzyme from Drosophila melanogaster, using a radioenzymatic activity assay and column chromatography. The isolated 29.5-kDa protein acetylates tryptamine, dopamine and serotonin with affinities of 0.89 to 0.97 mM, respectively. This suggests that the identified aaNAT may be involved in melatonin synthesis and scler

The hemagglutinin proteins of clades 1 and 2.3.4.4b H5N1 highly pathogenic avian influenza viruses exhibit comparable attachment patterns to avian and mammalian tissues.

UNLABELLED: The global spread of the A/goose/Guangdong/1/96-lineage H5N1 highly pathogenic avian influenza (HPAI) viruses is accompanied by an expanded host range and the establishment of sustained viral transmission among dairy cattle. To evaluate if the evolving H5N1 viruses have changed tissue tropism over time, we compared the binding patterns of recombinant hemagglutinin (HA) proteins derived from clade 1 (A/Vietnam/1203/04, H5VN) and circulating clade 2.3.4.4b viruses detected from a wild bird (A/Eurasian Teal/Hong Kong/AFCD-HKU-23- 14009-01020/2023, H5HK) and dairy cattle (A/bovine/Ohio

'Silencing of coatomer subunit beta disrupts development and causes mortality in the citrus root weevil Diaprepes abbreviatus'.

BACKGROUND: Citrus root weevil, Diaprepes abbreviatus is one of the most destructive pests of citrus, causing extensive root damage that predisposes trees to secondary infection with phytophthora and substantial yield losses. In the present study, we identified and functionally characterized the coatomer subunit beta gene (DaCOPB), a core component of the COPI vesicle trafficking machinery responsible for two-way transport between the endoplasmic reticulum and Golgi apparatus. RESULTS: The full-length DaCOPB transcript was obtained from the de novo transcriptome of D. abbreviatus and exhibite

Lipophorin receptor knockdown reduces hatchability of kissing bug Rhodnius prolixus eggs.

Lipophorin is the primary lipoprotein present in the hemolymph of insects, responsible for the lipids' transport between organs. It interacts with specific sites on cell membranes in an essential process for transferring lipids. The lipophorin receptor is the protein responsible for the interaction between lipophorin and cell membranes. In the kissing bug Rhodnius prolixus, much information on the interaction of lipophorin with organs is available. However, molecular data on the lipophorin receptor and its functions is still needed. Here, we explored lipophorin receptor gene expression and fun

Fat body lipolysis connects poor nutrition to hypopharyngeal gland degradation in Apis mellifera.

The hypopharyngeal glands (HGs) of honey bee nurse workers secrete the major protein fraction of jelly, a protein and lipid rich substance fed to developing larvae, other worker bees, and queens. A hallmark of poorly nourished nurses is their small HGs, which actively degrade due to hormone-induced autophagy. To better connect nutritional stress with HG degradation, we looked to honey bees and other insect systems, where nutrient stress is often accompanied by fat body degradation. The fat body contains stored lipids that are likely a substrate for ecdysteroid synthesis, so we tested whether s

RNA interference-mediated functional characterization of aquaporin genes in Tribolium castaneum.

An obvious challenge faced by most terrestrial insects is maintaining water homeostasis in an arid environment. Current research suggests aquaporins may be evolved to meet the challenge. However, up to now, this suggestion has not been verified in any insect that feeds upon solid food with mandibulate mouthparts. In the present paper, nine putative aquaporin genes [Tribolium castaneum big brain, T. castaneum Drosophila integral protein (TcDrip), T. castaneum Pyrocoelia rufa integral protein (TcPrip), T. castaneum aquaporin 12-like, T. castaneum entomoglyceroporin 1 (TcEglp1), TcEglp2, TcEglp3,

Combined effects of temperature and avermectins on life history and stress response of the western flower thrips, Frankliniella occidentalis.

Temperature and pesticide are two important factors that affect survival, reproduction and other physiological processes of insects. To determine interactions of elevated temperature and avermectins treatment on the western flower thrips, Frankliniella occidentalis, newly emerged adults were exposed to combinations of three temperatures (21, 26 and 33 °C) and two avermectins concentrations (0, 45 ppm), and survival rate, reproduction, longevity, antioxidant enzymes activities and heat shock proteins (hsps) induction were analyzed. The results showed that the survival, longevity and reproductio

Extraction-dependent changes in immunoglobulin E binding profiles of processed lesser mealworm (Alphitobius diaperinus) proteins.

BACKGROUND: Alphitobius diaperinus, the lesser mealworm, is an edible insect considered safe for human consumption; however, its proteins show documented cross-reactivity with crustacean and house dust mite (HDM) allergens, raising potential concerns for allergic populations. In this study, different lesser mealworm materials were extracted using three protein extraction protocols to evaluate how methodological choices and food processing treatments influence immunoglobulin E (IgE)-binding profiles. RESULTS: All protocols yielded satisfactory protein recovery, with protocols 1 and 2 enabling

Impacts of testis-specific serine/threonine protein kinase 3 on eupyrene spermatogenesis and contributes to male sterility in Plutella xylostella.

BACKGROUND: The genetic-based sterile insect technique (gSIT) is a sustainable, eco-friendly pest control strategy, whose development hinges on identifying key fertility-associated molecular targets-particularly those regulating spermatogenesis. Spermatid development relies not only on testis-specific proteins but also on post-translational modifications such as phosphorylation, yet such targets and their regulatory pathways remain uncharacterized in the diamondback moth, Plutella xylostella, a major worldwide pest characterized by rapid reproduction. RESULTS: The testis-specific serine/threo

Molecular docking and dynamics simulation studies uncover the host-pathogen protein-protein interactions in soybean (Glycine max (L.) Merr.) and Groundnut bud necrosis virus: first report.

BACKGROUND: Soybean is a widely consumed oilseed crop with economic importance. It is enriched with numerous bioactive compounds that have health-promoting properties. Groundnut bud necrosis virus (GBNV) affects many agriculturally important crops, such as soybean, resulting in crop losses. GBNV is a single-stranded RNA virus from the genus Orthotospovirus and family Bunyaviridae. It is transmitted by insect vectors (thrips) and is further aggravated by secondary transmission from infected plants to others in the same field. There are no commercially available drugs against GBNV; thus, there i

Copy Number Variation and Expression Analysis Reveals a Nonorthologous Pinta Gene Family Member Involved in Butterfly Vision.

Vertebrate (cellular retinaldehyde-binding protein) and Drosophila (prolonged depolarization afterpotential is not apparent [PINTA]) proteins with a CRAL-TRIO domain transport retinal-based chromophores that bind to opsin proteins and are necessary for phototransduction. The CRAL-TRIO domain gene family is composed of genes that encode proteins with a common N-terminal structural domain. Although there is an expansion of this gene family in Lepidoptera, there is no lepidopteran ortholog of pinta. Further, the function of these genes in lepidopterans has not yet been established. Here, we explo

Binding site for S-adenosyl-L-methionine in a central region of mammalian reovirus lambda2 protein. Evidence for activities in mRNA cap methylation.

One or more proteins in mammalian reovirus core particles mediate two RNA methylation activities, (guanosine-7-N)-methyltransferase and (guanosine-2'-O)-methyltransferase, that contribute to forming the 5' cap 1 structure on viral mRNA. We used UV irradiation to identify core proteins that bind S-adenosyl-L-methionine (SAM), the methyl-group donor for both methyltransferases. A [methyl-3H]SAM-binding site was observed among the reovirus lambda proteins; was shown to be specific by competition with low levels of S-adenosyl-L-homocysteine, the product of methyl-group transfer from SAM; and was s

PirAB toxin from Photorhabdus asymbiotica as a larvicide against dengue vectors.

We have evaluated Photorhabdus insect-related protein (Pir) from Photorhabdus asymbiotica against dengue vectors. PirAB shows larvicidal activity against both Aedes aegypti and Aedes albopictus larvae but did not affect the Mesocyclops thermocyclopoides predator. PirAB expressed the strongest toxicity compared to PirA, PirB, or the mixture of PirA plus PirB. Whether the presence of an enterobacterial repetitive intergenic consensus sequence in PirAB, but not in PirA, PirB, or the mixture of PirA plus PirB, has any impact on biological control efficacy needs further investigation.

Three polymorphic genes encoding a depressant toxin from the Egyptian scorpion Leiurus quinquestriatus quinquestriatus.

Four clones encoding the insect depressant toxin LqqIT2 have been isolated from the Egyptian scorpion Leiurus quinquestriatus quinquestriatus using RT-PCR. The four clones have been sequenced and their deduced amino acid sequences have been compared with the original amino acid sequence determined from the purified LqqIT2 protein and polymorphisms have been shown. This study succeeded in isolating more than one copy of the LqqIT2 gene, although only one amino acid sequence has been identified from the purified LqqIT2 toxin.

Development and applications of transgenesis in the yellow fever mosquito, Aedes aegypti.

Transgenesis technology has been developed for the yellow fever mosquito, Aedes aegypti. Successful integration of exogenous DNA into the germline of this mosquito has been achieved with the class II transposable elements, Hermes, mariner and piggyBac. A number of marker genes, including the cinnabar(+) gene of Drosophila melanogaster, and fluorescent protein genes, can be used to monitor the insertion of these elements. The availability of multiple elements and marker genes provides a powerful set of tools to investigate basic biological properties of this vector insect, as well as the materi

Glucose-6-phosphate dehydrogenase. Characterization of a reactive lysine residue in the Pichia jadinii enzyme reveals a limited structural variation in a functionally significant segment.

Glucose-6-phosphate dehydrogenase from the yeast Pichia jadinii has a reactive lysine residue in a segment of amino acid sequence Ile-Asp-His-Tyr-Leu-Gly-Lys*-Glu-Met-Val-Lys. This structure differs from that of other characterized glucose-6-phosphate dehydrogenases, but outside yeasts the segment is invariant in known mammalian, insect and bacterial forms. Thus, limited structural variation is now defined within yeasts for a part of the protein otherwise strictly conserved, and for which stringent structural requirements probably relate to enzymic mechanisms.

Alternative splicing originates different domain structure organization of Lutzomyia longipalpis chitinases.

BACKGROUND The insect chitinase gene family is composed by more than 10 paralogs, which can codify proteins with different domain structures. In Lutzomyia longipalpis, the main vector of visceral leishmaniasis in Brazil, a chitinase cDNA from adult female insects was previously characterized. The predicted protein contains one catalytic domain and one chitin-binding domain (CBD). The expression of this gene coincided with the end of blood digestion indicating a putative role in peritrophic matrix degradation. OBJECTIVES To determine the occurrence of alternative splicing in chitinases of L. lo

Genome-wide analysis of differential mRNA expression of Amsacta moorei entomopoxvirus, mediated by the gene encoding a viral protein kinase (AMV197).

Insect-born entomopoxviruses (Fam: Poxviridae) are potentially important bio-pesticide against insect pests and expression vectors as well as vectors for transient human gene therapies including recombinant viral vaccines. For these reasons, it is necessary to understand the regulatory genes functions to improve its biotechnological potential. Here, we focused on the characterization of serine/threonine (Ser/Thr; ORF AMV197) protein kinase gene from the Amsacta moorei entomopoxvirus (AMEV), the type species of the genus Betaentomopoxvirus. Transcription of the parental and an amv197-null recom

Psocoptera spp. (book louse): a new major household allergen in Mumbai.

BACKGROUND: Exposure and sensitization to indoor allergens of house dust mites and insects is well established. However, there are no reports on the allergenicity of Psocoptera spp. (book louse). These insects are known to inhabit various terrestrial habitats such as tree bark (bark lice), old papers, books, stored food products, etc and gain entry into human dwellings through these modes. They have the potential to sensitize individuals with an occupational exposure to them. OBJECTIVE: Our objective is to use clinical and immunologic techniques to evaluate the extent of sensitization to Psoc

Development of a recombinant membrane protein ELISA for analyzing antibody responses against SARS-CoV-2 envelope proteins.

Recombinant surface antigens from enveloped viruses are commonly produced without the transmembrane domains to avoid difficulties associated with membrane protein expression and purification. Here, we overcame these challenges by fusing several SARS-CoV-2 envelope proteins to green fluorescent protein and a Strep-tag to facilitate protein detection and purification. This approach simplified the insect cell expression optimization and purification of recombinant prefusion stabilized full-length spike (rS-GFP), membrane (rM-GFP) and envelope (rE-GFP) proteins from the ancestral SARS-CoV-2 strain
