The Impact of Heat Stress on Dairy Cattle: Effects on Milk Quality, Rumination Behaviour, and Reticulorumen pH Response Using Machine Learning Models.

Heat stress has a major impact on dairy cow health and productivity, especially during early lactation. Conventional heat stress monitoring methods frequently rely on single indicators, such as the temperature-humidity index (THI), which may miss subtle physiological and metabolic responses. This study presents a novel threshold-based classification framework that integrates biologically meaningful combinations of environmental, behavioural, and physiological variables to detect early-stage heat stress responses in dairy cows. Six composite heat stress conditions (C1-C6) were developed using r

Plant metabolites in nanotechnology: are we missing the link to stress resilience and sustainability?

Plant metabolites used in green nanoparticle synthesis are biologically active compounds whose functions may be retained or altered during nanoparticle formation, with important implications for plant stress resilience, plant-environment interactions, and environmental sustainability. Plant-derived metabolites are increasingly used in green nanoparticle synthesis, primarily for their reducing, capping, and stabilising properties. However, their biological functions within plants are rarely considered when evaluating nanoparticle-mediated effects on plant stress responses or environmental susta

Quercetin Antagonizes Doxorubicin-Induced Cardiotoxicity via HO-1/PGC-1α-ALOX5 Axis: Arachidonic Acid Metabolism-Ferroptosis Crosstalk as a Therapeutic Target.

Myocardial injury caused by Doxorubicin has limited its clinical application. Our research has demonstrated that doxorubicin induces various forms of cell death, oxidative stress, and metabolic abnormalities. We have confirmed, through both in vitro and in vivo studies, that apoptosis, iron overload, inflammatory responses, and arachidonic acid metabolism contribute to doxorubicin's cardiotoxic effects. Treatment with quercetin effectively reduces iron accumulation and preserves mitochondrial structural integrity by inhibiting oxidative stress and inflammatory responses. Using molecular dockin

CBP-HSF2 structural and functional interplay in Rubinstein-Taybi neurodevelopmental disorder.

Patients carrying autosomal dominant mutations in the histone/lysine acetyl transferases CBP or EP300 develop a neurodevelopmental disorder: Rubinstein-Taybi syndrome (RSTS). The biological pathways underlying these neurodevelopmental defects remain elusive. Here, we unravel the contribution of a stress-responsive pathway to RSTS. We characterize the structural and functional interaction between CBP/EP300 and heat-shock factor 2 (HSF2), a tuner of brain cortical development and major player in prenatal stress responses in the neocortex: CBP/EP300 acetylates HSF2, leading to the stabilization o

Leptin regulates dopamine responses to sustained stress in humans.

Neural systems that identify and respond to salient stimuli are critical for survival in a complex and changing environment. In addition, interindividual differences, including genetic variation and hormonal and metabolic status likely influence the behavioral strategies and neuronal responses to environmental challenges. Here, we examined the relationship between leptin allelic variation and plasma leptin levels with DAD2/3R availability in vivo as measured with [(11)C]raclopride PET at baseline and during a standardized pain stress challenge. Allelic variation in the leptin gene was associat

The Influence of Psychological Stress on HPV Infection Manifestations and Carcinogenesis.

Psychological stress is an important factor involved in disease manifestations of human papillomavirus (HPV) infection, and it can participate in HPV-associated carcinogenesis. The impact or effect which stress can have (exert) depends on a person's genetic pool, experiences and behaviors. Due to inconsistencies in some study results, this issue remains a subject of research. Concerning the course of HPV manifestations, it has been observed that a higher number of life stressors in at least the previous 6 months, the absence of social support and the types of personal coping mechanisms employe

An Integrated Multi-Biomarker Perspective on Acute Exercise: Differential Orexin, Alpha-Amylase, and Cortisol Responses Across Modalities.

Purpose: Acute exercise elicits coordinated neuroendocrine and autonomic responses that are strongly influenced by exercise modality and intensity. While cortisol and salivary alpha-amylase are widely used as peripheral indicators of hypothalamic-pituitary-adrenal (HPA) axis and sympathetic nervous system activation, respectively, salivary orexin remains an exploratory biomarker whose relationship with central orexinergic activity is not yet fully established, particularly in response to different exercise conditions. This study aimed to compare the acute salivary responses of orexin, alpha-am

Exercise alone or combined with functional foods in aging and age-related diseases: targeting DNA damage, repair pathways, and genome integrity.

Aging is fundamentally associated with the gradual accumulation of DNA damage, which is instrumental in the initiation and advancement of age-associated pathologies, including neurodegenerative conditions such as Alzheimer's disease and Parkinson's disease. Recent studies indicate that lifestyle modifications-especially the consumption of functional foods and participation in consistent physical exercise-can influence cellular and molecular processes related to DNA repair, reduction of oxidative stress, and maintenance of genomic integrity. This review consolidates contemporary research regard

Genetic interaction between F-box motif encoding YDR131C and retrograde signaling-related RTG1 regulates the stress response and apoptosis in Saccharomyces cerevisiae.

The retrograde signaling pathway is well conserved from yeast to humans, which regulates cell adaptation during stress conditions and prevents cell death. One of its components, RTG1 encoded Rtg1p in association with Rtg3p communicates between mitochondria, nucleus, and peroxisome during stress for adaptation, by regulation of transcription. The F-box motif protein encoded by YDR131C  constitutes a part of SCF Ydr131c -E3 ligase complex, with unknown function; however, it is known that retrograde signaling is modulated by the E3 ligase complex. This study reports epistasis interaction between

Inflammasome-independent NLRP3 function enforces ATM activity in response to genotoxic stress.

NLRP3 is a pattern recognition receptor with a well-documented role in inducing inflammasome assembly in response to cellular stress. Deregulation of its activity leads to many inflammatory disorders including gouty arthritis, Alzheimer disease, and cancer. Whereas its role in the context of cancer has been mostly explored in the immune compartment, whether NLRP3 exerts functions unrelated to immunity in cancer development remains unexplored. Here, we demonstrate that NLRP3 interacts with the ATM kinase to control the activation of the DNA damage response, independently of its inflammasome act

Smartphone-assisted upconversion nanoparticles-aptamer-based probe functionalized hydrogel microneedles for rapid interstitial fluid lactate profiling.

Rapid assessment of lactate dynamics is important for evaluating tissue hypoxia, metabolic stress, and exercise-induced physiological responses. However, conventional lactate measurements predominantly rely on blood sampling and enzyme-based assays, making them poorly suited for rapid and minimally invasive assessment of localized lactate dynamics in interstitial fluid (ISF). Here, we developed a rare-earth nanomaterial-aptamer probe-functionalized composite hydrogel microneedle patch for rapid ISF lactate profiling. The sensing system integrates fluorescence resonance energy transfer (FRET)-b

Exosomal microRNAs as Central Regulators of Cancer Cachexia: Multi-Omics Insights into Muscle Wasting and Adipose Browning.

Cancer cachexia is a multifactorial syndrome marked by involuntary weight loss, skeletal muscle wasting, adipose tissue remodeling, and systemic metabolic dysfunction. Exosome-derived microRNAs (miRNAs) have emerged as key mediators, reprogramming host tissues and driving these hallmarks. However, no integrated framework has linked exosomal miRNAs to the proteomic and metabolomic alterations that characterize cachexia. This review critically synthesizes evidence on exosomal miRNAs in muscle atrophy, adipose browning, and systemic metabolic disruption. Tumor-secreted exosomal miRNAs activate pr

Lunatin-1: A peptide derived from the venom of the Hadruroides lunatus scorpion modulates signaling pathways in HL60 tumor cells to induce cytotoxic effects.

Lunatin-1 is a 13-residue cytotoxic peptide derived from the venom of the scorpion Hadruroides lunatus. This study investigated its early effects on cellular signaling in the human promyelocytic leukemia cell line HL-60 using integrated proteomics and phosphoproteomics. Lunatin-1 regulated key mediators of apoptosis, such as caspase-2 (CASP2) and MEK1 (MAP2K1), and impacted major signaling pathways such as MAPK and PI3K/AKT. Lunatin-1 induced caspase-dependent and -independent apoptotic signaling, reduced AKT1 phosphorylation, and promoted BAX activation, consistent with mitochondrial apoptosi

Engineering epicardium-integrated human iPSC-derived heart tissue for modelling Hutchinson-Gilford progeria syndrome.

Engineered heart tissues (EHTs) have been used in cardiovascular research for nearly three decades, addressing limitations of animal models and oversimplified two-dimensional single-cell systems in recapitulating cardiac (patho) physiology. Recently, epicardium-integrated EHTs have gained attention as therapeutic tools for remuscularizing the failing heart. The epicardium, the outermost mesothelial layer of the heart, plays a crucial role in cardiac development and disease by serving as a source of progenitor cells and by providing essential paracrine signalling. Hutchinson-Gilford progeria sy

Superior osmotic stress tolerance in oilseed rape transformed with wild-type Rhizobium rhizogenes.

Natural transformation with R. rhizogenes enhances osmotic stress tolerance in oilseed rape through increasing osmoregulation capacity, enhancing maintenance of hydraulic integrity and total antioxidant capacity. Transformation of plants using wild strains of agrobacteria is termed natural transformation and is not covered by GMO legislation in, e.g., European Union and Japan. In this study, offspring lines of Rhizobium rhizogenes naturally transformed oilseed rape (Brassica napus), i.e., A11 and B3 (termed root-inducing (Ri) lines), were investigated for osmotic stress resilience. The opposite effects of stringent response on phage infection of Pseudomonas putida.

Guanosine tetra- and pentaphosphate ((p)ppGpp) are one of the key players in the stress response of bacteria. Accumulation of these alarmones activates the stringent response, usually triggered by different nutritional stresses. For Pseudomonas putida, there is only limited data available on the importance of the stringent response in stress situations. Also, in recent years, different specific phage defence systems have received much attention, but little is known about the involvement of stringent response in phage infection. Here, we show that P. putida PaW85 (p)ppGpp0 is prototrophic and t

Protein persulfidation: a ubiquitous modification regulating a broad spectrum of biological processes.

Hydrogen sulfide signaling occurs mainly through protein persulfidation, an important post-translational modification and a highly dynamic process in plants. Beyond enzyme activity, persulfidation affects protein localization, structure, and interactions. This review provides an overview of the mechanisms governing this modification and the pivotal role of the thioredoxin system mediating depersulfidation, which is essential for maintaining redox homeostasis. Proteomic studies in Arabidopsis thaliana and crop species reveal that protein persulfidation is a widespread post-translational modific

Review: Additives delivery through drinking water to support gut health in swine and poultry - a systematic literature review.

In swine and poultry, early-life stressors such as weaning and heat stress can impair gut health and growth performance. While additives are commonly added to feed, reduced feed intake under stress may limit their effectiveness. In contrast, water intake remains relatively constant, making drinking water a promising alternative delivery route. This systematic review assessed the impact of water-delivered additives-including probiotics, plant extracts and essential oils, amino acids, and vitamins-on gut health and performance in piglets and broilers. A structured search was performed from Febru
