Release of insulin and pancreatic somatostatin in response to increased circulating growth hormone (GH) levels.

In an experimental study of 5 anaesthetized pigs the effects of increased circulating levels of growth hormone (GH) on the release of insulin and somatostatin from the pancreas was investigated. Blood was sampled simultaneously from the pancreatic venous effluent and from mixed venous blood. As measured by radioimmunoassay infusion of GH (20 micrograms x kg-1 x h-1) resulted in a significant (p less than 0.01) increase both in insulin and somatostatin concentration in pancreatic venous blood. In mixed venous blood no significant changes in hormone levels were found. The results illustrate one

B-cell nesidioblastosis in the overweight newborn.

The endocrine pancreata of five overweight newborns (birthweight ranging from 3,800 g to 4,500 g) have been studied. Basal blood glucose and insulin levels were normal in these newborns. A marked and widespread B-cell nesidioblastosis was observed in relation to ten controls (birthweight ranging from 2,900 to 3,500 g). Morphometric analysis showed in the overweight newborns an increase in endocrine tissue, with special reference to B cells. These morphological aspects of the endocrine pancreas in relation to the birthweight were discussed.

Pancreatic polypeptide (PP)-like immunoreactivity in the pancreatic islets of the anglerfish (Lophius americanus) and the channel catfish (Ictalurus punctatus).

Pancreatic polypeptide (PP) immunoreactivity was demonstrated in the endocrine pancreas of the anglerfish (Lophius americanus) and the channel catfish (Ictalurus punctatus) using immunohistochemistry. In both species, PP-immunoreactive cells were localized at the periphery of endocrine tissue. In Lophius americanus, PP-immunoreactivity could be localized in the principal islet, the pyloric or secondary islet, and the mesenteric or tertiary islets. A peripheral localization of PP-immunoreactive cells having characteristic angular, or dendritic, appearance demonstrated in the above two species a

[Endocrine pancreas function and diabetes mellitus].

Carbohydrate metabolism is regulated by the anabolic action of insulin and by the catabolic effect of glucagon. Catabolic actions predominate in diabetes mellitus. Somatostatin exeits a diabetogenic action by inhibition of insulin release. The physiologic role of pancreatic polypeptide is unknown.

Recovery of pancreatic beta cells in response to long-term normoglycemia after pancreas or islet transplantation in severely streptozotocin diabetic adult rats.

In the well-established, high-dose streptozotocin diabetic rat model, it is unknown whether normoglycemia after pancreas or islet transplant … diabetes mellitus in adult rats due to a selective destruction of the pancreatic beta cells in the islets of Langerhans. At doses between 50 and 60 mg/kg of body weight, only very few beta cells survive in the pancreas (1-3). The diabetic state is irreversible and insulin-dependent, thus representing an experimental animal model for type I diabetes (2). Because of th …

Inhibition of pancreatic glucagon gene expression in mice bearing a subcutaneous glucagon-producing GLUTag transplantable tumor.

Transgenic mice that express a glucagon gene-simian virus-40 large T-antigen (GLUTag) fusion gene develop neuroendocrine carcinoma of the la … rrying the GLUTag tumor for 6-8 weeks demonstrated a marked reduction in the number and size of the islets of Langerhans and a disproportionately greater decrease in the number of cells exhibiting glucagon immunoreactivity. By electron microscopy, the residual A-cells were small, compressed at the periphery of the islets, and had poorly developed cytoplasmic organelle …

Local and Systemic Immunologic Profiles Differentiate Accepted and Rejected Islet Grafts in a Rat Anterior Chamber Model.

The anterior chamber of the eye (ACE) provides a minimally invasive, immune-privileged site for pancreatic islet engraftment. However, loss of immune privilege during revascularization can trigger inflammation and graft rejection. In this study, we compared ocular and systemic immunologic responses associated with accepted and rejected islet allografts in the ACE. Lewis rat recipients underwent ACE allogeneic islet transplantation without immunosuppressive agents. Ocular grafts and spleens were examined by histology and immunofluorescence for effector and regulatory T-cell (Treg) populations.

Transplantation of chemically induced pluripotent stem-cell-derived islets under abdominal anterior rectus sheath in a type 1 diabetes patient.

We report the 1-year results from one patient as the preliminary analysis of a first-in-human phase I clinical trial (ChiCTR2300072200) assessing the feasibility of autologous transplantation of chemically induced pluripotent stem-cell-derived islets (CiPSC islets) beneath the abdominal anterior rectus sheath for type 1 diabetes treatment. The patient achieved sustained insulin independence starting 75 days post-transplantation. The patient's time-in-target glycemic range increased from a baseline value of 43.18% to 96.21% by month 4 post-transplantation, accompanied by a decrease in glycated

The Effect of a Fast-Releasing Hydrogen Sulfide Donor on Vascularization of Subcutaneous Scaffolds in Immunocompetent and Immunocompromised Mice.

Islet transplantation into subcutaneous polymer scaffolds has shown to successfully induce normoglycemia in type 1 diabetes models. Vascularization of these scaffolds is imperative for optimal control of glucose levels. We studied the effect of the vascular stimulator hydrogen sulfide (H2S) on the degree of vascularization of a scaffold and the role of the immune system in this process. Scaffolds were subcutaneously implanted in immunocompetent C57BL/6 and immunocompromised nude mice. Mice received twice-daily intraperitoneal injections of the fast-releasing H2S donor sodium hydrosulfide (NaHS
