Pycnodysostosis is a rare disease with very few reported cased all over the world. It was first described in 1963 by Maroteaux and Lamy. It is also known as Toulouse-Lautrec syndrome, after a French artist, Henri de Toulouse Lautrec. The affected gene, CTSK, was first isolated in 1996. It is an autosomal recessive osteochondrodysplasia, characterized by disrupted function of osteoclasts. Incidence of this disease is 1.7 per 1 million births with a male to female ratio of 1:1 30% cases arise from consanguineous marriages.

A 6-year-old boy with Down's syndrome presented with recurrent episodes of asymptomatic peeling of the skin from the trunk, palms, soles, and face since he was 2 years old. He was the first child of a non-consanguineous marriage. There was no history of any seasonal aggravation or variation. The peeling occurred once or twice a month. Several pediatricians and dermatologists diagnosed it to be either a staphylococcal scalded skin syndrome or a drug reaction. A temporal correlation could not be established with any drugs, including cefixime, amoxicillin, and paracetamol.

Segregation analysis and consanguinity analysis were applied to 37 albinism families ascertained from Shandong province (P. R. China) to study the genetic heterogeneity of the disease. Our results show that albinism is an autosomal recessive disease with multilocus. The least number of the detrimental loci is 8, The average frequence of detrimental genes is 0.0023, and 3.83% individuals in general population are carriers. The incidence among the offsprings of the first cousin marriages would be 22 times higher than that in the general population.

The impact of rheumatoid arthritis (RA) on the families of people with the disease is poorly understood. A qualitative study was undertaken on 22 patients with rheumatoid arthritis, their respective well partners and their 40 children. For most children the effect of living with a parent suffering from a painful, chronic illness was not detrimental, but a minority suffered verbal and physical abuse.
