Maternal Bisphenol S Exposure Impairs Testicular Development and Sperm Function in Male Offspring by Disrupting the Immune-Endocrine Network.

Bisphenol S (BPS), a widely used substitute for bisphenol A, is an environmental contaminant with endocrine-disrupting properties. However, the transgenerational impact of maternal BPS exposure during gestation on male offspring reproductive health and the underlying molecular mechanisms remains unknown. This study shows that gestational exposure of mice to environmentally relevant concentrations of BPS (3, 30, and 300 µg/kg) dose- and time-dependently impairs testicular development and sperm function in male offspring. Integrated transcriptomic, physiological, and molecular analyses revealed

Vaccine- and infection-induced pneumococcal and DENV-NS1 antibodies reveal differential transplacental transfer and maternal-infant early exposure burden.

BACKGROUND: Pregnant women and their infants are vulnerable to severe infections with S. pneumoniae and dengue virus (DENV), for which early protection depends on maternally derived antibodies. Infection- and vaccine-induced responses may differ in quality and transfer, with implications for maternal immunization strategies. We evaluated isotype-specific antibody responses to pneumococcal vaccination and natural DENV infection in pregnant women and their infants and quantified early postnatal DENV exposure. METHODS: In a longitudinal cohort in southern Colombia, a DENV-hyperendemic region, 14

Maternal Dipyrone Exposure and Risk of Major Congenital Malformations, Adverse Perinatal and Postnatal Outcomes: A Population-Based Cohort Study.

Pain and fever are common in pregnancy. Dipyrone is widely used in Europe, Latin America, and parts of Asia, but is banned in other countries due to concerns about agranulocytosis. Evidence on its safety in pregnancy remains limited and inconsistent. This study aimed to evaluate whether maternal dipyrone use during the first- and third-trimesters is associated with major congenital malformations (MCMs) and adverse perinatal or early neonatal outcomes. A population-based cohort of all pregnancies that delivered or underwent elective termination at Soroka University Medical Center from 1998 to 2

Maternal exposure to Helicobacter pylori impairs spatial and recognition memory in rat offspring: Ameliorative effect of monoamine oxidase inhibition.

During recent years, researchers have extensively investigated various health issues associated with prenatal challenges and the subsequent outcomes emerging in the next generation. In this respect, prenatal exposure to infections is correlated with the development of cognitive deficits in offspring. There is evidence indicating that disturbance of maternal microbiota adversely affects the gut-brain axis, which in turn could compromise fetal development and cause persistent physiological defects. Previous studies have reported that Helicobacter pylori infection is correlated with cognitive dec

Gestational GenX Exposure Induces Maternal Hepatotoxicity by Disrupting the Lipid and Bile Acid Metabolism Distinguished from PFOA-Induced Pyroptosis.

Perfluorooctanoic acid (PFOA) and its replacement, GenX, are per- and polyfluoroalkyl substances (PFASs) widely used in industrial and consumer applications. Pregnant women are a vulnerable population to environmental pollutants. The maternal effects of GenX and PFOA exposure during pregnancy have not been fully elucidated. In this study, pregnant mice received daily oral doses of GenX (2 mg/kg/day), PFOA (1 mg/kg/day), or Milli-Q water (control) throughout gestation. Histopathological analyses revealed significant liver abnormalities in both exposure groups, including hepatocyte swelling, cel

Association of early pregnancy warm season exposure and neighborhood heat vulnerability with adverse maternal outcomes: A retrospective cohort study.

INTRODUCTION: Rising ambient temperatures threaten vulnerable populations such as pregnant women, with urban populations bearing a greater risk due to the urban heat island effect. Here, we assessed the independent effects of trimester-specific warm season exposure during pregnancy and neighborhood heat vulnerability on maternal outcomes, including gestational diabetes, hypertensive disorders of pregnancy, genitourinary infections, and operative delivery. METHODS: This retrospective study analyzed 819 participants from the Stress in Pregnancy Study (2009-2014), a longitudinal birth cohort stu

Impact of maternal stress and COVID-19 exposure during pregnancy on offspring neurodevelopment: signature cohort 12 month follow-up.

The COVID-19 pandemic brought unprecedented global challenges. Amid the crisis, the potential impact of COVID-19 exposure on the neurodevelopment of offspring born to infected mothers emerged as a critical concern. This is a prospective cohort study of pregnant women and their offspring enrolled in the Signature project at Hospital Universitario Virgen del Rocio in Seville, Spain, between 01/01/2024 and 08/31/2022. Participants were subsequently classified into SARS-CoV-2 exposed (mild or severe COVID-19) and non-exposed groups. Maternal stress was assessed through the Perceived Stress Scale;

In utero exposure to maternal diabetes exacerbates dietary sodium intake-induced endothelial dysfunction by activating cyclooxygenase 2-derived prostanoids.

Prenatal exposure to maternal diabetes has been recognized as a significant cardiovascular risk factor, increasing the susceptibility to the emergence of conditions such as high blood pressure, atherosclerosis, and heart disease in later stages of life. However, it is unclear if offspring exposed to diabetes in utero have worse vascular outcomes on a high-salt (HS) diet. To test the hypothesis that in utero exposure to maternal diabetes predisposes to HS-induced vascular dysfunction, we treated adult male wild-type offspring (DM_Exp, 6 mo old) of diabetic Ins2+/C96Y mice (Akita mice) with HS (

Think zinc: Transient nutritional deficiency related to novel maternal SLC30A2 mutation potentially precipitated by antenatal proton pump inhibitor exposure.

A second-born breastfed infant presented with zinc deficiency. His mother had a novel heterozygous mutation in SLC30A2. A previous baby did not have zinc deficiency but the mother had taken a proton pump inhibitor (PPI) during the second pregnancy. Antenatal PPI exposure may plausibly contribute to transient infantile zinc deficiency.

In utero exposure to maternal anti-aquaporin-4 antibodies alters brain vasculature and neural dynamics in male mouse offspring.

The fetal brain is constantly exposed to maternal IgG before the formation of an effective blood-brain barrier (BBB). Here, we studied the consequences of fetal brain exposure to an antibody to the astrocytic protein aquaporin-4 (AQP4-IgG) in mice. AQP4-IgG was cloned from a patient with neuromyelitis optica spectrum disorder (NMOSD), an autoimmune disease that can affect women of childbearing age. We found that embryonic radial glia cells in neocortex express AQP4. These cells are critical for blood vessel and BBB formation through modulation of the WNT signaling pathway. Male fetuses exposed

Impact of prenatal maternal cytokine exposure on sex differences in brain circuitry regulating stress in offspring 45 years later.

Stress is associated with numerous chronic diseases, beginning in fetal development with in utero exposures (prenatal stress) impacting offspring's risk for disorders later in life. In previous studies, we demonstrated adverse maternal in utero immune activity on sex differences in offspring neurodevelopment at age seven and adult risk for major depression and psychoses. Here, we hypothesized that in utero exposure to maternal proinflammatory cytokines has sex-dependent effects on specific brain circuitry regulating stress and immune function in the offspring that are retained across the lifes

Maternal non-steroidal anti-inflammatory drug exposure during pregnancy and risk of miscarriage: a systematic review and meta-analysis.

BACKGROUND: Numerous studies have suggested that non-steroidal anti-inflammatory drugs (NSAIDs) might be associated with increased risk of miscarriage. However, these results are conflicting and inconclusive. METHODS: We performed this systematic review and meta-analysis to assess the relationship between NSAIDs exposure and risk of miscarriage. A systematic literature search was conducted to identify relevant studies published from the time of database inception until June 2021. RESULTS: A total of ten studies involving 207,341 pregnant women were subjected to meta-analysis. There was no st

Association of uterine activity and maternal volatile anesthetic exposure during open fetal surgery for spina bifida: a retrospective analysis.

BACKGROUND: Recent warnings postulate a possible damaging effect of volatile anesthetics on the fetus. In our archive of fetal surgeries, we found wide variation in dosing of volatile anesthetics during spina bifida surgeries. We hypothesized that there was an association between volatile anesthetic exposure and uterine activity. METHODS: Sixty anesthesia records from spina bifida operations were assessed. We analyzed the course of the administered volatile anesthetic during surgery and calculated from each patient's anesthesia record the volatile anesthetic exposure expressed in vol%h. We di

Measuring the Outcomes of Maternal COVID-19-related Prenatal Exposure (MOM-COPE): study protocol for a multicentric longitudinal project.

INTRODUCTION: COVID-19 is a highly infectious respiratory disease that rapidly emerged as an unprecedented epidemic in Europe, with a primary hotspot in Northern Italy during the first months of 2020. Its high infection rate and rapid spread contribute to set the risk for relevant psychological stress in citizens. In this context, mother-infant health is at risk not only because of potential direct exposure to the virus but also due to high levels of stress experienced by mothers from conception to delivery. Prenatal stress exposure associates with less-than-optimal child developmental outcome

Maternal di-(2-ethylhexyl) phthalate exposure alters hepatic insulin signal transduction and glucoregulatory events in rat F1 male offspring.

Di-(2-ethylhexyl) phthalate (DEHP) is a commonly used plasticizer with endocrine disrupting properties. Its widespread use resulted in constant human exposure including fetal development and postnatal life. Epidemiological and experimental data have shown that DEHP has a negative influence on glucose homeostasis. However, the evidence regarding the effect of maternal DEHP exposure on hepatic glucose homeostasis is scarce. Hence, we investigated whether DEHP exposure during gestation and lactation disrupts glucose homeostasis in the rat F1 male offspring at adulthood. Pregnant rats were divided

Maternal exposure to an n-3 polyunsaturated fatty acid diet decreases mammary cancer risk of female offspring in adulthood.

Maternal exposure to dietary factors during pregnancy influences the risk of many adult-onset diseases in the later life of offspring. Here, we investigated the effects of maternal n-3 polyunsaturated fatty acid (PUFA) diet on breast cancer risk of female offspring. Pregnant C57BL/6J mice were fed a normal diet (control group), or a high-fat diet rich in safflower oil (SO), fish oil (FO) or flaxseed oil (FSO) (n = 10) throughout gestation and lactation. Their female offspring were fed an AIN-93G diet from weaning. Tumor incidences in offspring induced by 7,12-dimethylbenz[α]anthracene (DMBA) w…

Experimental maternal treatment with dexamethasone during lactation induces neonatal testicular and epididymal oxidative stress; Implications for early postnatal exposure.

Maternal treatment with dexamethasone during lactation alters reproductive functions and increases serum corticosterone in the male offspring. Excess corticosterone may induce oxidative stress. This study was designed to evaluate the effects of maternal treatment with dexamethasone during lactation on oxidative stress indices in the testis and epididymis of a male offspring. Twenty lactating dams were divided into 4 groups (n=5). Group 1 was administered 0.02ml/100g/day normal saline subcutaneously at lactation days 1-21. Groups 2, 3, and 4 were administered 100μg/kg/day dexamethasone (Dex) su

Gestational and lactational exposure to bisphenol AF in maternal rats increases testosterone levels in 23-day-old male offspring.

During prenatal and postnatal development, exposure to environmental chemicals with estrogenic activity, such as bisphenol AF (BPAF), may result in reproductive disorders. Currently, the mechanisms behind such disorders in male offspring induced by gestational and lactational exposure to BPAF remain poorly understood. Here, female rats from gestational day (GD) 3-19 were exposed to 100 mg BPAF/kg/day by oral gavage. On the day of birth (postnatal day (PD) 0), cross-fostering took place between treated and control litters, and cross-fostered mother rats were given BPAF 100 mg/kg/day during the
