Longitudinal Analysis of the Phenotype, Transcriptional Profile, and Anatomic Location of Memory CD8 T Cell Subsets after Acute Viral Infection.

Increased demand for novel, highly effective vaccination strategies necessitates a better understanding of long-lived memory CD8 T cell differentiation. To achieve this understanding, we used the mouse model of acute lymphocytic choriomeningitis virus (LCMV) infection. We reexamined classical memory CD8 T cell subsets and performed in-depth, longitudinal analysis of their phenotype, transcriptional programming, and anatomic location within the spleen. All analyses were performed at multiple time points from 8 days to 1 year postinfection. Memory subsets are conventionally defined by their expr

Precise memory for pure tones is predicted by measures of learning-induced sensory system neurophysiological plasticity at cortical and subcortical levels.

Despite identical learning experiences, individuals differ in the memory formed of those experiences. Molecular mechanisms that control the neurophysiological bases of long-term memory formation might control how precisely the memory formed reflects the actually perceived experience. Memory formed with sensory specificity determines its utility for selectively cueing subsequent behavior, even in novel situations. Here, a rodent model of auditory learning capitalized on individual differences in learning-induced auditory neuroplasticity to identify and characterize neural substrates for sound-s

Longitudinal effects of pediatric concussion into adulthood.

Each year, millions of children are diagnosed with concussions. This review summarizes current evidence on the long-term effects of pediatric concussion into adulthood. Findings regarding long-term neuropsychiatric outcomes are heterogeneous. Advanced neuroimaging studies provide limited evidence of lasting neurobiological alterations, and long-term studies remain scarce. Rodent models that mimic adolescent mild traumatic brain injury consistently demonstrate chronic neuroinflammation, neuronal loss, and memory deficits. Overall, conclusions are constrained by substantial methodological hetero

Intranasal unadjuvanted LcrV boosts parental Yersinia OMV primed lung immunity against pneumonic plague in mice.

Pneumonic plague remains a high-consequence respiratory disease, highlighting the need for vaccines that induce durable lung-localized immunity. We show that intramuscular prime-boost vaccination with Alum-absorbed OMV46-LcrV (OMV46-LcrV/Alum) is safe but provides limited long-term protection against respiratory Yersinia pestis challenge in mice. To enhance lung immunity, we deliver an adjuvant-free intranasal LcrV "spike" following IM OMV46-LcrV/Alum priming. This "prime-spike" regimen is well tolerated and confers complete short- and long-term protection against high-dose pulmonary Y. pestis

Neuroanatomical correlates of memory deficits in tuberous sclerosis complex.

Tuberous sclerosis complex (TSC) is a multisystem syndrome classically associated with the occurrence of focal brain dysplasias. We used structural magnetic resonance imaging to test for neuroradiological abnormalities in TSC (tubers, white matter lesions, and subependymal nodules) and to explore the relationships between these lesions and computational morphometric abnormalities of gray and white matter distribution. We tested memory function in TSC and investigated the relationship between memory function and both morphometric variation and lesion load. Patients demonstrated deficits bilater

Case Report: acute and long-term effects of a massive beta-mercaptoethanol inhalation.

INTRODUCTION: β-mercaptoethanol is widely used in biochemistry and industrial applications. Animal toxicity has been demonstrated but very limited data exists regarding its human effects. CASE DESCRIPTION: A 40-year-old male patient was exposed to β-mercaptoethanol vapours after an accidental occupational spillage. He developed dizziness, blurred vision, a headache and epistaxis within minutes alongside dyspnoea and cough in the following hours. Lung function tests displayed bronchial hyperreactivity and increased resistances, consistent with a reactive airways dysfunction syndrome. Neuropsyc

Rescue of astrocyte activity by the calcium sensor STIM1 restores long-term synaptic plasticity in female mice modelling Alzheimer's disease.

Calcium dynamics in astrocytes represent a fundamental signal that through gliotransmitter release regulates synaptic plasticity and behaviour. Here we present a longitudinal study in the PS2APP mouse model of Alzheimer's disease (AD) linking astrocyte Ca2+ hypoactivity to memory loss. At the onset of plaque deposition, somatosensory cortical astrocytes of AD female mice exhibit a drastic reduction of Ca2+ signaling, closely associated with decreased endoplasmic reticulum Ca2+ concentration and reduced expression of the Ca2+ sensor STIM1. In parallel, astrocyte-dependent long-term synaptic pla

Collaborative object handling supports autobiographical recall in Alzheimer's disease.

Autobiographical memory is impaired in Alzheimer's disease (AD), particularly episodic richness and phenomenological detail. Grounded in embodied cognition, this study examined whether collaborative object handling, combining sensorimotor and social interaction, could support autobiographical recall in AD. We recruited 22 patients with mild-to-moderate AD and 20 matched older controls to complete two autobiographical memory conditions in a within-subject design: a condition involving autobiographical recall without objects and a condition involving autobiographical recall supported by collabor

Neural conflict-control mechanisms improve memory for target stimuli.

According to conflict-monitoring models, conflict serves as an internal signal for reinforcing top-down attention to task-relevant information. While evidence based on measures of ongoing task performance supports this idea, implications for long-term consequences, that is, memory, have not been tested yet. Here, we evaluated the prediction that conflict-triggered attentional enhancement of target-stimulus processing should be associated with superior subsequent memory for those stimuli. By combining functional magnetic resonance imaging (fMRI) with a novel variant of a face-word Stroop task t

Functional memory T cells are derived from exhausted clones and expanded by checkpoint blockade.

Immune checkpoint blockade can facilitate tumor clearance by T cells, resulting in long term patient survival. However, the capacity of exhausted CD8+ T cells (Tex), present during chronic antigen exposure, to form memory after antigen clearance remains unclear. Here, we performed longitudinal single cell RNA/T cell receptor sequencing and ATAC-sequencing on antigen-specific T cells after the clearance of chronic lymphocytic choriomeningitis virus (LCMV) infection. These data revealed the formation of a robust population of memory CD8+ T cells that transcriptionally, epigenetically, and functi

Effects of Transcranial Direct Current Stimulation on Episodic Memory in Older Adults: A Meta-analysis.

OBJECTIVES: In the last two decades, the number of intervention studies using transcranial direct current stimulation (tDCS) has grown enormously. Though some studies have shown positive influences on episodic memory among older adults, disagreement exists in the literature. Therefore, the current meta-analysis aimed to provide a quantitative assessment of the efficacy of tDCS in modulating episodic memory functions in older adults. METHOD: Eligible studies were sham-controlled trials examining the effects of anodal tDCS on episodic memory in older adults. Twenty-four articles comprising 566

Disruption of hippocampal mitochondrial function underlies opioid-induced postoperative cognitive dysfunction in aged rats.

Postoperative cognitive dysfunction (POCD) is a common and persistent complication in aging individuals following surgery, particularly when opioids are used for perioperative pain management. Although opioids are widely administered in the perioperative setting, the mechanisms by which they contribute to long-term cognitive impairment remain poorly understood. Here, using an aged rat model of surgery with perioperative morphine administration, we investigated how synaptic, axonal, and mitochondrial abnormalities contribute to persistent hippocampal memory deficits, and evaluated therapeutic s

Prospective Memory, Sustained Attention and Response Inhibition in Poly-Substance Users Stable on Methadone Maintenance Treatment.

Background: Prospective memory and response inhibition are interrelated constructs, though studied separately in the drug addiction literature. Also, although sustained attention underlies response inhibition, its role in the relation between these functions has been largely neglected. The limited research on the cognitive effects of methadone-maintenance treatment (MMT) further stresses the importance of investigating these effects. Objective: Therefore, the current study focused on possible effects of MMT combined with long-term drug abuse on all these functions. Thirty five long term opiate

Choice-predictive activity in parietal cortex during source memory decisions.

Neurobiological research has classically focused on perceptual decision-making, although many real-life decisions are based on information that is not currently available but stored in long-term memory. Previous studies have suggested that the lateral parietal cortex encodes decision-related signals during item recognition judgments. In the present fMRI study, we employed a parametric manipulation of evidence for source memory judgments and tested several hypotheses concerning memory decision signals in parietal cortex. As expected, the mean BOLD signal in several parietal regions was modulate

Reducing hippocampal extracellular matrix reverses early memory deficits in a mouse model of Alzheimer's disease.

Alzheimer's disease is caused by increased production or reduced clearance of amyloid-β, which results in the formation amyloid-β plaques and triggers a cascade of downstream events leading to progressive neurodegeneration. The earliest clinical symptoms of Alzheimer's disease, i.e., memory loss, are however poorly understood from a molecular and cellular perspective. Here we used APPswe/PS1dE9 (APP/PS1) transgenic mice to study the early pre-pathological effects of increased amyloid-β levels on hippocampal synaptic plasticity and memory. Using an unbiased proteomics approach we show that the

The visual and semantic features that predict object memory: Concept property norms for 1,000 object images.

Humans have a remarkable fidelity for visual long-term memory, and yet the composition of these memories is a longstanding debate in cognitive psychology. While much of the work on long-term memory has focused on processes associated with successful encoding and retrieval, more recent work on visual object recognition has developed a focus on the memorability of specific visual stimuli. Such work is engendering a view of object representation as a hierarchical movement from low-level visual representations to higher level categorical organization of conceptual representations. However, studies

Unique amnestic syndrome after isolated left anterolateral thalamic stroke: a case report.

BACKGROUND: The thalamus plays a key role in motor and sensory processes of the brain. Though thalamic stroke is among the less frequent, even a minimal lesion can result in a serious impairment and long-lasting disability. After thalamic stroke, cases of dysexecutive syndrome and so-called diencephalic amnesia have been observed, yet the precise characterisation or categorisation of such amnestic syndrome is not available. Pure amnesia can be the one and only indication of a thalamic lesion. We present a unique case of a patient after left anterolateral thalamic stroke suffering from isolated

Effects of lysine-vasopressin and 1-deamino-8-D-arginine-vasopressin on memory in healthy individuals and diabetes insipidus patients.

Central diabetes insipidus (DI) patients showed impairments in short- and long-term memory functions, but not in attention and concentration, as compared to healthy individuals. A single i.m. injection or sub-chronic intranasal administration of either lysine-vasopressin (LVP) or 1-deamino-8-D-arginine-vasopressin (DDAVP) normalized the disturbed memory functions in DI patients. These peptides also improved memory functions in healthy individuals.

Memory-matches evoke human gamma-responses.

BACKGROUND: Human brain activity in the gamma frequency range has been shown to be a correlate of numerous cognitive functions like attention, perception and memory access. More specifically, gamma activity has been found to be enhanced when stimuli are stored in or match with short-term memory (STM). We tested the hypothesis that gamma activity is also evoked when stimuli match representations in long-term-memory (LTM). EEG was recorded from 13 subjects performing a choice reaction task. Visual stimuli were either known real-world objects with a memory representation or novel configurations n

Delayed episodic memory recall after one week is associated with executive functions and divided attention in pediatric epilepsy patients.

AIM: Recent studies suggest that although children with epilepsy may show normal learning and memory performance, accelerated long-term forgetting (ALF) may become evident over time. Our study examined associations between delayed episodic memory performance (recall 1-week after learning) and executive functions. METHOD: A consecutive sample of children with a diagnosis of idiopathic epilepsy with focal or generalized seizures, without morphologic or metabolic abnormalities (n = 20, mean age: 11.70 years) was compared to an IQ-matched healthy control group (n = 20, mean age: 11.55 years).
