Isorhamnetin (ISO) is an active flavonoid compound mainly isolated from the fruits of Hippophae rhamnoides L. and the leaves of Ginkgo biloba L. Previous studies have revealed the antifibrotic action of ISO in the liver and lungs, although its potential protective effects against renal fibrosis and the underlying mechanisms are still poorly understood. Given that many actions of ISO could be similarly attained by hydrogen sulfide (H2S), we speculated that ISO may work through the induction of endogenous H2S. To test the hypothesis, we established the unilateral ureteral obstruction (UUO) renal

The current COVID-19 pandemic, caused by the severe acute respiratory syndrome-coronavirus-2 (SARS-CoV-2) virus, represents the largest medical challenge in decades. It has exposed unexpected cardiovascular vulnerabilities at all stages of the disease (pre-infection, acute phase, and subsequent chronic phase). The major cardiometabolic drivers identified as having epidemiologic and mechanistic associations with COVID-19 are abnormal adiposity, dysglycemia, dyslipidemia, and hypertension. Hypertension is of particular interest, because components of the renin-angiotensin system (RAS), which are

Dyslipidemia is a common risk factor of chronic kidney disease (CKD). Current notion suggests that insufficient intracellular fatty acid oxidation (FAO) and subsequently enhanced fatty acid esterification within renal resident cells, a process termed as renal lipotoxicity, is the key pathogenic event responsible for dyslipidemia-induced kidney injury. However, the detailed mechanism is not fully elucidated. Recently, accumulating data indicated that acetylation modification is an important regulating manner for both mitochondrial function and energy metabolism, while whether acetylation modifi

BACKGROUND: Uteroglobin (UG) is a multifunctional protein with anti-inflammatory properties. The aim of this study was to first evaluate the role of UG gene G38A polymorphism in childhood idiopathic nephrotic syndrome (INS), and determine whether this variation may be related to the occurrence of INS or a steroid response.
METHODS: One hundred and thirty-six children diagnosed with INS in Gaziantep University, Department of Pediatric Nephrology, and 70 healthy volunteers were included. Children with INS were divided into two groups: steroid-sensitive (n = 84), and steroid-resistant (n = 52).

Manic-depression, or bipolar affective disorder, is a prevalent mental disorder with a global impact. Mood stabilizers have acute and long-term effects and at a minimum are prophylactic for manic or depressive poles without detriment to the other. Lithium has significant effects on mania and depression, but may be augmented or substituted by some antiepileptic drugs. The biochemical basis for mood stabilizer therapies or the molecular origins of bipolar disorder is unknown. One approach to this problem is to seek a common target of all mood stabilizers. Lithium directly inhibits two evolutiona

OBJECTIVE: For celiac disease (CD), screening a trend has recently emerged to measure tissue transglutaminase antibodies (tTGA) by immunoassays instead of the more laborious endomysial antibodies (EmA), as they recognize the same target, tissue transglutaminase (tTG). However, a high rate of false-positive results has been reported in some patient series with diseases known to be associated with CD. Moreover, tTG is a ubiquitous, multifunctional enzyme, overexpressed in experimental models of heart failure. Therefore, we assessed the specificity of tTGA assays in a large series of EmA-negative

BACKGROUND: Endothelin (ET) is an endothelium-derived multifunctional peptide that produces a potent, long-lasting vasoconstriction. Nitric oxide (NO), besides being the most important endothelium-derived relaxant factor in blood vessels, is supposed to be involved in regulating the interactions among endothelium, adhesive molecules, and leukocytes.
METHODS: We investigated the possible occurrence and distribution of ET and constitutive nitric oxide synthase (NOs), the enzyme that generates NO from L-arginine, in bovine lymphatic vessels and primary culture of lymphatic endothelium by using i

BACKGROUND AND PURPOSE: Transforming growth factor-beta 1 (TGF-beta 1) is a multifunctional polypeptide that controls the production of extracellular matrix protein. Platelets store a large quantity of TGF-beta 1, which is released at hemorrhage. We recently reported that human recombinant TGF-beta 1 induced communicating hydrocephalus in mice. The aim of this study was to determine whether TGF-beta 1 is related to the development of communicating hydrocephalus after subarachnoid hemorrhage (SAH).
METHODS: TGF-beta 1 in the cerebrospinal fluid of 24 patients with SAH was measured with enzyme-

DNA damage can stall the DNA replication machinery, leading to genomic instability. Thus, numerous mechanisms exist to complete genome duplication in the absence of a pristine DNA template, but identification of the enzymes involved remains incomplete. Here, we establish that Primase-Polymerase (PrimPol; CCDC111), an archaeal-eukaryotic primase (AEP) in eukaryotic cells, is involved in chromosomal DNA replication. PrimPol is required for replication fork progression on ultraviolet (UV) light-damaged DNA templates, possibly mediated by its ability to catalyze translesion synthesis (TLS) of thes

Ataxia with ocular apraxia type 2 (AOA2) is an autosomal recessive, early onset ataxia caused by mutations in the senataxin (SETX) gene. Ocular apraxia and increased levels of alpha-fetoprotein are characteristic but not obligate markers of the disease. AOA2 is allelic with ALS4, a motor neuron disorder of early onset and autosomal dominant inheritance. We observed a two generation family with ataxia which started at age 14 and 17 in two sibs and at age 23 in their paternal uncle.Oculomotor disturbances included strabismus, saccadic pursuit and gaze evoked nystagmus. MRI revealed severe cerebe

Malus micromalus Makino, a fruit crop with a millennium-long cultivation heritage in China, has hitherto received limited systematic investigation regarding its bioactive principles and the mechanistic underpinnings of its glucose-modulating effects. Sixteen compounds were isolated and structurally identified from the fruit. The bioactive components and action modes were systematically explored via network pharmacology, in vitro bioassays, enzyme kinetics, molecular simulation, and in vivo validation. ESR1 and MAPK3 were predicted as potential hub targets involved in insulin resistance and oxi

R-loops are RNA-DNA-hybrid-containing nucleic acids with important cellular roles. Deregulation of R-loop dynamics can lead to DNA damage and genome instability1, which has been linked to the action of endonucleases such as XPG2-4. However, the mechanisms and cellular consequences of such processing have remained unclear. Here we identify a new population of RNA-DNA hybrids in the cytoplasm that are R-loop-processing products. When nuclear R-loops were perturbed by depleting the RNA-DNA helicase senataxin (SETX) or the breast cancer gene BRCA1 (refs. 5-7), we observed XPG- and XPF-dependent cy

Cisplatin chemotherapy is standard care for many cancers but is toxic to the kidneys. How this toxicity occurs is uncertain. In this study, we identified apurinic/apyrimidinic endonuclease 2 (APE2) as a critical molecule upregulated in the proximal tubule cells (PTC) following cisplatin-induced nuclear DNA and mitochondrial DNA damage in cisplatin-treated C57B6J mice. The APE2 transgenic mouse phenotype recapitulated the pathophysiological features of C-AKI (acute kidney injury, AKI) in the absence of cisplatin treatment. APE2 pulldown-MS analysis revealed that APE2 binds myosin heavy-Chain 9

Autoimmune hepatitis (AIH) is a chronic liver disease characterized by progressive inflammation, female preponderance and seropositivity for autoantibodies such as anti-smooth muscle actin and/or anti-nuclear, anti-liver kidney microsomal type 1 (anti-LKM1) and anti-liver cytosol type 1 (anti-LC1) in more than 80% of cases. AIH is linked strongly to several major histocompatibility complex (MHC) alleles, including human leucocyte antigen (HLA)-DR3, -DR7 and -DR13. HLA-DR4 has the second strongest association with adult AIH, after HLA-DR3. We investigated the role of HLA-DR4 in the development

We present the developmental changes of peroxisomal enzymes, catalase, L-bifunctional protein (L-BF) and D-bifunctional protein (D-BF), in the normal brains, and patients with D-BF deficiency, a new peroxisomal disease. D-BF immunoreactivity was observed in controls as early as 13 gestational weeks (GW) and increased with maturation. The adult pattern with fine granule staining of somata and dendrites became apparent in adolescence. L-BF appeared at 20 GW in the cerebral cortex and Purkinje cells and positive glia appeared early in the white matter at 17 GW, and then increased with age. Catala

BACKGROUND: Thermal spring waters are known for their distinctive mineral compositions, which provide health-promoting, antioxidant, immunomodulatory and skin barrier-reinforcing effects. Historically valued for therapeutic use, these waters have recently gained renewed interest as active components in cosmetic and dermatological formulations. Previous works characterized one of Portugal's most renowned thermal waters from Chaves, revealing its potential as a dermatological base ingredient due to its anti-inflammatory effects, namely the reduction of interleukin-6 in stimulated human keratinoc

Diseases caused by trypanosomatid parasites are among the most pressing neglected illnesses. Chagas disease, caused by Trypanosoma cruzi, and visceral Leishmaniasis, caused by Leishmania infantum, have a severe health impact in developing countries. Searching for prospective metal-based drugs against these diseases, five multifunctional fac-[Mn(CO)3(CTZ)(NN)](PF6) compounds, including four new derivatives, were synthesized and thoroughly characterized, featuring NN polypyridyl derivatives and Clotrimazole (CTZ) as bioactive ligands. The biological behavior was compared with that previously rep

BACKGROUND: Nattokinase (NK), a serine protease renowned for its thrombolytic activity, holds immense promise for addressing cardiovascular diseases and metabolic disorders. However, its clinical and industrial translation has been severely hampered by low yields in conventional expression systems, which rely on inefficient solid-state fermentation or suboptimal recombinant platforms. Meanwhile, the global burden of obesity and its associated cardiovascular diseases remain prominent. To this end, implementing a fed-batch fermentation strategy for precise nutrient control is a critical approach

Aluminum (Al), a pervasive environmental neurotoxicant, has been strongly implicated in the onset and progression of Alzheimer's disease (AD)-like pathology. Chronic and sub-chronic exposure to aluminum chloride (AlCl3) induces cognitive deficits, oxidative stress, cholinergic dysfunction, neuroinflammation, and neuronal damage, making it a widely used agent for modeling AD in preclinical research. This study aimed to evaluate the neuroprotective efficacy of TKM01, a novel 4-anilinoquinazoline derivative, in an AlCl3-induced AD-like zebrafish model. Adult zebrafish were exposed to AlCl3 (11 mg

Swine influenza viruses (SIVs), particularly the H1N1 and H3N2 subtypes, pose ongoing threats to the swine industry and global public health. Current commercial vaccines generally provide limited cross-protection against heterologous viruses due to the frequent antigenic drift and shift in SIVs, highlighting the need for broad-spectrum vaccines. Self-assembling nanoparticles (NPs) can elicit strong and broad immune responses, representing a promising platform for the development of broad-spectrum nanovaccines. In this study, two newly-screened conserved epitopes (H1-3 and H1-5) and (H3-1 and H
