Genomic damage in chronic renal failure--potential therapeutic interventions.

In end-stage renal failure, genomic damage is enhanced. This has been shown both in the predialysis and dialysis phase by various biomarkers, such as micronuclei frequency and single cell gel electrophoresis in lymphocytes as well as with 8-hydroxy-2'-deoxyguanosine in leukocytes. There are also data about mitochondrial DNA deletions and chromosomal abnormalities. Genomic damage may be induced by a multitude of toxic factors and mutagens, in particular via enhanced generation of reactive oxygen species. In in vitro studies, incubation of tubular cells with various AGEs (carboxymethyllysine-BSA…

Metals and metal compounds in carcinogenesis.

Several metals and metal containing compounds are potent mutagens and carcinogens. The most often blamed are chromium, arsenic, nickel, vanadium, iron, copper and manganese. Although each of them has its own mechanism of action, it is believed that most of their mechanisms of action involve reactive oxygen species (ROS). Furthermore, nickel modulates gene expression by induction of DNA methylation and/or suppression of histone acetylation. Arsenic activity on cell metabolism is multiple; it seems that cell transformation is induced by long-term exposure to a low level of arsenic. The paradox o…

Review of the genotoxicity of nitrogen oxides.

Nitrogen oxides (NOx) are formed in combustion processes and are major pollutants in urban air. Relatively few studies on the genotoxicity of NO2 and NO have been performed. These studies indicate that NO2 is genotoxic in vitro, but the effect of NO seems to be very slight. One in vivo study showed chromosome aberrations and mutations in lung cells after inhalation of NO2 (and NO), but tests for chromosome aberrations in lymphocytes and spermatocytes or micronuclei in bone marrow were negative after inhalation of NO2. Based on present studies, there is no clear evidence of a carcinogenic poten

A review of the toxicology of salbutamol (albuterol).

This paper reviews the published toxicology of salbutamol. Salbutamol is a relatively selective beta 2-adrenoreceptor stimulant with rapid, potent bronchodilator activity and only minor inotropic or chronotropic effects. It was not found to be mutagenic. LD50 values and other acute studies indicated low toxicity. Findings published for repeat dose studies were mainly uneventful. Tachycardia and flushing of the skin were observed in dogs. There were several findings peculiar to the rat--growth of the salivary gland, enlargement of the Harderian gland, an increase in colloid in the pituitary, an

3D Liver Models for Genotoxicity Testing.

Genotoxicity testing is a vital aspect of the safety assessment of new products (e.g., pharmaceuticals, foodstuffs, industrial chemicals). The ability of an agent to induce DNA damage is linked to long-term cancer risk as DNA damage is known to drive genome instability and carcinogenesis. There is much focus these days on the 3Rs (replacement, reduction, and refinement of animal testing), and in vitro tests are being sought to replace animal approaches. These new approach methodologies (NAMs) include the use of 3D human tissue models. Liver 3D models are particularly valuable as liver enzyme me

Read-across for the prediction of genotoxicity of dimethenamid-P groundwater metabolites: A case study using synthetic chemistry and computational toxicology.

This work presents a read-across case study to predict the genotoxicity of two previously untested groundwater metabolites of dimethenamid-P (α-chloroacetamide herbicide). Due to synthesis challenges, direct testing of the metabolites was not feasible. Structural space alerts and matched molecular pairs analysis were employed to identify analogues and assess structural features. These analyses indicated that these features were not associated with genotoxicity. However, neither method provided evidence regarding the genotoxic potential of the lactone/hemiacetal moiety unique to the metabolites

Effects of 13 developmentally toxic chemicals on the migration of rat cephalic neural crest cells in vitro.

The inhibition of neural crest cell (NCC) migration has been considered as a possible pathogenic mechanism underlying chemical developmental toxicity. In this study, we examined the effects of 13 developmentally toxic chemicals on the migration of rat cephalic NCCs (cNCCs) by using a simple in vitro assay. cNCCs were cultured for 48 h as emigrants from rhombencephalic neural tubes explanted from rat embryos at day 10.5 of gestation. The chemicals were added to the culture medium at 24 h of culture. Migration of cNCCs was measured as the change in the radius (radius ratio) calculated from the c

[Comparison of photomutagenic activities of 5-mop (bergapten) and 8-mop (xanthotoxin) in chlamydomonas reinhardii (author's transl)].

The photomutagenic and phototoxic activities of 5-MOP and 8-MOP were compared in arg- cells of Chlamydomonas reinhardii and their ability to revert to Arg+ mutants. We determined the dependency of induction of Arg+ revertants from cell density, substance concentrations, UV-A fluence, and UV-A radiation intensities. Both furocoumarins were also tested for photomutagenicity under white-light conditions. In all experiments, 5-MOP was more active than 8-MOP. Only on the basis of saturation did both compounds show similarly strong mutagenic activities. When compared on the basis of equivalent cytot

Resistance of high fitness hepatitis C virus to lethal mutagenesis.

Viral fitness quantifies the degree of virus adaptation to a given environment. How viral fitness can influence the mutant spectrum complexity of a viral quasispecies subjected to lethal mutagenesis has not been investigated. Here we document that two high fitness hepatitis C virus populations display higher resistance to the mutagenic nucleoside analogues favipiravir and ribavirin than their parental, low fitness HCV. All populations, however, exhibited a mutation transition bias indicative of active mutagenesis. Resistance to the analogues was associated with a limited expansion of mutant sp

Key role of local acetaldehyde in upper GI tract carcinogenesis.

Ethanol is neither genotoxic nor mutagenic. Its first metabolite acetaldehyde, however, is a powerful local carcinogen. Point mutation in ALDH2 gene proves the causal relationship between acetaldehyde and upper digestive tract cancer in humans. Salivary acetaldehyde concentration and exposure time are the two major and quantifiable factors regulating the degree of local acetaldehyde exposure in the ideal target organ, oropharynx. Instant microbial acetaldehyde formation from alcohol represents >70% of total ethanol associated acetaldehyde exposure in the mouth. In the oropharynx and achlorhydr

DNA triplet repeat expansion and mismatch repair.

DNA mismatch repair is a conserved antimutagenic pathway that maintains genomic stability through rectification of DNA replication errors and attenuation of chromosomal rearrangements. Paradoxically, mutagenic action of mismatch repair has been implicated as a cause of triplet repeat expansions that cause neurological diseases such as Huntington disease and myotonic dystrophy. This mutagenic process requires the mismatch recognition factor MutSβ and the MutLα (and/or possibly MutLγ) endonuclease, and is thought to be triggered by the transient formation of unusual DNA structures within the exp

Evaluation of the toxic effects of arsenite, chromate, cadmium, and copper using a battery of four bioassays.

The sensitivities of four different kinds of bioassays to the toxicities of arsenite, chromate, cadmium, and copper were compared. The different bioassays exhibited different sensitivities, i.e., they responded to different levels of toxicity of each of the different metals. However, with the exception of the α-glucosidase enzyme activity, arsenite was the most toxic compound towards all the tested organisms, exhibiting the highest toxic effect on the seeds of Lactuca, with an EC(50) value of 0.63 mg/L. The sensitivities of Lactuca and Raphanus were greater than the sensitivities of two other

The restriction site mutation (RSM) method has been developed over the past 13 years as a sensitive mutation test which can detect mutations … h, colon and bladder. We can also use these clinical mutation data to speculate as to the causative mutagens involved in these cancer conditions. We here use an example of mutations detected in gastric tissue and those induced in vitro by hydrogen peroxide.

Nephrotoxic and genotoxic N-acetyl-S-dichlorovinyl-L-cysteine is a urinary metabolite after occupational 1,1,2-trichloroethene exposure in humans: implications for the risk of trichloroethene exposure.

Excretion of mercapturic acids in the urine is indicative of the formation of electrophiles in the metabolism of xenobiotics. The determination of these mercapturic acids thus may be a useful method to estimate the exposure. We identified the nephrotoxic and mutagenic mercapturic acids N-acetyl-S-(1,2-dichlorovinyl)-L- cysteine and N-acetyl-S-(2,2-dichlorovinyl)-L-cysteine in the urine of workers exposed to 1,1,2-trichloroethene. A method to quantify these mercapturic acids by gas chromatography-mass spectrometry-selected ion monitoring was developed and appreciable amounts (2.8-3.8 mumole/L w
