The role of methylglyoxal detoxification in Mycobacterium tuberculosis fitness and pathogenesis.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), constitutes a major global health challenge. The pathogenesis of Mtb can be largely attributed to its sophisticated survival strategies within host macrophages. Methylglyoxal (MGO), a ubiquitous reactive dicarbonyl metabolite that spontaneously reacts with biopolymers forming advanced glycation end products (AGEs), togethers with reactive oxygen species (ROS) damage cellular events. The glyoxalase system serves as the primary metabolic pathway for MGO detoxification, mitigating carbonyl stress induced by excess MGO. The mycobacteri…

EccC3 is essential for pathogenesis of rough Mycobacterium abscessus in zebrafish.

Mycobacterium abscessus (Mab) is an emerging human pathogen causing severe lung infections in cystic fibrosis patients and displays either a smooth (S) or a rough (R) morphotype, the latter being associated with a strong inflammatory response. In contrast to most other mycobacteria, it possesses only two ESX secretion systems, ESX-3 and ESX-4. Recent work showed that infection of Mab S lacking the eccC3-encoding ATPase component of ESX-3 in mice was associated with reduced bacterial loads in lungs and no mortality. While these studies emphasize the importance of ESX-3 in the virulence of Mab S…

Immunological characteristics and functions of macrophages in Mycobacterium leprae infection.

OBJECTIVE: Our research aimed to clarify the roles of M1 and M2 macrophages in leprosy, focusing on their polarization states, phagocytic capacities, and the survival of M. leprae within these macrophage subsets. METHODS: M1-like macrophages were induced by IFN-γ, IL-4 induced M2-like macrophages, and then they were compared with M. leprae-induced macrophages regarding cell-surface antigen expression and cytokine secretion. The phagocytic capabilities of M1-like and M2-like cells were assessed using a laser-scanning confocal microscope. Simultaneously, their bactericidal capacities were evalu

EmbB and EmbC regulate the sensitivity of Mycobacterium abscessus to echinomycin.

Treatment of Mycobacterium abscessus (Mab) infections is very challenging due to its intrinsic resistance to most available drugs. Therefore, it is crucial to discover novel anti-Mab drugs. In this study, we explored an intrinsic resistance mechanism through which Mab resists echinomycin (ECH). ECH showed activity against Mab at a minimum inhibitory concentration (MIC) of 2 µg/ml. A ΔembC strain in which the embC gene was knocked out showed hypersensitivity to ECH (MIC: 0.0078-0.0156 µg/ml). The MICs of ECH-resistant strains screened with reference to ΔembC ranged from 0.25 to 1 µg/ml. Mutatio…

Durlobactam in combination with β-lactams to combat Mycobacterium abscessus.

Mycobacterium abscessus (Mab) presents significant clinical challenges. This study evaluated the synergistic effects of a β-lactam and β-lactamase inhibitor combination against Mab and explored the underlying mechanisms. Synergy was assessed through MIC tests and time-kill studies, and binding affinities of nine β-lactams and BLIs to eight target receptors (L,D-transpeptidases [LDT] 1-5, D,D-carboxypeptidase, penicillin-binding protein [PBP] B, and PBP-lipo) were assessed using mass spectrometry and kinetic studies. Thermal stability and morphological changes were determined. Imipenem demonstr…

Methods and Models for Studying Mycobacterium tuberculosis in Respiratory Infections.

Respiratory infections, including tuberculosis, constitute a major global health challenge. Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains one of the leading causes of mortality worldwide. The disease's complexity is attributed to Mtb's capacity to persist in latent states, evade host immune defenses, and develop resistance to antimicrobial treatments, posing significant challenges for diagnosis and therapy. Traditional models, such as animal studies and two-dimensional (2D) in vitro systems, often fail to accurately recapitulate human-specific immune processes, particu…

BACKGROUND: There are hundreds of pathogens that cause lung infections. Compared to infections caused by a single pathogen, mixed infections … cted by mNGS. The findings showed that rare pathogen Scedosporium boydii (S. boydii, reads: 18) and Mycobacterium avium complex (MAC, reads: 19) were detected, and the patient was subsequently transferred to another hospital for the same mNGS with the same results as the first detection. Therefore, combined treatment with voriconazole, ethambutol, azithromycin, …

[Detection of Mycobacterium tuberculosis in sputum by polymerase chain reaction].

A method based on DNA amplification (PCR) and hybridization for the detection of M. tuberculosis was used to test 86 sputum specimens from 52 patients in whom tuberculosis was suspected. M. tuberculosis was detected in 35 specimens, with 1 false positive. The finding was negative in 51 specimens, of which 4 were false negative. Thirty-two specimens were positive by standard microbiological criteria (acid-fast stain and culture), and 54 specimens were negative (including 6 false negative). Of 38 specimens with definite diagnosis of tuberculosis, 89.5% (34/38) were positive by DNA amplification

Differential Mycobacterium bovis BCG vaccine-derived efficacy in C3Heb/FeJ and C3H/HeOuJ mice exposed to a clinical strain of Mycobacterium tuberculosis.

The global epidemic caused by the bacterial pathogen Mycobacterium tuberculosis continues unabated. Moreover, the only available vaccine against tuberculosis, Mycobacterium bovis bacillus Calmette-Guérin (BCG), demonstrates variable efficacy. To respond to this global threat, new animal models that mimic the pathological disease process in humans are required for vaccine testing. One new model, susceptible C3Heb/FeJ mice, is similar to human tuberculosis in that these animals are capable of forming necrotic tubercle granulomas, in contrast to resistant C3H/HeOuJ mice. In this study, we evaluat…

Mycobacterium bovis BCG-mediated protection against W-Beijing strains of Mycobacterium tuberculosis is diminished concomitant with the emergence of regulatory T cells.

Despite issues relating to variable efficacy in the past, the Mycobacterium bovis BCG vaccine remains the basis for new-generation recombinant vaccines currently in clinical trials. To date, vaccines have been tested mostly against laboratory strains and not against the newly emerging clinical strains. In this study, we evaluated the ability of BCG Pasteur to protect mice from aerosol infections with two highly virulent W-Beijing clinical strains, HN878 and SA161. In a conventional 30-day protection assay, BCG was highly protective against both strains, but by day 60 of the assay, this protect…

Recombinant Mycobacterium smegmatis expressing an ESAT6-CFP10 fusion protein induces anti-mycobacterial immune responses and protects against Mycobacterium tuberculosis challenge in mice.

The currently used vaccine against tuberculosis, Bacille Calmette-Guérin (BCG), has variable efficacy, so new vaccine development is crucial. In this study, we evaluated a recombinant vaccine prepared from non-pathogenic Mycobacterium smegmatis (rMS) that expresses a fusion of early secreted antigenic target 6-kDa antigen (ESAT6) and culture filtrate protein 10 (CFP10). C57BL/6 mice were immunized with the rMS expressing the ESAT6-CFP10 fusion protein (rM.S-e6c10) or with BCG. The mice in the rM.S-e6c10 group had a significantly higher titre of anti-ESAT6-CFP10 antibodies than did animals in t…

Gamma interferon production in response to Mycobacterium bovis BCG and Mycobacterium tuberculosis antigens in infants born to human immunodeficiency virus-infected mothers.

In utero sensitization to infectious pathogens can establish immunological memory and may influence the immune response to unrelated antigens. Little is known about the influence of intrauterine human immunodeficiency virus (HIV) exposure on the cellular immune response to mycobacterial antigens. Whole-blood culture gamma interferon (IFN-gamma) production in response to mycobacterial antigens was measured at birth and 6 weeks of age to determine the characteristics of the IFN-gamma response in HIV-exposed infants to Mycobacterium bovis BCG and mycobacterial antigens. At birth, we observed an i…

Mycobacterium bovis BCG substrains confer different levels of protection against Mycobacterium tuberculosis infection in a BALB/c model of progressive pulmonary tuberculosis.

Mycobacterium bovis BCG is the only available vaccine against tuberculosis. Reasons for why diverse BCG substrains induce different levels of protection in clinical trials remain unclear. The aim of this study was to compare the effectiveness of 10 BCG substrains in a mouse model of pulmonary tuberculosis. BALB/c mice were subcutaneously vaccinated and 2 months later were challenged with Mycobacterium tuberculosis H37Rv by intratracheal injection. Two and 4 months after challenge, delayed-type hypersensitivity (DTH) response, lung tissue affected by pneumonia, CFU, T-cell counts, and cytokine …

Virulence, immunogenicity, and protective efficacy of two recombinant Mycobacterium bovis bacillus Calmette-Guérin strains expressing the antigen ESAT-6 from Mycobacterium tuberculosis.

We constructed two recombinant Mycobacterium bovis BCG (rBCG) strains expressing ESAT-6 of Mycobacterium tuberculosis, named rBCG-1 and rBCG-2. rBCG-1 contained the ESAT-6 gene linked to BCG hsp60 and expressed a fusion protein, while rBCG-2, with a secretory sequence, could secret ESAT-6 into the culture medium. There was no evidence for increased virulence of the two rBCG strains when we made a comparison between them and BCG with regard to organ bacterial loads, lung histology, and survival time. rBCG-1 induced significantly higher specific antibody titers and stronger cellular immune respo…

[Possible presence of a booster effect in people repeatedly exposed to Mycobacterium tuberculosis demonstrated in a Mycobacterium tuberculosis-specific interferon-gamma release assay].

INTRODUCTION: Recently, several reports have described a waning response of Mycobacterium tuberculosis (TB)-specific interferon-gamma (IFN-gamma) release assays (IGRAs) performed years after the initial TB infection. Since a considerable part of elderly people in Japan was infected with TB in their youth, it was hypothesized that if the general population was screened with IGRAs, the mean IFN-gamma value of positive subjects (POSITIVES) would be reduced as the age of the subjects increased. Conversely, in subjects who were recently exposed to TB (CONTACTS), it was assumed that the POSITIVES wo…
