Lead chromate was investigated for its carcinogenic potential in both rats and mice. Results show that this compound is a very potent carcinogen in rats when administered i.m. Sixty-four % of the animals treated developed malignant tumors at the injection site. Three renal carcinomas were also found after i.m. treatment with lead chromate. INTRODUCTION: Peritoneal metastatic carcinoma (PMC) faces limited therapeutic efficacy due to the peritoneal-plasma barrier and heterogeneous drug distribution. Current standard therapy (cytoreductive surgery combined with hyperthermic intraperitoneal chemotherapy, CRS + HIPEC) carries high complication rates. While pressurized intraperitoneal aerosol chemotherapy (PIPAC) improves distribution, its penetration depth remains suboptimal. Cold atmospheric plasma (CAP) induces tumor cell apoptosis via reactive oxygen and nitrogen species (RONS) but has not been applied in intraperitoneal therapy.

The synthesis of representative seco-nucleoside analogues of 5-fluorouracil (5-FU)/nitrosourea molecular combinations having uracil (U) as base instead of 5-FU is described. The anti-tumour activity of corresponding pairs of drugs is compared in experimental tumours of the mouse colon, lung and breast. These studies have demonstrated that the presence of a hydrogen or fluorine atom at pyrimidine C-5 is unimportant for the activity shown against most of the experimental tumours employed, rather that the pyrimidine cyclic urea and/or alkylthio functionalities may constitute the critical factors.

Experimental colonic carcinoma in a dog was induced by anal insertion of an N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG) suppository (each cone containing 50 mg of ENNG) for 17 months. The dog was autopsied 20 months after the initiation insertion of the suppository. Ependymomas were produced in 3 of 11 newborn hamsters inoculated intracerebrally with a human papovavirus (MMV) isolated from a malignant lymphoma of the brain of a child with Wiskott-Aldrich syndrome. One of the tumors was serially transplanted into weanling hamsters. Because mammary glands are surrounded by adipose tissues, we hypothesize that the ultra-lipophilic endogenous estrogen-17beta-fatty acid esters may have preferential hormonal and carcinogenic effects in mammary tissues compared to other target organs (such as the uterus and pituitary). Inoculation with a crude suspension prepared from autologous liver cell carcinoma inhibited the development of liver tumor in cirrhotic rats exposed to 3'-methyl-4-diethylaminoazobenzene (3'-Me-DAB). The suspension had also an inhibitory effect on the growth of tumor in rats bearing liver cell carcinoma. The results obtained suggest that a close relationship exists between tumor-associated antigens and onco-development.

Three male control B6C3F1 mice, one male and one female F344 rat were found to have lung tumours with unusual sites of distant metastases. The neoplasms spread to the regional lymph nodes, left heart, kidney, prostate and testes. One mouse showed metastases to the brain.
