Cross-leg free flaps for difficult cases of leg defects: indications, pitfalls, and long-term results.

BACKGROUND: Cross-leg free flaps have been described for reconstruction of leg defects in cases of trauma with no suitable recipient vessels in the vicinity of the defect. The pitfalls, however, have not been well documented in the literature. The technique of transfer and the length of time required before pedicle division depend on the nature of the flap and the contact surface. METHODS: Eight cross-leg free flap reconstructions were performed on seven patients between 1986 and 1993. These included three latissimus dorsi muscle flaps, three fibula osteomyocutaneous flaps, one tensor fascia

3D bioprinted human vascular organoid sheets promote functional ischemic repair and exhibit adaptive in vivo remodeling.

BACKGROUND: Ischemic vascular diseases remain a major clinical challenge, creating a need for engineered vascular tissues that can establish functional vascular networks and promote durable tissue repair. Conventional vascular organoids offer limited control over cellular composition and spatial organization, while the fate and adaptive remodeling of graft-derived human vascular cells after transplantation remain poorly understood. METHODS: Human vascular organoid sheets (hVOS) were constructed by extrusion-based three-dimensional (3D) bioprinting of human pluripotent stem cell (hPSC)-derived

Near-Infrared Region I-Responsive Composite Scaffolds Promote Bone Defect Repair by Jointly Regulating Angiogenic and Osteogenic Differentiation through Magnesium Ions and Metformin.

Currently, the repair of bone defects faces multiple challenges, including insufficient angiogenesis and limited osteogenic differentiation capacity, which pose significant obstacles to treatment. In this study, a ternary composite scaffold composed of PCL-nHA-nMgO (PHM) was fabricated using 3D printing technology. After surface modification with a photothermal polydopamine (PDA) coating, a PHM@PDA composite scaffold was obtained. Metformin (Met) and PDA were loaded into GelMA hydrogels to produce GPM hydrogels. The GPM hydrogels were then packed into the PHM@PDA composite scaffolds, resulting

On-demand hydrogen sulfide-releasing hydrogel reprogramming the diabetic wound microenvironment to accelerate healing.

Chronic diabetic wounds often progress to infection, amputation, and high mortality due to excessive reactive oxygen species (ROS), persistent inflammation, and impaired angiogenesis. Hydrogen sulfide (H2S) can promote angiogenesis, enhance antioxidant defenses, and drive macrophages toward reparative M2 phenotypes, but its clinical use is limited by instability and toxicity. We developed a multifunctional hydrogel (CCT@HSD) composed of collagen, carboxymethyl chitosan, and tannic acid, incorporating a ROS-responsive H2S donor (HSD) for on-demand release. This hydrogel integrated antibacterial

Regulatory Role of miR-196a-5p in Angiogenesis-Related Markers in Endothelial Cells Exposed to Hypertensive Pregnancies.

Offspring of hypertensive disorders of pregnancies (HDP) exhibit early-life endothelial dysfunction and have an elevated susceptibility to hypertension during adulthood which is potentially mediated by microRNA (miRNA), a key regulator of gene expression. RNA sequencing showed that miR-196a-5p was significantly upregulated in HUVEC exposed to HDP and may regulate angiogenesis in endothelial cells. Therefore, this study aims to elucidate the role of miR-196a-5p in regulating angiogenesis in HUVEC exposed to HDP. miR-196a-5p expression was validated by stem-loop RT-qPCR. Predicted target genes w

Trophoblast-derived factors drive human mesenchymal stem cell differentiation along an endothelial lineage: A model of early placental vasculogenesis.

Mechanisms controlling the process and patterning of blood vessel development in the placenta remain largely unknown. The close physical proximity of early blood vessels observed in the placenta and the cytotrophoblast, as well as the reported production of vasculogenic growth factors by the latter, suggests that signalling between these two niches may be important. Here, we have developed an in vitro model to address the hypothesis that the cytotrophoblast, by the secretion of soluble factors, drives differentiation of resident sub-trophoblastic mesenchymal stem cells (MSCs) along a vascular

Protocol for modeling blood flow recovery and angiogenesis in response to hindlimb ischemia.

Here, we present a protocol that models peripheral artery disease using the surgical induction of hindlimb ischemia (HLI) in mice. We list the steps to surgically excise the femoral artery and assess blood flow recovery through laser Doppler imaging. We then detail procedures for visualizing immune cell recruitment and blood vessel growth to explore the role of inflammatory macrophages in angio/arteriogenesis. For complete details on the use and execution of this protocol, please refer to Mantsounga et al.1.

Tobacco ethanol extract accelerates wound healing through anti-inflammation and promoting angiogenesis.

OBJECTIVE: Wound repair remains a significant challenge in the field of regenerative medicine. Notably, while numerous studies have investigated the effects of isolated nicotine or chlorogenic acid, the systematic evaluation of tobacco ethanol extract (TEE) on cutaneous wound healing has remained uncharted territory. Therefore, this study aims to systematically explore TEE's multifaceted effect on wound healing. METHODS: The hepatorenal toxicity of 5% TEE was evaluated in 6-7-week-old ICR mice. Subsequently, full-thickness 6 mm wounds were created, and images were captured on days 0, 3, 6, 9,

N-(3-trimethoxysilylpropyl)ethylenediamine-crosslinked sodium alginate hydrogel: applications in angiogenesis and wound healing across avian and murine models.

Managing wounds remains a significant challenge in modern medicine. Biopolymer hydrogels provide a moist environment conducive to tissue healing. In this study, N-(3-trimethoxysilylpropyl)ethylenediamine (N-3-TMSPED) crosslinked sodium alginate hydrogels were synthesized via lyophilization, with varying concentrations of polyethylene glycol (PEG 600) to evaluate their role in angiogenesis and wound healing. Scanning electron microscopy confirmed porous structures essential for angiogenesis. The hydrogels showed maximum swelling at neutral and basic pH, and enhanced thermal stability with incre

Effects of the gut microbiota on placental angiogenesis and intrauterine growth in gnotobiotic mice.

Environmental causes of intrauterine growth restriction (IUGR) remain poorly characterized. Here, we compare germ-free (GF) and conventionally raised (CONV-R) mice to assess the effects of the gut microbiota on placental/fetal development at embryonic day (E)11.5 (end of placentation) and E17.5 (near term). Pregnancy- and microbiota-associated changes in gene expression occur along the gut, including those related to angiogenesis, while bacterial composition and fermentation activity remain stable. Placental weights at E11.5 and fetal weights at E17.5 are significantly reduced in GF animals. C

Exploring the antiangiogenic effects of Phospholipases A2 from Bothrops diporus venom.

Angiogenesis, the formation of new blood vessels from pre-existing vasculature, is a crucial process in both physiological and pathological contexts, including cancer. Phospholipases A2 (PLA2s), enzymes found in snake venoms, have attracted attention due to their potential antiangiogenic properties. In this study, we explored the antiangiogenic effects of PLA2 isoforms isolated from Bothrops diporus venom using a combination of in vivo and ex vivo models. The chorioallantoic membrane (CAM) assay revealed a significant reduction in vascular density and branching following PLA2s treatment, with

Injectable gelatin-pectin hydrogel for dental tissue engineering: Enhanced angiogenesis and antibacterial efficacy for pulpitis therapy.

Pulpitis is inflammation of the dental pulp, often caused by bacterial infection from untreated cavities, leading to pain. The main challenge in treatment is eliminating infection while preserving tooth vitality. This study aims to address this challenge by developing a hydrogel for convenient insertion into the root canal system, securely attaching to dentin walls. An injectable hydrogel system is developed by chemically cross-linking natural polysaccharide pectin with gelatin (GPG) through reversible Schiff base reaction. The GPG system was then used to encapsulate and release drugs, such as

Matrix Viscoelasticity Controls Differentiation of Human Blood Vessel Organoids into Arterioles and Promotes Neovascularization in Myocardial Infarction.

Stem cell-derived blood vessel organoids are embedded in extracellular matrices to stimulate vessel sprouting. Although vascular organoids in 3D collagen I-Matrigel gels are currently available, they are primarily capillaries composed of endothelial cells (ECs), pericytes, and mesenchymal stem-like cells, which necessitate mature arteriole differentiation for neovascularization. In this context, the hypothesis that matrix viscoelasticity regulates vascular development is investigated in 3D cultures by encapsulating blood vessel organoids within viscoelastic gelatin/β-CD assembly dynamic hydrog

An Injectable Black Phosphorus Hydrogel for Rapid Tooth Extraction Socket Healing.

The excess production of reactive oxygen species (ROS) will delay tooth extraction socket (TES) healing. In this study, we developed an injectable thermosensitive hydrogel (NBP@BP@CS) used to treat TES healing. The hydrogel formulation incorporated black phosphorus (BP) nanoflakes, recognized for their accelerated alveolar bone regeneration and ROS-scavenging properties, and dl-3-n-butylphthalide (NBP), a vasodilator aimed at enhancing angiogenesis. In vivo investigations strongly demonstrated that NBP@BP@CS improved TES healing due to antioxidation and promotion of alveolar bone regeneration

The Role of RASGRP2 in Vascular Endothelial Cells-A Mini Review.

RAS guanyl nucleotide-releasing proteins (RASGRPs) are important proteins that act as guanine nucleotide exchange factors, which activate small GTPases and function as molecular switches for intracellular signals. The RASGRP family is composed of RASGRP1-4 proteins and activates the small GTPases, RAS and RAP. Among them, RASGRP2 has different characteristics from other RASGRPs in that it targets small GTPases and its localizations are different. Many studies related to RASGRP2 have been reported in cells of the blood cell lineage. Furthermore, RASGRP2 has also been reported to be associated w

Urethral reconstruction using an amphiphilic tissue-engineered autologous polyurethane nanofiber scaffold with rapid vascularization function.

Reconstruction and functional rehabilitation of the long urethra in males is one of the difficult tasks in urological treatment. Although many kinds of tissue-engineered urethra scaffold grafts have been successfully used in animals and even clinical research of urethra reconstruction, they all have the disadvantages of slow vascularization in scaffolds, which may lead to complications such as stricture and blockage of the urethra. Here, an amphiphilic polyurethane tubular nanofiber scaffold with a hierarchical structure was designed as a urethral scaffold. The scaffold can regulate the phenot

Intracavernous delivery of Dickkopf3 gene or peptide rescues erectile function through enhanced cavernous angiogenesis in the diabetic mouse.

BACKGROUND: Severe peripheral angiopathy in patients with diabetes is a major contributing factor for low response rate to phosphodiesterase-5 inhibitors. OBJECTIVES: To examine whether and how Dickkopf3 (DKK3), a secreted modulator of the Wnt pathway that known to be involved in endothelial cell repair and vascular progenitor cell migration, restores erectile function in diabetic mice. METHODS: Eight-week-old C57BL/6 mice received intraperitoneal injections of streptozotocin (50 mg/kg for 5 days). Eight weeks after the diabetes was induced, the efficacy of DKK3 was determined by three indep
