Cell-autonomous inhibition of alpha 7-containing nicotinic acetylcholine receptors prevents death of parasympathetic neurons during development.

Neurotrophic molecules are key retrograde influences of cell survival in the developing nervous system, but other influences such as activity are also emerging as important factors. In the avian ciliary ganglion, half the neurons are eliminated between embryonic day 8 (E8) and E14, but it is not known how cell death is initiated. Because systemic application of alpha7-nicotinic acetylcholine receptor (nAChR) antagonists prevents this cell loss, we examined differences in receptor densities and responses of intracellular calcium to nicotine using the calcium-sensitive dye fura-2. In addition, w

Genioglossus reflex inhibition to upper-airway negative-pressure stimuli during wakefulness and sleep in healthy males.

During wakefulness, obstructive sleep apnoea patients appear to compensate for an anatomically narrow upper airway by increasing upper airway dilator muscle activity, e.g. genioglossus, at least partly via a negative-pressure reflex that may be diminished in sleep. Previous studies have assessed the negative-pressure reflex using multi-unit, rectified, moving-time-average EMG recordings during brief pulses of negative upper-airway pressure. However, moving-time averaging probably obscures the true time-related reflex morphology, potentially masking transient excitatory and inhibitory component

Abnormal modulatory influence of diffuse noxious inhibitory controls in migraine and chronic tension-type headache patients.

The aim of this study was to evaluate the function of pain modulating systems subserving diffuse noxious inhibitory controls (DNICs) in primary headaches. DNICs were examined in 24 migraineurs, 17 patients with chronic tension-type headache (CTTH) and 20 healthy subjects by means of nociceptive flexion RIII reflex and the cold pressor test (CPT) as heterotopic noxious conditioning stimulation (HNCS). The subjective pain thresholds (Tp) and the RIII reflex threshold (Tr) were significantly lower in CTTH vs. controls. In controls a significant inhibition of the RIII reflex was observed during th

GABAergic modulation of diffuse noxious inhibitory controls (DNIC): a test by use of lorazepam.

Diffuse noxious inhibitory controls (DNIC) are an important supra-spinal mechanism of pain inhibition. Neurotransmitters and modulators involved in DNIC are serotonin and endogenous opioids. The influence of substances binding to the GABA(A) receptor complex, which has been suggested to play an important role in descending pain inhibition on DNIC has not yet been investigated. The aim of the present study was to find out whether the inhibitory action of DNIC might also be mediated by GABAergic mechanisms. Therefore, DNIC modulation via GABAergic mechanisms was studied in a double blind, placeb

Spinal cord transection inhibits HR reduction in anesthetized rats immersed in an artificial CO2-hot spring bath.

Like humans, the heart rate (HR) of anesthetized rats immersed in CO(2)-water is lower than that when immersed in tap water at the same temperature. To investigate the afferent signal pathway in the mechanism of HR reduction, Wistar rats were anesthetized with urethane and then the spinal cord was transected between T(4) and T(5). The animals were immersed up to the axilla in a bathtub of tap-water (CO(2) contents: 10-20 mg.l(-1)) or of CO(2)-water (965-1,400 mg.l(-1)) at 35 degrees C while recording HR, arterial blood pressure, and arterial blood gas parameters (PaCO(2), PaO(2), pH). Affects of aging on receptive fields in rat primary auditory cortex layer V neurons.

Advanced age is commonly associated with progressive cochlear pathology and central auditory deficits, collectively known as presbycusis. The present study examined central correlates of presbycusis by measuring response properties of primary auditory cortex (AI) layer V neurons in the Fischer Brown Norway rat model. Layer V neurons represent the major output of AI to other cortical and subcortical regions (primarily the inferior colliculus). In vivo single-unit extracellular recordings were obtained from 114 neurons in aged animals (29-33 mo) and compared with 105 layer V neurons in young-adu

Stress-induced attenuation of brain stem activation following intestinal anaphylaxis in the rat.

Intestinal anaphylaxis triggers neuronal activation in the nucleus tractus solitarius (nTS) of the rat brain stem. Stress may modulate reflex circuitry in the brain stem and facilitate intestinal inflammatory responses. We hypothesized that stress would modulate central neuronal activation during intestinal anaphylaxis. NTS neurons were activated following intestinal antigen challenge in sensitized Hooded Lister rats but not in negative controls (P < 0.05). The number of Fos-positive neurons following intestinal anaphylaxis decreased in animals exposed to water-avoidance stress (P < 0.05), alt

Tonic inhibitory control exerted by opioid peptides in the paraventricular nuclei of the hypothalamus on regional hemodynamic activity in rats.

1. Systemic and regional cardiovascular changes were measured following bilateral microinjection of specific and selective opioid-receptor antagonists into the paraventricular nuclei of the hypothalamus (PVN) of awake, freely moving rats. 2. PVN microinjection of increasing doses of the specific opioid antagonist naloxone - methiodide (1 - 5.0 nmol), or a selective mu-opioid receptor antagonist, beta-funaltrexamine (0.05 - 0.5 nmol), evoked important cardiovascular changes characterized by small and transient increases in heart rate (HR) and mean arterial pressure (MAP), vasoconstriction in re

Spinal inhibitory neurons that modulate cutaneous sensory pathways during locomotion in a simple vertebrate.

During locomotion, reflex responses to sensory stimulation are usually modulated and may even be reversed. This is thought to be the result of phased inhibition, but the neurons responsible are usually not known. When the hatchling Xenopus tadpole swims, responses to cutaneous stimulation are modulated. This occurs because sensory pathway interneurons receive rhythmic glycinergic inhibition broadly in phase with the motor discharge on the same side of the trunk. We now describe a new whole-cell recording preparation of the Xenopus tadpole CNS. This has been used with neurobiotin injection to d

Oculomotor abnormalities in boys with tourette syndrome with and without ADHD.

OBJECTIVE: To assess saccadic eye movements in boys with Tourette syndrome (TS) with and without attention-deficit hyperactivity disorder (ADHD), comparing performance with that of an age-matched group of male controls. METHOD: Three different saccade tasks (prosaccades, antisaccades, and memory-guided saccades) were used to examine functions necessary for the planning and execution of eye movements, including motor response preparation, response inhibition, and working memory. The study included 14 boys with TS without ADHD (TS-only), 11 boys with TS and ADHD (TS+ADHD), and 10 male controls.

Disinhibition of lower midbrain neurons enhances non-shivering thermogenesis in anesthetized rats.

UNLABELLED: The hypothesis that the lower midbrain, specifically in and around the retrorubral field (RRF) and/or rubrospinal tract (rs), contains a tonic inhibitory mechanism on non-shivering thermogenesis (NST) was examined in urethane-anesthetized rats. It has been proposed that removal of the tonic inhibitory mechanism in the lower midbrain causes body temperature increase through disinhibition-induced activation of the central sympathetic nervous system. The present experiments were carried out to examine whether and where the proposed midbrain region contains cell bodies that tonically i

Conditioned freezing (generalized motor inhibition) in several rat strains: its usefulness in assessing somato-vegetative responses to nociceptive stress.

Conditioned initial freezing duration was timed in the light-dark box test. Four genetically diverse groups of rats (Wistar, Long Evans, Brattleboro heterozigous for diabetes insipidus, Brattleboro homozygous for diabetes insipidus) were employed. 0.6 mA and 1.8 mA footshocks were administered as nociceptive stressors. The results showed that freezing duration easily and conveniently measures the animal's capacity of responding to stress. This finding is discussed in terms of genetically determined hormonal make-up and of CNS receptorial mechanisms. Initial freezing duration is proposed as a t

Cutaneous effects on presynaptic inhibition of flexor Ia afferents in the human forearm.

1. In six subjects, H reflexes obtained in the flexor muscles in the forearm were inhibited by single motor threshold shocks to the radial nerve in the spiral groove. The first two phases of the inhibitory time course were studied: with intervals between the radial and median nerve shocks of -1 to +1 ms, and +5 to +30 ms. These two phases are thought to be due respectively to disynaptic inhibition between radial Ia afferents and flexor alpha-motoneurones, and to presynaptic inhibition of flexor Ia afferents. 2. Single or short trains (10 ms, 400 Hz) of cutaneous stimuli to the dorsal or palmar

Studies of the inhibitory non-adrenergic neuromuscular transmission in the smooth muscle of the normal human intestine and from a case of Hirschsprung's disease.

A modified sucrose-gap method was used to study both non-adrenergic inhibitory neuromuscular transmission and effects of adenosine 5'-triphosphate (ATP) on isolated smooth muscle preparations from the human intestine. It was found that non-adrenergic inhibition in the circular smooth muscle layer was of larger amplitude than in the longitudinal layer. Study of the ionic mechanisms underlying non-adrenergic inhibition indicated that an increase in K+ conductance was responsible for the generation of non-adrenergic inhibitory junction potentials (IJPs). The results suggest that the inhibitory ac

Low-magnesium epilepsy in rat hippocampal slices: inhibitory postsynaptic potentials in the CA1 subfield.

Spontaneous, synchronous epileptiform discharges were recorded in both CA3 and CA1 subfields of rat hippocampal slices perfused with Mg2+-free medium. Surgical separation of the two areas abolished the spontaneous discharges only in the CA1 subfield. However, epileptiform responses in the isolated CA1 subfield could still be evoked by orthodromic stimulation. Intracellularly these stimulus-induced responses were characterized by a depolarization associated with a burst of action potentials. Stimulation of the alveus still evoked a hyperpolarizing potential, presumably a recurrent inhibitory po

Inhibition of cardiopulmonary input to thoracic spinothalamic tract cells by stimulation of the subcoeruleus-parabrachial region in the primate.

Effects of electrically stimulating the subcoeruleus-parabrachial (SC-PB) region on 31 spinothalamic tract neurons which receive excitatory input from cardiopulmonary sympathetic afferent fibers were studied in 21 monkeys anesthetized with chloralose. A conditioning stimulus to the SC-PB region inhibited the activity of 5 cells responding to the test stimulus applied to sympathetic afferent fibers. At a conditioning-test interval of 10 ms, test responses were maximally reduced to 47 +/- 6% of the control. Inhibitory effects were present at conditioning-test intervals up to 150 ms. Excitatory e

[Role of cortical inhibition in heterosensory interaction of neurons of the sensomotor cortex of the cat].

Background and evoked neuronal activity of the sensorimotor cortex was studied in cats under chloralose anaesthesia. Click, flashes and shocks applied to the contralateral forepaw were used as stimuli. Paired heteromodal stimuli were presented with interstimulus intervals of 0, 100, 200, 300 and 400 ms. Stimuli of different modality completely or partly blocked the response to the testing stimulus when the interstimulus interval was 100-200 ms. The maximum response to the second stimulus followed 200-300 ms after the first stimulus. To study the intercortical mechanisms of heterosensory intera

Interactions between naloxone and GABA in the control of locomotor activity in the rat.

It was found that naloxone causes a small but significant reduction of motility. The GABAB agonist baclofen and the GABA transaminase inhibitor gamma-acetylenic GABA (GAG) also reduced locomotor activity. When a subeffective dose of baclofen was combined with naloxone 0.8 or 3.2 mg/kg, baclofen significantly inhibited motility beyond the inhibition caused by naloxone + saline. GAG, in a dose of 12.5 mg/kg, was also potentiated by naloxone, 3.2 mg/kg. The locomotion reducing effects of naloxone could be blocked by either picrotoxin or bicuculline. It is concluded that GABAergic mechanisms parti

Synaptic perturbation and entrainment of gastric mill rhythm of the spiny lobster.

The gastric mill rhythm of the lobster stomatogastric ganglion was perturbed with short trains of synaptic input from the inferior ventricular nerve (IVN) through fibers. The stimulus was delivered randomly for phase-response curve analysis or repetitively to examine entrainment. The responses depend on the phase of the stimulus in the endogenous rhythm. The stimulus may alter the internal coordination of the motor pattern. Stimuli that occur during a lateral gastric nerve-anterior lateral nerve-E-neuron (LG-GM-E) burst perturb the burst internally and produce a prolonged LG-GM-E burst, while
