Aberrant triple-network connectivity patterns discriminate biotypes of first-episode medication-naive schizophrenia in two large independent cohorts.

Schizophrenia is a complex disorder associated with aberrant brain functional connectivity. This study aims to demonstrate the relation of heterogeneous symptomatology in this disorder to distinct brain connectivity patterns within the triple-network model. The study sample comprised 300 first-episode antipsychotic-naive patients with schizophrenia (FES) and 301 healthy controls (HCs). At baseline, resting-state functional magnetic resonance imaging data were captured for each participant, and concomitant neurocognitive functions were evaluated outside the scanner. Clinical information of 49 F

Hub-connected functional connectivity within social brain network weakens the association with real-life social network in schizophrenia patients.

Hubs in the brain network are the regions with high centrality and are crucial in the network communication and information integration. Patients with schizophrenia (SCZ) exhibit wide range of abnormality in the hub regions and their connected functional connectivity (FC) at the whole-brain network level. Study of the hubs in the brain networks supporting complex social behavior (social brain network, SBN) would contribute to understand the social dysfunction in patients with SCZ. Forty-nine patients with SCZ and 27 healthy controls (HC) were recruited to undertake the resting-state magnetic r

Distinct alterations of amygdala subregional functional connectivity in early- and late-onset obsessive-compulsive disorder.

BACKGROUND: Age of onset may be an important feature associated with distinct subtypes of obsessive-compulsive disorder (OCD). The amygdala joined neurocircuitry models of OCD for its role in mediating fear and regulating anxiety. The present study aims to identify the underlying pathophysiological specifics in OCD with different onset times by assessing amygdala subregional functional connectivity (FC) alterations in early-onset OCD (EO-OCD) and late-onset OCD (LO-OCD). METHODS: Resting-state functional magnetic resonance imaging data were acquired from 88 medication-free OCD patients (inclu

Resting-state functional connectivity of the human hypothalamus.

Communication pathways of the hypothalamus with other brain regions and the periphery are critical to successfully control key physiological and psychological processes. With advanced functional magnetic resonance imaging (fMRI) techniques, it is possible to target hypothalamic function and infer discrete hypothalamus networks. Resting-state functional connectivity (RSFC) is a promising tool to study the functional organization of the brain and may act as a marker of individual differences and dysfunctions. Based on recent fMRI findings, the hypothalamus is mostly connected to parts of the str

Organization of Afferents along the Anterior-posterior and Medial-lateral Axes of the Rat Orbitofrontal Cortex.

The orbitofrontal cortex (OFC) has been anatomically divided into a number of subregions along its medial-lateral axis, which behavioral research suggests have distinct functions. Recently, evidence has emerged suggesting functional diversity is also present along the anterior-posterior axis of the rodent OFC. However, the patterns of anatomical connections that underlie these differences have not been well characterized. Here, we use the retrograde tracer cholera toxin subunit B (CTB) to simultaneously label the projections into the anterior lateral (ALO), posterior lateral (PLO), and posteri

The Anatomy and Physiology of Claustrum-Cortex Interactions.

The claustrum is one of the most widely connected regions of the forebrain, yet its function has remained obscure, largely due to the experimentally challenging nature of targeting this small, thin, and elongated brain area. However, recent advances in molecular techniques have enabled the anatomy and physiology of the claustrum to be studied with the spatiotemporal and cell type-specific precision required to eventually converge on what this area does. Here we review early anatomical and electrophysiological results from cats and primates, as well as recent work in the rodent, identifying the

Cortico-basal ganglia circuits underlying dysfunctional control of motor behaviors in neuropsychiatric disorders.

Neuropsychiatric disorders often manifest with abnormal control of motor behavior. Common symptoms include restricted and repetitive patterns of behavior (RRBs). Cortico-basal ganglia circuits have been implicated in the etiology of RBBs. However, there is a vast range of behaviors encompassed in RRBs, from simple explosive motor tics to rather complex ritualized compulsions. In this review, we highlight how recent findings about the function of specific basal ganglia circuits can begin to shed light into defined motor symptoms associated with neuropsychiatric disorders. We discuss recent stud

Effect of Wnt signaling pathway on neurogenesis after cerebral ischemia and its therapeutic potential.

Neurogenesis process in the chronic phase of ischemic stroke has become the focus of research on stroke treatment recently, mainly through the activation of related pathways to increase the differentiation of neural stem cells (NSCs) in the brain sub-ventricular zone (SVZ) and subgranular zone (SGZ) of hippocampal dentate gyrus (DG) areas into neurons, promoting neurogenesis. While there is still debate about the longevity of active adult neurogenesis in humans, the SVZ and SGZ have the capacity to upregulate neurogenesis in response to cerebral ischemia, which opens discussion about potential

New insights into cortico-basal-cerebellar connectome: clinical and physiological considerations.

The current model of the basal ganglia system based on the 'direct', 'indirect' and 'hyperdirect' pathways provides striking predictions about basal ganglia function that have been used to develop deep brain stimulation approaches for Parkinson's disease and dystonia. The aim of this review is to challenge this scheme in light of new tract tracing information that has recently become available from the human brain using MRI-based tractography, thus providing a novel perspective on the basal ganglia system. We also explore the implications of additional direct pathways running from cortex to ba

Neural Circuit Mechanisms That Underlie Parental Care.

In mammals, parental care is essential for the survival of the young; therefore, it is vitally important to the propagation of the species. These behaviors, differing between the two sexes, are innate, stereotyped, and are also modified by an individual's reproductive experience. These characteristics suggest that neural mechanisms underlying parental behaviors are genetically hardwired, evolutionarily conserved as well as sexually differentiated and malleable to experiential changes. Classical lesion studies on neural control of parental behaviors, mostly done in rats, date back to the 1950s.

Dysregulation of external globus pallidus-subthalamic nucleus network dynamics in parkinsonian mice during cortical slow-wave activity and activation.

KEY POINTS: Reciprocally connected GABAergic external globus pallidus (GPe) and glutamatergic subthalamic nucleus (STN) neurons form a key network within the basal ganglia. In Parkinson's disease and its models, abnormal rates and patterns of GPe-STN network activity are linked to motor dysfunction. Using cell class-specific optogenetic identification and inhibition during cortical slow-wave activity and activation, we report that, in dopamine-depleted mice, (1) D2 dopamine receptor expressing striatal projection neurons (D2-SPNs) discharge at higher rates, especially during cortical activatio

Negative Parenting Affects Adolescent Internalizing Symptoms Through Alterations in Amygdala-Prefrontal Circuitry: A Longitudinal Twin Study.

BACKGROUND: The synergic interaction of risk genes and environmental factors has been thought to play a critical role in mediating emotion-related brain circuitry function and dysfunction in depression and anxiety disorders. Little, however, is known regarding neurodevelopmental bases underlying how maternal negative parenting affects emotion-related brain circuitry linking to adolescent internalizing symptoms and whether this neurobehavioral association is heritable during adolescence. METHODS: The effects of maternal parenting on amygdala-based emotional circuitry and internalizing symptoms

Changes in the anterior cingulate cortex in Crohn's disease: A neuroimaging perspective.

INTRODUCTION: Evidence suggests that Crohn's disease (CD) pathophysiology goes beyond the gastrointestinal tract and is also strongly associated with the brain. In particular, the anterior cingulate cortex (ACC), which plays an integral role in the first brain as part of the default mode network (DMN) and pain matrix, shows abnormalities using multiple neuroimaging modalities. This review summarizes nine related studies that investigated changes in the ACC using structural magnetic resonance imaging, resting-state functional magnetic resonance imaging, and magnetic resonance spectroscopy. What's in a Hub?-Representing Identity in Language and Mathematics.

Hubs emerge in structural and resting state network analysis as areas highly connected to other parts of the brain and have been shown to respond to several task domains in functional imaging studies. A cognitive explanation for this multi-functionality is still wanting. We propose, that hubs subserve domain-general meta-cognitive functions, relevant to a variety of domain-specific networks and test this hypothesis for the example of processing explicit identity information. To isolate this meta-cognitive function from the processing of domain-specific context, we investigate the overlapping a

Visualizing Functional Network Connectivity Difference between Healthy Control and Major Depressive Disorder Using an Explainable Machine-learning Method.

Major depressive disorder (MDD) is a complex mental disorder characterized by a persistent sad feeling and depressed mood. Recent studies reported differences between healthy control (HC) and MDD by looking to brain networks including default mode and cognitive control networks. More recently there has been interest in studying the brain using advanced machine learning-based classification approaches. However, interpreting the model used in the classification between MDD and HC has not been explored yet. In the current study, we classified MDD from HC by estimating whole-brain connectivity usi

Brainstem Circuits Controlling Action Diversification.

Neuronal circuits that regulate movement are distributed throughout the nervous system. The brainstem is an important interface between upper motor centers involved in action planning and circuits in the spinal cord ultimately leading to execution of body movements. Here we focus on recent work using genetic and viral entry points to reveal the identity of functionally dedicated and frequently spatially intermingled brainstem populations essential for action diversification, a general principle conserved throughout evolution. Brainstem circuits with distinct organization and function control s

Routing of Hippocampal Ripples to Subcortical Structures via the Lateral Septum.

The mnemonic functions of hippocampal sharp wave ripples (SPW-Rs) have been studied extensively. Because hippocampal outputs affect not only cortical but also subcortical targets, we examined the impact of SPW-Rs on the firing patterns of lateral septal (LS) neurons in behaving rats. A large fraction of SPW-Rs were temporally locked to high-frequency oscillations (HFOs) (120-180 Hz) in LS, with strongest coupling during non-rapid eye movement (NREM) sleep, followed by waking immobility. However, coherence and spike-local field potential (LFP) coupling between the two structures were low, sugge

Brain functional connectivity in individuals with callosotomy and agenesis of the corpus callosum: A systematic review.

In the absence of the corpus callosum due to either surgical transection or congenital agenesis, the interhemispheric exchange of information is disrupted, as emphasized by several clinical studies. In such cases, a reduction of interhemispheric functional connectivity, that is, an increased independence of the functional signals of the two disconnected hemispheres, is expected to occur. A growing literature has investigated this hypothesis, and a number of studies were able to confirm it. However, this increased independence is not always observed, especially in congenital agenesis, in which

Neural mechanisms for sigh generation during prenatal development.

The respiratory network of the preBötzinger complex (preBötC), which controls inspiratory behavior, can in normal conditions simultaneously produce two types of inspiration-related rhythmic activities: the eupneic rhythm composed of monophasic, low-amplitude, and relatively high-frequency bursts, interspersed with sigh rhythmic activity, composed of biphasic, high-amplitude, and lower frequency bursts. By combining electrophysiological recordings from transverse brainstem slices with computational modeling, new advances in the mechanisms underlying sigh production have been obtained during pre
