Microbial phenolic metabolites at the neurovascular interface: a precision-nutrition framework for brain aging.

Brain aging is increasingly recognized as a neurovascular and inflammatory process involving blood-brain barrier dysfunction, endothelial activation, pericyte impairment, astrocyte-microglia reactivity, oxidative stress, white matter vulnerability, and cognitive decline. Although polyphenol-rich diets are widely associated with brain-health benefits, many parent polyphenols have limited systemic bioavailability, suggesting that gut microbiota-derived microbial phenolic metabolites may represent more biologically relevant circulating exposures. However, systemic MPM exposure is also conditioned

Vascular remodelling and fibrotic changes in the joint capsule during periprosthetic knee infections.

AIMS: Periprosthetic joint infection (PJI) represents one of the most severe complications following joint arthroplasty, often associated with a high recurrence rate despite appropriate therapeutic interventions. The underlying mechanisms contributing to this persistent risk remain incompletely understood. We hypothesize that alterations in joint capsule vascularization and fibrotic remodelling contribute to the pathophysiology of PJI and its recurrence. METHODS: A total of 69 patients undergoing joint arthroplasty surgery were included in the study (21 controls: primary total knee arthroplas

Transcriptional profile of native CD271+ multipotential stromal cells: evidence for multiple fates, with prominent osteogenic and Wnt pathway signaling activity.

OBJECTIVE: Controversy surrounds the identity and functionality of rare bone marrow-derived multipotential stromal cells (BM-MSCs), including their differentiation capabilities, their relationship to pericytes and hematopoiesis-supporting stromal cells, and the relevance of their culture-expanded progeny in studies of skeletal biology and development of cell-based therapies. The aim of this study was to clarify the nature of candidate BM-MSCs by profiling transcripts that reflect different aspects of their putative functions in vivo. METHODS: Rare, sorted BM-derived CD45(-/low) CD271(bright) …

Hemangiopericytoma: Conducts and perioperative management of an extent sinonasal tumor in a Jehovah's Witnesses patient - Case report.

BACKGROUND: Hemangiopericytomas (HPCs) are rare tumors derived from mesenchymal cells with pericyte differentiation. About 5% of head and neck HPCs occur in the nasal cavity and paranasal sinuses. Due to its rarity, rich vascularity and variable biological behavior, its management is a challenge in itself. CASE: We report a case of sinonasal HPC in a Jehovah's Witness patient and discuss the obstacles and care related to the restrictions and therapeutic challenges involved in the approach to the patient. The patient was successfully treated by endoscopic endonasal approach with all per-operat

Malignant hemangiopericytoma of the condyle.

Hemangiopericytoma (HPC) is an uncommon vascular tumor arising from pericytic cells with variable malignant potential. Primary HPCs of the bone are extremely rare; however, involvement of the condylar process has not been reported. We presented a 21-year-old female patient with low-grade malignant HPC in the right mandibular condyle. Clinical examination and imaging findings revealed a well-defined soft mass, encapsulating the mandibular condyle from behind. The lesion and mandibular condyle were removed simultaneously; then, the costochondral graft was used to reconstruct the defect. Histopat

Multiple congenital hemangiopericytomas of the head and neck.

Hemangiopericytoma is a vascular tumor arising from contractile cells, the pericyte of Zimmermann. The neoplasms most frequently involve deep musculature; however, 16% occur in the head and neck region. The case presented here represents the rare occurrence of multiple congenital hemangiopericytoma of the head and neck. In older patients, these rather poorly differentiated tumors recur locally after excision in 50%-80% of cases depending on the organ site involved. Distant metastasis to lung or bone may follow. Congenital hemangiopericytomas appear to follow a more benign clinical course, poss

[Erythema elevatum diutinum. II. Immunoelectronmicroscopical study of leukocytoclastic vasculitis within the intracutaneous test reaction induced by streptococcal antigen (author's transl)].

A leukocytoclastic vasculitis was induced by intracutaneous injection of streptococcal antigen in a patient with erythema elevatum diutinum (E.e.d.). The immunoelectronmicroscopical demonstration of C3 was performed by use of the peroxidase-antiperoxidase multistep technique 24 h after the injection of the antigen. Deposits of C3 were found between endothelial cells, on the outer surface of endothelial cells, pericytes, and smooth muscle cells, as well as within the multilayered basal lamina of small vessels. Intact and disintegrating neutrophils accumulate within the vessel walls and in their…

Mapping disease-specific vascular cell populations responsible for obliterative arterial remodelling during the development of pulmonary arterial hypertension.

AIMS: Pulmonary arterial hypertension (PAH) is a lethal pulmonary vascular disease characterized by arteriolar pruning and occlusive vascula … lling in both the rat model and PAH patients, contributing to intimal occlusive lesions and SM-like pericytes forming bands of medial muscularization. CONCLUSION: Together these findings implicate disease-specific vascular cells in PAH progression and suggest that TM4SF1 may be a novel therapeutic target for arterial remodelling.

Umbilical cord mesenchymal stem cells from gestational diabetes show impaired ability to up-regulate paracellular permeability from sub-endothelial niche.

In vitro studies have shown that Wharton's jelly mesenchymal stem cells (WJ-MSCs) can cross umbilical and uterine endothelial barriers and up-regulate endothelial junctional integrity from sub-endothelial niches. This pericytic behaviour may be lost in pregnancies complicated by gestational diabetes (GDM), where increased vascular permeability and junctional disruption are reported. The aim of the present study was to investigate whether WJ-MSCs isolated from GDM pregnancies displayed any changes in morphology, proliferation, VEGF-A secretion, and their ability to influence paracellular juncti

Follicular dendritic cell differentiation is associated with distinct synovial pathotype signatures in rheumatoid arthritis.

Follicular dendritic cells (FDCs) fundamentally contribute to the formation of synovial ectopic lymphoid-like structures in rheumatoid arthr … cles. Moreover, PDGF-BB induced CNA.42+ FDC differentiation and CXCL13 secretion from NG2+ synovial pericytes, and together with TNF-α/LT-β conversely regulated early and late FDC differentiation genes in unsorted RA synovial fibroblasts (RASF) and this was confirmed in flow sorted stromal cell subsets. Furthermore, RASF TNF-αR expression was upregulated by …

Expression of angiogenic and vasculogenic factors in human lymphedematous tissue.

BACKGROUND: Lymphedema is the progressive distention of tissue due to lymphatic dysfunction. The affected area enlarges over time because of fibroadipose deposition, causing morbidity. Because increasing tissue mass requires neovascularization, we hypothesized that angiogenesis or vasculogenesis might be upregulated in lymphedema. METHODS AND RESULTS: Lymphedematous tissue was collected prospectively from nine patients after resection: upper extremity (n = 1), lower extremity (n = 3), penis/scrotum (n = 5). Neovascularization was compared to normal tissue. Specimens were analyzed using immuno

Cerebrospinal fluid biomarkers of neurovascular dysfunction in mild dementia and Alzheimer's disease.

Alzheimer's disease (AD) is the most common form of age-related dementias. In addition to genetics, environment, and lifestyle, growing evidence supports vascular contributions to dementias including dementia because of AD. Alzheimer's disease affects multiple cell types within the neurovascular unit (NVU), including brain vascular cells (endothelial cells, pericytes, and vascular smooth muscle cells), glial cells (astrocytes and microglia), and neurons. Thus, identifying and integrating biomarkers of the NVU cell-specific responses and injury with established AD biomarkers, amyloid-β (Aβ) and…

Glomerular Endothelial Cell Crosstalk With Podocytes in Diabetic Kidney Disease.

Diabetes is the main cause of renal failure worldwide. Complications of the kidney micro-and macro-circulation are common in diabetic patients, leading to proteinuria and can progress to end-stage renal disease. Across the complex interplays aggravating diabetes kidney disease progression, lesions of the glomerular filtration barrier appear crucial. Among its components, glomerular endothelial cells are known to be central safeguards of plasma filtration. An array of evidence has recently pinpointed its intricate relations with podocytes, highly specialized pericytes surrounding glomerular cap…

Hepatic gateways.

The intestinal mucosal barrier contributes to homeostasis by limiting systemic dissemination of microbes and toxins while allowing nutrients to pass through to the systemic circulation. In a recent issue of Science, Spadoni et al. demonstrated a novel mechanism to enable this selectivity: the existence of a gut-vascular barrier (GVB) as indicated by a series of studies on the interaction between murine and human intestine with Salmonella typhimurium species . They showed that (i) enteroglial cells and pericytes in contact with endothelial cells (ECs) form the GVB (ii) Salmonella typhimurium ca…

[Current concepts of perinatal ischemic injury in the brain neurovascular unit: molecular targets for neuroprotection].

Perinatal hypoxic-ischemic brain injury is a relevant medical and social problem. Among many pathological processes in the neonatal period perinatal hypoxic-ischemic injury is a major cause of further hemorrhage, necrotic and atrophic changes in the brain. This review presents recent data on the basic mechanisms of the hypoxic-ischemic brain injury along the concept of neurovascular unit (neurons, astrocytes, endothelial cells, pericytes) with the focus on alterations in cell-to-cell communication. Pathological changes caused by ischemia-hypoxia are considered within two phases of injury (isch…

Activated Ito cells of cirrhotic liver express M3 muscarinic receptor after thioacetamide exposure.

Ito cells or perisinusoidal stellate cells or hepatic fat storing cells are pericytes of normal liver sinusoidal endothelial cells. Activation of Ito cells by chemicals or toxins causes transdifferentiation into the main collagen-producing cells involving in the progression of liver cirrhosis. Quantitative analysis of Ito cell activation by immunohistochemistry has been shown to be useful in predicting the rate of progression of liver fibrosis in some clinical situations. The present study demonstrated that the activated Ito cells in thioacetamide-induced cirrhotic liver which were immunoposit…
