[Progress in studies of effects on brain-derived neurotrophic factor in degenerative fundus disease].

Degenerative fundus diseases are a group of outer retinal disease caused by dysfunction of photoreceptors, some of which have the inner layer defection. Brain-derived neurotrophic factor (BDNF) has been shown to play a key role in the neuronal survival, which accomplishes its protection via receptors, including a low affinity p75 and a high affinity tyrosine kinase receptor TrkB, retinal growth cone filopodia and cofactor of Zinc. It is proved that BDNF can induce differentiation of stem cells and retinal progenitor, in case for the effective retinal transplantation. The protection by BDNF has

Poly(ADP-ribose) polymerase regulates transient receptor potential channel M2-dependent calpain activation in rd1 mouse retinal degeneration.

Inherited retinal degeneration refers to untreatable blinding diseases characterized by progressive photoreceptor loss. Photoreceptor degeneration is often associated with an excessive activation of poly(ADP-ribose) polymerase and Ca2+-dependent calpain-type proteases. To explore the interplay between poly(ADP-ribose) polymerase and calpain activity, we employed organotypic retinal explant cultures derived from wild-type mice and from the rd1 mouse model for inherited retinal degeneration. Retinae were treated with the poly(ADP-ribose) polymerase inhibitors INO1001 or Olaparib, the poly(ADP-ri

The unspecified role of cones and rods in grating detection: a theoretical note.

Detection and resolution thresholds for two 50 ms square-wave gratings (1.5 and 6.7 cps) were determined as a function of background luminance. Results indicated differential cone and rod involvement in resolution of the two targets at low background levels. Implications for other studies employing similar stimuli in terms of retinal inhomogeneity were suggested.

Hemeralopia in an older adult.

An elderly male was referred for evaluation of hemeralopia when cataract extraction did not alleviate his symptom of difficulty seeing in bright illumination. Visual acuity was 20/40 OU. Funduscopic examination revealed marked arteriolar attenuation, disc pallor, and retinal pigment epithelial changes. Visual fields demonstrated relative paracentral scotomata. An ERG was consistent with widespread photoreceptor degeneration. A diagnosis of visual paraneoplastic syndrome was made and an extensive systemic evaluation revealed a squamous cell carcinoma of the lung. The presentation and evaluation

Constitutive opsin signaling: night blindness or retinal degeneration?

A subset of genetic mutations in photoreceptor-specific genes results in abnormally prolonged activation of transducin-mediated photosignaling in rod cells. In humans and animal models, these mutations cause visual dysfunctions ranging from a mild stationary night blindness to severe, early-onset retinal degeneration. There are mechanistic differences between mutations causing night blindness and those causing retinal degeneration. Here, we hypothesize that mutations causing continuous activation of the visual cascade as the result, for example, of the inability of the photoreceptor to regener

cGMP-dependent kinase regulates response sensitivity of the mouse on bipolar cell.

The visual system can adjust its sensitivity over a wide range of light intensities. Photoreceptors account for some of this adjustment, but there is evidence that postreceptoral processes also exist. To investigate the latter, we pharmacologically mimicked the effects of light stimulation on mouse On bipolar cells, thus avoiding confounding effects of receptoral mechanisms. Here, we report that cGMP selectively enhances responses to dim, but not bright, stimuli through a purely postsynaptic mechanism. This action of cGMP was completely blocked by inhibitors of cGMP-dependent kinase. We propos

Chloroquine retinopathy in the rhesus monkey.

Chloroquine was administered intramuscularly 5 days a week to rhesus monkeys for as long as 4 1/2 years. No clinical, fluorescein angiographic, or electrophysiological evidence of retinal damage was observed. Yet chloroquine/chloroquine byproduct analysis of the ocular tissues revealed an enormous binding capacity of the pigmented tissues of the eye (choroid plus RPE, ciliary body, and iris) with eventual accumulation observed in the retina. Despite the normal ophthalmic appearance and function, extensive pathological changes occurred in the retinas and choroids of these experimental monkeys.

Dynamics of experimental autoimmune uveoretinitis induced by adoptive transfer of S-antigen-specific T cell line.

Long-term S-antigen (S-Ag) specific T lymphocyte lines can induce experimental autoimmune uveoretinitis (EAU) when transferred into naive rats systemically or intravitreally. The uveitogenic lymphocyte line (ThS) stains positively for the ART 18 (IL-2 receptor) and W3/25 (T helper/inducer); negatively for OX6 (RT1B) and OX8 (T suppressor/cytotoxic cells). The inflammation induced by systemic or intravitreal transfer of the ThS line was studied with respect to the surface markers of the infiltrating cells, as well as the markers expressed by the resident ocular cells, by immunohistopathological

Protocol for the isolation, culturing, and evaluation of primary Müller glial cells from pig retina as cellular models in health and disease.

Müller glial cells maintain retinal homeostasis and provide structural and metabolic support. The pig eye closely resembles the human eye in morphology and physiology, offering strong translational potential. Here, we present a protocol for isolating and culturing primary Müller glial cells from pig retina to study their function in health and disease. We describe steps for eyeball dissection, retinal isolation, photoreceptor cell depletion, and retinal digestion. We then detail procedures for the selection, culture, and characterization of Müller glial cells.

Quantum Dots-caused Retinal Degeneration in Zebrafish Regulated by Ferroptosis and Mitophagy in Retinal Pigment Epithelial Cells through Inhibiting Spliceosome.

Quantum dots (QDs) are widely used, but their health impact on the visual system is little known. This study aims to elucidate the effects and mechanisms of typical metallic QDs on retinas using zebrafish. Comprehensive histology, imaging, and bulk RNA sequencing reveal that InP/ZnS QDs cause retinal degeneration. Furthermore, single-cell RNA-seq reveals a reduction in the number of retinal pigment epithelial cells (RPE) and short-wave cone UV photoreceptor cells (PR(UV)), accompanied by an increase in middle- and long-wave cone red, green, and blue photoreceptor cells [PR(RGB)]. Mechanistical…

Short pulse epiretinal stimulation allows focal activation of retinal ganglion cells.

Epiretinal implants suffer from a lack of spatial resolution, which is greatly influenced by the undesired co-activation of distal cells with their axons passing close to targeted somas. Short current pulses in the range of 50μs have been shown to preferentially activate somas, but the low specificity may limit practical applications. In this paper, we explored decreasing pulse durations down to 10μs for achieving focal activation, i.e., a large differentiation between axonal and somatic activation in epiretinal configuration. We determined thresholds for pulses ranging between 10 and 500μs in

Fundus Autofluorescence in Neovascular Age-Related Macular Degeneration: A Clinicopathologic Correlation Relevant to Macular Atrophy.

PURPOSE: Macular atrophy (MA) of retinal pigment epithelium (RPE) and photoreceptors leads to vision loss in neovascular age-related macular degeneration (nAMD) despite successful treatment with antiangiogenic agents. To enhance understanding of MA, fortify the cellular basis of fundus autofluorescence (FAF) imaging, and inform management of nAMD, we performed histologic analysis of an eye with multimodal clinical imaging and apparent prior exudation due to nAMD. DESIGN: Case study and clinicopathologic correlation. PARTICIPANT: A White woman in whom age-related macular degeneration (AMD) fi

Hypomorphic mutations identified in the candidate Leber congenital amaurosis gene CLUAP1.

PURPOSE: Leber congenital amaurosis (LCA) is an early-onset form of retinal degeneration. Six of the 22 known LCA genes encode photoreceptor ciliary proteins. Despite the identification of 22 LCA genes, the genetic basis of ~30% of LCA patients remains unknown. We sought to investigate the cause of disease in the remaining 30% by examining cilia-associated genes. METHODS: Whole-exome sequencing was performed on an LCA cohort of 212 unsolved probands previously screened for mutations in known retinal-disease genes. Immunohistochemistry using mouse retinas was used to confirm protein localizati

Contrast sensitivity and functional aspects of cone, rod, and melanopsin vision in humans.

The sensitivity of the visual system is best defined by its response to contrast. To this end, the visual system can be decomposed into pathways with unique contrast responses to the spatial and temporal content of the stimulus. In this chapter, we evaluate the contrast sensitivity of the individual mechanisms in the eye through the lens of silent substitution - a technique that exploits a high degree of spectral control to precisely set the stimulus contrast presented to each of the five photoreceptor classes. With the discovery of a fifth (melanopsin) photoreceptor in the human eye, new meth

Single-cell RNA sequencing of the retina in a model of retinitis pigmentosa reveals early responses to degeneration in rods and cones.

BACKGROUND: In inherited retinal disorders such as retinitis pigmentosa (RP), rod photoreceptor-specific mutations cause primary rod degeneration that is followed by secondary cone death and loss of high-acuity vision. Mechanistic studies of retinal degeneration are challenging because of retinal heterogeneity. Moreover, the detection of early cone responses to rod death is especially difficult due to the paucity of cones in the retina. To resolve heterogeneity in the degenerating retina and investigate events in both types of photoreceptors during primary rod degeneration, we utilized droplet

Inhibition of mTOR signaling by rapamycin protects photoreceptors from degeneration in rd1 mice.

Retinitis pigmentosa (RP) is an inherited retinal degenerative disease that begins with defective rod photoreceptor function, followed by impaired cone function, and complete blindness in its late stage. To date, however, there is no effective treatment for RP. By carrying a nonsense mutation in the Pde6b gene, rd1 mice display elevated cGMP in conjunction with higher intracellular Ca 2+ in their rod photoreceptors, resulting in fast retinal degeneration. Ca 2+ has been linked to activation of the mammalian target of rapamycin (mTOR) pathway. The mTOR pathway integrates extracellular and intra

PRCD Is a Small Disc-Specific Rhodopsin-Binding Protein of Unknown Function.

PRCD (progressive rod-cone degeneration) is a small ~6 kDa protein with unknown function that specifically resides in photoreceptor discs and interacts with rhodopsin. PRCD's discovery resulted from decades-long study of a canine retinal disease called progressive rod-cone degeneration which is one of the most frequent causes of blindness in dogs characterized by the slow, progressive death of rod photoreceptors followed by cones. A series of genetic studies eventually mapped the disease to a single point mutation in a novel gene which was then named Prcd. Highlighting the importance of this g

Functional optical coherence tomography enables in vivo physiological assessment of retinal rod and cone photoreceptors.

Transient intrinsic optical signal (IOS) changes have been observed in retinal photoreceptors, suggesting a unique biomarker for eye disease detection. However, clinical deployment of IOS imaging is challenging due to unclear IOS sources and limited signal-to-noise ratios (SNRs). Here, by developing high spatiotemporal resolution optical coherence tomography (OCT) and applying an adaptive algorithm for IOS processing, we were able to record robust IOSs from single-pass measurements. Transient IOSs, which might reflect an early stage of light phototransduction, are consistently observed in the
