Signaling pathway played by salicylic acid, gentisic acid, nitric oxide, polyamines and non-enzymatic antioxidants in compatible and incompatible Solanum-tomato mottle mosaic virus interactions.

Plants are exposed to a vast array of pathogens. The interaction between them may be classified in compatible and incompatible. Polyamines (PAs) are involved in defense responses, as well as salicylic acid (SA), gentisic acid (GA) and nitric oxide (NO), which can increase the content of reactive oxygen species (ROS), creating a harsh environment to the pathogen. ROS can also damage the host cell and they can be controlled by ascorbate and glutathione. Among phytopathogens, one of the major threats to tomato crops is tomato mottle mosaic virus (ToMMV). Resistance against this virus probably inv

The cytosolic/nuclear HSC70 and HSP90 molecular chaperones are important for stomatal closure and modulate abscisic acid-dependent physiological responses in Arabidopsis.

Cytosolic/nuclear molecular chaperones of the heat shock protein families HSP90 and HSC70 are conserved and essential proteins in eukaryotes. These proteins have essentially been implicated in the innate immunity and abiotic stress tolerance in higher plants. Here, we demonstrate that both chaperones are recruited in Arabidopsis (Arabidopsis thaliana) for stomatal closure induced by several environmental signals. Plants overexpressing HSC70-1 or with reduced HSP90.2 activity are compromised in the dark-, CO(2)-, flagellin 22 peptide-, and abscisic acid (ABA)-induced stomatal closure. HSC70-1 a

Harpin induces disease resistance in Arabidopsis through the systemic acquired resistance pathway mediated by salicylic acid and the NIM1 gene.

Harpin, the product of the hrpN gene of Erwinia amylovora, elicits the hypersensitive response and disease resistance in many plants. Harpin and known inducers of systemic acquired resistance (SAR) were tested on five genotypes of Arabidopsis thaliana to assess the role of SAR in harpin-induced resistance. In wild-type plants, harpin elicited systemic resistance to Peronospora parasitica and Pseudomonas syringae pv. tomato, accompanied by induction of the SAR genes PR-1 and PR-2. However, in experiments with transgenic Arabidopsis plants containing the nahG gene which prevents accumulation of

Chelidonium majus--an integrative review: traditional knowledge versus modern findings.

Chelidonium majus L. (family Papaveraceae), or greater celandine, is an important plant in western phytotherapy and in traditional Chinese medicine. Crude extracts of C. majus as well as purified compounds derived from it exhibit a broad spectrum of biological activities (antiinflammatory, antimicrobial, antitumoral, analgesic, hepatoprotective) that support some of the traditional uses of C. majus. However, herbal medicine also claims that this plant has several important properties which have not yet been scientifically studied: C. majus is supposed to have diuretic, antitussive and eye-rege

β-Aminobutyric Acid-Induced Resistance Against Root-Knot Nematodes in Rice Is Based on Increased Basal Defense.

The nonprotein amino acid β-aminobutyric acid (BABA) is known to protect plants against various pathogens. The mode of action is relatively diverse and specific in different plant-pathogen systems. To extend the analysis of the mode of action of BABA to plant-parasitic nematodes in monocot plants, we evaluated the effect of BABA against the root-knot nematode (RKN) Meloidogyne graminicola in rice. BABA treatment of rice plants inhibited nematode penetration and resulted in delayed nematode and giant cell development. BABA-induced resistance (BABA-IR) was still functional in mutants or transgen

Necrotrophic pathogens use the salicylic acid signaling pathway to promote disease development in tomato.

Plants use different immune pathways to combat pathogens. The activation of the jasmonic acid (JA)-signaling pathway is required for resistance against necrotrophic pathogens; however, to combat biotrophic pathogens, the plants activate mainly the salicylic acid (SA)-signaling pathway. SA can antagonize JA signaling and vice versa. NPR1 (noninducible pathogenesis-related 1) is considered a master regulator of SA signaling. NPR1 interacts with TGA transcription factors, ultimately leading to the activation of SA-dependent responses. SA has been shown to promote disease development caused by the

Assays of Antifungal Activity of an Antimicrobial Peptide Against Botrytis cinerea.

Antimicrobial peptides (AMPs) represent the first line of host immune defense against a broad spectrum of microorganisms. Whether naturally occurring or synthetically designed, these small peptides can potently inhibit microbial growth and remain stable under diverse conditions. Most AMPs act by directly disrupting fungal or bacterial membranes. In this study, we identified a glycine-rich peptide with strong antifungal activity against Botrytis cinerea. We further investigated its mode of action (MOA) and demonstrated its preventive antifungal activity in plants. Our work focuses primarily on

Usage of celery root exosome as an immune suppressant; Lipidomic characterization of apium graveolens originated exosomes and its suppressive effect on PMA/ionomycin mediated CD4+ T lymphocyte activation.

Diseases such as autoimmune, cancer, neurodegenerative diseases or obesity have a serious impact on the lives of patients all rise from a common point; the immune system. Various in vitro and in vivo studies on regulating the immune system have been made to correct these diseases. As one of the key effector cells of the immune system, T lymphocytes are the focus of many of these studies. In this study, exosomes isolated from a known anti-inflammatory plant, celery, were used to suppress the inflammatory response of T lymphocytes. Celery-derived exosomes (C-Exo) were isolated using an aqueous t

Smart Silica Nanocomposites Activating Plant Defenses via Enhanced Biomolecule Deposition on Superhydrophobic Monocot Surfaces.

Sustainable management of plant diseases is vital for maintaining food security, particularly due to the significant importance of monocots in global agriculture. The utilization of conventional pesticides, however efficacious, raises concerns regarding environmental contamination, hazards to human health, and the emergence of disease resistance. Herein we propose smart nanocomposites (SNPPECs) that integrate biomolecules to activate the plant immune system, encapsulated in nanoparticles within a polyelectrolyte complex. The polyelectrolyte complex, composed of oppositely charged polymers, aug

A single component two-valent LcrV-F1 vaccine protects non-human primates against pneumonic plague.

Yersinia pestis continues to pose a threat as a potential biological weapon and is recognized by public health experts as a re-emerging infectious disease. Therefore there is great interest in developing a safe and effective vaccine. Vaccines against plague containing both the Fraction 1 (F1) and V antigens of Y. pestis have shown promise in protecting animal models against pneumonic plague, the deadliest form of the disease. Here we report on a plague vaccine consisting of the F1 and LcrV antigens fused to a single carrier molecule, the thermostable enzyme lichenase from Clostridium thermocel

Melatonin in edible plants identified by radioimmunoassay and by high performance liquid chromatography-mass spectrometry.

Melatonin, the chief hormone of the pineal gland in vertebrates, is widely distributed in the animal kingdom. Among many functions, melatonin synchronizes circadian and circannual rhythms, stimulates immune function, may increase life span, inhibits growth of cancer cells in vitro and cancer progression and promotion in vivo, and was recently shown to be a potent hydroxyl radical scavenger and antioxidant. Hydroxyl radicals are highly toxic by-products of oxygen metabolism that damage cellular DNA and other macromolecules. Herein we report that melatonin, in varying concentrations, is also fou

Populus simonii × Populus nigra overexpressing PsnWRKY70 recruits phyllosphere bacterial strains that inhibit Alternaria alternata.

WRKY transcription factors have been implicated in the regulation of disease resistance associated with plant immune responses, which has crucial implications for defense responses against stress in plants. The role played by the PsnWRKY70 gene of Populus (Populus simonii × P. nigra) in triggering the mechanism between the phyllosphere microbiome and plant defense against foliar pathogens remains unclear. Molecular ecological network analysis demonstrated that the stability and complexity of the phyllosphere bacterial community of Populus were influenced by Alternaria alternata infection. Spec

Recent advances in progesterone receptor-targeted therapy for uterine fibroids: A systematic review of synthetic and natural modulators.

Progesterone receptor (PR) signaling plays a pivotal role in uterine fibroids (UFs) development and progression. Selective progesterone receptor modulators (SPRMs) and plant-derived PR modulators have emerged as promising therapeutic strategies. This review synthesizes current evidence on novel synthetic SPRMs and natural compounds with PR-modulating activity. Following the PRISMA guidelines, literature searches in PubMed, Scopus, and Google Scholar identified 22 studies that met the eligibility criteria. Synthetic SPRMs act mainly through PR antagonism, with differences in receptor selectivit

Low dietary sodium potentially mediates COVID-19 prevention associated with whole-food plant-based diets.

Compared with an omnivorous Western diet, plant-based diets containing mostly fruits, vegetables, grains, legumes, nuts and seeds, with restricted amounts of foods of animal origin, are associated with reduced risk and severity of COVID-19. Additionally, inflammatory immune responses and severe acute respiratory symptoms of COVID-19, including pulmonary oedema, shortness of breath, fever and nasopharyngeal infections, are associated with Na toxicity from excessive dietary Na. High dietary Na is also associated with increased risks of diseases and conditions that are co-morbid with COVID-19, in

Novel mode of action of plant defense peptides - hevein-like antimicrobial peptides from wheat inhibit fungal metalloproteases.

The multilayered plant immune system relies on rapid recognition of pathogen-associated molecular patterns followed by activation of defense-related genes, resulting in the reinforcement of plant cell walls and the production of antimicrobial compounds. To suppress plant defense, fungi secrete effectors, including a recently discovered Zn-metalloproteinase from Fusarium verticillioides, named fungalysin Fv-cmp. This proteinase cleaves class IV chitinases, which are plant defense proteins that bind and degrade chitin of fungal cell walls. In this study, we investigated plant responses to such p

Molecular Mechanisms of Natural Antiprotozoal Compounds: Emerging Strategies for Neglected Tropical Diseases.

In numerous tropical countries, outbreaks of protozoan parasites are major contributors to illness and mortality. These include protozoa from the genus Trypanosoma, which cause Human African Trypanosomiasis (HAT) and Chagas Disease, and Leishmania, which causes several forms of Leishmaniasis. The World Health Organization (WHO) has identified seventeen "Neglected Tropical Diseases" (NTDs). While not formally classified as an NTD, malaria (caused by Plasmodium spp.) presents similar therapeutic challenges and is included due to its global burden. Numerous bioactive compounds found in plants, in

Short-Term Exposure to Nitrogen Dioxide Provides Basal Pathogen Resistance.

Nitrogen dioxide (NO2) forms in plants under stress conditions, but little is known about its physiological functions. Here, we explored the physiological functions of NO2 in plant cells using short-term fumigation of Arabidopsis (Arabidopsis thaliana) for 1 h with 10 µL L-1 NO2. Although leaf symptoms were absent, the expression of genes related to pathogen resistance was induced. Fumigated plants developed basal disease resistance, or pattern-triggered immunity, against the necrotrophic fungus Botrytis cinerea and the hemibiotrophic bacterium Pseudomonas syringae Functional salicylic acid an

Metal Homeostasis in Land Plants: A Perpetual Balancing Act Beyond the Fulfilment of Metalloproteome Cofactor Demands.

One of life's decisive innovations was to harness the catalytic power of metals for cellular chemistry. With life's expansion, global atmospheric and biogeochemical cycles underwent dramatic changes. Although initially harmful, they permitted the evolution of multicellularity and the colonization of land. In land plants as primary producers, metal homeostasis faces heightened demands, in part because soil is a challenging environment for nutrient balancing. To avoid both nutrient metal limitation and metal toxicity, plants must maintain the homeostasis of metals within tighter limits than the
