Sequence of the gene encoding the major outer membrane protein of the mouse pneumonitis biovar of Chlamydia trachomatis.

The gene encoding the major outer membrane protein of the Chlamydia trachomatis mouse pneumonitis biovar was sequenced and the amino acid sequence deduced. The primary structure of this protein is similar to that of the lymphogranuloma venereum and trachoma biovars in that it consists of four variable domains interspersed with five constant domains. This protein may be an ideal candidate for a vaccine in chlamydia-infected mouse experimental models.

Immunogenicity and immunoprotective potential of recombinant OmpC and TrxA-OmpC from Vibrio parahaemolyticus.

Studies exploring the potential of outer membrane protein C (OmpC) as a vaccine candidate and diagnostic antigen for Vibrio parahaemolyticus remain limited. In this study, native OmpC and a thioredoxin-fused form (TrxA-OmpC) were successfully expressed in a prokaryotic system and evaluated for their surface exposure and immunogenicity. Surface accessibility of OmpC in the presence of bacterial surface polysaccharides was investigated using antisera raised against polysaccharide-deficient mutant strains. Immune responses induced by both recombinant proteins were assessed through immunoassays, W

Bioinformatics-based design of a fusion vaccine with CTLA-4 variable region to combat Brucella.

Brucellosis has become a global zoonotic disease, seriously endangering the health of people all over the world. Vaccination is an effective strategy for protection against Brucella infection in livestock in developed countries. However, current vaccines are pathogenic to humans and pregnant animals, which limits their use. Therefore, it is very important to improve the safety and immune protection of Brucella vaccine. In this study, different bioinformatics approaches were carried out to predict the physicochemical properties, T/B epitope, and tertiary structure of Omp2b and Omp31. Then, thes

OmpC, a novel factor H-binding surface protein, is dispensable for the adherence and virulence of Salmonella enterica serovar Typhimurium.

Salmonella enterica serovar Typhimurium utilizes a series of strategies to evade host innate immune defenses, including the serum complement system. Many microbial pathogens have evolved the ability to bind the complement regulatory protein factor H (FH) through their surface factor H-binding proteins (FHBPs) to circumvent the complement-mediated bactericidal effect. However, the roles of FHBPs in Salmonella pathogenesis are not well understood. In this study, we demonstrated that the survival of S. Typhimurium in human serum was decreased in a time and concentration dependent manner. Pre-incu

Outbreak strain characterisation and pharyngeal carriage detection following a protracted group B meningococcal outbreak in adolescents in South-West England.

Between April 2016 and September 2017, four cases of group B meningococcal disease were reported among sixth-form college students in Bristol, UK. Culture and non-culture whole genome sequencing was utilised and demonstrated that the four genomes of the responsible ST-41 strains clustered closely on a sub-lineage of ST-41/44 clonal complex. The outbreak resulted in two fatalities. A distinct social group associated with one of the cases was selected for vaccination with 4CMenB and pharyngeal swabbing. In vitro culturing, multiple real-time PCR assays (sodC, ctrA and siaDB) and a PorA PCR-seque

Persistent occurrence of serogroup Y/sequence type (ST)-23 complex invasive meningococcal disease among patients aged five to 14 years, Italy, 2007 to 2013.

In Italy, the incidence of invasive meningococcal disease (IMD) has remained stable since 2007 (around 0.3 cases/100,000 inhabitants). However, as reported for other European countries, an increase of serogroup Y Neisseria meningitidis has been observed. In this study we report IMD cases from 2007 to 2013 in Italy and investigate the clinical and epidemiological features of cases affected by serogroup Y. Molecular characteristics of serogroup Y strains are also described. During the study period, the proportion of IMD cases due to serogroup Y increased, ranging from 2% in 2007 to 17% in 2013 (

Antigenic shift and increased incidence of meningococcal disease.

BACKGROUND: The incidence of serogroup C and Y meningococcal disease increased in the United States during the 1990s. The cyclical nature of endemic meningococcal disease remains unexplained. The purpose of this study was to investigate the mechanisms associated with the increase in the incidence of meningococcal disease. METHODS: We characterized an increasing incidence of invasive serogroup C and Y meningococcal disease using population-based surveillance from 1992 through 2001. Isolates were characterized by multilocus sequence typing and antigen sequence typing of 3 outer membrane protein

Dendritic cells pulsed with a recombinant chlamydial major outer membrane protein antigen elicit a CD4(+) type 2 rather than type 1 immune response that is not protective.

Chlamydia trachomatis is an obligate intracellular bacterium that infects the oculogenital mucosae. C. trachomatis infection of the eye causes trachoma, the leading cause of preventable blindness. Infections of the genital mucosae are a leading cause of sexually transmitted diseases. A vaccine to prevent chlamydial infection is needed but has proven difficult to produce by using conventional vaccination approaches. Potent immunity to vaginal rechallenge in a murine model of chlamydial genital infection has been achieved only by infection or by immunization with dendritic cells (DC) pulsed ex v

Chlamydia trachomatis infection in the female reproductive tract of the rat: influence of progesterone on infectivity and immune response.

As the most common cause of sexually transmitted disease in women, chlamydial infections can lead to pelvic inflammatory disease, infertility, and ectopic pregnancy. To better understand the role played by sex hormones in modulating the immune response of the genital tract to microbial infections, we have developed a rat model to study Chlamydia trachomatis infection. Inbred female Lewis rats were primed with progesterone and inoculated by intrauterine instillation of C. trachomatis (mouse pneumonitis strain MoPn) into each uterine horn. When infected animals were examined for the presence of

Anti-OmpC antibodies in Crohn's disease and ulcerative colitis: evidence from a systematic review and meta-analysis.

OBJECTIVE: Anti-OmpC antibodies target the Escherichia coli outer membrane porin C. Their diagnostic and prognostic roles in inflammatory bowel disease (IBD) remain uncertain. We systematically assessed their prevalence, diagnostic accuracy, and clinical associations in Crohn's disease (CD) and ulcerative colitis (UC). METHODS: We performed a systematic review and meta-analysis (PROSPERO CRD420251066959) of 31 studies including ∼11 000 participants with CD, UC, and controls. Random-effects models estimated pooled prevalence, odds ratios (OR), sensitivity, specificity, diagnostic odds ratios (

Fluoroquinolone resistance in ESKAPE pathogens: evolutionary pathways, one health transmission, and clinical surveillance.

Fluoroquinolones (FQs) remain important treatments for many Gram-negative and some Gram-positive infections, but rapid resistance development is steadily reducing their clinical usefulness. This review integrates biological and epidemiologic evidence through a One Health perspective focused on the ESKAPE group: Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter spp. At the molecular level, resistance often begins with changes in quinolone-resistance determining regions of DNA gyrase and topoisomerase IV, followe

Structure-activity of outer membrane proteins in native bacterial membrane vesicles by solid-state NMR.

The outer membranes (OMs) of bacterial pathogens are potent virulence factors and serve as the first line of defense against host immunity. Their striking bilayer asymmetry is essential for function but poses an exceptional challenge for reconstitution in vitro, limiting structure-activity analysis to artificial non-native platforms that can interfere with structure and function. Here, we describe bacterial OM vesicles (OMVs), natively secreted from the cellular OM during bacterial cell growth and development, as an effective vehicle for structure-activity analysis in situ based on solid-state

Emergence, evolution, and global dissemination of antimicrobial resistance: A One Health review.

Antimicrobial resistance (AMR) is a critical global health threat that undermines the treatment of infections and compromises medical interventions. AMR develops when microorganisms evolve mechanisms to survive antimicrobial exposure, a process accelerated by misuse and overuse of antibiotics in human medicine, agriculture, and veterinary settings. Bacterial resistance poses the greatest immediate concern, contributing to an estimated 1.27 million deaths in 2019 and nearly 5 million deaths associated with resistant infections worldwide. Without urgent intervention, Projections suggest that AMR

High prevalence of carbapenem-resistant Pseudomonas spp. in the pork production chain: a potential reservoir for zoonotic transmission.

Multidrug-resistant Pseudomonas aeruginosa is currently one of the primary threats to human health, accounting for a significant proportion of difficult-to-treat infections globally. Although pork meat is frequently contaminated with Pseudomonas spp., well-known spoilage organisms, their potential role as reservoirs of antibiotic-resistant strains and associated risks to human health remains unclear. This study investigated the prevalence of carbapenem-resistant Pseudomonas spp. (CRP) throughout the same pork processing chain, and assessed their pathogenicity and genetic relatedness. We sample

The sRNA-encoded peptide TimP rewires the Escherichia coli envelope via LamB to sensitize bacteria to polymyxins.

The rise of multidrug-resistant (MDR) Escherichia coli has marginalized last-line polymyxins (PMs) owing to evolving resistance and narrow therapeutic windows. Revitalizing these agents requires the identification of novel regulatory circuits governing the integrity of the gram-negative envelope. In this study, we identified a stress-responsive peptide TimP and demonstrated that its intracellular overexpression (TimP+), a synthetic variant (FT-Pep), and biogenic TimP+-derived extracellular vesicles (TimP EVs) exhibited equivalent functionality in sensitizing E. coli to PMs, consistently induci
