Combining Radiation-Treated Tumor Vaccines With Mn-MOF Nanoadjuvants to Amplify Radiation Induced Anti-Tumor Immune Responses.

Radiotherapy-induced mutations are critical for neoantigen generation and the initiation of in situ vaccine mediated immune responses; however, their efficacy is severely limited by the immunosuppressive tumor microenvironment. Here, genomic analysis demonstrated that B16F10 tumor cells subjected to in vitro irradiation faithfully recapitulate part of the radiotherapy-induced mutational landscape observed in subcutaneous tumor models. Based on these findings, a biomimetic cancer vaccine was developed through the integration of RT-treated tumor cell membranes and manganese-based metal organic f…

Risk of radiation-induced skin injury in patients undergoing SIRT: Evaluation of trigger levels for radiation protection.

INTRODUCTION: Selective internal radiation therapy (SIRT) is a fluoroscopically-guided interventional procedure used in the treatment of primary and secondary hepatic tumors. This study evaluated the potential risk of radiation-induced skin injury in patients undergoing both the pre-SIRT work-up and the therapeutic administration, by assessing whether international trigger levels were exceeded for cumulative air kerma (Ka,r), defined as the air kerma measured at the patient entrance reference point. If trigger levels are exceeded, future checks are activated for patients. METHODS: We retrospe…

Radiation dose and risk assessment in pediatric dental CBCT: DAP-based evaluation and implications for radiation protection.

Pediatric patients are more sensitive to ionising radiation, making radiation protection a critical concern in dental cone-beam computed tomography (CBCT). This study aimed to assess organ and effective doses and associated radiation risk in pediatric imaging, to validate the PCXMC-20Rotation Monte Carlo code against optically stimulated luminescence dosimeters (OSLDs), and to evaluate dose-area product (DAP) as an indicator for effective dose (E) and risk of exposure-induced cancer death (REID). Age-specific 5 and 10 year-old head-and-neck phantoms were manufactured by three-dimensional (3D) …

Effectiveness of radiation protection educational material during angiography using visualization of scattered radiation by augmented reality technique.

In medical settings, radiation exposure among radiation workers is a significant concern, and understanding radiation protection is crucial. We developed and evaluated radiation protection educational materials using an augmented reality application for visualizing scatter radiation. The evaluation included a true/false quiz, a questionnaire based on the ARCS (Attention, Relevance, Confidence, and Satisfaction) model, and open-ended responses. The correct response rates for the true/false quiz were 65.5% and 72.4% for two questions regarding the effect of C-arm angle changes on scatter radiati…

Modulated Arc Therapy for hippocampal-avoidance whole brain radiation therapy: planning comparison with intensity modulated Radiation Therapy.

This study aimed to evaluate the modulated arc therapy (mARC) technique as a planning and treatment option for hippocampal sparing whole brain radiotherapy (HS-WBRT) following the Radiation Therapy Oncology Group (RTOG) 0933 dosimetric criteria. Computed tomography (CT) and magnetic resonance imaging (MRI) were selected retrospectively for 15 patients. Two types of plans were created for each patient, namely an intensity-modulated radiation therapy (IMRT) and a mARC plan. IMRT and mARC plans were compared in terms of plan quality indices, absorbed dose to organs at risk (OARs), number of monit…

Radiation dose reduction during venous access port implantation: the importance of upgrading equipment and radiation-protection training.

BACKGROUND: Implantable central venous port systems are widely used in oncology. We upgraded our fluoroscopy machines, and all anesthetists completed two training courses focusing on the risks of ionizing radiation for patients and health workers. AIMS: This study aimed to evaluate the impact of upgrading the machines and the radiation-protection training on ionizing radiation exposure during venous port system implantation. METHODS: We retrospectively analyzed consecutive venous port implantations between 2019 and 2022. The older fluoroscopy machines were replaced by two new machines. A fir…

Transcriptional Changes in Radiation-Induced Lung Injury: A Comparative Analysis of Two Radiation Doses for Preclinical Research.

In a recent stereotactic body radiation therapy animal model, radiation pneumonitis and radiation pulmonary fibrosis were observed at around 2 and 6 weeks, respectively. However, the molecular signature of this model remains unclear. This study aimed to examine the molecular characteristics at these two stages using RNA-seq analysis. Transcriptomic profiling revealed distinct transcriptional patterns for each stage. Inflammatory response and immune cell activation were involved in both stages. Cell cycle processes and response to type II interferons were observed during the inflammation stage.…

Optimizing lung cancer radiation therapy: A systematic review of multifactorial risk assessment for radiation-induced lung toxicity.

BACKGROUND: Radiation therapy (RT) is essential in treating advanced lung cancer, but may lead to radiation pneumonitis (RP). This systematic review investigates the use of pulmonary function tests (PFT) and other parameters to predict and mitigate RP, thereby improving RT planning. METHODS: A systematic review sifted through PubMed and on BioMed Central, targeting articles from September 2005 to December 2022 containing the keywords: Lung Cancer, Radiotherapy, and pulmonary function test. RESULTS: From 1153 articles, 80 were included. RP was assessed using CTCAEv.4 in 30 % of these. Six stu…

Targeting ATM enhances radiation sensitivity of colorectal cancer by potentiating radiation-induced cell death and antitumor immunity.

INTRODUCTION: The efficacy of radiotherapy in colorectal cancer (CRC) is often limited by radiation resistance. Ataxia telangiectasia mutated (ATM) is well known for its role in repairing double-strand DNA breaks within the DNA damage response (DDR) pathway. However, whether ATM mediates other mechanisms contributing to radiation resistance remains insufficiently investigated. OBJECTIVES: This study investigates how targeting ATM enhances CRC radiation sensitivity and evaluates combination strategies to improve radiotherapy outcomes. METHODS: Clinical specimens were analyzed to correlate ATM…

Reduced Mortality and Radiation Necrosis After Surgery With Postoperative Stereotactic Radiation in Patients With Multiple Brain Metastases.

BACKGROUND AND OBJECTIVES: Although stereotactic radiation has frequently supplanted whole-brain radiation therapy (WBRT) in treating patients with multiple brain metastases, the role of surgery for these patients remains unresolved. No randomized trials have compared surgical resection with postoperative stereotactic radiosurgery (SRS) or stereotactic radiotherapy (SRT) to SRS/SRT alone. Previous studies addressing surgery for patients with multiple brain metastases are often limited by small sample sizes, a lack of appropriate comparison groups, or a focus on patients treated before recent a…
